[Effect of histamine-blocking substances on mitogen-induced lympho proliferation in rheumatic diseases].

Peripheral blood lymphocytes of patients with rheumatoid arthritis, psoriatic arthritis and healthy controls, isolated by a ficoll density gradient, were incubated with different concentrations of histamine, cimetidine and diphenhydramine (DPH) and simultaneously stimulated with phytohemagglutinine (PHA). In comparison to healthy control persons the lymphoproliferation rate in arthritic patients could be increased about 20% by addition of the histamine-blocking agents cimetidine and DPH. This suggests the existence of in vivo histamine-induced suppressor cells in arthritic patients, which can

Lymphoblastoid cell-induced suppression of human peripheral blood leukocyte mitogenic responses.

Two lymphoblastoid tumor cell lines, the Burkitt lymphoma derived BJAB cell line which is free of Epstein-Barr virus (EBV) and B95-8 cells, which are marmoset lymphocytes transformed by EBV isolated from an infectious mononucleosis patient, were studied in regards to their effects on the blastogenic responsiveness of normal human peripheral blood leukocytes stimulated in vitro with mitogens. Mitomycin C treated tumor cell suspensions, when cocultured with normal human blood leukocytes, markedly depressed the expected blastogenic responses in vitro to concanavalin A, pokeweed mitogen, and phyto

Some tetracycline drugs suppress mitogen-stimulated lymphocyte growth but others do not.

Eight tetracycline drugs were tested for inhibitory effect on phytohaemagglutinin (PHA) stimulated growth (measured by [3H]-TdR uptake) of peripheral blood mononuclear cells: at least 26 normal subjects were tested with each drug. Doxycycline reduced DNA synthesis appreciably at concentrations within and just above the therapeutic range of blood levels: it was the most potent inhibitor, but demeclocycline, methacycline and minocycline had similar, if less potent, effects. Tetracycline, oxytetracycline, chlortetracycline and clomocycline did not inhibit DNA synthesis of 3-day cultures even at c

Membrane fractions of trypanosomes mimic the immunosuppressive and mitogenic effects of living parasites on the host.

African trypanosomiasis in mice leads to profound changes in lymphoid tissues. In an attempt to define the nature of the trypanosome stimulus, we have studied the effect of radio-attenuated trypanosomes and their subcellular fractions in vivo. We find that relatively low doses of irradiated Trypanosoma brucei S42 injected into (CBA/H x C57B1/6)F1 mice mimicked the previously reported effects of infective parasites. 2 x 10(7) irradiated trypanosomes caused a greater than two-fold increase in spleen weight accompanied by a roughly 10-fold increase in background plaque forming cells (PFC) to shee

Mitogen stimulation and distribution of T- and B-lymphocytes during natural rubella infection.

The phytohaemagglutinin (PHA) response of lymphocytes from seven patients with natural rubella infection was investigated during the acute, early and late convalescence stages of disease. When the lymphocytes were cultured in autologous serum, a moderate depression of the response, following stimulation with PHA in both optimal and suboptimal concentrations was obtained two weeks after the onset of exanthema (early convalescence). Two months later (late convalescence), the lymphocyte stimulation response had returned to almost normal values. On the other hand, lymphocytes incubated in pooled h

The mitogenic effect of the lymphocytosis promoting factor from Bordetella pertussis on human lymphocytes.

The purified lymphocytosis promoting factor (LPF) from Bordetella pertussis was found to be a potent mitogen for peripheral blood lymphocytes (PBL) from normal adults as well as for cord blood lymphocytes. Proliferation occurred in autologous plasma or fetal calf serum, regardless of previous exposure to pertussis infection or immunization. Only one adult human serum, from a physician constantly working with B. pertussis, inhibited the mitogenic response to LPF and this serum was shown to contain precipitating antibody against LPF. The proliferative effect of LPF was characteristic of a "nonsp

Lymphocyte function in experimental african trypanosomiasis: mitogenic effects of trypanosome extracts in vitro.

