Neuroimmune interactions in painful TMD: Mechanisms and treatment implications.

The underlying mechanisms and treatment of painful temporomandibular disorders (TMDs) are important but understudied topics in craniofacial research. As a group of musculoskeletal diseases, the onset of painful TMD is proved to be a result of disturbance of multiple systems. Recently, emerging evidence has revealed the involvement of neuroimmune interactions in painful TMD. Inflammatory factors play an important role in peripheral sensitization of temporomandibular joint (TMJ), and neurogenic inflammation in turn enhances TMJs dysfunction in TMD. Furthermore, centralized neuroimmune communicat

Gut microbiome in neuroendocrine and neuroimmune interactions: The case of genistein.

The healthy and diverse microbes living in our gut provide numerous benefits to our health. It is increasingly recognized that the gut microbiome affects the host's neurobehavioral state through production of metabolites, modulation of intestinal immunity (e.g., cytokines) and other mechanisms (e.g., gut neuropeptides). By sending the sensed information (e.g., metabolic and immunologic mediators) about the state of the inner organs to the brain via afferent fibers, the vagus nerve maintains one of the connections between the brain and GI tract, and oversees many critical bodily functions (e.g.

Neuroimmunology: towards more-accurate diagnosis in neuromyelitis optica.

Neuromyelitis optica spectrum disorders (NMOSD) can mimic multiple sclerosis (MS). Avoiding misdiagnosis is crucial, because some disease-modifying drugs for MS can aggravate NMOSD, causing blindness and paraplegia. A recent study reports that misdiagnosis of NMOSD as MS occasionally occurs, and that a two-step antibody assay could improve differential diagnosis.

Neural regulation of inflammation in the airways and lungs.

Many pulmonary diseases are characterized by inflammatory pathologies which in turn are responsible for obstruction, mucus hypersecretion, dyspnea, cough and other clinical symptoms of lung disease. Understanding processes that regulate inflammation will therefore provide insights into mechanisms that contribute to pulmonary dysfunction. The airways and lungs are densely innervated by autonomic and sensory nerves which might regulate aspects of pulmonary inflammation. In this review we provide a critical appraisal of the available literature on the topic of neuro-immune interactions in the air

Neuroimmune mechanisms in fetal alcohol spectrum disorder.

Fetal alcohol spectrum disorder (FASD) is a major health concern worldwide and results from maternal consumption of alcohol during pregnancy. It produces tremendous individual, social, and economic losses. This review will first summarize the structural, functional, and behavior changes seen in FASD. The development of the neuroimmune system will be then be described with particular emphasis on the role of microglial cells in the normal regulation of homeostatic function in the central nervous system (CNS) including synaptic transmission. The impact of alcohol on the neuroimmune system in the

Treatment of HPV with hypnosis--psychodynamic considerations of psychoneuroimmunology: a brief communication.

There is increasing evidence that the hypnotic cure of warts (infection by the human papilloma virus or HPV) results from activation of an immune response, but whether this is cellular or systemic is unknown. The hypnosis can be by direct suggestion or analytical hypnotherapy when indicated. The evidence is reviewed, and 4 clinical cases suggesting cellular immune response are presented.

Is autism an autoimmune disease?

Autism spectrum disorder (ASD) is a spectrum of behavioral anomalies characterized by impaired social interaction and communication, often accompanied by repetitive and stereotyped behavior. The condition manifests within the first 3 years of life and persists into adulthood. There are numerous hypotheses regarding the etiology and pathology of ASD, including a suggested role for immune dysfunction. However, to date, the evidence for involvement of the immune system in autism has been inconclusive. While immune system abnormalities have been reported in children with autistic disorder, there i

["Sickness behavior"--mechanisms of origin and significance].

OBJECTIVES: Methotrexate (MTX) is a cornerstone in the treatment of rheumatoid arthritis (RA). Among its anti-proliferative activity, the anti-inflammatory mechanisms of MTX seem to play a major role in the treatment of RA. MTX reduces the production of pro-inflammatory cytokines such as interleukin (IL)-1, IL-2, IL-6 and interferon (INF)-γ, while the gene expression of anti-inflammatory Th2 cytokines like IL-4 and IL-10 is increased - altogether resulting in the anti-inflammatory effect. As little is known about the impact of MTX on other cytokines involved in the pathogenesis of RA, the pres

Involvement of cytokines in eating disorders: a critical review of the human literature.

A number of findings from clinical and animal studies indicate that pro-inflammatory cytokines may play roles in eating disorders. The measurement of pro-inflammatory cytokines (IL-1, IL-6, TNFalpha), which are known to decrease food intake, provides highly variable data from which firm conclusions cannot be drawn. In most of the longitudinal studies where pro-inflammatory cytokines have been shown to be impaired in anorexia or bulimia nervosa, a return to normal values was observed after renutrition. However these findings do not exclude the possibility that pro-inflammatory cytokines might b

The opioid-cytokine connection.

Opioids (exogenous opiates and endogenous opioid peptides) have a diversity of effects on the immune system. Although numerous studies have shown that opioid-induced immunosuppression can be mediated indirectly via the central nervous system (CNS) or through direct interactions with immunocytes, the precise cellular mechanisms underlying the immunomodulatory effects of opioids are largely unknown. In recent years, investigations from several laboratories have indicated that opioids can operate as cytokines, the principal communication signals of the immune system. All of the major properties o

Wharton's jelly-derived mesenchymal stem cells attenuate sepsis-induced organ injury partially via cholinergic anti-inflammatory pathway activation.

