Activity and Functional Connectivity of Lumbar Spinal Interneurons at Rest: Irregular, Sparse Yet Structured.

Spinal interneurons are central to sensorimotor integration. Even in the absence of overt sensory inputs or motor output, spinal interneurons remain active, yet their functional organization across spatial and temporal scales remains poorly understood. We characterized the discharge properties and functional connectivity of 2,847 lumbar spinal interneurons recorded in vivo from quiescent decerebrate cats (3 males, 5 females) using microelectrode arrays inserted into rostral (L3-4) and caudal (L5-7) spinal segments at superficial (0-1500 µm) and deep (1500-3000 µm) depths. Superficial neurons g

Hyaluronic acid-alginate hydrazone crosslinked hydrogels support the generation and maturation of V2a interneurons.

Injury to the central nervous system (CNS) causes inflammation, cell death, and glial scar formation that inhibits tissue repair. Injectable hydrogels modified with extracellular matrix (ECM)-derived peptides can provide biochemical cues to promote neural tissue repair and serve as a vehicle to deliver therapeutics across the blood-spinal/blood-brain barrier in a minimally invasive manner. We developed an injectable hydrazone crosslinked hyaluronic acid-alginate (HA-Alg) hydrogel for neural tissue repair. We fabricated hydrogels with a range of polymer concentrations and evaluated their physic

Parvalbumin interneurons regulate rehabilitation-induced functional recovery after stroke and identify a rehabilitation drug.

Motor disability is a critical impairment in stroke patients. Rehabilitation has a limited effect on recovery; but there is no medical therapy for post-stroke recovery. The biological mechanisms of rehabilitation in the brain remain unknown. Here, using a photothrombotic stroke model in male mice, we demonstrate that rehabilitation after stroke selectively enhances synapse formation in presynaptic parvalbumin interneurons and postsynaptic neurons in the rostral forelimb motor area with axonal projections to the caudal forelimb motor area where stroke was induced (stroke-projecting neuron). GABAergic signaling by VIP interneurons gates running-dependent visual recovery in the adult brain.

Experience-dependent plasticity in the adult visual cortex is enhanced by locomotion, a process mediated by vasoactive intestinal peptide (VIP)-expressing interneurons. While VIP interneurons are known to signal through both Gamma-aminobutyric acid (GABA) and VIP peptide, the specific contributions of these pathways during different forms of plasticity remain unclear. Monocular deprivation (MD) in adult mice alters cortical responses, though more slowly and differently than during a critical period in early life. Here, we used two-photon calcium imaging in awake adult mice to dissect the roles

Epigenetic regulation of cocaine intake through dopaminergic control of cholinergic interneurons in male mice.

Substance use disorders are chronic neuropsychiatric conditions influenced by multiple factors, shaping individuals' vulnerability to addictive drugs like cocaine. Here, we reveal that dopamine D2 receptor mediated inhibition of striatal cholinergic interneurons regulates the motivation for cocaine intake by modulating acetylcholine signaling in striatal circuits. This acetylcholine-dependent mechanism contributes to cocaine self-administration through the muscarinic M4 receptor and the histone acetyltransferase Kat5/Tip60. We discover that Kat5 is upregulated in response to cocaine and furthe

Roles of spinal V3 interneurons: Roles in controlling movement in healthy and injured conditions.

Spinal V3 interneurons are glutamatergic neurons that are distributed among the dorsal, intermediate, and ventral spinal cord. They are involved in broad neural circuit connections in the central nervous system. Functionally, they play important roles in locomotion, such as the maintenance of robust and balanced gaits during walking. More importantly, after spinal cord injury, these neurons maintain their excitability and facilitate proprioceptive sensory transmission to motor neurons, which are crucial for the initiation of complex coordinated reciprocal and rhythmic activities that resemble

Pathogenic LRRK2 mutations cause loss of primary cilia and Neurturin in striatal parvalbumin interneurons.

