Robust Saccharomyces cerevisiae by rational metabolic engineering for effective ethanol production from undetoxified steam-exploded corn stover hydrolysate.

Lignocellulosic bioethanol production by S.cerevisiae is severely hampered by xylose assimilation and inhibitors. Aiming to solve these barriers, the xylose isomerase pathway was heterologously introduced into parental strain, followed by conducting the adaptive laboratory evolution. Meanwhile, the reduced glutathione and NADPH synthesis systems to reduce excess intracellular reactive oxygen species (ROS) were further enhanced. Results indicated the bioethanol production from undetoxified steam-exploded corn stover hydrolysate (SECSH) without any nutrients supplementation was improved using th

Microbial upcycling of unrefined glycerol to beta-carotene by tailored metabolic engineering of extremophilic microorganism Deinococcus radiodurans R1.

Unrefined glycerol, an abundant byproduct of biodiesel production, has emerged as a promising carbon source for microbial biorefineries. Despite its potential, large-scale application remains limited by cytotoxic impurities such as residual methanol and peroxides. Here, we report the engineering of extremophilic microorganism Deinococcus radiodurans R1 for efficient β-carotene production from unrefined glycerol. To establish a base strain, the crtY gene was overexpressed and the crtLm genes were deleted to complete the β-carotene biosynthetic pathway, enabling production of 0.46 ± 0.03 mg/L (0

Synergistic strategy for high-yield 2,3-butanediol and acetoin production in Bacillus licheniformis MW03 based on metabolic engineering.

Bacillus licheniformis is an efficient platform for 2,3-butanediol (2,3-BD) and acetoin production due to its rapid glucose utilization rate and adaptability to industrial fermentation conditions. Here, we isolated the B. licheniformis strain MW03 with high yield of acetoin and 2,3-BD, which carried genetic mutations in acoR and budC, respectively encoding an acetoin dehydrogenase regulator and meso-2,3-BD dehydrogenase. To further confirm the physiological effects on acetoin and 2,3 BD biosynthesis, gene editing was performed using the CRISPR-Cas9 system, followed by phenotypic screening and

Metabolic engineering of artificially modified transcription factor SmMYB36-VP16 for high-level production of tanshinones and phenolic acids.

Tanshinones and phenolic acids are the two main chemical constituents in Salvia miltiorrhiza, which are used clinically for the treatment of hypertension, coronary heart disease, atherosclerosis, and many other diseases, and have broad medicinal value. The efficient synthesis of the target products of these two metabolites in isolated plant tissues cannot be achieved without the regulation and optimization of metabolic pathways, and transcription factors play an important role as common regulatory elements in plant tissue metabolic engineering. However, most of the regulatory effects are speci

Combinatorial Metabolic and Process Engineering for Enhanced GA4+7 Production and Tunable GA4/GA7 Ratio in Fusarium fujikuroi.

Gibberellins (GAs) are ubiquitous phytohormones that regulate plant growth and are widely used in agriculture. Among these, GA3 and GA4+7 are the only commercially available products, yet GA4+7 commands a much higher price than GA3, primarily due to its low titer in the industrial fungus Fusarium fujikuroi. To engineer a high-yielding GA4+7 producer, we first deleted p450-3 to block the conversion of GA4 and GA7 to GA1 and GA3. This led to the accumulation of GA4+7 at 0.934 g/L, a 37-fold increase over the wild-type strain, albeit with over half reduction in total GA accumulation. Using this Δ

Interleukin-21 engineering enhances CD19-specific CAR-NK cell activity against B-cell lymphoma via enriched metabolic pathways.

BACKGROUND: NK cells engineered to express interleukin-15 (IL-15) and a CD19-targeted chimeric antigen receptor (CAR) have been used to treat patients with relapsed and/or refractory B cell malignances, demonstrating encouraging outcomes and favorable safety profile. However, the effect of IL-21 in CAR-NK cell therapy remains unknown. METHODS: CD19-specific CAR with 4-1BB costimulatory domain and cytokine IL-21 or IL-15 was constructed and transduced into peripheral blood (PB)-derived NK cells to produce CD19-CAR-IL21 NK cells (CAR-21) or CD19-CAR-IL15 NK cells (CAR-15), respectively. Metabolic engineering of Saccharomyces cerevisiae for co-production of ethanol and 3-methyl-1-butanol from sugarcane molasses.

3-Methyl-1-butanol (3MB) is a promising renewable solvent, drop-in fuel, and precursor for various industrial products, including flavors, fragrances, and surfactants. Due to the myriad of intertwined biosynthetic pathways that share metabolic precursors, conventional metabolic engineering strategies to overproduce 3MB in yeast have typically resulted in yields that are far too low for economic viability. However, because 3MB is naturally produced by yeast, 100 million liter of 3MB are already produced annually as a byproduct of bioethanol fermentations. Despite its significant commercial valu

Efficient production of 2'-fucosyllactose in Pichia pastoris through metabolic engineering and constructing an orthogonal energy supply system.

2'-fucosyllactose (2'-FL) holds significant role in the infants' nutrition. While microbial production of 2'-FL has predominantly utilized Escherichia coli and Saccharomyces cerevisiae, the potential of Pichia pastoris, renowned for its robust NADPH regeneration capability, remains underexplored. Herein, we systematically engineered the metabolism of P. pastoris to develop an efficient 2'-FL-producing cell factory. We first constructed the de novo biosynthesis pathway for 2'-FL in P. pastoris, achieving an initial titer of 0.143 g/L. By optimizing enzyme selection and solubility of α-1,2-fucos

High-level co-production of 3-hydroxypropionic acid and 1,3-propanediol from glycerol: Metabolic engineering and process optimization.

