Cypermethrin alters the status of oxidative stress in the peripheral blood: relevance to Parkinsonism.

Parkinson's disease (PD) is a motor scarcity disorder characterized by the striatal dopamine deficiency owing to the selective degeneration of the nigrostriatal dopaminergic neurons. While oxidative stress is implicated in PD, prolonged exposure to moderate dose of cypermethrin induces Parkinsonism. The study aimed to investigate the status of oxidative stress indicators and antioxidant defence system of the polymorphonuclear leukocytes (PMNs), platelets and plasma to delineate the effect of Parkinsonian dose of cypermethrin in the peripheral blood of rats and its subsequent relevance to Parki

Nitric oxide evolution and perception.

Various experimental data indicate signalling roles for nitric oxide (NO) in processes such as xylogenesis, programmed cell death, pathogen defence, flowering, stomatal closure, and gravitropism. However, it still remains unclear how NO is synthesized. Nitric oxide synthase-like activity has been measured in various plant extracts, NO can be generated from nitrite via nitrate reductase and other mechanisms of NO generation are also likely to exist. NO removal mechanisms, for example, by reaction with haemoglobins, have also been identified. NO is a gas emitted by plants, with the rate of evolu

Carbon monoxide oxygenase activity of cytochrome cd1.

Cytochrome cd1 from the denitrifying bacterium Pseudomonas aeruginosa catalyzes the oxygenation of carbon monoxide by dioxygen. A minimum estimate of the turnover number for this activity is 7 mol of carbon dioxide produced per hour per mole of cytochrome subunit at 30 degrees C and pH 7. The reaction is 98% inhibited by 2.5 mM cyanide, but catalase has no effect. The reaction accounts for the unusual reduction of ferric cytochrome in the presence of carbon monoxide, but no additional reducing agent. The reaction is independent of the steady-state oxidation level of the cytochrome during turno

Magnesium-induced transporters remodel the ionome of mulberry leaves by promoting the nitrate reductase (NIA) gene in nitrogen metabolism.

Magnesium (Mg) is fundamental for diverse physiological, biochemical, and molecular processes in higher plants because its deficiency or toxicity severely restricts development. The supply of Mg remodels the plant ionome and transporter profiles while simultaneously activating the nitrate reductase enzyme, which catalyzes nitrate reduction to nitrite and enhances nitrogen (N) assimilation. However, the specific mechanisms involving ion interactions and transporters in Mg metabolism that improve N metabolism in mulberry remain largely unexplored. To address this research gap, we employed physio

Molybdenum Availability Is Key to Nitrate Removal in Contaminated Groundwater Environments.

The concentrations of molybdenum (Mo) and 25 other metals were measured in groundwater samples from 80 wells on the Oak Ridge Reservation (ORR) (Oak Ridge, TN), many of which are contaminated with nitrate, as well as uranium and various other metals. The concentrations of nitrate and uranium were in the ranges of 0.1 μM to 230 mM and <0.2 nM to 580 μM, respectively. Almost all metals examined had significantly greater median concentrations in a subset of wells that were highly contaminated with uranium (≥126 nM). They included cadmium, manganese, and cobalt, which were 1,300- to 2,700-fold hig

Gasotransmitters (NO and HCN) alter phytoglobin-NO cycle during dormancy alleviation of apple (Malus domestica Borkh.) embryos.

Nitric oxide (NO) and hydrogen cyanide (HCN) are N-containing gasotransmitters of bimodal action depending on the concentration. Both gases exhibit a beneficial effect on seed dormancy removal. The aim of this work was to compare the mode of action of NO and HCN on the elements of phytoglobin (Pgb)-NO cycle during seed dormancy alleviation (under aerobic conditions). Short-term fumigation of apple (Malus domestica Borkh.) embryos with HCN or NO increased the formation of reactive nitrogen species (RNS) in embryonic axes. The induction of nitrite/nitrate (NO2-/NO3-) signalling pathway, together

Congenital methemoglobinemia: Rare presentation of cyanosis in newborns.

Methemoglobin (MetHb) is an oxidized form on hemoglobin, which is unable to bind oxygen and consequently carry it to the tissues. Normally present in small quantities (<1%) without detrimental effects, its elevation produces hypoxemia which can be profound and even lethal. Methemoglobinemia is an abnormal increase of MetHb (>3%) of total hemoglobin. It can be classified in two types: hereditary and acquired. Acquired form is caused by exogenous oxidizing agents, such as nitrites or certain medications, while hereditary types of disease are the result of genetic deficiency in cytochrome B5 redu

The potential effects of pomegranate (Punica granatum) juice on carbon tetrachloride-induced nephrotoxicity in rats.

The purpose of this study was to evaluate the effect of pomegranate (Punica granatum) in inhibiting and reversing the nephrotoxicity of carbon tetrachloride, a potent oxidative stress inducer which induces cellular kidney damage. Rats were intraperitoneally injected with carbon tetrachloride (2 mL/kg body weight) which produced severe renal tissue damage, as demonstrated by decreased uric acid and dramatic elevation of urea and creatinine. In addition, carbon tetrachloride injection caused oxidative stress in rats, as evidenced by increased lipid peroxidation and nitrite/nitrate (NO x ) concen

Methemoglobinemia caused by the ingestion of courgette soup given in order to resolve constipation in two formula-fed infants.

