Production of nerve growth factor by brown fat in culture: relation with the in vivo developmental stage of the tissue.

The regulation of nerve growth factor (NGF) production is still poorly understood. We attempt here to determine whether brown adipose tissue (BAT), which is densely innervated by sympathetic nerve fibres and can be induced to grow in adult rats by simple cold exposure, has the ability to produce NGF and thus to stimulate the growth of its innervation and, if so, whether this NGF activity is developmentally regulated. BAT at various stages of development was cocultured with NGF-sensitive sympathetic ganglia in the presence or absence of antiserum to NGF. Both undifferentiated BAT from newborn h

Regeneration of calcitonin gene-related peptide immunoreactive nerves in replanted rat molars and their supporting tissues.

First maxillary right molars in 66 rats were elevated and replanted and the pulps allowed to regenerate for 1-90 days. The contralateral tooth served as control. Regeneration of nerves in the pulp and periodontium was studied by CGRP-immunohistochemistry and the avidin-biotin-peroxidase method. The pulp and periodontium of the controls were richly supplied with CGRP-labelled nerves. One day after replantation the pulp was completely devoid of CGRP-immunoreactive nerves. After 2 days, axon sprouts were present in the apical, regenerated pulp and in the periodontium. From 3-7 days CGRP-immunorea

Frozen agarose-assisted tissue preparation for stimulated Raman histology enables rapid intraoperative assessment of peripheral nerve integrity.

BACKGROUND: Intraoperative assessment of peripheral nerve viability during traumatic nerve repair is predominantly based on the surgeon's subjective macroscopic criteria. Frozen section histology can provide objective information, its routine, repeated use is however limited by processing time. Intraoperative Stimulated Raman Histology (SRH) enables rapid, label-free imaging of native tissue. Current intraoperative SRH workflows rely on squash preparations that do not adequately preserve peripheral nerve microarchitecture. OBJECTIVE: To establish a rapid tissue preparation protocol that prese

Preparation of Newborn Rat Brain Tissue for Ultrastructural Morphometric Analysis of Synaptic Vesicle Distribution at Nerve Terminals.

Our laboratory and many others have exploited the high resolving power of transmission electron microscopy to study the morphology and spatial organization of synaptic vesicles. In order to obtain high-quality electron micrographs that can yield the degree of morphological detail necessary for quantitative analysis of pre-synaptic vesicle distribution, optimal specimen preparation is critical. Chemical fixation is the first step in the process of specimen preparation, and of utmost importance to preserve fine ultrastructure. Vascular fixation with a glutaraldehyde-formaldehyde solution, follow

Experimental Glaucoma Causes Optic Nerve Head Neural Rim Tissue Compression: A Potentially Important Mechanism of Axon Injury.

PURPOSE: We tested the hypothesis that experimental glaucoma (EG) results in greater thinning of the optic nerve head (ONH) neural rim tissue than the peripapillary retinal nerve fiber layer (RNFL) tissue. METHODS: Longitudinal spectral-domain optical coherence tomography (SDOCT) imaging of the ONH and peripapillary RNFL was performed every other week under manometric IOP control (10 mm Hg) in 51 nonhuman primates (NHP) during baseline and after induction of unilateral EG. The ONH parameter minimum rim area (MRA) was derived from 80 radial B-scans centered on the ONH; RNFL cross-sectional are

The Connective Tissue Components of Optic Nerve Head Cupping in Monkey Experimental Glaucoma Part 1: Global Change.

PURPOSE: To characterize optic nerve head (ONH) connective tissue change within 21 monkey experimental glaucoma (EG) eyes, so as to identify its principal components. METHODS: Animals were imaged three to five times at baseline then every 2 weeks following chronic unilateral IOP elevation, and euthanized early through end-stage confocal scanning laser tomographic change. Optic nerve heads were serial-sectioned, three-dimensionally (3D) reconstructed, delineated, and quantified. Overall EG versus control eye differences were assessed by general estimating equations (GEE). Significant, animal-s

Apelin-13 microinjection into the paraventricular nucleus increased sympathetic nerve activity innervating brown adipose tissue in rats.

