Creutzfeldt-Jakob disease.

This review will explore the clinical and pathological findings of the various forms of Creutzfeldt-Jakob disease (CJD). Clinical findings of CJD are characterized by rapidly progressive cognitive dysfunction, diffusion-weighted magnetic resonance imaging (DWI) hyperintensity, myoclonus, periodic sharp-wave complexes on electroencephalogram and akinetic mutism state. Neuropathologic findings of CJD are characterized by spongiform changes in gray matter, gliosis-particularly hypertrophic astrocytosis-neuropil rarefaction, neuron loss and prion protein (PrP) deposition. The earliest pathological

Plasticity of the nigrostriatal system in MPTP-treated mice. A biochemical and morphological correlation.

In order to compare the recovery capacity of the nigrostriatal system between adult and old mice, MPTP hydrochloride was administered to 48 BL/C57 male mice, which were sacrificed 24 h or 10 d after the second dose. The animals were divided into four groups, based on age (adult or old) and moment of sacrifice (24 h or 10 d). The detailed morphology of the neurons and the cellular processes of the substantia nigra pars compacta and the striatum were studied using the Golgi method. Immunostaining with a polyclonal glial fibrillary acidic protein antiserum using the peroxidase-antiperoxidase tech

Cholecystokinin-like peptide is a modulator of a crustacean central pattern generator.

The presence, release, and physiological effects of a cholecystokinin(CCK)-like peptide within the stomatogastric ganglion (STG) of the lobster, Panulirus interruptus, are described. Indirect immunofluorescence with 2 antisera raised against CCK8 was used to determine the distribution of CCK-like immunoreactivity (CCKLI) in the stomatogastric nervous system. CCKLI was demonstrated in the input nerve and the neuropil of the STG and in neuropil and somata in the commissural ganglia (CGs), brain, and eyestalks. None of the somata within the STG displayed CCKLI. The cross-reactivities of the CCK a

Ultrastructural and immunohistochemical study of degenerate neurite-bearing ghost tangles.

Some ghost tangles in the brains of Alzheimer patients were accompanied by many small argyrophilic structures which were electron microscopically confirmed to be degenerate neurites. In these ghost tangles, roughly dispersed 15 nm straight and occasional twisted tubules were penetrated by proliferated astrocytic processes. Immunohistochemically, these ghost tangles lost immunoreactivities to anti-NFT, -tau and -ubiquitin antibodies, but were thioflavine-S fluorescent, though antigenicity to beta-protein was not proved. This similarity in composition of degenerate neurite-bearing ghost tangles

Diffuse deposition of immunohistochemically labeled prion protein in the granular layer of the cerebellum in a patient with Creutzfeldt-Jakob disease.

Amyloid plaques in Creutzfeldt-Jakob disease, kuru, and Gerstmann-Sträussler-Scheinker syndrome are known to contain an abnormal isoform of a cellular protein, the prion protein (PrP). The prion protein in its normal cellular isoform is a membrane-bound glycoprotein of unknown function. The mechanisms causing a modification of PrP and accumulation in amyloid plaques are unknown. Here we present a case of Creutzfeldt-Jakob disease with widespread deposition of immunohistochemically labeled PrP in the internal granular layer of the cerebellum. Immunohistochemically labeled PrP was deposited in d

Creutzfeldt-Jacob disease: a golgi study.

A cerebral biopsy of a patient with Creutzfeldt-Jacob (C-J) disease was examined with the Golgi method. Distortion of soma and neuronal processes associated with vacuolization of the neuropil was observed. The main findings were decreased numbers of basal dendrites and of branches of the apical dendrite of the pyramidal cells, marked loss of synaptic spines, and varicosities in the proximal segment of some apical and basal dendrites. These changes, though non-specific, may be interpreted as the result of deafferentation, although primary reactions related to C-J disease cannot be ruled out. Th

The distribution of acetylcholinesterase in the central nervous system of jumping spiders and wolf spiders (Arachnida, Araneida: Salticidae et Lycosidae).

