A relation between (NAD+)/(NADH) potentials and glucose utilization in rat brain slices.

Changes in several parameters involved in the control of metabolism were correlated with changes in glucose utilization in rat brain slices incubated under conditions which reduced glucose oxidation by 40 to 70%. The parameters included: the concentrations of ATP, ADP, AMP, and the adenylate energy charge; the cytoplasmic oxidation-reduction state ([NAD+]/[NADH]), determined from the [pyruvate]/[lactate] equilibrium; the mitochondrial oxidation-reduction state, determined from the [NH4+] ]2-oxoglutarate]/[glutamate] Equilibrium; the cytoplasmic and mitochondrial oxidation-reduction potentials

Kidney function during hydropenia nad water diuresis in patients with idiopathic recurring nephrolithiasis.

The kidney function of 41 patients with idiopathic recurring kidney stones was investigated by inulin- and PAH-clearances and by measuring the excretion of electrolytes during hydropenia and water diuresis. All patients had normal inulin- and PAH-clearances and normal concentrating capacity as indicated by free water reabsorption (Tc H2O). Seven patients, all of whom had previously been found to have low excretion of magnesium in urine, were unable to dilute their urine. During water diuresis these patients also had lower osmolar clearance and lower excretion of sodium than the other patients.

Genetic modulation of NAD(P)H homeostasis reveals distinct redox-coupled metabolic vulnerabilities.

An elevated NADH/NAD+ ratio (NADH-reductive stress) is a hallmark of mitochondrial dysfunction, yet how it drives pathologies ranging from primary mitochondrial disorders to ageing remains poorly understood1,2. Here, we show that mitochondrial respiratory chain inhibition in primary human fibroblasts induces NADH-reductive stress that coexists with profound NADPH depletion. Using the Escherichia coli soluble transhydrogenase (EcSTH)3 to model this redox coupling in cells with intact mitochondria, we demonstrate that perturbation of both NAD(H) and NADP(H) redox pairs elicits metabolomic and tr

Tyrosine phosphorylation and dimerization cooperatively activate NAMPT to enable NAD+ synthesis in cancer.

Nicotinamide phosphoribosyltransferase (NAMPT), the rate-limiting enzyme in the NAD⁺ salvage pathway, is frequently upregulated in cancer, yet mechanisms regulating its catalytic activity remain undefined. We identify NAMPT as a direct substrate of multiple proto-oncogenic tyrosine kinases, including ALK, insulin receptor, IGF1R, and PDGFRA. Phosphoproteomics identified NAMPT Y188 as the major phosphorylation site, including the oncogenic fusion kinase NPM1::ALK. NAMPT interacted with NPM1::ALK in the cytoplasm, nucleus, and mitochondria, while Y188 phosphorylation enhanced catalytic activity,

NAD+ sensing by PARP7 regulates the C/EBPβ-dependent transcription program during adipogenesis.

We identified poly(ADP-ribose) polymerase 7 (PARP7), a mono(ADP-ribosyl) transferase, as a regulator of C/EBPβ-dependent proadipogenic gene expression. PARP7 functions as a nuclear NAD+ sensor; at higher nuclear NAD+ concentrations in undifferentiated preadipocytes, PARP7 is catalytically active for auto-mono(ADP-ribosyl)ation (autoMARylation). As nuclear NAD+ concentrations decline upon differentiation, autoMARylation decreases dramatically. AutoMARylation promotes instability of PARP7 through an E3 ligase-ubiquitin-proteasome pathway mediated by the ubiquitin E3 ligases DTX2 and RNF114, whic

From NAD+ metabolism to skeletal muscle function: The emerging role of ADP-ribosylation.

Nicotinamide adenine dinucleotide (NAD+) is a metabolic coenzyme and substrate for enzymes catalyzing post-translational modifications such as ADP-ribosylation. This reversible modification, mediated by ADP-ribosyltransferases and hydrolases, regulates cellular processes. Although studied in cancer and DNA repair, its roles in skeletal muscle remain less defined. Emerging evidence shows ADP-ribosylation modifies structural proteins like actin and desmin, affecting filament organization, contractility, and calcium signaling. It contributes to muscle regeneration by regulating satellite cell act

Phage RyR-domain proteins degrade ADPR-based immune signals and fuel NAD+ synthesis.

