Effect of GNTI, a kappa opioid receptor antagonist, on MK-801-induced hyperlocomotion and stereotypy in mice.

AIM: To examine the effect of GNTI [5'-guanidinyl-17-(cyclopropylmethyl)-6,7- dehydro-4,5alpha-epoxy-3,14-dihydroxy-6,7-2',3'-indolomorphinan], a selective antagonist for the kappa opioid receptor, in the MK-801 (dizocilpine maleate)-induced behavioral model of psychosis in schizophrenia as a way to explore the involvement of the kappa opioid receptor in modulating psychotic symptoms of schizophrenia. METHODS: Two doses of MK-801 (0.3 mg/kg and 0.6 mg/kg) were administered by systemic injection in mice to induce psychosis-like behavior as a rodent schizophrenia model, preceded by an injection

[Optimum dose of intravenous butorphanol as a supplemental drug during regional anesthesia].

The optimum doses according to age of butorphanol tartrate as a supplemental drug during epidural and spinal anesthesia were investigated in 60 patients without complications. We classified patients into 4 groups by age as A, B, C and D-group. A-group consisting of patients between 20 to 40 years received 1.5mg of butorphanol. B-group 41-60, C-group 61-75 and D-group over 76 years received 1.0mg, 0.5mg and 0.25mg respectively. After operation was started without pain, and during the stable period, we determined by age, doses of butorphanol injected intravenously in one minute. HRs, mean BPs, r

Nalbuphine reverses fentanyl-related EEG-changes in man.

Nalbuphine, chemically related to naloxone however, with mixed agonist-antagonist activity, was considered to be of clinical interest in reversing opioid anesthesia. In an open-end study 15 patients (mean age 40 years) undergoing orthopedic surgery received nalbuphine (0.1 mg/kg) after fentanyl (total dose 17.4 micrograms/kg over a mean of 95 min). In order to establish the state of vigilance, continuous EEG-recording of the posterior region of the cortex (P3-O1) was done with the aid of power spectral analysis using fast Fourier transformation (FFT). Additionally blood pressure and heart rate

Cardiorespiratory effects of nalbuphine and morphine premedication in adult cardiac surgical patients.

Premedication for cardiac surgical patients attempts to achieve relief of anxiety with minimal or no cardiorespiratory changes. Nalbuphine (NUBAIN) and morphine produced similar decreases in respiratory rate (1-3 breaths per min) and PaO2 (averaging 1.34 kPa), and increases in arterial PaCO2 (averaging 0.56 kPa). Cardiovascular changes were not seen with either agent. A similar degree of sedation and relief of anxiety was achieved with both drugs. Side effects were minimal and infrequent. Nalbuphine produces respiratory depression and sedation without cardiovascular changes which is comparable

Effects of three narcotic antagonists (naltrexone, diprenorphine, and S-20682) on blood pressure, heart rate and electrical cortical activity.

Three narcotic antagonist drugs (EN-1639 or naltrexone, M-5050 or diprenorphine and the 6-oxo analogue of oxilorphan, S-20682) were evaluated in increasing doses (1-80 micrograms/kg) in the unanaesthetised dog for possible effects on blood pressure, heart rate, respiratory rate, arterial blood gases, and convulsive EEG changes. Compared to the control-awake situation, diprenorphine and S-20682 induced hypotension (maximum fall 15%), bradycardia (maximum 12%) and bradypnoea (maximum 25%). These effects were not increased at doses higher than 20 micrograms/kg. Despite the induced bradypnoea, the

[Buprenorphine as a postoperative analgesic following halothane anesthesia. Hemodynamic and respiratory effects].

In 10 healthy patients, buprenorphine was given as the postoperative analgesic (dosage: peripheral venous injection of 5 microgram/kg BW) after traumatological interventions in the lower extremities, which had been performed under barbiturate-induced halothane anaesthesia. The haemodynamic investigations revealed that buprenorphine has only a minor effect on the high pressure system. In the area of the pulmonary circulation, there was a significant increase in mean pulmonary artery pressure from 15.9 mm Hg to 17.8 mm Hg (+12%), as well as an increase in pulmonary vascular resistance by 16.5%.