Extracts of Trypanosoma brucei and Trypanosoma congolense were incubated in vitro with nonimmune lymphocytes of mice, rats, guinea pigs, and rabbits in order to test for mitogenic effects or for other characteristics of polyclonal B lymphocyte activators. Trypanosome extracts (TE) were not mitogenic for spleen cells of mice, rats, and guinea pigs in vitro, nor did the parasite extracts alter the mitogenic responses of lymphocytes from these animals to known B- and T-cell mitogens. TE also failed to induce polyclonal antibody synthesis in mouse spleen cell cultures in an in vitro antibody respo

Mitogen-activated protein kinase signaling and drug-induced liver injury: A molecular connection.

Drug-induced liver injury (DILI) is a global health concern and one of the leading causes for the withdrawal of drugs from the market. DILI can exacerbate chronic liver injury to acute or chronic liver failure. The mitogen-activated protein kinase (MAPK) signaling cascade is the most conserved pathway that regulates key physiological processes, including cell proliferation, differentiation, inflammation, stress responses, and apoptosis. MAPK pathways such as extracellular signal-regulated kinase (ERK), c-Jun N-terminal kinase (JNK), and p38 are activated by growth factors, cytokines and oxidat

Hamayou () protein hydrolysate ameliorates depression by regulating the mitogen-activated protein kinase pathway.

OBJECTIVE: To clarify the effect of Hamayou (Oviductus Ranae) protein hydrolysate (ORPH) on depression and its exact underlying mechanism from a new perspective. METHODS: We used the Chronic Unpredictable Mild Stress (CUMS) method to prepare a mouse model of depression and lipopolysaccharide (LPS) to prepare a model of BV2 cellular inflammation to investigate the antidepressant effect and mechanism of action of ORPH. Behavioral changes in mice and cerebral blood flow were detected by behavioral experiments and scatter imaging. Levels of corticosterone (CORT), proinflammatory cytokines and neu

RAS/Mitogen-Activated Protein Kinase Signaling Pathway in Testicular Germ Cell Tumors.

Germ cell tumors (GCTs) are relatively rare tumors. However, they are the most diagnosed malignancies occurring in the testis among men aged between 15 and 40 years. Despite high aneuploidy and a paucity of somatic mutations, several genomic and transcriptomic assays have identified a few significantly mutated somatic genes, primarily KIT and K-RAS. The receptor Tyrosine Kinase (RTK) pathway and the downstream related Mitogen-Activated Protein Kinase (MAPK) cascades are crucial signal transduction pathways that preside over various cellular processes, including proliferation, differentiation,

Mitogen-Activated Protein Kinase Inhibitor-Induced Inflammatory Alopecia in Woman With Ovarian Cancer.

Inflammatory alopecia is an increasingly reported side effect of targeted cancer therapies. Here we report one case of inflammatory alopecia secondary to mitogen-activated protein kinase kinase (MEK) inhibitor agent Trametinib in a woman with ovarian cancer. Biopsies of the scalp were consistent with early scarring alopecia compatible with drug-induced alopecia. Significant improvement in hair loss occurred after treatment with intralesional Kenalog (ILK) injections and oral isotretinoin. Though acute alopecia has been described in patients using MEK inhibitors, this is the first reported case

Immunomodulatory effects of copper nanoparticles against mitogen-stimulated rat splenic and thymic lymphocytes.

In the present study, we have evaluated the effects of copper (Cu) nanoparticles (NPs) on the primary B-and T-lymphocytes proliferation, cytokine levels, and bio-distribution through in vitro, in vivo and ex-vivo studies to allow the possible exploitations of CuNPs in biomedical applications. CuNPs were characterized by UV-Visible spectroscopy, transmission electron microscopy (TEM), and nanoparticle tracking analysis (NTA). The proliferative response of lymphocytes was studied by 3H-thymidine incorporation assay and lymphocyte viability through trypan blue assay. The bio-distribution of CuNPs

Systematic review of p38 mitogen-activated kinase and its functional role in reproductive tissues.