The distribution and activity pattern of acetylcholinesterase in the central nervous system of salticid and lycosid spiders has been studied. Enzyme activity was limited to the neuropile mass. The salticid and lycosid species investigated showed different intensities of enzyme reactions in the protocerebrum. The differences observed may be related to the somewhat contrasting habits of these spider families. Reactions were strong especially in the optic ganglia (lamina glomerularis, corpora pedunculata) and in the cerebral ganglion, while the central body showed only weak to moderate activity.

Capillary connections between sensory circumventricular organs and adjacent parenchyma enable local volume transmission.

Among contributors to diffusible signaling are portal systems which join two capillary beds through connecting veins. Portal systems allow diffusible signals to be transported in high concentrations directly from one capillary bed to the other without dilution in the systemic circulation. Two portal systems have been identified in the brain. The first was discovered almost a century ago and connects the median eminence to the anterior pituitary gland. The second was discovered a few years ago and links the suprachiasmatic nucleus to the organum vasculosum of the lamina terminalis, a sensory ci

The distribution and compartmental organization of the cholinergic neurons in nucleus accumbens of the rat.

In this study the distribution of the cholinergic neurons was examined in relation to the compartmental organization of nucleus accumbens. This was accomplished by charting the location of the choline acetyltransferase-immunoreactive neurons and mapping their distribution in relation to cytoarchitectural features and the patterns of acetylcholinesterase activity and enkephalin immunoreactivity. Choline acetyltransferase-containing perikarya are inhomogeneously distributed in nucleus accumbens. Their density is lowest at the rostral pole and highest, caudomedially, at the septal pole. The cells

The neuropathological landscape of Hispanic and non-Hispanic White decedents with Alzheimer disease.

Despite the increasing demographic diversity of the United States' aging population, there remain significant gaps in post-mortem research investigating the ethnoracial heterogeneity in the neuropathological landscape of Alzheimer Disease (AD). Most autopsy-based studies have focused on cohorts of non-Hispanic White decedents (NHWD), with few studies including Hispanic decedents (HD). We aimed to characterize the neuropathologic landscape of AD in NHWD (n = 185) and HD (n = 92) evaluated in research programs across three institutions: University of California San Diego, University of Californi

Pathological changes induced by Alzheimer's brain inoculation in amyloid-beta plaque-bearing mice.

Alzheimer's disease (AD) is characterized by intracerebral accumulations of extracellular amyloid-β (Aβ) plaques and intracellular tau pathology that spread in the brain. Three types of tau lesions occur in the form of neuropil threads, neurofibrillary tangles, and neuritic plaques i.e. tau aggregates within neurites surrounding Aβ deposits. The cascade of events linking these lesions and synaptic or memory impairments are still debated. Intracerebral infusion of human AD brain extracts in Aβ plaque-bearing mice that do not overexpress pathological tau proteins induces tau pathologies followin

Evidence that 3,3',5-triiodothyronine is concentrated in and delivered from the locus coeruleus to its noradrenergic targets via anterograde axonal transport.

Recent immunohistochemical studies of rat brain triiodothyronine reveal heaviest localization in locus coeruleus perikarya. The cellular distribution is similar to that observed in concomitant studies of tyrosine hydroxylase immunohistochemistry: heavy clumps of immunoreactive triiodothyronine are distributed within locus coeruleus cytosol and in cell processes, leaving cell nuclei unstained. At the same time, in locus coeruleus targets, cell nuclei as well as surrounding neuropil are prominently triiodothyronine labeled. These observations, combined with diverse evidence linking thyroid hormo

Distribution and functional significance of Met-enkephalin-Arg6-Phe7- and Met-enkephalin-Arg6-Gly7-Leu8-like peptides in the blowfly Calliphora vomitoria. I. Immunocytochemical mapping of neuronal pathways in the brain.

Neuronal pathways immunoreactive to antisera against the extended-enkephalins, Met-enkephalin-Arg6-Phe7 (Met-7) and Met-enkephalin-Arg6-Gly7-Leu8 (Met-8), have been identified in the brain of the blowfly Calliphora vomitoria. Co-localisation with other enkephalins in certain neurons suggests that a precursor similar to preproenkephalin A exists in insects and that differential enzymatic processing occurs as in vertebrates. Co-localisations of the extended-enkephalin-like peptides with other vertebrate-type peptides, including cholecystokinin and pancreatic polypeptide, also occur. The enkephal

Regressive Neuroblastoma: Widely Accepted, Underreported Histology.

