1. The present study was carried out to determine the effects of feeding ensiled alfalfa leaves (ALS) as an alternative protein source to laying hens under the terms of an organic diet. Due to the occurrence of unexpected negative health effects and undesirable egg yolk pigmentation in the test groups the trial was prematurely stopped and further analysis was conducted to evaluate the responsible substances.2. Body weights of the test groups decreased significantly already in week 2 of the trial. Performance variables dropped. Olive green pigmented egg yolks were found in groups fed diets cont

Four multiparous Holstein cows in midlactation were used in a 4 x 4 Latin square to evaluate whether source of forage influenced salivary secretion during eating in lactating dairy cows. The forages were allocated separately from the pelleted concentrates. Cows were offered 1 of 4 forages each period: barley silage, alfalfa silage, long-stemmed alfalfa hay, or chopped barley straw. Saliva secretion was measured during the morning meal by collecting masticates through the rumen cannula at the cardia of each cow. Rate of salivation (213 g/min) was not affected by forage source. However, the fora

CUF 101, a resistant cultivar of alfalfa, was exposed to 15 clones of Acyrthosiphon pisum Harris collected from alfalfa fields in three regions of France (east, south, central west) to determine whether the level of resistance varied across the different clones. The survival of alfalfa seedlings infested at the cotyledon stage was assessed using a standardized method. Although no difference in seedling mortality was detected between clones grouped by region, there was a significant variation among the 15 pea aphid clones. In particular, two clones of southern origin were more aggressive. In ad

Using ion-selective microelectrodes, the problem of how signals coming from symbiotic partners or from potential microbial intruders are distinguished was investigated on root hairs of alfalfa (Medicago sativa). The Nod factor, NodRm-IV(C16:2,S), was used to trigger the symbiotic signal and (GlcNAc)(8) was selected from (GlcNAc)(4-8), to elicit defense-related reactions. To both compounds, root hairs responded with initial transient depolarizations and alkalinizations, which were followed by a hyperpolarization and external acidification in the presence of (GlcNAc)(8). We propose that alfalfa

The slow rate of natural attenuation of organic pollutants, together with unwanted environmental impacts of traditional remediation strategies, has necessitated the exploration of plant-microbe systems for enhanced bioremediation applications. The identification of microorganisms capable of promoting rhizoremediation through both plant growth-promoting and hydrocarbon-degrading processes is crucial to the success and adoption of plant-based remediation techniques. In this study, through successive enrichments of soil samples from a historic oil-contaminated site in Wietze, Germany, we isolated

Seed physical dormancy is a crucial adaptive trait conferred by the water-impermeable seed coat, particularly its specialized palisade layer. Although the structural components of this barrier, including the cuticle and cell wall polysaccharides, are well characterized, the molecular mechanisms coordinating their formation and remodeling remain poorly understood. Here, using single-nucleus RNA sequencing of Medicago truncatula, we identify a regulatory module essential for palisade layer formation and the establishment of physical dormancy. We show that MtKNOX4 interacts with cytoplasmic MtSAW

This article follows-up on our recently published work, which evaluated the impact of the addition of an alfalfa leaf-derived adsorbent in the aflatoxin B1 (AFB1)-contaminated diet in regard to the production parameters, blood cell count, serum biochemistry, liver enzymes, and liver histology of turkey poults. This paper presents complementary results on microbial community, ileal morphology, barrier function, and immunity. For this purpose, 350 1-day-old female turkey poults were randomly distributed into five groups: (1) Control, AFB1-free diet; (2) AF, AFB1-contaminated diet at 250 ng/g; (3

AIMS: The purpose of the research is to study the effects of different fibre types and sources on the intestinal flora of geese.
METHODS AND RESULTS: A total of 48 geese (males: 35 days old) were divided into four groups, each of which included three replicates of four geese. Groups 1-4 were fed a diet containing 5% corn stover Crude fibre (CF, the LJ group), 8% corn stover CF (the HJ group), 5% alfalfa CF (the LM group) or 8% alfalfa CF (the HM group), respectively. After 42 days of feeding, the intestinal flora of each group was determined by 16SrRNA gene sequencing. In the duodenum, the di

The utilization of forages grown on metal-contaminated soil can increase the risk of heavy metals entering the food chain and affecting human health because of elevated toxic metal concentrations. Meanwhile, hydrogen sulfide (H2S) and nitric oxide (NO) as signaling molecules are known to promote plant growth in metal-contaminated soils. However, the regulatory mechanisms of such molecules in plant physiology and soil biochemistry have not been well-documented. Hence, we investigate the role of the exogenous application of H2S and NO on alfalfa growth in lead/cadmium (Pb/Cd)-contaminated soil.

