New role for histamine in interleukin-3-induced proliferation of hematopoietic stem cells.

This study investigated the effect of histamine generated by murine bone marrow cells in response to IL-3 on one particular biological activity of this growth factor, i.e., triggering of cells forming colonies in spleen (CFU-S) into S phase. Evidence is provided that i) IL-3-induced day-8 CFU-S cell cycling, evaluated by hydroxy-urea suicide, is completely abrogated when the binding of histamine to its H2 receptors is blocked by the specific antagonist oxmetidine, whereas cetirizine, a H1 receptor antagonist, is ineffective; and ii) the entry of day-8 CFU-S into S phase in response to IL-3 is

Assessment of protein-calorie malnutrition.

We critically review the various methods for assessing protein--calorie malnutrition. These tests are lacking in sensitivity and specificity, and currently no single one can be used as a reliable indicator of malnutrition. However, a combination of several of them can be used as prognostic indicators and are of help in selecting patients who may benefit from nutritional support. Body-composition analyses and functional tests hold the promise of greater applicability in the future. Newer and more nearly accurate tests for use in diagnosis of protein-calorie malnutrition as well as for objective

Anatomical, physiological, and pharmacological characteristics of histidine decarboxylase knock-out mice: evidence for the role of brain histamine in behavioral and sleep-wake control.

The hypothesis that histaminergic neurons are involved in brain arousal is supported by many studies. However, the effects of the selective long-term abolition of histaminergic neurons on the sleep-wake cycle, indispensable in determining their functions, remain unknown. We have compared brain histamine(HA)-immunoreactivity and the cortical-EEG and sleep-wake cycle under baseline conditions or after behavioral or pharmacological stimuli in wild-type (WT) and knock-out mice lacking the histidine decarboxylase gene (HDC-/-). HDC-/-mice showed an increase in paradoxical sleep, a decrease in corti

Serum amine-based metabolites and their association with outcomes in primary prevention implantable cardioverter-defibrillator patients.

AIMS: Heart failure patients are at increased risk of ventricular arrhythmias and all-cause mortality. However, existing clinical and serum markers only modestly predict these adverse events. We sought to use metabolic profiling to identify novel biomarkers in two independent prospective cohorts of patients with implantable cardioverter-defibrillators (ICDs) for primary prevention of sudden cardiac death (SCD). METHODS AND RESULTS: Baseline serum was quantitatively profiled for 42 known biologically relevant amine-based metabolites among 402 patients from the Prospective Observational Study o

Apomorphine-induced turning behavior in 6-hydroxydopamine lesioned rats is increased by histidine and decreased by histidine decarboxylase, histamine H1 and H2 receptor antagonists, and an H3 receptor agonist.

The role of histamine and its receptors in basal ganglia neurocircuitry was assessed in apomorphine-induced turning behavior. Rats with unilateral 6-hydroxydopamine lesions of the substantia nigra pars compacta and medial forebrain bundle were administered histaminergic agents, and apomorphine-induced turning behavior was tested on Days 7 and 14 post-lesion. Compared with saline-treated rats, histidine (500 mg/kg, i.p.), a precursor of histamine, increased turning behavior (p<0.05), while alpha-fluoromethylhistidine (alpha-FMH, 25 microg, i.c.v.), an irreversible inhibitor of histidine decarbo

Submucosal microinfusion of endothelin and adrenaline mobilizes ECL-cell histamine in rat stomach, and causes mucosal damage: a microdialysis study.

Rat stomach ECL cells release histamine in response to gastrin. Submucosal microinfusion of endothelin or adrenaline, known to cause vasoconstriction and gastric lesions, mobilized striking amounts of histamine. While the histamine response to gastrin is sustainable for hours, that to endothelin and adrenaline was characteristically short-lasting (1-2 h). The aims of this study were to identify the cellular source of histamine mobilized by endothelin and adrenaline, and examine the differences between the histamine-mobilizing effects of gastrin, and of endothelin and adrenaline. Postoperative enteral vs parenteral nutrition. A randomized controlled trial.

Twenty patients undergoing major upper-gastrointestinal-tract or pancreaticobiliary surgery were randomized to receive postoperative nutritional support by total parenteral nutrition (TPN) or elemental diet administered by needle-catheter jejunostomy (NCJ). Both routes of administration provided adequate nutritional support. No unexpected complications were encountered. The NCJ group compared favorably with the TPN group at the end of the seven-day trial. The NCJ group provided significant cost efficiency while maintaining adequate nutritional support.

FIA-MS/MS-Based Targeted Metabolomics of Amino Acids and Acylcarnitines Uncovers Network-Level Metabolic Reprogramming in Chronic Kidney Disease.

Background/Objectives: Chronic kidney disease (CKD) is a major global public health problem, and conventional biomarkers such as serum creatinine primarily reflect excretory function with limited sensitivity for early detection and for predicting progression. Amino acids (AAs) and acylcarnitines (ACs) reflect metabolic processes related to nitrogen metabolism and mitochondrial fatty-acid oxidation that are influenced by renal function. This study aimed to characterize CKD-associated alterations in AAs and ACs profiles using an integrated analytical framework combining differential analysis, su

Comparative study on changes in serum amino acid metabolic profiles in mice exposed to perfluorooctanoic acid and sodium ρ-perfluorous nonenoxybenzene sulfonate.

