Cutaneous Leishmaniasis: Update on Vaccine Development.

Leishmaniasis is an important disease mediated by the protozoan parasite Leishmania via the bite of the female sandfly insect vector. Leishmaniasis is endemic in the tropical and subtropical regions. The most common form of the disease is cutaneous leishmaniasis, which affects more than 10 million people worldwide and includes at least 1.5 million new cases every year. So far, treatment of the disease relies on unsatisfactory chemotherapy that can be complicated by the rising appearance of drug-resistant parasites. Furthermore, it is challenging to achieve solid control of the insect vector an

Application of quantitative PCR for quantization of densovirus genome.

Densovirus is classified as invertebrate virus belonging to the subfamily Densovirinae of Parvoviridae family. This group of viruses infects only insects and several densoviruses have been isolated from indigenous mosquitoes and mosquito cell lines. A number of mosquitoes, especially Aedes aegypti and Ae. albopictus are important vectors of viruses, which are the major causes of dengue hemorrhagic fever and yellow fever in humans. As densoviruses do not cause any pathology in humans, these viruses have been proposed to be a potential vector for use in biological control of mosquitoes and insec

Venereal transmission of canine visceral leishmaniasis.

Leishmania chagasi, the agent of visceral leishmaniasis in dogs in the Americas has a tropism to the male genital system, particularly the epididymis, prepuce, and glans penis, resulting in shedding of Leishmania in the semen. The goal of this study was to verify the possibility of venereal transmission of L. chagasi. Twelve Leishmania-free bitches, housed in the absence of the insect vector, copulated with multiple naturally infected dogs that were shedding Leishmania in the semen. PCR analysis of serially collected ejaculates indicated that shedding of Leishmania in the semen is intermittent

Immune interactions of entomopathogens with mosquito and tick vectors.

Entomopathogenic fungi and bacteria offer promising alternatives to chemical insecticides for controlling mosquitoes and ticks-key vectors of malaria and Lyme disease. However, their efficacy largely depends on immune interactions between pathogens and hosts. Here, we focus on mosquitoes and ticks as representative disease vectors and dissect conserved and divergent immune responses, from physical barriers to cellular defenses and key humoral pathways, with mechanistic insights informed by insect model systems. Furthermore, we elucidate diverse virulence and immune evasion strategies that enab

Arthropods as potential vectors of African swine fever virus outbreaks in pig farms in the Republic of Korea.

The seasonality of African swine fever (ASF) in the summers and outbreaks in farms with high biosecurity levels suggest that the ASF virus (ASFV) may be transmitted by arthropod vectors. Arthropods were collected in this study from 14 pig farms with ASF outbreaks in Korea in 2019 to explore the role of arthropods as potential ASFV vectors. A total of 28,729 arthropods, including 28,508 (99.2%) Diptera, were collected using blacklight traps, insect nets and yellow sticky strips. All arthropod samples were negative for ASFV genomic DNA according to laboratory tests using real-time polymerase cha

Structure and expression of Rhodnius prolixus GH18 chitinases and chitinase-like proteins: Characterization of the physiological role of RpCht7, a gene from subgroup VIII, in vector fitness and reproduction.

Chitinases are enzymes responsible for the hydrolysis of glycosidic linkages within chitin chains. In insects, chitinases are typically members of the multigenic glycoside hydrolase family 18 (GH18). They participate in the relocation of chitin during development and molt, and in digestion in detritivores and predatory insects, and they control the peritrophic membrane thickness. Chitin metabolism is a promising target for developing vector control strategies, and knowledge of the roles of chitinases may reveal new targets and illuminate unique aspects of their physiology and interaction with

Cellular and molecular basis of IR3535 perception in Drosophila.

BACKGROUND: IR3535 is among the most widely used synthetic insect repellents, particularly for the mitigation of mosquito-borne diseases such as malaria, yellow fever, dengue and Zika, as well as to control flies, ticks, fleas, lice and mites. These insects are well-known vectors of deadly diseases that affect humans, livestock and crops. Moreover, global warming could increase the populations of these vectors. RESULTS: Here, we performed IR3535 dose-response analyses on Drosophila melanogaster, a well-known insect model organism, using electrophysiology and binary food choice assays. Our fin

Analysis of vaccine-like lumpy skin disease virus from flies near the western border of China.

