Bioprinting of hair follicle germs for hair regenerative medicine.

Hair regenerative medicine is a promising approach to treat hair loss. The replication of in vivo tissue configurations and microenvironments, such as hair follicle germs, has been studied to prepare tissue grafts for hair regenerative medicine. However, such approaches should be scalable, because a single patient with alopecia requires thousands of tissue grafts. In this paper, we propose an approach for the scalable and automated preparation of highly hair-inductive tissue grafts using a bioprinter. Two collagen droplets (2 µL each) containing mesenchymal and epithelial cells were placed adj

Polygonatum polysaccharide microspheres encapsulated living Clostridium butyricum for mucosal barrier repair and immune regulation.

A collaborative intervention strategy based on probiotics represents a promising therapeutic approach against inflammatory bowel disease (IBD). However, enhancing probiotic efficacy within the IBD microenvironment while minimizing side effects remains a critical challenge. Polygonatum polysaccharide (PP), a natural plant-derived bioactive polymer, has attracted significant attention due to its ability to regulate oxidative and inflammatory stress with a favorable safety profile. Herein, PP was incorporated into calcium-alginate microgels (Mgel), resulting in enhanced mechanical strength and th

Spatially Defined Cell-Secreted Protein Detection Using Granular Hydrogels: μGeLISA.

Intercellular communication through secreted proteins is necessary in essential processes such as embryo and limb development, disease progression, and immune responses. There exist many techniques to study bulk solution protein concentrations, but there is a limited set of tools to study the concentrations of cell-secreted proteins in situ within diverse cell platforms while retaining spatial information. In this study, we have developed a microgel system that is able to quantitatively measure the cell-secreted protein concentration within defined three-dimensional culture configurations with

Ultrasound-treated water-dispersible myofibrillar protein microgel particles: An effective strategy for stable Pickering emulsion gels with enhanced interfacial properties, dysphagia-oriented texture modification and improved encapsulation capacity.

To improve the poor processing adaptability and weak interfacial activity of pale, soft and exudative (PSE)-like chicken breast meat myofibrillar protein (MP) under low-salt conditions, this study proposed a colloidal restructuring strategy. PSE-like MP was diluted with water and subjected to heating to form aggregates. These aggregates were converted into water-dispersible MP microgel particles (MMPs) by ultrasound treatment and used to prepare Pickering emulsion gels for developing meat protein-based dysphagia foods. The influence of ultrasound time (20 kHz, 450 W, 0-25 min) on the structura

Biocompatible Hybrid Organic/Inorganic Microhydrogels Promote Bacterial Adherence and Eradication in Vitro and in Vivo.

Self-assembling peptides and proteins have the potential to serve as multifunctional building blocks for the generation of versatile materials for a wide range of biomedical applications. In particular, supramolecular hydrogels comprised of self-assembled protein nanofibrils, have been used in contexts ranging from tissue engineering to drug delivery. Due to the rapid emergence of multidrug resistant bacteria, development of biomaterials with intrinsic antimicrobial properties has been continuously increasing. Here, we describe hybrid organic/inorganic nanofibrillar silk microgels decorated wi

Enzymatic biodegradation of hydrogels for protein delivery targeted to the small intestine.

Multiresponsive poly(methacrylic acid-co-N-vinylpyrrolidone) hydrogels were synthesized with biodegradable oligopeptide crosslinks. The oligopeptide crosslinks were incorporated using EDC-NHS zero-length links between the carboxylic acid groups of the polymer and free primary amines on the peptide. The reaction of the peptide was confirmed by primary amine assay and IR spectroscopy. The microgels exhibited pH-responsive swelling as well as enzyme-catalyzed degradation targeted by trypsin present in the small intestine, as demonstrated upon incubation with gastrointestinal fluids from rats. Utilizing a microfluidic platform to investigate drug-eluting beads: Binding and release of amphiphilic antidepressants.

Drug-eluting beads made of responsive polyelectrolyte networks are used in the treatment of liver cancer. Aggregates of loaded drugs in complex with the networks dissolve upon release, causing swelling of the network. According to a recent mechanism the release and swelling rates are controlled by the mass transport of drug through a depletion layer created in the microgel. We hypothesise that the mechanism, in which the stability of the drug aggregates and the swelling properties of the network play crucial roles, offers means to control the release profile also for other drugs. To test this,

Bioactive and Injectable Granular Hydrogels Incorporating Decellularized Extracellular Matrix.

Decellularized extracellular matrices (dECMs) provide bioactive cues that may be useful for the repair of fibrocartilaginous tissues, such as the temporomandibular joint disc (TMJd), which lacks a natural regenerative capacity. While potent in bioactivity, dECM hydrogels do not possess the mechanical properties necessary for joint repair, motivating the development of improved materials. Granular hydrogels provide a unique opportunity to repair tissues by mechanically stabilizing the defect with injectable, jammed hydrogel microparticles that exhibit microporosity to support cellular infiltrat

Mechanically graded granular scaffolds for osteochondral tissue engineering.

Engineered scaffolds designed to approximate the mechanical microenvironment of the osteochondral unit often address this complexity using discrete, two-phase architectures that introduce mechanical discontinuities and interfacial stress concentrations rather than a contiguous stiffness transition. To address this challenge, we created a photoannealed polyethylene glycol (PEG) granular scaffold with a spatially controlled stiffness gradient within a cell-permissive, macroporous architecture. Stiffness was dictated by photoannealing microgels using a photomask. We tuned void volume and availabl

Chitosan-coated miconazole as an effective anti-inflammatory agent for the treatment of postoperative infections in obstetrics and vaginal yeast infection control on in vitro evaluations.

