The compact (cp) phenotype in cucumber (Cucumis sativus L.) is an important plant architecture-related trait with a great potential for cucumber improvement. In this study, we conducted map-based cloning of the cp locus, identified and functionally characterized the candidate gene. Comparative microscopic analysis suggested that the short internode in the cp mutant is due to fewer cell numbers. Fine genetic mapping delimited cp into an 8.8-kb region on chromosome 4 harboring only one gene, CsERECTA (CsER) that encodes a leucine-rich repeat receptor-like kinase. A 5.5-kb insertion of a long ter

A novel, efficient maize genetic transformation system was developed using Agrobacterium-mediated transformation of embryo explants from mature seeds. Seeds from field grown plants were sterilized and crushed to isolate embryo explants consisting of the coleoptile, leaf primordia, and shoot apical meristem which were then purified from the ground seed bulk preparation. The infection of relevant tissues of seed embryo explants (SEEs) by Agrobacterium was improved by the centrifugation of the explants. Transgenic plants were obtained by multiple bud induction on high cytokinin media, followed by

BACKGROUND AND AIMS: The MADS-box transcription factor AGAMOUS (AG) is an important regulator of stamen and fruit identity as well as floral meristem determinacy in a number of core eudicots and monocots. However, its role outside of these groups has not been assessed explicitly. Examining its role in opium poppy, a basal eudicot, could uncover much about the evolution and development of flower and fruit development in the angiosperms.
METHODS: AG orthologues were isolated by degenerate RT-PCR and the gene sequence and structure examined; gene expression was characterized using in situ hybrid

Hydrogen sulfide (H2S) functions as a pivotal gaseous signaling molecule in plants, yet its role in promoting crop yield remains elusive. Here, we demonstrate that sodium hydrosulfide (NaHS) application, a donor of hydrogen sulfide (H2S), significantly accelerates growth, promotes flowering, and enhances grain yield in rice (Oryza sativa L.). Optimal NaHS treatment increased plant height, root length, and biomass accumulation, concomitant with elevated sucrose, starch, chlorophyll contents, and nitrate reductase activity. Integrated transcriptomic and proteomic analyses revealed that NaHS repr

PHD fingers are modular domains in chromatin-associated proteins that decode the methylation status of histone H3 tails. A PHD finger signature is found in plant vernalization (VEL) proteins, which function as accessory factors of the Polycomb system to control flowering in Arabidopsis through an epigenetic silencing mechanism. It has been proposed that VEL PHD fingers bind to methylated histone H3 tails to facilitate association of the Polycomb silencing machinery with target genes. Here, we use structural analysis by X-ray crystallography to show that the VEL PHD finger forms the central mod

A major limitation for the utilization of sorghum forage is the production of the cyanogenic glycoside dhurrin in its leaves and stem that may cause the death of cattle feeding on it at the pre-flowering stage. Therefore, we attempted to develop transgenic sorghum plants with reduced levels of hydrogen cyanide (HCN) by antisense mediated down-regulation of the expression of cytochrome P450 CYP79A1, the key enzyme of the dhurrin biosynthesis pathway. CYP79A1 cDNA was isolated and cloned in antisense orientation, driven by rice Act1 promoter. Shoot meristem explants of sorghum cultivar CSV 15 we

Diverse forms of leaves are present in nature. However, the regulatory mechanisms that underpin the development of diverse leaf forms remain enigmatic. The initiation of leaf primordia from the periphery of shoot apical meristem (SAM) requires downregulation of the class 1 knotted-like homeobox KNOXI proteins. In plants with simple leaves, this downregulation is permanent, consistent with leaves being determinant organs. In most of plants with compound leaves, the KNOXI proteins are reactivated in developing leaf primordia, and this reactivation is required for the development of compound leav

Elongation of the Arabidopsis hypocotyl pushes the shoot-producing meristem out of the soil by rapid expansion of cells already present in the embryo. This elongation process is shown here to be impaired by as much as 35% in mutants lacking ABCB19, an ATP-binding cassette membrane protein required for polar auxin transport, during a limited time of fast growth in dim white light beginning 2.5 days after germination. The discovery of high ectopic expression of a cyclin B1;1-based reporter of mitosis throughout abcb19 hypocotyls without an equivalent effect on mitosis prompted investigations of

Plant responses to abiotic stress significantly affect the development of cells, tissues and organs. However, no studies correlating Pb-induced mitotic inhibition and DNA damage and the alterations in redox homeostasis during root division per se were found in the literature. Therefore, an experiment was conducted to evaluate the impact of Pb on mitotic activity and the associated changes in the oxidative metabolism in onion roots. The cytotoxic effect of Pb on cell division was assessed in the root meristems of Allium cepa (onion). The mitotic index (MI) was calculated and chromosomal abnorma

UNLABELLED: The key to successful cryopreservation lies in the induction of tolerance towards dehydration/desiccation and freezing. The accumulation of osmo-active compounds, which can be induced by drought and cold stress, is therefore important. In the present study, three-week old shoots from in vitro plantlets of the cultivated potato Solanum tuberosum and its frost-resistant relative Solanum commersonii were submitted to osmotic stress (by using sucrose) and chilling (6°C). After 14days of exposure, shoot tips were sampled in order to gain an insight into changes of the proteome and solub

