miR-151a-5p in Neuron-Derived Extracellular Vesicles Mediates Antidepressant Response.

Major depressive disorder (MDD) lacks accessible molecular markers that reflect brain pathology and monitor treatment response. Using the neuron-specific protein SNAP25, we investigated the cargo of neuron-derived extracellular vesicles (NEVs) isolated from plasma in relation to antidepressant treatment and identified miR-151a-5p as a mediator of treatment response, increasing selectively in responders while remaining low in non-responders. Plasma levels mirrored deficits in human post-mortem brain tissue from the ventral anterior cingulate cortex, a cortical area implicated in MDD. In mice, e

Social isolation recruits amygdala-medial prefrontal cortex projections to escalate alcohol drinking in male mice.

Social isolation profoundly alters motivation and increases vulnerability to alcohol misuse in humans, yet the underlying neural mechanisms remain unclear. Here we show that isolation escalates alcohol drinking in male mice but suppresses it in females. Whole-cell recordings revealed that neurons in the basolateral amygdala projecting to the medial prefrontal cortex (BLA-mPFC) track alcohol intake in both sexes. Isolation increased BLA-mPFC excitability in males but decreased it in females, mirroring their opposite behavioral adaptations. Given this divergence, we focused subsequent mechanisti

Human spinal cord organoids recapitulate developmental and disease-associated oligodendrocyte lineage signatures.

Oligodendrocytes play essential roles in central nervous system development and homeostasis, and their dysfunction is a hallmark of numerous neurological disorders. However, human in vitro systems that support oligodendrocyte lineage progression while enabling the study of disease-relevant states remain limited. Here, we establish human spinal cord organoids (hSpO) and cortico-motor assembloids as platforms to model oligodendrocyte development, neuron-glia interactions, and cytokine-induced dysfunction. We show that hSpO generate oligodendrocyte lineage populations that transcriptionally resem

Identification of nonvisual photomotor response cells in the vertebrate hindbrain.

Nonvisual photosensation enables animals to sense light without sight. However, the cellular and molecular mechanisms of nonvisual photobehaviors are poorly understood, especially in vertebrate animals. Here, we describe the photomotor response (PMR), a robust and reproducible series of motor behaviors in zebrafish that is elicited by visual wavelengths of light but does not require the eyes, pineal gland, or other canonical deep-brain photoreceptive organs. Unlike the relatively slow effects of canonical nonvisual pathways, motor circuits are strongly and quickly (seconds) recruited during th

Functional neuroanatomy and the rationale for using EEG biofeedback for clients with Asperger's syndrome.

This paper reviews the symptoms of Asperger's Syndrome (AS), a disorder along the autism continuum, and highlights research findings with an emphasis on brain differences. Existing theories concerning AS are described, including theory of mind (Hill and Frith in Phil Trans Royal Soc Lond, Bull 358:281-289, 2003), mirror neuron system (Ramachandran and Oberman in Sci Am 295(5):62-69, 2006), and Porges' (Ann N Y Acad Sci 1008:31-47, 2003, The neurobiology of autism, Johns Hopkins University Press, Baltimore, 2004) polyvagal theory. Injury-associated PACAP expression in rat sensory and motor neurons is induced by endogenous BDNF.

Peripheral nerve injury results in dramatic upregulation in pituitary adenylate cyclase activating polypeptide (PACAP) expression in adult rat dorsal root ganglia and spinal motor neurons mirroring that described for the neurotrophin brain derived neurotrophic factor (BDNF). Thus, we posited that injury-associated alterations in BDNF expression regulate the changes in PACAP expression observed in the injured neurons. The role of endogenous BDNF in induction and/or maintenance of PACAP mRNA expression in injured adult rat motor and sensory neurons was examined by intrathecally infusing or intra

Preservation of segmental hindbrain organization in adult frogs.

To test for possible retention of early segmental patterning throughout development, the cranial nerve efferent nuclei in adult ranid frogs were quantitatively mapped and compared with the segmental organization of these nuclei in larvae. Cranial nerve roots IV-X were labeled in larvae with fluorescent dextran amines. Each cranial nerve efferent nucleus resided in a characteristic segmental position within the clearly visible larval hindbrain rhombomeres (r). Trochlear motoneurons were located in r0, trigeminal motoneurons in r2-r3, facial branchiomotor and vestibuloacoustic efferent neurons i

Specific somatic sensory relays in the mammalian diencephalon.

There are five major domains within the diencephalon that receive input from ascending somatic sensory pathways serving the limbs, trunk and viscera. These domains include the ventroposterolateral nucleus (VPL), the posterior group complex (PO), the zona incerta (ZI), the intralaminar complex and the thalamic reticular nucleus. Of these five domains, VPL is commonly considered a "specific" somatic "relay". The reasons for this view are that VPL receives dense, synaptically secure and precisely organized input from the equally precisely organized dorsal column nuclei (DCN), that most of VPL's n

Neuronal Gtf2i deletion alters mitochondrial and autophagic properties.

Gtf2i encodes the general transcription factor II-I (TFII-I), with peak expression during pre-natal and early post-natal brain development stages. Because these stages are critical for proper brain development, we studied at the single-cell level the consequences of Gtf2i's deletion from excitatory neurons, specifically on mitochondria. Here we show that Gtf2i's deletion resulted in abnormal morphology, disrupted mRNA related to mitochondrial fission and fusion, and altered autophagy/mitophagy protein expression. These changes align with elevated reactive oxygen species levels, illuminating Gt

Abdominal vagal signalling: a novel role for cholecystokinin in circulatory control?

