We previously identified an operon involved in an arginine deiminase (ADI) pathway (arc operon) on a CTX-M-producing plasmid from an O102-ST405 strain of Escherichia coli As the ADI pathway was shown to be involved in the virulence of various Gram-positive bacteria, we tested whether the ADI pathway could be involved in the epidemiological success of extended-spectrum-β-lactamase (ESBL)-producing E. coli strains. We studied two collections of human E. coli isolated in France (n = 493) and England (n = 1,509) and show that the prevalence of the arc operon (i) is higher in ESBL-producing strains

Ochrobactrum and Brucella are closely related bacteria that populate different habitats and differ in their pathogenic properties. Only little is known about mobile genetic elements in these genera which might be important for survival and virulence. Previous studies on Brucella lysogeny indicated that active phages are rare in this genus. To gain insight into the presence and nature of prophages in Ochrobactrum, temperate phages were isolated from various species and characterized in detail. In silico analyses disclosed numerous prophages in published Ochrobactrum genomes. Induction experimen

Most bacteria are polylysogens that carry multiple prophages integrated into the chromosome. These prophages confer advantages to their bacterial host, yet also pose a lethal threat as they can reactivate and enter a lytic cycle. DNA damage of the bacterial host is a common trigger of prophage lytic cycles. Because DNA damage is frequently experienced by bacterial pathogens exposed to host immune defenses, prophage activation may be common during infection. Investigating the consequences of prophage induction in Salmonella, we discover a prophage competition element in the Gifsy-1 prophage tha

A gene library constructed from large (approximately 20 kb) fragments of total DNA from the geldananmycin-producing strain Streptomyces hygroscopicus 3602 cloned in the plasmid vector pIJ61 were used to transform S. lividans TK24. Three transformants of about 800 tested were found to have acquired the ability to produce an antibiotic lethal to a geldanamycin-sensitive strain of Bacillus subtilis. The plasmids isolated from these transformants, pIA101, pIA102 and pIA103, each contained an insert of approximately 15 kb. A 4.5 kb DNA fragment from the insert in pIA102 hybridised to DNA from S. hy

Since the discovery of the first Listeria bacteriophage by Schultz in 1945, more than 219 phages were isolated for L. monocytogenes, L. invanovii, L. innocua, L. seeligeri and L. welshimeri. To date, no phage for L. murrayi and L. grayi is described in the literature. Unless two phages found by Jasińska in a water sample, all the other phages, when the origin is mentioned, were isolated from lysogenic strains, with or without induction. Electron microscopic examination of 55 Listeria phages revealed that two L. innocua phages belonged to the Myoviridae family (tail with contractile sheath) whe

The increasing occurrence of hospital-associated infections, particularly bacteremia, caused by extensively drug-resistant (XDR) carbapenemase-producing colistin-resistant Klebsiella pneumoniae highlights a critical requirement to discover new therapeutic alternatives. Bacteriophages having host-specific bacteriolytic effects are promising alternatives for combating these pathogens. Among 12 phages isolated from public wastewater in Thailand, two phages-vB_kpnM_05 (myovirus) and vB_kpnP_08 (podovirus) showed broad-host range, producing bacteriolytic activities against 81.3% (n = 26) and 78.1%

Prophages are important drivers of bacterial evolution and can provide pathogens with additional factors that enhance virulence and/or resistance properties. However, they are not considered when assessing the dynamics of outbreak populations using phylogenetic approaches. Recently, we showed that BAL114, an extensively antibiotic-resistant Acinetobacter baumannii isolate from Vietnam that carries KL58 at the capsular polysaccharide (CPS) biosynthesis locus, produced CPS with two types of linkage between KU58 tetrasaccharide units that were both different from the linkage formed by the WzyKL58

The human bacterial pathogen group A Streptococcus (GAS) causes many different diseases including pharyngitis, tonsillitis, impetigo, scarlet fever, streptococcal toxic shock syndrome, necrotizing fasciitis and myositis, and the post-infection sequelae glomerulonephritis and rheumatic fever. The frequency and severity of GAS infections increased in the 1980s and 1990s, but the cause of this increase is unknown. Recently, genome sequencing of serotype M1, M3 and M18 strains revealed many new proven or putative virulence factors that are encoded by phages or phage-like elements. Importantly, the

A Shiga toxin (Stx)-encoding temperate bacteriophage of Shigella sonnei strain CB7888 was investigated for its morphology, DNA similarity, host range, and lysogenization in Shigella and Escherichia coli strains. Phage 7888 formed plaques on a broad spectrum of Shigella strains belonging to different species and serotypes, including Stx-producing Shigella dysenteriae type 1. With E. coli, only strains with rough lipopolysaccharide were sensitive to this phage. The phage integrated into the genome of nontoxigenic S. sonnei and laboratory E. coli K-12 strains, which became Stx positive upon lysog