Bacterial, plant, and animal cells synthesize nucleotide immune signals as a conserved strategy to defend against viral infection1-4. In bacteria, Thoeris anti-phage defense systems convert nicotinamide adenine dinucleotide (NAD+) into the cyclic ADP-ribose signals 2'cADPR and 3'cADPR to activate downstream effectors and restrict viral replication5-8. Phage proteins can bind and sequester Thoeris signals6,9-13, but no mechanisms are known to degrade the exceptionally stable 2'cADPR and 3'cADPR molecules and terminate immune activation. Here we use a forward biochemical screen to discover the m

Differential NAD + availability may drive asymmetric SIRT7 activity in tumor and immune cells.

Metabolic competition within the tumor microenvironment shapes the availability of nutrients and metabolites that regulate both tumor progression and antitumor immunity. Among the molecules linking metabolism to gene regulation, the NAD + -dependent enzyme SIRT7 has emerged as an important regulator of ribosome biogenesis, genome stability, chromatin organization, and metabolic adaptation. Although SIRT7 is frequently associated with tumor progression, emerging evidence indicates that it also supports the metabolic fitness and effector functions of immune cells, suggesting that its biological

Human whole-blood NAD+ levels do not vary with age or lifestyle interventions.

Nicotinamide adenine dinucleotide (NAD+) levels in blood and tissues are widely proposed to decline with age, yet evidence in human blood is inconsistent. Using a rigorously validated ultra-high-performance liquid chromatography coupled with high-resolution mass spectrometry system that accounts for real-world analytical variability, we quantify NAD+ across seven independent human cohorts. We find that whole-blood NAD+ levels remain remarkably stable with age and across lifestyle interventions, but change in response to nicotinamide riboside supplementation, as expected. Our results challenge

Targeting the NAD+-SIRT3 axis to mitigate metabolic memory in diabetic kidney disease.

Metabolic memory-the persistent risk of diabetic complications after early hyperglycemia-drives progressive renal injury in diabetic kidney disease (DKD) via sustained oxidative stress, inflammation, and epigenetic reprogramming. We synthesize clinical and experimental evidence showing the nicotinamide adenine dinucleotide (NAD+)-SIRT3 (sirtuin 3) axis as a central mechanistic hub linking mitochondrial dysfunction to epigenetic and inflammatory programs in DKD metabolic memory, while evaluating restoration strategies. Integrating data from preclinical, cellular, and human studies, we review SI

A metabolic constraint in de novo NAD+ synthesis drives mucosal inflammation in IBD.

BACKGROUND: Inflammatory bowel disease (IBD) is associated with energy deficiency and perturbed metabolism of the essential amino acid tryptophan (Trp). OBJECTIVE: We aimed to determine whether excessive Trp degradation fuels or compensates for inflammation through de novo nicotinamide adenine dinucleotide (NAD+) synthesis. DESIGN: A prospective systems medicine approach (metabolomics, transcriptomics) was employed longitudinally in patients with advanced IBD therapy. Findings were validated with targeted Trp metabolomics in experimental colitis and in vitro in fibroblasts, intestinal epithe

Spatiotemporal multi-omics analysis uncovers NAD-dependent immunosuppressive niche triggering early gastric cancer.

Understanding the cellular origins and early evolutionary dynamics that drive the initiation of carcinogenesis is critical to advancing early detection and prevention strategies. By characterizing key molecular, cellular and niche events at the precancerous tipping point of early gastric cancer (EGC), we aimed to develop more precise screening tools and design targeted interventions to prevent malignant transformation at this stage. We utilized our AI models to integrate spatial multimodal data from nine EGC endoscopic submucosal dissection (ESD) samples (covering sequential stages from normal

NAD+ rescues Phosphatidylinositol-induced granulosa cell Pyroptosis to restore follicular development in PCOS.