Neuroblastoma accounts for approximately 8% of all pediatric cancers and up to 15% of pediatric cancer-related deaths in malignant cases; neuroblastoma is the most common malignancy in infancy. Notably, up to 50% of tumors may undergo spontaneous regression without treatment. However, detailed histological descriptions of regressed neuroblastoma remain limited in the literature. We report the histological features of a particularly illustrative case, characterized by edema, mild collagen deposition, evidence of prior hemorrhage, and calcifications. Cellularity was minimal, consisting of rare,

Microscopic Study of Nervous System Plasticity: Interactions of Sympathetic Nerves with Neurons of Intraocular Hippocampal Transplants.

Functional interactions of sympathetic fibers innervating the iris with the neurons of central origin in intraocular transplants of the rat hippocampus were studied by optic, confocal, and electron microscopy. After formaldehyde fixation, fluorescent dye Dil was applied to the upper cervical ganglion; the dye migrated to the transplant by lateral diffusion via axons. Sympathetic nerves labeled with fluorescent dye grew into the neurotransplants along perivascular membranes of blood vessels. In addition, some fluorescent axons were identified in the transplant parenchyma. Electron microscopy sh

Extraventricular neurocytoma with atypical features and ganglionic differentiation.

Extraventricular neurocytoma (EVN) is an unusual variant of central neurocytoma located outside the ventricular system. Both tumours share a similar histology characterised by monotonous populations of round-to-oval cells with scanty cytoplasm separated by neuropil and branching capillaries. We report an EVN arising from the right frontal lobe near the olfactory tract in a 34-year-old male with worsening chronic epilepsy. Our patient's tumour exhibited many uncommon features including ganglionic differentiation, increased mitotic activity and a high proliferative index. We discuss the importan

Structure of the Brain of the Smallest Coleoptera.

The structure of the brain of the smallest coleopteran, Scydosella musawasensis Hall, 1999, is described for the first time. As in other extremely small beetles, the brain of S. musawasensis displays signs of miniaturization: displacement to the thorax, compactization, and a small number and size of the neurons. The body size of the studied smallest beetle is similar to that of the minute hymenopteran Megaphragma, which has a nearly anucleate nervous system. However, the structure of the brain of the studied smallest beetle is similar to that of large representatives of the order and is charac

Bleeding in the subarachnoid space: a possible complication during laser therapy for equine progressive ethmoid haematoma.

A 10-year-old KWPN (Royal Warmblood Studbook of the Netherlands) gelding was euthanized after developing severe neurological symptoms preceded by severe epistaxis during laser treatment for progressive ethmoid haematoma (PEH) in the right nasal cavity. Postmortem examination of the head revealed a large amount of clotted blood between the right ventral and dorsal conchae in the nasal cavity and acute haemorrhage in the right subarachnoid space. Histologically, there was moderate, acute polioencephalomalacia in the neuropil adjacent to the haemorrhage. The haemorrhages were most likely caused b

Central projections of the parapineal organ of the adult rainbow trout (Oncorhynchus mykiss).

Neural connections of the small parapineal organ of the adult rainbow trout were experimentally investigated by using a lipophilic carbocyanine dye as a tracer. The dye was applied to the parapineal organ, to the pineal organ, or to the left or right habenular ganglion. The parapineal organ mainly projected via a coarse parapineal tract to a conspicuous neuropil in the rostrodorsal part of the left habenular ganglion. A small accessory parapineal tract projecting to the right habenular ganglion was also found in some animals. No pineal afferents were observed in the parapineal, nor was any neu

Tau protein and the neurofibrillary pathology of Alzheimer's disease.

Abundant neurofibrillary tangles, neuropil threads and senile plaque neurites constitute the neurofibrillary pathology of Alzheimer's disease. They form in the nerve cells that undergo degeneration in the disease, in which their regional distribution correlates with the degree of dementia. Each lesion contains the paired helical filament (PHF) as its major component. PHFs are composed of the microtubule-associated protein tau in a hyperphosphorylated state. PHF-tau is hyperphosphorylated on all six adult brain isoforms. As a consequence, tau is unable to bind to microtubules and is believed to