Parkinson's disease-associated, activating mutations in the LRRK2 kinase block primary cilium formation in cell culture and in specific cell types in the brain. In the striatum that is important for movement control, about half of astrocytes and cholinergic interneurons, but not the predominant medium spiny neurons, lose their primary cilia. Here, we show that mouse and human striatal parvalbumin interneurons that are inhibitory regulators of movement also lose primary cilia. Without cilia, these neurons are not able to respond to Sonic hedgehog signals that normally induce the expression of P

Fast-spiking parvalbumin-positive interneurons: new perspectives of treatment and future challenges in dementia.

Central nervous system parvalbumin-positive interneurons (PV-INs) are crucial and highly vulnerable to various stressors. They also play a significant role in the pathological processes of many neuropsychiatric diseases, especially those associated with cognitive impairment, such as Alzheimer's disease (AD), vascular dementia (VD), Lewy body dementia, and schizophrenia. Although accumulating evidence suggests that the loss of PV-INs is associated with memory impairment in dementia, the precise molecular mechanisms remain elusive. In this review, we delve into the current evidence regarding the

The fate of interneurons, GABAA receptor sub-types and perineuronal nets in Alzheimer's disease.

Alzheimer's disease (AD) is the most common neurological disease, which is associated with gradual memory loss and correlated with synaptic hyperactivity and abnormal oscillatory rhythmic brain activity that precedes phenotypic alterations and is partly responsible for the spread of the disease pathology. Synaptic hyperactivity is thought to be because of alteration in the homeostasis of phasic and tonic synaptic inhibition, which is orchestrated by the GABAA inhibitory system, encompassing subclasses of interneurons and GABAA receptors, which play a vital role in cognitive functions, includin

Chemogenetic Activation of Cortical Parvalbumin-Positive Interneurons Reverses Noise-Induced Impairments in Gap Detection.

Exposure to loud noises not only leads to trauma and loss of output from the ear but also alters downstream central auditory circuits. A perceptual consequence of noise-induced central auditory disruption is impairment in gap-induced prepulse inhibition, also known as gap detection. Recent studies have implicated cortical parvalbumin (PV)-positive inhibitory interneurons in gap detection and prepulse inhibition. Here, we show that exposure to loud noises specifically reduces the density of cortical PV but not somatostatin (SOM)-positive interneurons in the primary auditory cortex in mice (C57B

Pitx2 cholinergic interneurons are the source of C bouton synapses on brainstem motor neurons.

Cholinergic neuromodulation has been described throughout the brain and has been implicated in various functions including attention, food intake and response to stress. Cholinergic modulation is also thought to be important for regulating motor systems, as revealed by studies of large cholinergic synapses on spinal motor neurons, called C boutons, which seem to control motor neuron excitability in a task-dependent manner. C boutons on spinal motor neurons stem from spinal interneurons that express the transcription factor Pitx2. C boutons have also been identified on the motor neurons of spec

RORβ Spinal Interneurons Gate Sensory Transmission during Locomotion to Secure a Fluid Walking Gait.

Animals depend on sensory feedback from mechanosensory afferents for the dynamic control of movement. This sensory feedback needs to be selectively modulated in a task- and context-dependent manner. Here, we show that inhibitory interneurons (INs) expressing the RORβ orphan nuclear receptor gate sensory feedback to the spinal motor system during walking and are required for the production of a fluid locomotor rhythm. Genetic manipulations that abrogate inhibitory RORβ IN function result in an ataxic gait characterized by exaggerated flexion movements and marked alterations to the step cycle. I

Cultured networks of excitatory projection neurons and inhibitory interneurons for studying human cortical neurotoxicity.

Translating neuroprotective treatments from discovery in cell and animal models to the clinic has proven challenging. To reduce the gap between basic studies of neurotoxicity and neuroprotection and clinically relevant therapies, we developed a human cortical neuron culture system from human embryonic stem cells or human inducible pluripotent stem cells that generated both excitatory and inhibitory neuronal networks resembling the composition of the human cortex. This methodology used timed administration of retinoic acid to FOXG1(+) neural precursor cells leading to differentiation of neurona

From Balance to Breakdown: striatal PV interneurons in Huntington's disease and Autism Spectrum Disorder.