Our analyses of lipopolysaccharide mutants of Sinorhizobium meliloti offer insights into how this bacterium establishes the chronic intracellular infection of plant cells that is necessary for its nitrogen-fixing symbiosis with alfalfa. Derivatives of S. meliloti strain Rm1021 carrying an lpsB mutation are capable of colonizing curled root hairs and forming infection threads in alfalfa in a manner similar to a wild-type strain. However, developmental abnormalities occur in the bacterium and the plant at the stage when the bacteria invade the plant nodule cells. Loss-of-function lpsB mutations,

Seventy-five New Zealand White x Californian rabbits were used to study the influence of the chemical composition of lucerne hay on caecal and caecotrophy characteristics. Five lucerne hays varying in chemical composition were ground and formed into pellets. These were the sole form of nutrition during the experiment. The type of lucerne hay did not affect caecal volatile fatty acid concentration, pattern of fermentation of pH. However, the caecal ammonia concentration decreased linearly (by 30% between extreme diets, P = 0.002) when dietary fibre proportion increased. The weight of caecum and

Forty-four multiparous and 43 primiparous Holstein cows were used to study the effect of dietary alfalfa silage: grain ratio on digestion, passage of digesta, and energy balance in a complete lactation experiment. Cows were placed on one of five treatments with forage contents from 38.2 to 98.2% (DM basis) during the first 12 wk of lactation. Forage content was increased during wk 13 to 26 of lactation to give diets containing 48.2 to 98.2% forage and again during wk 27 to 44 to give diets containing 68.2 to 98.2% forage. Both DMI and DM digestibilities decreased as proportion of dietary forag

Chalcone isomerase (CHI) catalyzes the cyclization of three-ring scaffolds of flavonoids, a class of plant-based natural products important for nutrition and disease prevention. A persistent question has been whether the enzyme uses protein dynamics to facilitate conformational rearrangements of substrates within the active site. To help resolve this question, CHI from Medicago truncatula was crystallized with phloretin, a flexible substrate analogue that cannot undergo cyclization, and solved to 2.0-Å resolution. The crystal structure possesses eight protein molecules per asymmetric unit, rev

Alfalfa (Medicago sativa) is the most widely cultivated forage crop worldwide, owing to its high protein content, balanced amino acid profile, and abundance of vitamins and minerals. However, weed infestation severely threatens alfalfa yield and quality. The lack of herbicide‑resistant cultivars, combined with the diversity of weed species, has made herbicide tolerance a key breeding objective. To address this challenge, we developed a highly optimized prime editing system and, for the first time, achieved prime editing in alfalfa by enhancing the expression efficiency of its components. There exist commonalities between symbiotic Sinorhizobium meliloti and pathogenic Brucella bacteria in terms of extensive gene synteny and the requirements for intracellular survival in their respective hosts. The RNA chaperone Hfq is essential for virulence for several bacterial groups, including Brucella; however, its role in S. meliloti has not been investigated. Our studies of an S. meliloti loss-of-function hfq mutant have revealed that Hfq plays a key role in the establishment of the symbiosis between S. meliloti and its host Medicago sativa. S. meliloti Hfq is involved in controlling th

Many plant endosymbionts are facultative, switching between host-associated and free-living stages. Extensive genomic and experimental studies suggest that adaptation during the saprophytic, off-host phase, rather than adaptation to hosts, primarily constrains the biogeographic distribution of these microbes. To test this hypothesis, we analyzed the growth capacities and genomic features of 38 Sinorhizobium and Ensifer strains isolated from the nodules of Medicago lupulina (black medic), collected from two regions with distinct thermal environments. The warmer region is predominantly inhabited

This paper reports the studies on the elaboration of new environmentally friendly fertilizer obtained by valorization of post-extraction biomass residues of alfalfa (Medicago) and goldenrod (Solidago), after extraction with supercritical carbon dioxide, via biosorption process. The performance and controlled release properties of fertilizer were assessed in laboratory under in vitro and in vivo conditions, as well as on the field. In vitro tests show high bioavailability of micronutrients (Cu, Mn, Zn) administered on the biological carrier - between 60 and 80%, in relation to 100% availability

Biosurfactant-assisted rhizosphere remediation is a promising strategy for polycyclic aromatic hydrocarbon (PAH)-contaminated soils; however, the mechanisms by which biosurfactants regulate host plant responses to PAH stress and drive rhizosphere microbiome assembly remain unclear. Here, we constructed an alfalfa-assisted rhizosphere remediation system using novel trehalose tetraesters (TTEs) and TTE-producing Rhodococcus erythropolis KB1 to remediate phenanthrene-contaminated soil and decoded mechanisms via multi-omics. The colonization of KB1 and TTE-mediated rhizosphere regulation enhanced