It is generally accepted that the gastrointestinal circulation is primarily under the control of the enteric nervous system. However, recent studies have demonstrated that the sympathetic nervous system may play a greater role in postprandial gastrointestinal circulatory function than was thought previously. Cholecystokinin (CCK) is a gastrointestinal hormone released from enteroendocrine cells lining the intestinal mucosa in response to feeding. Systemic administration of CCK induces gastrointestinal vasodilation mediated by withdrawal of sympathetic vasomotor drive. CCK differentially influe

Aplasics born without hands mirror the goal of hand actions with their feet.

The premotor and parietal mirror neuron system (MNS) is thought to contribute to the understanding of observed actions by mapping them onto "corresponding" motor programs of the observer [1-24], but how would the MNS respond to the observation of hand actions if the observer never had hands? Would it not show changes of blood-oxygen-level dependent (BOLD) signal, because the observer lacks motor programs that can resonate [12, 25, 26], or would it show significant changes because the observer has motor programs for the foot or mouth with corresponding goals [15, 17, 19, 27, 28]? We scanned two

Temporal dynamics of cortical representation for action.

Brain-imaging studies have shown that the human Broca's region and precentral motor cortex are activated both during execution of hand actions and during observation of similar actions performed by other individuals. We aimed to clarify the temporal dynamics of this cortical activation by neuromagnetic recordings during execution, on-line imitation, and observation of right-hand reaching movements that ended with a precision pinch of the tip of a manipulandum. During execution, the left inferior frontal cortex [Brodmann's area (BA) 44] was activated first (peak approximately 250 ms before the

Distinct and additive effects of visual and vibratory feedback for motor rehabilitation: an EEG study in healthy subjects.

INTRODUCTION: The use of visual and proprioceptive feedback is a key property of motor rehabilitation techniques. This feedback can be used alone, for example, for vision in mirror or video therapy, for proprioception in focal tendon vibration therapy, or in combination, for example, in robot-assisted training. This Electroencephalographic (EEG) study in healthy subjects explored the distinct neurophysiological impact of adding visual (video therapy), proprioceptive (focal tendinous vibration), or combined feedback (video therapy and focal tendinous vibration) to a motor imagery task. METHODS

Topographic organization, number, and laminar distribution of callosal cells connecting visual cortical areas 17 and 18 of normally pigmented and Siamese cats.

The callosal connections between visual cortical areas 17 and 18 in adult normally pigmented and "Boston" Siamese cats were studied using degeneration methods, and by transport of WGA-HRP combined with electrophysiological mapping. In normal cats, over 90% of callosal neurons were located in the supragranular layers. The supragranular callosal cell zone spanned the area 17/18 border and extended, on average, some 2-3 mm into both areas to occupy a territory which was roughly co-extensive with the distribution of callosal terminations in these areas. The region of the visual field adjoining the

Superficial NK1 expressing spinal dorsal horn neurones modulate inhibitory neurotransmission mediated by spinal GABA(A) receptors.

Lamina 1 projection neurones which express the NK1 receptor (NK1R+) drive a descending serotonergic pathway from the brainstem that enhances spinal dorsal horn neuronal activity via the facilitatory spinal 5-HT3 receptor. Selective destruction of these cells via lumbar injection of substance P-saporin (SP-SAP) attenuates pain behaviours, including mechanical and thermal hypersensitivity, which are mirrored by deficits in the evoked responses of lamina V-VI wide dynamic range (WDR) neurones to noxious stimuli. To assess whether removing the origin of this facilitatory spino-bulbo-spinal loop re

The deltoid muscle and the pattern of paresis in ALS.

There is neuroanatomical and clinical evidence that the corticospinal tract governs the patterns of pareses in sporadic ALS. These patterns are mirrored by phylogenetically young monosynaptic corticomotor neuronal connections. It is well known that, clinically, dysfunction of the deltoid muscle contributes considerably to the early disability of the ALS patient. In this study, we prospectively compared the degree of pareses of the deltoid muscle with the triceps and biceps brachii in N = 71 patients (426 muscles). We could show that the extent of involvement of the deltoid muscle early in the

Effects of Microstimulation in the Anterior Intraparietal Area during Three-Dimensional Shape Categorization.

The anterior intraparietal area (AIP) of rhesus monkeys is part of the dorsal visual stream and contains neurons whose visual response properties are commensurate with a role in three-dimensional (3D) shape perception. Neuronal responses in AIP signal the depth structure of disparity-defined 3D shapes, reflect the choices of monkeys while they categorize 3D shapes, and mirror the behavioral variability across different stimulus conditions during 3D-shape categorization. However, direct evidence for a role of AIP in 3D-shape perception has been lacking. We trained rhesus monkeys to categorize d

Binocular deficits associated with early alternating monocular defocus. II. Neurophysiological observations.

Experiencing binocularly conflicting signals early in life dramatically alters the binocular responses of cortical neurons. Because visual cortex is highly plastic during a critical period of development, cortical deficits resulting from early abnormal visual experience often mirror the nature of interocular decorrelation of neural signals from the two eyes. In the preceding paper, we demonstrated that monkeys that experienced early alternating monocular defocus (-1.5, -3.0, or -6.0 D) show deficits in stereopsis that generally reflected the magnitude of imposed monocular defocus. Because thes

Agent-based representations of objects and actions in the monkey pre-supplementary motor area.

Information about objects around us is essential for planning actions and for predicting those of others. Here, we studied pre-supplementary motor area F6 neurons with a task in which monkeys viewed and grasped (or refrained from grasping) objects, and then observed a human doing the same task. We found "action-related neurons" encoding selectively monkey's own action [self-type (ST)], another agent's action [other-type (OT)], or both [self- and other-type (SOT)]. Interestingly, we found "object-related neurons" exhibiting the same type of selectivity before action onset: Indeed, distinct sets