Methemoglobinemia is not a rare condition arising from the exposure to hemoglobin-oxidizing agents such as nitrates-nitrites present in well water or vegetables. Infants < 3 months of age are more susceptible than adults because of lower amounts of a key enzyme, NADH-cytochrome b5 reductase, which converts methemoglobin back to hemoglobin. We report 2 infants, aged respectively 2 and 1 months, suffering from methemoglobinemia, fed with a formula that was reconstituted with a high concentration of courgette soup to resolve constipation. They developed a severe cyanosis with methemoglobinemia (r

Nitrous oxide reduction by two partial denitrifying bacteria requires denitrification intermediates that cannot be respired.

Denitrification is a form of anaerobic respiration wherein nitrate (NO3-) is sequentially reduced via nitrite (NO2-), nitric oxide, and nitrous oxide (N2O) to dinitrogen gas (N2) by four reductase enzymes. Partial denitrifying bacteria possess only one or some of these four reductases and use them as independent respiratory modules. However, it is unclear if partial denitrifiers sense and respond to denitrification intermediates outside of their reductase repertoire. Here, we tested the denitrifying capabilities of two purple nonsulfur bacteria, Rhodopseudomonas palustris CGA0092 and Rhodobact

Silicon Nanoparticles Alter Soybean Physiology and Improve Nitrogen Fixation Potential Under Atmospheric Carbon Dioxide (CO2).

The interactive effects between nano-silicon dioxide (n-SiO2) and elevated CO2 (eCO2; 645 ppm) on soybean physiology, nitrogen fixation, and nutrient dynamics under climate stress remain underexplored. This study elucidates their combined effects under ambient (aCO2; 410 ppm) and eCO2 conditions. eCO2 + n-SiO2 synergistically enhanced shoot length (30%), total chlorophyll (112.15%), and photosynthetic rate (103.23%), alongside improved stomatal conductance and intercellular CO2 (17.19%), optimizing carbon assimilation. Nodulation efficiency increased, with nodule number and biomass rising by 4

Integration of Non-Invasive Micro-Test Technology and 15N Tracing Reveals the Impact of Nitrogen Forms at Different Concentrations on Respiratory and Primary Metabolism in Glycyrrhiza uralensis.

Glycyrrhiza uralensis is a highly valued medicinal species worldwide. However, a paradox arises in its cultivation in that high nitrogen fertilization boosts yield at the expense of root quality, a problem linked to nitrogen's regulation of tricarboxylic acid (TCA) cycle-driven respiration. It remains unclear how different nitrogen forms coordinate respiratory and primary metabolism. We examined the regulatory mechanisms of nitrate (NO3-) versus ammonium (NH4+) on these processes in cultivated G. uralensis by supplying seedlings with varying concentrations of K15NO3 or (15NH4)2SO4 in a modifie

Effects of nitric oxide modulators on cardiovascular risk factors in mild hyperhomocysteinaemic rat model.

Hyperhomocysteinaemia is considered to be an independent risk factor in atherosclerosis. In the present article, we observed the effect of nitric oxide modulators on cardiovascular risk factors in mild hyperhomocysteinaemic rats. A rat model of mild hyperhomocysteinaemia was established by administering methionine (1 g/kg body weight, orally) for 4 weeks. The other groups were concomitantly treated with sodium nitroprusside (SNP) and N(omega)-nitro-l-arginine (LNNA) during the induction of hyperhomocysteinaemia. Lipid profile, total antioxidant capacity and the level of homocysteine and NO(x)

1H NMR spectroscopy of cytochrome cd1 derivatives.

Proton nuclear magnetic resonance spectra are reported for cytochrome cd1 from Pseudomonas aeruginosa (ATCC 19429) in several forms including complexes of the ferricytochrome with cyanide, azide, and fluoride, a quasi-apo form in which the noncovalently associated heme d1 has been removed but the covalently bound heme c is retained, and the reduced state of both native and the quasi-apo forms. Comparisons are made to the previously reported spectrum of ferricytochrome cd1. The following points are made. The spectra of the azide and fluoride complexes and the ferric quasi-apo form show perturba

Haldane, hot dogs, halitosis, and hypoxic vasodilation: the emerging biology of the nitrite anion.

While it has long been known that the reduction of nitrite to nitric oxide (NO) forms iron-nitrosyl-myoglobin and is the basis of meat curing, a greater biological activity of the nitrite anion has only recently been appreciated. In the stomach, NO is formed from acidic reduction of nitrite and increases mucous barrier thickness and gastric blood flow (see the related study beginning on page 106). Nitrite levels in blood reflect NO production from endothelial NO synthase enzymes, and recent data suggest that nitrite contributes to blood flow regulation by reaction with deoxygenated hemoglobin