All of 41 naturally occurring coagulase-positive methicillin-resistant strains of Staphylococcus aureus isolated in various laboratories were resistant to several antibiotics and were lipase-negative. Most strains produced hemolysins, and 38 strains produced enterotoxin B. Acriflavine treatment of four strains resulted in elimination of resistance to methicillin and mercury; in one strain, resistance to cadmium was also lost. Production of enterotoxin B and beta-hemolysin was eliminated in all four strains and penicillinase production was eliminated in one strain. In transduction experiments,

Our analysis of the plague diagnostic phage L-413C genome sequence and structure reveals that L-413C is highly similar and collinear with enterobacteriophage P2, though important differences were found. Of special interest was the mosaic nature of the tail fiber protein H in L-413C, given the differentiating specificity of this phage for Yersinia pestis vs. Yersinia pseudotuberculosis. While the N-terminal 207 and C-terminal 137 amino acids of L-413C display significant homology with the P2 H protein, a large (465 amino acid) middle section appears to be derived from a T4-related H protein, wi

Actinobacillus actinomycetemcomitans is a suspected etiologic agent in destructive periodontal diseases. The detection of bacteriophages in A. actinomycetemcomitans in the subgingival plaque of patients with rapidly destructive forms of periodontitis led to the hypothesis that bacteriophage infection might increase the virulence of this bacterium (19). A. actinomycetemcomitans was isolated from 68 subjects from the Netherlands and Switzerland with localized juvenile periodontitis, rapidly progressing periodontitis, or adult periodontitis, and was tested for the presence of temperate bacterioph

Mutation of the c-myc gene locus has occurred during in vitro culture of the Burkitt lymphoma cell line Namalwa. Gene cloning and sequencing of this c-myc gene locus has revealed the insertion of incomplete copies of the Squirrel Monkey Retrovirus-Human (SMRV-H) proviral genome. Insertions of the SMRV-H genome were seen in all Namalwa cell lines tested, with multiple insertions being seen in some sublines. Viral particles with the morphology of type-D retrovirus, budding from these cells, were seen by transmission EM. New insertions of the genome were produced spontaneously in these cells. The

Shiga toxin-producing Escherichia coli (STEC) is an important foodborne pathogen, and beef-associated contamination remains a major food-safety concern. STEC in beef products is difficult to control when cells persist as biofilms on food-contact surfaces and in processing environments. Here, lytic phage vB-EcoP-B10 was isolated from cattle-farm wastewater using cattle-farm-derived STEC B10 as the host. Its genomic safety, biological properties, antibiofilm activity, and raw-beef efficacy were evaluated. TEM and genome analyses assigned vB-EcoP-B10 to a novel species within Kayfunavirus. Its 40

Phage therapy has emerged as a promising alternative for combating multidrug-resistant hypervirulent Klebsiella pneumoniae (MDR-hvKP) infections. Recent advances suggest that tailoring phage cocktails based on genomic surveillance of circulating strains might yield more effective regimens (Koncz et al., 2024). Here, we combined publicly available epidemiological records with regional strain analysis to examine the distribution of the ST23-K1 lineage and to place the selected target in genomic and clinical context. Among the 227 isolates in our Henan collection, ST23-K1 accounted for 11.01% of

We have isolated and characterized a novel bacteriophage termed Jab, with lytic activity against multidrug-resistant clinical isolates of Escherichia coli. Phage Jab was identified from liquid manure by means of metagenome sequencing of a phage community enrichment using an E. coli clinical isolate ECH07 as host. The initial enrichment was composed of four phages, of which phage Jab represented only a minute fraction (less than 1%). Jab isolation strategy comprised a targeted approach using iterative replication rounds while equipping ECH07 with resistance against the numerically dominant phag

Enterococcus faecalis is a Gram-positive opportunistic pathogen commonly associated with hospital-acquired and chronic wound infections. Its capacity for biofilm formation and increasing resistance to standard antibiotics, particularly ciprofloxacin (CIP), complicates treatment. The emergence of CIP-resistant E. faecalis (EFR) strains limits existing antibiotic options and requires alternative treatment approaches. Our study evaluated the antibacterial efficacy of a lytic bacteriophage (φR12EFP) and its combination with CIP against E. faecalis. Phage φR12EFP was isolated from environmental sam
