Brain Stimulation for Improving Sleep and Memory.

Given the critical role of sleep, particularly sleep slow oscillations, sleep spindles, and hippocampal sharp wave ripples, in memory consolidation, sleep enhancement represents a key opportunity to improve cognitive performance. Techniques such as transcranial electrical and magnetic stimulation and acoustic stimulation can enhance slow oscillations and sleep spindles and potentially improve memory. Targeted memory reactivation in sleep may enhance or stabilize memory consolidation. Each technique has technical considerations that may limit its broader clinical application. Therefore, neurost

Designing for memory: how pathway configuration shapes long-term navigational representations.

BACKGROUND: Spatial memories undergo systems consolidation, typically transforming from flexible, hippocampal-dependent representations of spatial relations, to rigid, extra-hippocampal route representations. This study investigated whether environmental design influences this consolidation process to enhance long-term wayfinding. METHODS: We compared navigational performance in a virtual shopping centre featuring either a "Grid" configuration, which promotes flexible navigational representations, or a "Star" configuration, which encourages route-based sequence learning. In Experiment 1 (N =

Prevention of recurrent affective episodes using extinction training in the reconsolidation window: A testable psychotherapeutic strategy.

Stressors may initially precipitate affective episodes, but with sufficient numbers of recurrences, episodes can occur more autonomously. It is postulated the memory engram for these recurrent depressions moves from the conscious representational memory system to the unconscious habit memory system encoded in the striatum. If this were the case, cognitive behavior therapy targeted toward extinction of habit memories could be an effective maneuver for helping reverse the automaticity of affective episode recurrence. Extinction training in the reconsolidation window (which opens about 5min to 1h

Effects of Multisession Prefrontal Transcranial Direct Current Stimulation on Long-term Memory and Working Memory in Older Adults.

Transcranial direct current stimulation (tDCS) is a noninvasive form of electrical brain stimulation popularly used to augment the effects of working memory (WM) training. Although success has been mixed, some studies report enhancements in WM performance persisting days, weeks, or even months that are actually more reminiscent of consolidation effects typically observed in the long-term memory (LTM) domain, rather than WM improvements per se. Although tDCS has been often reported to enhance both WM and LTM, these effects have never been directly compared within the same study. However, given

Acute pain impairs retention of locomotor learning.

Despite abundant evidence that pain alters movement performance, considerably less is known about the potential effects of pain on motor learning. Some of the brain regions involved in pain processing are also responsible for specific aspects of motor learning, indicating that the two functions have the potential to interact, yet it is unclear if they do. In experiment 1, we compared the acquisition and retention of a novel locomotor pattern in young, healthy individuals randomized to either experience pain via capsaicin and heat applied to the lower leg during learning or no stimulus. On day

The many faces of amnesia.

Results from studies of retrograde amnesia provide much of the evidence for theories of memory consolidation. Retrograde amnesia gradients are often interpreted as revealing the time needed for the formation of long-term memories. The rapid forgetting observed after many amnestic treatments, including protein synthesis inhibitors, and the parallel decay seen in long-term potentiation experiments are presumed to reveal the duration of short-term memory processing. However, there is clear and consistent evidence that the time courses obtained in these amnesia experiments are highly variable with

The relationship between protein synthesis and protein degradation in object recognition memory.

For decades there has been a consensus that de novo protein synthesis is necessary for long-term memory. A second round of protein synthesis has been described for both extinction and reconsolidation following an unreinforced test session. Recently, it was shown that consolidation and reconsolidation depend not only on protein synthesis but also on protein degradation by the ubiquitin-proteasome system (UPS), a major mechanism responsible for protein turnover. However, the involvement of UPS on consolidation and reconsolidation of object recognition memory remains unknown. Here we investigate

Medial temporal lobe functional network architecture supports sleep-related emotional memory processing in older adults.

Memory consolidation occurs via reactivation of a hippocampal index during non-rapid eye movement slow-wave sleep (NREM SWS) which binds attributes of an experience existing within cortical modules. For memories containing emotional content, hippocampal-amygdala dynamics facilitate consolidation over a sleep bout. This study tested if modularity and centrality-graph theoretical measures that index the level of segregation/integration in a system and the relative import of its nodes-map onto central tenets of memory consolidation theory and sleep-related processing. Findings indicate that great

Sleep forms flexible context representations in toddlers.

Sleep consolidates episodic memory through a hippocampus-dependent process in adults. Whether and how sleep supports memory consolidation during early life, when hippocampal function is immature, remains unclear. Here, we examined effects of sleep on the consolidation of spatial context, a core component of episodic memory, in toddlers aged 2-3 years. Toddlers were familiarized with two spatial contexts, followed by a ~ 90-min nap or equivalent wake period. Afterwards, using a hide-and-seek game we tested their ability to relocate toys within these contexts. Only after post-familiarization sle

Spontaneous slow oscillation-slow spindle features predict induced overnight memory retention.

STUDY OBJECTIVES: Synchronization of neural activity within local networks and between brain regions is a major contributor to rhythmic field potentials such as the EEG. On the other hand, dynamic changes in microstructure and activity are reflected in the EEG, for instance slow oscillation (SO) slope can reflect synaptic strength. SO-spindle coupling is a measure for neural communication. It was previously associated with memory consolidation, but also shown to reveal strong interindividual differences. In studies, weak electric current stimulation has modulated brain rhythms and memory reten

ACQUISITION AND CONSOLIDATION OF TWO-WAY ACTIVE AVOIDANCE IN THE HIGH IMMOBILIZATION "DEPRESSIVE" RATS.