Outer cell layers of rice roots, which comprise epidermis, exodermis and sclerenchyma, have been proposed to protect the roots from various stresses in soil. Here, we report a mutant which is defective in the specification of outer cell layers, and examined the role of these layers in Al and other metal resistance. Morphological and histochemical observations revealed that the mutant isolated based on Al sensitivity frequently showed a disordered pattern of periclinal cell division in the epidermal layers at a region close to the root apical meristem. The lateral root caps in the mutant became

Lateral root initiation in Arabidopsis thaliana occurs in xylem pole pericycle (Px) cells, yet the structural features associated with founder-cell competence in differentiated root tissues remain insufficiently defined. Here, we combined spatial cell biology approaches, including three-dimensional analysis of cell geometry, nuclear morphometry, EdU labeling, and auxin-induction experiments, to determine whether Px cells exhibit a structural prepattern before visible organogenesis. Px cells were already distinguishable in the root apical meristem (RAM) by their shorter length along the longitu

Plant cell type diversity often arises from transcriptional regulatory programs that co-opt conserved transcription factors (TFs) involved in other developmental programs. For example, the basic helix-loop-helix (bHLH) TF FAMA directs the differentiation of guard cells (GCs), and part of the FAMA regulatory network was co-opted to direct the differentiation of Brassicales-specific idioblasts known as myrosin cells (MCs). In this review, we explore how new cell types and lineage-specific innovations can be specified by the same conserved bHLH TFs through different sets of downstream targets. We

Background/Objectives: The green synthesis of gold nanoparticles (AuNPs) using plant-derived flavonoids offers a sustainable alternative to traditional methods. This study aimed to synthesize, characterize, and evaluate the biological activities and cytogenotoxicity of AuNPs functionalized with naringin (NG) and its aglycone, naringenin (NGN). Methods: Synthesis was optimized by varying pH, HAuCl4 concentration, reagent ratios, temperature, and stirring time. The resulting AuNPs-NG and AuNPs-NGN were characterized via Ultraviolet-Visible (UV-Vis) Spectroscopy, Fourier-Transform Infrared (FTIR)

Goji is recognized as a nutritionally rich superfruit with medicinal and industrial importance. However, its inherently indeterminate growth habit causes asynchronous flowering and uneven fruit ripening, resulting in harvesting inefficiency with high labour costs and low fruit yield. To elucidate the molecular basis of shoot determinacy and early flowering, comparative transcriptome analysis of indeterminate and determinate shoot apices in an F1 hybrid population of Lycium barbarum identified 15 candidate genes associated with shoot growth termination and early flowering, including an SP-like

The oil extracted from the mesocarp of oil palm (Elaeis guineensis, Jacq.) drupes is the world's leading source of vegetable oil. Oil palm cultivation requires large amounts of mineral nutrients supplied through fertilization, representing a major production cost. Among essential nutrients, potassium (K⁺) plays a critical role in plant growth and fruit development. However, the mechanisms underlying K⁺ transport and its relationship with yield remain poorly understood. Here, we identified and characterized EgKT2-1, a Shaker-like potassium channel belonging to the AKT2-like subfamily. Quantitat

Latent viral and viroid infections represent a major phytosanitary threat to pome fruit production because infected propagation material facilitates the long-term spread of pathogens in orchards. This study investigated the prevalence of viral and viroid pathogens in apple and pear orchards of southern and southeastern Kazakhstan and evaluated the efficiency of integrated in vitro sanitation approaches. A total of 34 samples collected from commercial orchards were analyzed using RT-qPCR. Apple stem pitting virus (ASPV) was the most prevalent pathogen detected across all surveyed regions, follo

Nectar spurs are tubular outgrowths of perianth tissue-a key innovation that has evolved many times independently in angiosperms. These novel floral structures are tightly linked to pollination and pollinator shifts may explain their role in increasing diversification rates. In the tribe Antirrhineae (Plantaginaceae), nectar spurs on the ventral petal have convergently evolved four times, in the Kickxia clade, Cymbalaria, Chaenorhinum, and Linaria. Here we explore the molecular mechanism underpinning this convergence, by investigating two Class I KNOX genes (HIRZ and INA) across Antirrhineae.

Rho-like GTPases from plants (ROPs) constitute a plant-specific subfamily of Rho-family small GTPases. The Rho family is conserved among all eukaryotic cells, and includes the Rho, Rac and Cdc42 subfamilies found in fungi and animals. Like other GTPases, ROPs act as molecular switches that cycle between the inactive GDP-bound and active GTP-bound states, and are typically involved in cell signaling. Functional studies have demonstrated that ROPs play key roles in regulating processes such as the establishment of cell polarity, polar growth of cells (e.g., pollen tubes and root hairs), cell sha

Hybrid breeding based on male sterility requires the removal of male parents, which is time- and labor-intensive; however, the use of female sterile male parent can solve this problem. In the offspring of distant hybridization between Brassica oleracea and Brassica napus, we obtained a mutant, 5GH12-279, which not only fails to generate gynoecium (thereby causing female sterility) but also has serrated leaves that could be used as a phenotypic marker in seedling screening. Genetic analysis revealed that this trait was controlled by a single dominant gene. Further analysis revealed that Bo2g005