Lumpy skin disease (LSD) is a devastating viral disease that occurs in cattle. In China, it was first detected in the Xin-Jiang autonomous region, near the border with Kazakhstan, in August 2019. As there were no new occurrences of LSD in either country following the first detection, the initial introduction of the virus remains unknown. Arthropod vectors were considered as potential vectors. Consequently, to identify the arthropod vectors involved in transmitting LSD virus (LSDV), an insect surveillance campaign was launched at four different sites scattered along the border, and samples from

Household model of Chagas disease vectors (Hemiptera: Reduviidae) considering domestic, peridomestic, and sylvatic vector populations.

ABSTRACT Disease transmission is difficult to model because most vectors and hosts have different generation times. Chagas disease is such a situation, where insect vectors have 1-2 generations annually and mammalian hosts, including humans, can live for decades. The hemataphagous triatominae vectors (Hemiptera: Reduviidae) of the causative parasite Trypanosoma cruzi (Kinetoplastida: Trypanosomatidae) usually feed on sleeping hosts, making vector infestation of houses, peridomestic areas, and wild animal burrows a central factor in transmission. Because of difficulties with different generatio

Wolbachia pipientis: an expanding bag of tricks to explore for disease control.

Wolbachia pipientis are maternally inherited, endosymbiotic bacteria that are widespread among insects. Two recent studies have demonstrated that Wolbachia inhibits the ability of medically significant pathogens, including filarial nematodes, dengue virus and Plasmodium to form infections in the mosquito vector, Aedes aegypti. We highlight the added value of these traits for Wolbachia based biocontrol strategies and evaluate the evidence for the idea that the insect immune response is responsible for the pathogen inhibition.

Putting behaviors into context for vector-borne diseases: Examining behaviors that may reduce exposure to disease vectors.

Understanding why people adopt or ignore vector-borne disease (VBD) preventive measures is key to better risk assessment and control. However, little is known about why some households adopt preventive practices while others do not, which is particularly striking in this era of One Health's attention to human, environmental, and animal health. We examined what household factors are linked to practices that may reduce exposure to disease vectors, with a focus on Chagas disease (CD) and American Cutaneous Leishmaniasis (ACL) vectors. We surveyed 204 households in 12 rural communities in Coclé Pr

Productivity and genetic stability of a novel baculovirus vector for multigene expression from independent transgene loci.

The baculovirus expression vector system is an established platform for large-scale production of (glyco)proteins, subunit vaccines, virus-like particles, and recombinant adeno-associated virus (rAAV) vectors. We engineered a novel bacmid vector (BAC6) to improve genetic stability by deletion of the non-homologous repeat (hr) origin of DNA replication (ori) and to preserve product integrity and recovery through deletion of chitinase and cathepsin. Tn7-based transposition in E. coli and homologous recombination in insect cells were combined in BAC6 to drive expression from the odv-e56 and polyh

Epidemiology of Andean cutaneous leishmaniasis: incrimination of Lutzomyia ayacuchensis (Diptera: psychodidae) as a vector of Leishmania in geographically isolated, upland valleys of Peru.

The southernmost limit of the distribution of endemic Andean cutaneous leishmaniasis (CL), commonly known as Uta, is localized in the western Andean valleys of Ayacucho, Peru. This area is completely isolated from other regions endemic for this disease. Identification of the insect vector for Andean CL was carried out by combining entomologic and parasitologic approaches. Two Lutzomyia species were captured: Lutzomyia ayacuchensis and Lu. noguchii. The former species was considered responsible for transmission of Leishmania because 1) there was a coincidence in space and time between the prese

Controlled Transmission of a Fijivirus Under Field Conditions Using Mass-Reared Planthoppers.