Perfluorinated compounds (PFCs) such as perfluorooctanoic acid (PFOA) and sodium ρ-perfluorous nonenoxybenzene sulfonate (OBS) are toxic to organisms, but their toxicological differences in affecting serum amino acid metabolism remain unclear. To investigate the effects of subchronic PFOA and OBS exposure on mice’s serum amino acid metabolic profile and explore their toxicological differences. Fifteen healthy male C57BL/6N mice were randomly divided into three groups: control (CON) group, PFOA group (exposed to 3 mg/(kg·d) PFOA), and OBS group (exposed to 3 mg/(kg·d) OBS). The experiment was c

Diagnostic value of computed tomography-based muscle quality and metabolic signatures in heart failure with sarcopenia.

AIMS: Sarcopenia is a major comorbidity in heart failure (HF) patients, but its comprehensive evaluation remains challenging due to resource limitations. Computed tomography (CT) imaging allows opportunistic assessment of both muscle quantity and quality; however, its diagnostic utility and relationships with physical performance and underlying metabolic alterations remain unclear. We investigated the diagnostic utility of single-slice abdominal CT for sarcopenia assessment and explored its association with plasma amino acid profiles in HF patients. METHODS AND RESULTS: We retrospectively ana

Alteration of the follicular fluid amino acid profile reveals the important roles of several amino acids in embryo quality in patients with polycystic ovary syndrome.

BACKGROUND: Polycystic ovary syndrome (PCOS) is widely acknowledged as the prevailing reproductive endocrine disorder accompanied by numerous metabolic dysfunctions. However, there has been a lack of systematic examination regarding the amino acids profile in PCOS. There is a dearth of evidence in regard to the examination of connections between amino acid metabolites found in follicular fluid (FF) and the quality of embryos during in vitro fertilization (IVF). OBJECTIVE: Our objective was to assess amino acid signatures associated with PCOS, as well as identify potential amino acid markers f

Adaptation of body protein metabolism in adult and aging man.

Aging causes a selective loss in lean body mass, muscle being most extensively eroded. This presentation is an analysis of the consequences of this age-related loss for protein and amino acid metabolism. The use of stable isotopes allows us to measure the dynamics of protein metabolism in the intact human subject, while the output of 3-methylhistidine permits estimates of muscle turnover to be made which show a smaller reduction in body protein turnover per kg body weight in elderly subjects. This is due to the reduced contribution by muscle protein turnover, which decreases from 30 per cent o

Combined metagenomic and metabolomic analyses reveal gut microbiota dysbiosis and metabolic dysfunction in pediatric neurodevelopmental disorders.

INTRODUCTION: Neurodevelopmental disorders (NDDs) are chronic brain diseases linked to innate immune signaling abnormalities, affecting children with complex gut-brain axis etiologies and limited targeted therapies. While infant microbes/metabolites may predict childhood NDDs, their landscape and host-metabolism interactions in NDDchildren remain unclear. METHODS: This study enrolled 40 NDDchildren (mean age: 5.18 ± 1.77, F:M = 11:29) and 60 healthy controls (HCs; mean age:5.11 ± 1.42, F:M = 25:35) from Gansu Province Hospital Rehabilitation Center. Frailty is characterized by biomarker patterns reflecting inflammation or muscle catabolism in multi-morbid patients.

BACKGROUND: Frailty development is partly dependent on multiple factors like low levels of nutrients and high levels of oxidative stress (OS) and inflammation potentially leading to a muscle-catabolic state. Measures of specific biomarker patterns including nutrients, OS and inflammatory biomarkers as well as muscle related biomarkers like 3-methylhistidine (3MH) may improve evaluation of mechanisms and the complex networks leading to frailty. METHODS: In 220 multi-morbid patients (≥ 60 years), classified as non-frail (n = 104) and frail (n = 116) according to Fried's frailty criteria, we mea

Biomarkers related to fatty acid oxidative capacity are predictive for continued weight loss in cachectic cancer patients.

BACKGROUND: Cachexia is characterized by a negative protein and energy balance leading to loss of adipose tissue and muscle mass. Cancer cachexia negatively impacts treatment tolerability and prognosis. Supportive interventions should be initiated as early as possible. Biomarkers for early prediction of continuing weight loss during the course of disease are currently lacking. METHODS: In this pilot, observational, cross-sectional, case-control study, cachectic cancer patients undergoing systemic first-line cancer treatment were matched 2:1 with healthy controls according to age, gender and b

Responsiveness of muscle protein synthesis to growth hormone administration in HIV-infected individuals declines with severity of disease.

This study was undertaken to determine if human recombinant growth hormone (hrGH, 6 mg/d for 2 wk) would stimulate muscle protein synthesis in AIDS wasting. Healthy controls were compared with patients who were HIV+, had AIDS without weight loss, and had AIDS with > 10% weight loss. Before hrGH, rates of skeletal muscle protein synthesis, measured with l-[2H5]phenylalanine, were the same in controls and in all stages of disease. Rates of myofibrillar protein degradation, however, assessed from urinary excretion of 3-methyl histidine, were higher in AIDS and AIDS wasting than in HIV+ or healthy