The aim of present study is to clarify the role of apelin in regulating energy homeostasis in brown adipose tissue (BAT). We examined the central effects of apelin-13 on the brain c-fos like immunoreactivity (c-FLI), BAT temperature and the activity of the sympathetic nerve activity innervating BAT in rats. In the hypothalamus, central infusion into the third cerebral ventricle (i3vt) of apelin-13 caused induction of c-FLI in the paraventricular nucleus (PVN) compared with the controls (PBS-treated) group. In addition, microinjection of apelin-13 into the PVN produced significant increases in

Unilateral stellate ganglion block produces bidirectional changes in tissue oxygen tension of the mental nerve in rabbits.

PURPOSE: The purpose of this study was to investigate changes in the tissue oxygen tension (PO(2)) of the mental nerve bilaterally before and after unilateral stellate ganglion block (SGB). MATERIALS AND METHODS: Nine male Japan white rabbits were used. Anesthesia was maintained by a continuous infusion of propofol under mechanical ventilation with room air. For the SGB, the tip of a 26-gauge needle was placed on the left transverse process of the cervical vertebra; 0.2 mL of 1% lidocaine solution was injected. Data were recorded immediately before SGB and when the maximal change in PO(2) aft

Distribution and significance of nerve growth factor receptor (NGFR/p75NTR) in normal, benign and malignant breast tissue.

Nerve growth factor receptor (NGFR) is a transmembrane glycoprotein without intrinsic tyrosine kinase activity, whose expression is not restricted to neural cells. NGFR is reported to act as a tumour suppressor, negatively regulating cell growth and proliferation. NGFR expression was immunohistochemically analysed in normal breast tissue and in 140 benign, biphasic and preinvasive breast lesions, in 22 tumours with myoepithelial differentiation and in two cohorts of breast cancer patients: a series of 245 invasive breast carcinomas studied with tissue microarrays and 37 high-grade invasive duc

Retinoic acid increases tissue and plasma contents of nerve growth factor and prevents neuropathy in diabetic mice.

BACKGROUND: Decreased production of nerve growth factor (NGF) may contribute to diabetic neuropathy; however, exogenous administration of NGF induces only a modest benefit. Retinoic acid (RA) promotes the endogenous expression of nerve growth factor and its receptor. We studied the effects of RA on diabetic neuropathy in mice with streptozotocin-induced diabetes. MATERIAL AND METHODS: One hundred and twenty National Institutes of Health (NIH) albino mice randomly separated into three groups (A, n = 30; B, n = 30; C, n = 60). Diabetes mellitus was induced with streptozotocin in groups A and B.

A comparative study of embedded nerve tissue in six NF2-associated schwannomas and 17 nonassociated NF2 schwannomas.

BACKGROUND: Neurofibromatosis-2 (NF2) is an autosomal dominant disorder in which patients typically show bilateral acoustic tumors, and they are usually diagnosed histopathologically as schwannomas. The nerve of origin of a schwannoma is often demonstrated on the periphery along the capsule but not penetrating the substance of the tumor. However, there is a possibility that NF2 schwannomas and solitary schwannomas differ from participation in nerve components. METHODS: In this study, the authors noted the relationship between the tumor and the original nerves. To detect whether there were emb

Positive effect of ulnar nerve fascicle transfer to musculocutaneous nerve seeded with allogeneic adipose tissue derived stem cells on nerve regeneration for repairing upper brachial plexus injury in a rat model: A preliminary study.

BACKGROUND: Traumatic peripheral nerve injury, with an annual incidence reported to be approximately 13-23 per 100,000 people, is a serious clinical condition that can often lead to significant functional impairment and permanent disability. Although nerve transfer has become increasingly popular in the treatment of brachial plexus injuries, satisfactory results cannot be obtained even with total nerve root transfer, especially after serious injuries. To overcome this problem, we hypothesize that the application of stem cells in conjunction with nerve transfer procedures may be a viable altern

Bridging the gap: towards a digital twin to optimize therapeutic cell-seeding strategies in nerve tissue engineering.

Peripheral nerve injuries impose a substantial strain on the lives of millions of people worldwide. Engineered neural tissues provide a promising avenue to improve peripheral nerve repair strategies, potentially allowing the precise seeding of cells and materials that support regenerative processes. Current optimization of cell-seeding strategies relies on testing the impact of relevant parameters in vivo, requiring considerable time and resources. We propose an alternative approach to the design of cellular hydrogels based on a mathematical cell-solute model, informed via in vitro experiment,