Oxidative stress (OS) plays a role in uterine tissue remodeling during pregnancy and parturition. While p38 MAPK is an OS-response kinase, a precise functional role is unknown. Therefore, we conducted a systematic review of literature on p38 MAPK expression, activation, and function in reproductive tissues throughout pregnancy and parturition, published between January 1980 and August 2017, using four electronic databases (Web of Science, PubMed, Medline, and CoCHRANE). We identified 418 reports; 108 were selected for full-text evaluation and 74 were included in final review. p38 MAPK was inve

Toxoplasma gondii mitogen-activated protein kinases are associated with inflammasome activation in infected mice.

Toxoplasma gondii can activate the nucleotide-binding domain and leucine-rich repeat-containing proteins NLRP1/3 inflammasomes, which mediate host resistance to the infection. Here we showed that deletion of mitogen-activated protein kinases MAPK1 and MAPK2 of type I parasite decreases acute virulence in mice, characterized by low levels of interleukin (IL)-18, NLRP1/3, ASC, and caspase-1, and high levels of IL-10 and interferon (IFN)-β transcripts. Additionally, the mutants increased phosphorylation of STAT1, and decreased phosphorylation of STAT3. These findings suggest that MAPKs are associ

Activin A induces skeletal muscle catabolism via p38β mitogen-activated protein kinase.

BACKGROUND: Activation of type IIB activin receptor (ActRIIB) in skeletal muscle leads to muscle atrophy because of increased muscle protein degradation. However, the intracellular signalling mechanism that mediates ActRIIB-activated muscle catabolism is poorly defined. METHODS: We investigated the role of p38β mitogen-activated protein kinases (MAPK) in mediating ActRIIB ligand activin A-activated muscle catabolic pathways in C2C12 myotubes and in mice with perturbation of this kinase pharmacologically and genetically. RESULTS: Treatment of C2C12 myotubes with activin A or myostatin rapidly

Deletion of mitogen-activated protein kinase 1 inhibits development and growth of Toxoplasma gondii.

Mitogen-activated protein kinases (MAPKs) regulate key signaling events in a variety of eukaryotic cells. Toxoplasma gondii, the causative agents of toxoplasmosis, possesses a p38α MAPK homologue, MAPK1, which is an important manipulator of host immunity and virulence in mice. In this work, we showed an increased transcript level of MAPK1 in T. gondii during bradyzoite differentiation induced by alkaline treatment and heat shock in vitro, suggesting that MAPK1 may be associated with bradyzoite differentiation. The biological roles of MAPK1 of T. gondii were investigated by construction of a MA

Mitogen-Activated Protein Kinase Phosphatase (MKP)-1 in Nervous System Development and Disease.

Mitogen-activated protein kinase phosphatase (MKP)-1 provides a negative feedback mechanism for regulating mitogen-activated protein kinase (MAPK) activity and thus a variety of cellular processes such as proliferation, differentiation, growth and apoptosis. MKP-1 is established as a central regulator of a variety of functions in the immune, metabolic and cardiovascular systems, and it is now increasingly acknowledged as having a role to play in the nervous system. It has been implicated in regulating processes of neuronal cell development and death as well as in glial cell function. Reduced M

A role for mitogen kinase kinase 3 in pulmonary inflammation validated from a proteomic approach.

Proteomics is a powerful tool to ascertain which proteins are differentially expressed in the context of disease. We have used this approach on inflammatory cells obtained from patients with asthma to ascertain whether novel drugs targets could be illuminated and to investigate the role of any such target in a range of in vitro and in vivo models of inflammation. A proteomic study was undertaken using peripheral blood mononuclear cells from mild asthmatic subjects compared with healthy subjects. The analysis revealed an increased expression of the intracellular kinase, mitogen activated protei