Once relegated to the background of striatal circuitry, parvalbumin-expressing interneurons are now emerging as central players in health and disease. Acting as true gatekeepers, striatal PV interneurons are well-described for their role in synchronizing striatal output and balancing excitation and inhibition to sustain coordinated motor and cognitive functions. In this review, we highlight recent advances in understanding their developmental origins, molecular identity, physiological properties, and their roles in striatal function. Furthermore, we examine converging evidence implicating PV i

Alterations in GABAergic Interneurons, Neuroinflammation, and Epigenetics: Potential Targets for Therapeutic Strategies in Schizophrenia.

BACKGROUND: Schizophrenia (SZ) is a long-term psychiatric condition that affects cognition, thought processing, emotional regulation, and behavior. Epigenetic modifications during pregnancy or early postnatal development increase vulnerability to SZ by disrupting the balance of GABAergic-glutamatergic neurotransmission. OBJECTIVE: The association between GABAergic interneurons and inflammation has been increasingly investigated. Systemic inflammation alters the expression of GABAergic interneuron markers, including somatostatin and parvalbumin, in various brain regions. These modifications co

Parvalbumin interneurons and dentate gyrus homeostatic dysregulation shape epileptogenesis in Angelman syndrome model mice.

Understanding how neural circuits transition from seizure-resistant to seizure-prone is essential for developing improved epilepsy therapies. Here, we study this process by leveraging the heightened susceptibility to seizure kindling of Angelman syndrome (AS) model mice, which lack the maternal Ube3a (mUbe3a) allele. We identify parvalbumin-expressing (PV+) interneurons as critical gatekeepers; selective mUbe3a deletion in PV+ neurons phenocopies enhanced AS epileptogenesis, whereas restoring UBE3A broadly in GABAergic neurons confers seizure resistance. Further, pathological remodeling of the

Background optic flow modulates responses of multiple descending interneurons to object motion in locusts.

Animals flying within natural environments are constantly challenged with complex visual information. Therefore, it is necessary to understand the impact of the visual background on the motion detection system. Locusts possess a well-identified looming detection pathway, comprising the lobula giant movement detector (LGMD) and the descending contralateral movement detector (DCMD). The LGMD/DCMD pathway responds preferably to objects on a collision course, and the response of this pathway is affected by the background complexity. However, multiple other neurons are also responsive to looming st

Alcohol Attenuates CRF-Induced Excitatory Effects from the Extended Amygdala to Dorsostriatal Cholinergic Interneurons.

Alcohol relapse is associated with corticotropin-releasing factor (CRF) signaling and altered reward pathway function, though the precise mechanisms remain unclear. Here, we investigated how CRF modulates cholinergic interneurons (CINs) in the dorsal striatum, a region critical in mediating cognitive flexibility and action selection. Using monosynaptic and retrograde circuit tracing, we identified direct inputs from CRF-expressing (CRF⁺) neurons in the central amygdala (CeA) and bed nucleus of the stria terminalis (BNST) to dorsal striatal CINs. We showed that CINs express CRF receptor 1 (CRFR

Dynamic recruitment of PV interneurons in the somatosensory cortex induced by experience dependent plasticity.

Adaptive circuit plasticity plays crucial roles in the brain during development, learning, sensory experience, and after injury. During chronic whisker trimming, a well-studied paradigm for inducing experience dependent plasticity, whisker representations in the somatosensory barrel cortex (S1BF) undergo remapping, with expansion of maps for spared whiskers and contraction of maps for trimmed whiskers. At the cellular level, excitatory pyramidal cells in Layer 2/3 shift their whisker tuning, increasing selectivity to spared whiskers and away from deprived whiskers. While these changes are well

KCa1.1 channels contribute to optogenetically driven post-stimulation silencing in cerebellar molecular layer interneurons.

Using cell-attached recordings from molecular layer interneurons (MLI) of the cerebellar cortex of adult mice expressing channel rhodopsin 2, we show that wide-field optical activation induces an increase in firing rate during illumination and a firing pause when the illumination ends (post-stimulation silencing; PSS). Significant spike rate changes with respect to basal firing rate were observed for optical activations lasting 200 ms and 1 s as well as for 1 s long trains of 10 ms pulses at 50 Hz. For all conditions, the net effect of optical activation on the integrated spike rate is signifi