Polycystic ovary syndrome (PCOS) is a systemic endocrine metabolic disorder that seriously affects women's reproductive health. This study aimed to investigate the mechanism by which NAD+ precursors inhibits granulosa cell (GCs) pyroptosis induced by increased phosphatidylinositol (PI) levels in PCOS. Metabolomic profiling of PCOS patients demonstrated significant lipid metabolic disturbances, with differentially metabolites significantly enriched in phosphatidylinositol signaling, pyroptosis and inflammatory pathways (p < 0.05). Furthermore, PI levels were higher in the follicular fluid of PC

NAD+-Boosters Improve Mitochondria Quality Control In Parkinson's Disease Models Via Mitochondrial UPR.

Serving as a pivotal hub for cellular metabolism and intracellular signaling, the mitochondrion has emerged as a crucial organelle whose dysfunction is linked to many human diseases, including neurodegenerative disorders, particularly Parkinson's disease (PD). However, whether mitochondrial quality control (MQC) can be targeted for therapeutic interventions remains uncertain. This study uses clinical samples, molecular biology techniques, pharmacological interventions, and genetic approaches to investigate the significance of NAD+ levels in cross-species models of PD. These results reveal that

NAD+ modulation of intestinal macrophages renders anti-inflammatory functionality and ameliorates gut inflammation.

Macrophages can maintain gut immune homeostasis by driving clearance of infection, but also can prevent chronic inflammation and induce tissue repair. Reduced nicotinamide adenine dinucleotide (NAD+) levels in macrophages have been reported to be associated with the onset of severe colitis. Given that dysregulation of gut macrophages plays a significant role in inflammatory bowel disease (IBD), they represent a potential target for novel therapies. Here we show an IBD therapeutic candidate LMT503, a substrate that modulates NADH quinone oxidoreductase (NQO1), which induces anti-inflammatory ma

Photoelectrochemical/fluorescent detection of NAD+ based on gold nanoparticles acted as ethanol dehydrogenase.

A dual-signal photoelectrochemical-fluorescent (PEC-FL) sensing method was developed for the detection of nicotinamide adenine dinucleotide (NAD+), utilizing gold nanoparticles (AuNPs) as nanozymes. Acting as ethanol dehydrogenase mimics, AuNPs catalyzed the reduction of NAD+ to NADH in the presence of ethanol. The generated NADH, serving as an electron donor, was oxidized at the surface of the ITO/BiVO4/AuNPs electrode, leading to changes in the PEC signal. Furthermore, NADH emitted fluorescence at 470 nm, enabling specific detection of NAD+. The method demonstrated excellent linearity for NA

Tubular insulin-induced gene 1 deficiency promotes NAD+ consumption and exacerbates kidney fibrosis.

Profibrotic proximal tubules (PT) were identified as a unique phenotype of proximal tubule cells (PTCs) in renal fibrosis by single-cell RNA sequencing (scRNA-seq). Controlling the process of renal fibrosis requires understanding how to manage the S1 subset's branch to the S3 subset rather than to the profibrotic PT subset. Insulin-induced gene 1 (Insig1) is one of the branch-dependent genes involved in controlling this process, although its role in renal fibrosis is unknown. Here, we discovered that tubular Insig1 deficiency, rather than fibroblast Insig1 deficiency, plays a detrimental role

NAD salvage pathway machinery expression in normal and glaucomatous retina and optic nerve.

Glaucoma is the leading cause of irreversible blindness and is a major health and economic burden. Current treatments do not address the neurodegenerative component of glaucoma. In animal models of glaucoma, the capacity to maintain retinal nicotinamide adenine dinucleotide (NAD) pools declines early during disease pathogenesis. Treatment with nicotinamide, an NAD precursor through the NAD salvage pathway, robustly protects against neurodegeneration in a number of glaucoma models and improves vision in existing glaucoma patients. However, it remains unknown in humans what retinal cell types ar