People with immune deficiency are at risk of developing infections caused by several bacterial and fungal species. In this work, chitosan-coated miconazole was developed by a simple sol-gel method. Miconazole is considered an effective drug to treat vaginal infection-causing bacteria and fungi. The coating of chitosan with miconazole nitrate showed the highest drug loading efficiency (62.43%) and mean particle size (2 μm). FTIR spectroscopic analysis confirmed the entrapment of miconazole nitrate into chitosan polymer. The antifungal result demonstrated that MN@CS microgel possessed notable an

Granular Hydrogels as Brittle Yield Stress Fluids.

While granular hydrogels are increasingly used in biomedical applications, methods to capture their rheological behavior generally consider shear-thinning and self-healing properties or produce ensemble metrics (e.g., dynamic moduli) while neglecting transient yielding and unyielding processes. Combining oscillatory shear testing with Brittility (Bt) via the Kamani-Donley-Rogers (KDR) model, this work shows that granular hydrogels behave as brittle yield stress fluids. This work quantifies steady and transient rheology as a function of microgel properties and granular composition for polyethyl

Bioprinting of miRNA-Induced Spheroids for Vascularized, Heterocellular Bone Regeneration.

Successful bone regeneration requires coupled osteogenic and vascular development; however, achieving simultaneous multicellular differentiation within engineered tissues remains challenging. Here, we developed a microRNA (miR)-guided spheroid platform to induce dual osteogenic and endothelial differentiation of human adipose-derived stem cells (hASCs) for vascularized bone regeneration. hASCs were transfected with miR-148b or miR-210 to promote osteogenic and vascular-associated phenotypes, respectively, and assembled into spheroids that were bioprinted within an nHA-containing GelMA microgel

Acellular matrix microgel assembly integrated with curcumin-loaded ZIF-8 nanoparticles and simvastatin promotes microfracture-based articular cartilage repair via pyroptosis inhibition, endogenous BMSC recruitment, and hyaline chondrogenic differentiation.

Articular cartilage repair remains a major clinical challenge. Although microfracture is widely employed, it often leads to fibrocartilage formation rather than functional hyaline cartilage regeneration. Herein, we develop an injectable cartilage acellular matrix (CACM) microgel assembly integrating curcumin-loaded ZIF-8 nanoparticles (Cur@ZIF-8) and simvastatin (SIM) to improve microfracture-based cartilage repair. The dynamically crosslinked microgel assembly exhibits good injectability, tissue adhesion, shape adaptability, and an interconnected microporous structure, thereby facilitating ma

Compression, expansion and relaxation of soft colloidal monolayers at the air/water interface.

The phase behavior of soft and deformable microgels at fluid interfaces is typically studied with a Langmuir trough and using uniaxial compression. In situ investigations that shine light on the structural arrangements and changes during compression are scarce. Knowledge on the phase behavior is mostly generated from ex situ observations after transfer of monolayers from the fluid interface onto a substrate. Similarly, little is known about the impact of the compression geometry and potentially occurring relaxation processes after compression. Here, we use small-angle light scattering implemen

Dynamic granular hydrogels to assess pancreatic cancer cell fate.

Granular hydrogels are an emerging biomaterial platform increasingly used in biomedical applications, including therapeutic delivery and tissue regeneration. Assembled from micron-scale hydrogel particles through physical assembly or chemical cross-linking, granular hydrogels possess micro- and macroscopic pores that facilitate molecular transport and cell migration. However, current granular hydrogels are typically fabricated with defined stiffness, porosity, and compositions that do not recapitulate the dynamic nature of native tissues, including the tumor microenvironment. To address this c

A Balance between Pro-Inflammatory and Pro-Reparative Macrophages is Observed in Regenerative D-MAPS.

Microporous annealed particle scaffolds (MAPS) are a new class of granular materials generated through the interlinking of tunable microgels, which produce an interconnected network of void space. These microgel building blocks can be designed with different mechanical or bio-active parameters to facilitate cell infiltration and modulate host response. Previously, changing the chirality of the microgel crosslinking peptides from L- to D-amino acids led to significant tissue regeneration and functional recovery in D-MAPS-treated cutaneous wounds. In this study, the immunomodulatory effect of D-

Particle Hydrogels Based on Hyaluronic Acid Building Blocks.

The extracellular matrix (ECM) provides tissues with the mechanical support, space, and bioactive signals needed for homeostasis or tissue repair after wounding or disease. Hydrogel based scaffolds that can match the bulk mechanical properties of the target tissue have been extensively explored as ECM mimics. Although the addition of microporosity to hydrogel scaffolds has been shown to enhance cell/tissue-material integration, the introduction of microporosity often involves harsh chemical methods, which limit bioactive signal incorporation and injectability. Particle hydrogels are an emergin

Machine learning-driven single-cell phenotyping in size-controlled microenvironments via parallel deterministic droplet microfluidics.

Understanding how individual cells respond to distinct physical microenvironments is critical for mechanobiology, cell therapy, and tissue engineering. Current single-cell encapsulation methods are often limited by Poisson loading and fixed droplet sizes, preventing parallel generation of multiple, size-specific microenvironments and constraining high-resolution phenotypic analyses. Here, we present a droplet microfluidic platform that enables deterministic single-cell encapsulation within microgels of multiple sizes from a single precursor stream, achieved through parallelized flow-focusing c

Poroelasticity influences the compression stiffening of model soft tissues.

The material properties of connective tissues are often considered to be dominated by the extracellular matrix. However, cells within tissues contribute significantly to the mechanics, particularly when considering volume-changing deformations such as axial compression. Recent studies have considered the compression response of model tissues at long times, where the role of the interstitial fluid is negligible. Here, we experimentally study the interplay of matrix, cell and fluid mechanics in the short time response of model tissues based on fibrous networks with embedded microgel inclusions a