Mal de Río Cuarto disease, caused by a Fijivirus, is a major constraint for maize production in Argentina. The traditional evaluation of resistant hybrids is limited by the low efficiency of natural virus transmission and the lack of standardized field inoculation methods. We developed a protocol that combines laboratory mass-rearing of the planthopper vector Delphacodes kuscheli with a controlled field transmission system. The method involves the synchronized production of large insect populations, acquisition of viruliferous vectors under controlled conditions, and their safe transport to th

Metagenomic analysis of taxa associated with Lutzomyia longipalpis, vector of visceral leishmaniasis, using an unbiased high-throughput approach.

BACKGROUND: Leishmaniasis is one of the most diverse and complex of all vector-borne diseases worldwide. It is caused by parasites of the genus Leishmania, obligate intramacrophage protists characterised by diversity and complexity. Its most severe form is visceral leishmaniasis (VL), a systemic disease that is fatal if left untreated. In Latin America VL is caused by Leishmania infantum chagasi and transmitted by Lutzomyia longipalpis. This phlebotomine sandfly is only found in the New World, from Mexico to Argentina. In South America, migration and urbanisation have largely contributed to th

The dynamic of Trypanosoma cruzi transmissibility in field-caught Mepraia spinolai (Hemiptera: Reduviidae) of contrasting seasons: A developmental stage-dependent study.

A competent vector can harbor a pathogen and infect a high number of hosts. Scarce information is available on how triatomine vectors accomplish this task in nature, particularly without knowing the number of parasites they excrete. We study the dynamics of Trypanosoma cruzi transmissibility in the triatomine Mepraia spinolai collected in summer (December 2018) and winter (July 2019) of an austral endemic area. The insects received two feedings in the laboratory, the day after arrival and 45 days after the first feeding. Excreta samples were collected, DNA was extracted, and parasite loads wer

Transcriptomic Insights into the Insect Immune Response to Nematode Infection.

Insects in nature interact with a wide variety of microbial enemies including nematodes. These include entomopathogenic nematodes that contain mutualistic bacteria and together are able to infect a broad range of insects in order to complete their life cycle and multiply, filarial nematodes which are vectored by mosquitoes, and other parasitic nematodes. Entomopathogenic nematodes are commonly used in biological control practices and they form excellent research tools for understanding the genetic and functional bases of nematode pathogenicity and insect anti-nematode immunity. In addition, cl

Evidence for transovarial transmission of tomato yellow leaf curl virus by its vector, the whitefly Bemisia tabaci.

The whitefly Bemisia tabaci is the only vector of the tomato yellow leaf curl geminivirus (TYLCV). The insect transmits the virus in a persistent-circulative manner. TYLCV DNA was detected by polymerase chain reaction and by Southern blot hybridization in progeny (eggs, first and second instars, adults) of single viruliferous whiteflies that developed on eggplant or on cotton (two TYLCV nonhost plants). Furthermore, TYLCV DNA was present in the progeny of insects that had acquired the virus through the egg. The adult progeny of the viruliferous insects and their own progeny were able to infect

Glossina spp. gut bacterial flora and their putative role in fly-hosted trypanosome development.

Human African trypanosomiasis (HAT) is caused by trypanosomes transmitted to humans by the tsetse fly, in which they accomplish their development into their infective metacyclic form. The crucial step in parasite survival occurs when it invades the fly midgut. Insect digestive enzymes and immune defenses may be involved in the modulation of the fly's vector competence, together with bacteria that could be present in the fly's midgut. In fact, in addition to the three bacterial symbionts that have previously been characterized, tsetse flies may harbor additional bacterial inhabitants. This revi

Endophytic Entomopathogenic Fungi Shape Herbivore Behavior and Plant-Insect Interactions: Implications for Biological Control.

Entomopathogenic fungi (EPF) are well established as biological control agents, but their emerging role as endophytes reveals a broader and more powerful function in crop protection. By colonizing plant tissues, endophytic entomopathogenic fungi (EEPF) create a dynamic tripartite interaction between plants, fungi, and herbivores, enabling systemic, plant-mediated pest suppression. This review synthesizes current knowledge on the behavioral and ecological responses of herbivorous arthropods to EEPF-colonized plants, with an emphasis on the mechanisms and implications for integrated pest managem