Present study investigated possible changes in acquisition and consolidation of associative memory in high immobilization "depressive" and low immobilization "non-depressive" rats. Question is very topical because understanding the character of learning and memory disturbances, one of symptoms of major depressive disease, is very significant for more intimate definition of the pathophysiology of major depressive disorder and appropriate searching the ways of its correction. Selection of rats according to the level of immobilization was made by means of forced swim test. Learning and memory dis

Fine-Tuning the Details: Post-encoding Music Differentially Impacts General and Detailed Memory.

Music can effectively induce emotional arousal, which is associated with the release of stress hormones that are important for the emotional modulation of memory. Thus, music may serve as a powerful modulator of memory and mood, making it a promising therapeutic tool for memory and mood disorders such as Alzheimer's disease or depression. However, music's impact on memory depends on its features, timing, and ability to elicit emotional arousal. In the current study, we manipulated various features of music played during post-encoding memory consolidation to elicit emotional arousal and impact

Impaired procedural memory in narcolepsy type 1.

OBJECTIVES: Sleep enhances the consolidation of memories. Here, we investigated whether sleep-dependent memory consolidation differs between healthy subjects and narcolepsy type 1 (NT1) patients. MATERIAL AND METHODS: We recruited 18 patients with NT1 and 24 healthy controls. The consolidation of spatial (declarative memory; 2-dimensional object location) and procedural (non-declarative memory; finger sequence tapping) memories was examined across one night of at-home sleep. Sleep was measured by an ambulatory sleep recording device. RESULTS: The overnight gain in the number of correctly rec

An evolutionary conserved division-of-labor between archicortical and neocortical ripples organizes information transfer during sleep.

Systems-level memory consolidation during sleep depends on the temporally precise interplay between cardinal sleep oscillations. Specifically, hippocampal ripples constitute a key substrate of the hippocampal-neocortical dialog underlying memory formation. Recently, it became evident that ripples are not unique to archicortex, but constitute a wide-spread neocortical phenomenon. To date, little is known about the morphological similarities between archi- and neocortical ripples. Moreover, it remains undetermined if neocortical ripples fulfill distinct functional roles. Leveraging intracranial

Consolidation/reconsolidation therapies for the prevention and treatment of PTSD and re-experiencing: a systematic review and meta-analysis.

Translational research highlights the potential of novel 'memory consolidation/reconsolidation therapies' to treat re-experiencing symptoms and post-traumatic stress disorder (PTSD). This systematic review and meta-analysis assessed the efficacy of so-called memory consolidation/reconsolidation therapies in randomised controlled trials (RCTs) for prevention and treatment of PTSD and symptoms of re-experiencing in children and adults (PROSPERO: CRD42020171167). RCTs were identified and rated for risk of bias. Available data was pooled to calculate risk ratios (RR) for PTSD prevalence and standa

Excitatory repetitive transcranial magnetic stimulation to left dorsal premotor cortex enhances motor consolidation of new skills.

BACKGROUND: Following practice of skilled movements, changes continue to take place in the brain that both strengthen and modify memory for motor learning. These changes represent motor memory consolidation a process whereby new memories are transformed from a fragile to a more permanent, robust and stable state. In the present study, the neural correlates of motor memory consolidation were probed using repetitive transcranial magnetic stimulation (rTMS) to the dorsal premotor cortex (PMd). Participants engaged in four days of continuous tracking practice that immediately followed either excit

How coupled slow oscillations, spindles and ripples coordinate neuronal processing and communication during human sleep.

Learning and plasticity rely on fine-tuned regulation of neuronal circuits during offline periods. An unresolved puzzle is how the sleeping brain, in the absence of external stimulation or conscious effort, coordinates neuronal firing rates (FRs) and communication within and across circuits to support synaptic and systems consolidation. Using intracranial electroencephalography combined with multiunit activity recordings from the human hippocampus and surrounding medial temporal lobe (MTL) areas, we show that, governed by slow oscillation (SO) up-states, sleep spindles set a timeframe for ripp

Artificial sharp-wave-ripples to support memory and counter neurodegeneration.

Information processed in our sensory neocortical areas is transported to the hippocampus during memory encoding, and between hippocampus and neocortex during memory consolidation, and retrieval. Short bursts of high-frequency oscillations, so called sharp-wave-ripples, have been proposed as a potential mechanism for this information transfer: They can synchronize neural activity to support the formation of local neural networks to store information, and between distant cortical sites to act as a bridge to transfer information between sensory cortical areas and hippocampus. In neurodegenerative

Towards Optimization of Oscillatory Stimulation During Sleep.

BACKGROUND: Oscillatory rhythms during sleep, such as slow oscillations (SOs) and spindles and, most importantly, their coupling, are thought to underlie processes of memory consolidation. External slow oscillatory transcranial direct current stimulation (so-tDCS) with a frequency of 0.75 Hz has been shown to improve this coupling and memory consolidation; however, effects varied quite markedly between individuals, studies, and species. In this study, we aimed to determine how precisely the frequency of stimulation must match the naturally occurring SO frequency in individuals to best improve

Coordinated activities of retrosplenial ensembles during resting-state encode spatial landmarks.

The brain likely uses offline periods to consolidate recent memories. One hypothesis holds that the hippocampal output provides a unique, global linking or 'index' code for each memory, and that this code is stored in the cortex in association with locally encoded attributes of each memory. Activation of the index code is hypothesized to evoke coordinated memory trace reactivation thus facilitating consolidation. Retrosplenial cortex (RSC) is a major recipient of hippocampal outflow and we have described populations of neurons there with sparse and orthogonal coding characteristics that resemb