3-Hydroxypropionic acid (3-HP) and 1,3-propanediol (1,3-PDO) are value-added chemicals with versatile applications in the chemical, pharmaceutical, and food industries. Nevertheless, sustainable production of 3-HP and 1,3-PDO is often limited by the lack of efficient strains and suitable fermentation configurations. Herein, attempts have been made to improve the co-production of both metabolites through metabolic engineering of Escherichia coli and process optimization. First, the 3-HP and 1,3-PDO co-biosynthetic pathways were recruited and optimized in E. coli, followed by coupling the pathwa

Daucus carota DcPSY2 and DcLCYB1 as Tools for Carotenoid Metabolic Engineering to Improve the Nutritional Value of Fruits.

Carotenoids are pigments with important nutritional value in the human diet. As antioxidant molecules, they act as scavengers of free radicals enhancing immunity and preventing cancer and cardiovascular diseases. Moreover, α-carotene and β-carotene, the main carotenoids of carrots (Daucus carota) are precursors of vitamin A, whose deficiency in the diet can trigger night blindness and macular degeneration. With the aim of increasing the carotenoid content in fruit flesh, three key genes of the carotenoid pathway, phytoene synthase (DcPSY2) and lycopene cyclase (DcLCYB1) from carrots, and carot

Metabolically engineered chimeric probiotic-cancer vesicles orchestrate anti-tumor systemic immunity.

The paradigm shift toward patient-centric precision medicine has established personalized cancer vaccines (PCVs) as a key strategy for eliciting tumor-selective immunity. However, traditional cell-based PCVs face significant translational barriers, including manufacturing complexity and safety concerns. While tumor-derived extracellular vesicles (TDEVs) offer a promising cell-free alternative that recapitulates the parental tumor's antigenic landscape, their therapeutic efficacy is frequently antagonized by intrinsic immunosuppressive cargo. Here, we report ChimeraVax, a chimeric vesicular vac

Gene-programmed micro-nano metabolic engine drives coupled osteogenic-angiogenic regeneration.

Effective bone healing is accompanied by coupled osteogenic-angiogenic regeneration, a process that critically depends on metabolic communication between bone marrow mesenchymal stem cells (BMSCs) and endothelial cells through an endogenous metabolic axis in which endothelial nitric oxide synthase (eNOS) catalyzes the conversion of l-arginine into nitric oxide (NO). However, within hypoxic microenvironments such as those present in large bone defects, the restricted expression of eNOS and the depletion of its substrate synergistically suppress NO biosynthesis, thereby impairing osteogenic-angi

Engineering of 1,4-Butanediol and Adipic Acid Metabolism in Pseudomonas taiwanensis for Upcycling to Aromatic Compounds.

The overwhelming amount of plastic produced is an unprecedented challenge for humanity due to the lack of end-of-life solutions for heterogeneous plastic wastes. One possibility is feedstock recycling of mixed plastics and complex polymers with subsequent biological funnelling and upcycling. Major depolymerisation products of common plastics such as polyurethanes, polyesters and polyamides include aliphatic dicarboxylic acids or diols such as adipic acid (AA) and 1,4-butanediol (BDO), which can be metabolised by engineered Pseudomonas putida strains. However, the spectrum of upcycled compounds

Biosynthesis of chiral diols from alkenes using metabolically engineered type II methanotroph.

Methanotrophs are environmentally friendly microorganisms capable of converting gas to liquid using methane monooxygenases (MMOs). In addition to methane-to-methanol conversion, MMOs catalyze the conversion of alkanes to alcohols and alkenes to epoxides. Herein, the efficacy of epoxidation by type I and II methanotrophs was investigated, and type II methanotrophs were observed to be more efficient in converting alkenes to epoxides. Subsequently, three (Epoxide hydrolase) EHs of different origins were overexpressed in the type II methanotroph Methylosinus trichosporium OB3b to produce 1,2-diols

Engineering principles in plant metabolism: integrating control, mechanics, and transport under dynamic environments.

Plant responses to dynamic environmental conditions remain challenging to describe within a unified mechanistic framework. This is largely because metabolism operates under simultaneous and interacting regulatory, mechanical, and transport constraints. Here, we synthesize three engineering perspectives as complementary frameworks to quantify these constraints on plant metabolism: (i) control engineering, for dynamic regulation of the biochemical pathways; (ii) structural and mechanical engineering, to quantify load-bearing constraints and geometry-dependent scaling relationships in plant tissu

Engineered Nestin+ TSPC-Derived Exosomes Promote Tendon Repair Via Metabolic Reprogramming.

Tendon repair remains a substantial clinical problem with limited treatment options. While Nestin+ tendon stem/progenitor cells (TSPCs) are known to regulate tendon healing, their clinical translation faces challenges. Here, we show that exosomes derived from engineered Nestin-overexpressing TSPCs (Nes-EVs) significantly ameliorate tendinopathy in multiple mouse models. We isolated Nes-EVs following lentiviral Nestin overexpression and demonstrated their capacity to reduce cellular senescence and promote tenogenic differentiation in vitro. Engineered Lactate Catabolizing Probiotics Reveal Timescale Dependent Microbiome-Host Metabolic Coupling.

The exchange of lactate, a metabolic substrate and regulator, between the gut lumen and systemic circulation for use in host and microbial processes is well documented, but tools capable of uncovering whether this process influences host metabolic status across acute and chronic contexts are lacking. In our prior work, we engineered probiotic Bacillus subtilis PY79 to produce lactate oxidase (LOX) intracellularly, allowing it to rapidly convert intestinal lactate to pyruvate. Following oral administration, LOX reduced systemic lactate concentrations at rest and under challenge conditions, prov