Sepsis induces organ dysfunction due to overexpression of the inflammatory host response, resulting in cardiopulmonary and autonomic dysfunction, thus increasing the associated morbidity and mortality. Wharton's jelly-derived mesenchymal stem cells (WJ-MSCs) express genes and secrete factors with anti-inflammatory properties, neurological and immunological protection, as well as improve survival in experimental sepsis. The cholinergic anti-inflammatory pathway (CAP) is mediated by α7-nicotinic acetylcholine receptors (α7nAChRs), which play an important role in the control of systemic inflammat

The neuroimmune axis in cardiovascular disease: from immune activation to inflammatory resolution.

Cardiovascular diseases (CVDs) are driven not only by hemodynamic and metabolic stress but also by persistent sterile inflammation and defective resolution. Neuroimmune regulation has emerged as an important focus in cardiovascular research. In this review, we synthesize current evidence on the neuroimmune axis in CVDs and highlight how dysregulated autonomic immune communication promotes persistent inflammation, defective resolution, and adverse cardiovascular remodeling. We further emphasize that the neuroimmune axis may influence not only inflammatory activation but also the transition towa

TDP-43 Acetylation at the Neuroimmune Interface: A Hypothesis-Driven Framework for Peripheral Inflammatory Stratotypes in ALS.

Transactive Response Deoxyribonucleic Acid-Binding Protein-43 (TDP-43) acetylation may couple motor-neuron degeneration to systemic immune orchestration in Amyotrophic Lateral Sclerosis (ALS). Upon nuclear clearance and mislocalisation, TDP-43 enters the periphery; acetylation shapes its conformation, trafficking and immunogenicity. This narrative review synthesises single-cell transcriptomics, proteomic immunoprofiling and clinical inflammatory phenotyping to examine whether site-specific acetylated TDP-43 species may be associated with peripheral inflammatory signatures relevant to ALS immun

Enteric neuroimmune interactions in health and disease.

The intestine contains a dense intrinsic nervous system that is closely integrated with local immune cells to coordinate motility, barrier defense, and tissue repair. Recent studies, primarily in mice and supported by emerging human evidence, have identified diverse enteric neuroimmune pathways through which enteric neurons shape immune cell responses, while immune cells, in turn, influence neuronal survival and function across distinct intestinal compartments. These bidirectional interactions are increasingly implicated in intestinal disorders, including Hirschsprung's disease, irritable bowe

Renal sensory nerves in injury: more than just sensing.

Recent basic research suggests that the neuroimmune axis plays an important role in the pathogenesis of acute kidney injury. Although it is established that the kidney is innervated by both afferent and efferent nerves, there are few studies on the role of renal sensory nerves. Zhang et al. postulated that renal sensory nerves activate a high IL4Rα expression macrophage subset, resulting in the amelioration of kidney injury. This finding highlights a potential neuroimmune pathway that may represent a novel therapeutic target for acute kidney injury.

The neuroimmune axis in breast cancer: from mechanistic insights to clinical applications.

BACKGROUND: Breast cancer is the most commonly diagnosed malignancy in women worldwide, and its marked heterogeneity together with the complexity of the tumor microenvironment contributes to substantial variability in therapeutic responses. Although conventional studies have mainly focused on tumor cell-intrinsic alterations, increasing evidence has identified the neuroimmune axis as a critical regulator of breast cancer progression and treatment resistance. MAIN BODY: The neuroimmune axis involves bidirectional crosstalk between the nervous system and the immune system, mediated by neurotran

Neurosurgery as an immune anchor point: a translational framework for perioperative immunoengineering.

Neurosurgical diseases-including brain tumors, hemorrhage/trauma, ischemia, infection, epilepsy, and spinal cord injury-share convergent neuro-immune mechanisms. In the acute phase, sterile inflammation and barrier disruption trigger innate immune cascades. During the subacute phase, immune resolution and clearance determine the quality of tissue repair. In the chronic phase, persistent immune-glia interactions and synaptic remodeling influence epileptogenesis and long-term cognitive outcomes. Recent discoveries-such as the meningeal immune niche, meningeal lymphatic system, and glymphatic cle

Microglial checkpoint collapse in Alzheimer's disease: a tri-axial framework for biomarker-informed neuroimmune therapy.

BACKGROUND: Anti-amyloid antibodies have validated amyloid-β (Aβ) as a disease-relevant target in Alzheimer's disease (AD), but their modest clinical effect, efficacy largely restricted to early disease, and amyloid-related imaging abnormalities (ARIA) indicate that Aβ removal alone does not resolve the glial, lipid, and inflammatory programmes that sustain neurodegeneration. Microglia sit at the centre of this therapeutic gap. Single-nucleus and spatial profiling has resolved several AD-associated microglial states, yet state labels remain descriptive and do not explain why adaptive engagemen

Rethinking interleukin-31: A neuroimmune orchestrator of itch beyond Th2 immunity.

Interleukin (IL)-31 has emerged as a pivotal mediator of pruritus. Since its initial identification, substantial progress has been made in elucidating the role of IL-31 in itch pathophysiology, particularly its direct effects on peripheral sensory neurons. Elevated IL-31 expression has been documented in a wide range of pruritic skin diseases, including atopic dermatitis, prurigo nodularis, and systemic disorders. Beyond its pruritogenic activity, IL-31 is increasingly recognized as a neuroimmune cytokine that links immune-cell activation to sensory-neuronal circuits and, in some contexts, may
