Evaluation of semi-generic PBTK modeling for emergency risk assessment after acute inhalation exposure to volatile hazardous chemicals.

BACKGROUND: Physiologically Based Toxicokinetic Models (PBTK) may facilitate emergency risk assessment after chemical incidents with inhalation exposure, but they are rarely used due to their relative complexity and skill requirements. We aimed to tackle this problem by evaluating a semi-generic PBTK model built in MS Excel for nine chemicals that are widely-used and often released in a chemical incident. MATERIAL & METHODS: The semi-generic PBTK model was used to predict blood concentration-time curves using inhalation exposure scenarios from human volunteer studies, case reports and hypothe

Nasal toxicity of manganese sulfate and manganese phosphate in young male rats following subchronic (13-week) inhalation exposure.

Growing evidence suggests that nasal deposition and transport along the olfactory nerve represents a route by which inhaled manganese and certain other metals are delivered to the rodent brain. The toxicological significance of olfactory transport of manganese remains poorly defined. In rats, repeated intranasal instillation of manganese chloride results in injury to the olfactory epithelium and neurotoxicity as evidenced by increased glial fibrillary acidic protein (GFAP) concentrations in olfactory bulb astrocytes. The purpose of the present study was to further characterize the nasal toxici

Glutaraldehyde inhalation exposure of rats: effects on lung morphology, Clara-cell protein, and hyaluronic acid levels in BAL.

Glutaraldehyde (GA) is a biocide widely used in hospital and laboratory practice. GA is a volatile substance and, under certain circumstances, significant airborne concentrations may be generated at room temperature. Occupational exposure to GA by inhalation is suspected of causing delayed irritating effects. In recent years, GA has emerged as the main cause of occupational asthma among health-care workers. The aim of the present study was to evaluate effects of GA inhalatory exposure (0.025 ppm or 0.1 ppm, for 28 days) in rats exposed corresponding to the occupational shift cycle, at time poi

Pulmonary carcinogenic effects of subchronic inhalation exposure with long-term follow-up to polyhexamethylene guanidine phosphate in rats.

Polyhexamethylene guanidine phosphate (PHMG-p) is a biocidal compound linked to respiratory toxicity following exposure to humidifier disinfectants, but long-term pulmonary outcomes after inhalation remain insufficiently characterized. This study evaluated the pulmonary carcinogenic effects of chronic PHMG-p inhalation in a rat model under environmentally relevant exposure conditions. Sprague-Dawley rats were exposed to aerosolized PHMG-p at concentrations of 0.01, 0.03, or 0.09 mg/m³ via whole-body inhalation for approximately 90 days and were serially assessed at 26, 52, 78, and 96 weeks usi

Identification of micro- and nanoplastic particles in postnatal sprague-dawley rat offspring after maternal inhalation exposure throughout gestation.

Micro-nanoplastic particulates (MNPs) have been identified in both indoor and outdoor environments. From these real-world exposures, MNPs have been identified in human fluids and organ tissues, including the placenta and breastmilk. Laboratory studies have identified MNPs are capable of crossing the placental barrier and depositing in fetal tissues; however, it remained unclear if MNPs persist in offspring tissues after birth. Six pregnant Sprague-Dawley rats were divided equally into two groups: control and exposed to polyamide-12 (PA-12) MNP aerosols (11.46 ± 3.78 mg/m3) over an average of 4

Inhalation exposure to airborne PM2.5 attenuates hepatic metabolic pathways through S-nitrosylation of the primary ER stress sensor.

Inhalation exposure to airborne fine particulate matter (aerodynamic diameter: <2.5 µm, PM2.5) is known to cause metabolic dysfunction-associated steatohepatitis (MASH) and the associated metabolic syndrome. Hepatic lipid accumulation and inflammation are the key characteristics of MASH. However, the mechanism by which PM2.5 exposure induces lipid accumulation and inflammation in the liver remains to be further elucidated. In this study, we revealed that inhalation exposure to PM2.5 induces nitrosative stress in mouse livers by suppressing hepatic S-nitrosoglutathione reductase activities, whi

Opportunities and challenges in reducing personal inhalation exposure to air pollution among electronic waste recovery workers in Ghana.

BACKGROUND: Informal sector electronic waste (e-waste) recovery produces toxic emissions resulting from burning e-waste to recover valuable metals. OBJECTIVES: To identify high-risk worker groups by measuring relative levels of personal inhalation exposure to particulate matter (PM) of fine (≤2.5 µm) and coarse (2.5-10 µm) fractions (PM2.5 and PM2.5-10, respectively) across work activities among e-waste workers, and to assess how wind conditions modify levels of PM by activity and site location. METHODS: At the Agbogbloshie e-waste site, 170 partial-shift PM samples and time-activity data we

An Air-liquid Interface Bronchial Epithelial Model for Realistic, Repeated Inhalation Exposure to Airborne Particles for Toxicity Testing.