Benzomorphan interactions with acetylcholine receptor complexes from Torpedo.

Benzomorphan interactions with acetylcholine receptor complexes from Torpedo californica electric organ were investigated using radiolabelled probes. All of the benzomorphans had minimal affinity for the acetylcholine binding site. Four benzomorphans (N-allylnormetazocine, cyclazocine, phenazocine and pentazocine) inhibited the binding of tritiated perhydrohistrionicotoxin and phencyclidine to ion channel sites with IC50 values between 0.4 and 5 microM. Two other benzomorphans, ketazocine and ethylketocyclazocine, were much less active. Thus, ion channel affinity is not limited to benzomorphan

Sublingual buprenorphine used postoperatively: ten hour plasma drug concentration analysis.

1 A 10 h study of plasma drug concentrations of the opiate buprenorphine after use was designed because a previous 3 h study had shown that peak plasma drug concentrations in some patients had not occurred by 3 h after the sublingual dose. 2 Fifteen postoperative patients were studied: at 3 h after a 0.3 mg intravenous dose five patients received a sublingual preparation of 0.4 mg of buprenorphine, five 0.8 mg of buprenorphine and five placebo. Plasma drug concentrations of buprenorphine were measured by specific radioimmuno-assay. 3 Plasma drug concentrations after sublingual buprenorphine we

Reversible suppression of spermiogenesis in beagle dogs following overdosage with the narcotic analgesic cogazocine lactate.

Acute single overdosage (1.0 mg/kg, i.m.) with the novel benzmorphan narcotic analgesic agent Cogazocine lactate lowered serum LH and testosterone concentrations of Beagle dogs. The effect was rapidly reversible and persisted for less than 24 h after drug administration. Repeated administration (1.0 mg/kg, i.m.) induced reversible suppression of spermatid maturation with subsequent effects on semen characteristics. Examination of circulating LH and testosterone concentrations, together with pituitary and prostate morphology, suggested that the changes were due to inhibition of LH release. It i

[Comparison between the side-effects of buprenorphine and morphine in conscious man (author's transl)].

Side-effects of increasing doses of morphine (0.100, 0.150, 0.200 mg/kg) and buprenorphine (0.0015, 0.003, 0.006 mg/kg), given intramuscularly, are clinically observed in conscient subjects suffering from intense pain in the facial or trigeminal nerves territory. Buprenorphine induces a drop in ventilation even with the lower doses, which persists for 120-180 minutes. This effect is more important after the injection of an equianalgesic dose of morphine. The same phenomenon is observed for the drop in heart rate and for the hypotension, which remained in all cases very slight. The central side

Effects of the μ-opioid peptide and nociceptin/orphanin FQ peptide receptor agonist BU08028 on cocaine self-administration in male rhesus monkeys.

Cocaine use disorder (CUD) persists as a major public health concern. The lack of US Food and Drug Administration-approved pharmacotherapies for CUD underscores the critical need for developing novel pharmacological treatments. Evidence from rodent models indicates that stimulating brain receptors for the nociceptin/orphanin FQ peptide can decrease self-administration of abused drugs, including cocaine. In the present study, effects of the buprenorphine analog (2S)-2-[(5R,6R,7R,14S)-N-cyclopropylmethyl-4,5-epoxy-6,14-ethano-3-hydroxy-6 methoxy morphinan- 7-yl]-3,3-dimethylpentan-2-ol (BU08028)

Comparison of the μ-Opioid Receptor Antagonists Methocinnamox and Naloxone to Reverse and Prevent the Ventilatory Depressant Effects of Fentanyl, Carfentanil, 3-Methylfentanyl, and Heroin in Male Rats.

The number of opioid overdose deaths has increased significantly over the past decade. The life-threatening effect of opioids is hypoventilation, which can be reversed by the μ-opioid receptor (MOR) antagonist naloxone; however, because of the very short duration of action of naloxone, reemergence of MOR agonist-induced hypoventilation can occur, requiring additional doses of naloxone. The MOR antagonist methocinnamox (MCAM) antagonizes hypoventilation by the nonmorphinan fentanyl and the morphinan heroin in laboratory animals with an unusually long duration of action.