For toxicity testing of airborne particles, air-liquid interface (ALI) exposure systems have been developed for in vitro tests in order to mimic realistic exposure conditions. This puts specific demands on the cell culture models. Many cell types are negatively affected by exposure to air (e.g., drying out) and only remain viable for a few days. This limits the exposure conditions that can be used in these models: usually relatively high concentrations are applied as a cloud (i.e., droplets containing particles, which settle down rapidly) within a short period of time. Such experimental condit

Acute inhalation exposure to cyclohexane and schedule-controlled operant performance in rats: comparison to d-amphetamine and chlorpromazine.

Adult male rats pressed a lever on a multiple fixed ratio-fixed interval (FR20-FI120 sec) schedule of food presentation, and after attaining a stable baseline subjects received an acute inhalation exposure to cyclohexane vapor (0 ppm, 500 ppm, 2000 ppm, or 7000 ppm) for 6 hr. During the operant session that began 30 min after termination of exposure, FR running rate for the 7000 ppm group decreased 11% relative to performance on the previous day. FR post-reinforcement pause duration and the rate and pattern of FT performance were unaffected. Cyclohexane exposures of 500 or 2000 ppm had no dete

An overlooked source of inhalational and dermal exposure to bisphenol A and its chlorinated derivatives in disposable face masks.

The presence of emerging and toxic chlorinated bisphenol A derivatives (ClxBPA) in disposable face masks (DFMs) remains unconfirmed. A total of 123 DFMs of various types, sourced primarily from China with additional samples from other Asian regions, North America, and Europe were analyzed. The results illustrated bisphenol A (BPA) was detected in all samples, while 3-monochloro bisphenol A (MCBPA) and 3,3',5,5'-tetrachloro-bisphenol A (TCBPA) were found in over 65% of DFMs. BPA exhibited the highest concentrations (geometric mean: 11.6 ng g-1), followed by TCBPA (0.0446 ng g-1), MCBPA (0.0234

Could e-cigarette devices generate inhalable micro- and nanoplastics? Exposure plausibility and reproductive relevance.

Electronic cigarettes are widely used by individuals in their reproductive years, yet polymer materials in pods, wicks, seals, and casings remain underexamined as possible contributors to inhaled particulate exposure. This narrative review evaluates whether e-cigarette device components could plausibly generate microplastics or nanoplastics during use and whether such particles, if present, could have reproductive relevance. Evidence from device-material studies, polymer degradation chemistry, aerosol physics, inhalation toxicology, and reproductive microplastic models was synthesized. Current

Co-exposure to inhaled tungsten particles and low-dose gamma rays: neurotoxicological outcome in rats.

Throughout their lives, individuals are exposed to various pollutants, potentially including co-exposure to radiological and chemical stressors. Yet, existing literature about these combinations is scarce. We selected tungsten and ionizing radiations. Tungsten is an emerging contaminant present as aerosolized particles in several scenarios, potentially concurrently with low-dose irradiation, causing a co-exposure. The cerebral toxicity of this co-exposure was studied after 24 h and 28 days in the frontal cortex and olfactory bulb of male Sprague-Dawley rats exposed to gamma irradiation (50 mGy

Why so blue? A novel presentation of methaemoglobinaemia secondary to an inhaled occupational nitric acid exposure.

INTRODUCTION: Nitric and hydrofluoric acids are commonly used in the commercial cleaning industry. We are unaware of reports of nitric acid inhalation forming methaemoglobin. Additionally, methaemoglobinaemia and treatment with methylthioninium chloride (methylene blue) may precipitate clinical uncertainty due to similar wavelengths of absorbance in pulse oximetry. CASES: We report two patients with respiratory distress from symptomatic methaemoglobinaemia following a prolonged, inhaled occupational exposure to nitric acid in the context of industrial cleaning. Their methaemoglobinaemia was s

Past and current asbestos exposure and future mesothelioma risks in Britain: The Inhaled Particles Study (TIPS).

BACKGROUND: Occupational and environmental airborne asbestos concentrations are too low and variable for lifetime exposures to be estimated reliably, and building workers and occupants may suffer higher exposure when asbestos in older buildings is disturbed or removed. Mesothelioma risks from current asbestos exposures are therefore not known. METHODS: We interviewed and measured asbestos levels in lung samples from 257 patients treated for pneumothorax and 262 with resected lung cancer, recruited in England and Wales. Average lung burdens in British birth cohorts from 1940 to 1992 were estim

In vitro genotoxicity data of nanomaterials compared to carcinogenic potency of inorganic substances after inhalational exposure.

Literature data of epidemiological studies, carcinogenicity studies and in vitro studies on inorganic substances were surveyed with the aim to determine sensitivity and specificity of in vitro tests of nanomaterials. Asbestos, quartz and chromium and cadmium compounds were assigned to classes of highest carcinogenic potency. After 20 years of occupational exposure to long-term average concentrations of 0.5mg/m(3) of these dusts - or to even lower concentrations - an epidemiologically detectable increased lung cancer risk has to be expected. In contrast, diesel engine emissions, some nickel spe
