Studies on retrograde and anterograde amnesia of olfactory memory after denervation of the hippocampus by entorhinal cortex lesions.

The effect of hippocampal denervation on olfactory memory in rats was tested after interrupting the lateral olfactory tract projections at the level of the entorhinal cortex. When lesioned animals were trained to learn new odors, they showed no evidence of retention 3 h after acquisition. These results confirm earlier data on rapid forgetting in rats after hippocampal deafferentation and are in parallel to the anterograde amnesia typically found in humans with hippocampal damage. On the other hand, preoperatively learned information was minimally impaired after hippocampal deafferentation even

Numerical and Machine Learning Analysis of the Parameters Affecting the Regionally Delivered Nasal Dose of Nano- and Micro-Sized Aerosolized Drugs.

The nasal epithelium is an important target for drug delivery to the nose and secondary organs such as the brain via the olfactory bulb. For both topical and brain delivery, the targeting of specific nasal regions such as the olfactory epithelium (brain) is essential, yet challenging. In this study, a numerical model was developed to predict the regional dose as mass per surface area (for an inhaled mass of 2.5 mg), which is the biologically most relevant dose metric for drug delivery in the respiratory system. The role of aerosol diameter (particle diameter: 1 nm to 30 µm) and inhalation flow

Extensive immunolesions of basal forebrain cholinergic system impair offspring recognition in sheep.

The involvement of the basal forebrain cholinergic system has been extensively investigated in instrumental learning but little is known of its participation in social memory, especially in the memorization of individual traits of a conspecific. The present study tested in sheep its contribution to both instrumental learning and individual offspring recognition. Six weeks before parturition, ewes received injections of a specific cholinergic immunotoxin (ME20.4 IgG-saporin) into the lateral ventricles (150 microg) and in some cases additional immunotoxin injections into the nucleus basalis (11

Olfaction in rats with extensive lesions of the olfactory bulbs: implications for odor coding.

Rats were initially trained on a series of odor detection tasks and then received a unilateral olfactory bulbectomy and removal of different parts of the contralateral bulb. After postoperative recovery they were tested for detection of different concentrations of four odors, on a series of odor discrimination tasks and for their ability to acquire a relatively easy and a more difficult odor mixture discrimination task. Groups were formed based on which region of the bulb was intact (regional savings score) and on amount of bulb intact (bulbar savings score). In general, only rats with bulbar

Smell and Taste Disorders in Primary Care.

Disorders of smell and taste are reported by approximately one-fifth of people 40 years and older, and one-third of people 80 years and older. These disorders affect quality of life and the ability to identify smoke and toxins. Smell and taste disorders can be early signs of dementia or Parkinson disease and are associated with increased mortality. Dysfunction may be apparent or may develop insidiously. Screening questionnaires are available, but many patients are unaware of their disorder. Most smell and taste disorders are due to sinonasal disease but also could be caused by smoking, medicat

Esthesioneuroblastoma: the complete picture - case report and review of the literature.

Esthesioneuroblastoma (ENB), also called olfactory neuroblastoma, is a cancerous tumor originating from the olfactory neuroepithelial cells frequently invading the brain through the cribriform plate. The optimal therapy is the multimodality treatment involving a group of physicians trained in different medical specialties. Establishing a careful histopathological diagnostic and treatment planning based on a multidisciplinary approach is of paramount importance. The treatment of ENB correlates with the extent of the lesion, with surgery being the mainstay of therapy followed by postoperative ir

Management of Anosmia in COVID-19: A Comprehensive Review.

With the evolving understanding of COVID-19, a thorough analysis of the effects of this unique coronavirus on the affected people's olfactory abilities could highlight the disease's specific course of treatment. Researchers have discovered that the neurological side effects of SARS-CoV-2 infection include acute anosmia and ageusia. This work aims to review the relevant mechanisms, provide information on COVID-19-related anosmia, and suggest a novel approach to treating long-term anosmia brought on by coronavirus disease. For that, we did a thorough literature assessment of the subject from var

A Randomized Controlled Trial on the Management of Post-COVID-19 Olfactory Dysfunction in a Pediatric Population.

Olfactory impairment is an important sequela of SARS-CoV-2 infection with potential long-term implications. Of treatments posited for post-COVID-19 olfactory dysfunction (OD), the strongest evidence supports olfactory training (OT) and topical steroids; however, a direct comparison of these treatments has yet to be published. Our objective was to directly compare the efficacy of OT versus budesonide irrigations for the treatment of post-COVID-19 OD in a pediatric population.  Methods: Pediatric patients aged six to 21 years presenting to the University of Pittsburgh, a tertiary children's hosp

Multicomponent water proton transverse relaxation and T2-discriminated water diffusion in myelinated and nonmyelinated nerve.

The influence of compartmental boundaries on water proton transverse relaxation and diffusion measurements was investigated in three distinct excised nerves, namely, the non-myelinated olfactory nerve, the Schwann cell myelinated trigeminal nerve, and the oligodendrocyte myelinated optic nerve of the garfish. The transverse relaxation decay curves were multiexponential and their decomposition yielded three primary components with T2 values approximately 30-50, 150, and 500 ms, which were subsequently assigned to water protons in the myelin, axoplasm, and interaxonal compartments. The short T2

Navigating the efficacy, safety and outcome of intranasal platelet-rich plasma for persistent olfactory loss: a systematic review and meta-analysis.

BACKGROUND: Persistent olfactory dysfunction (OD) poses a significant clinical challenge with limited therapeutic options. Intranasal platelet-rich plasma (PRP), an autologous source of regenerative growth factors, presents a biologically plausible intervention, yet its efficacy across various OD etiologies remains synthetically underexplored. METHODS: This systematic review and meta-analysis adhered to PRISMA guidelines. A comprehensive search of PubMed/MEDLINE, Web of Science, and Cochrane Library (2010-2025) identified studies assessing intranasal PRP for persistent OD (> 6 months). Random

Nostril-specific and structure-based olfactory learning of chiral discrimination in human adults.

Practice makes perfect. In human olfaction, such plasticity is generally assumed to occur at the level of cortical synthetic processing that shares information from both nostrils. Here we present findings that challenge this view. In two experiments, we trained human adults unirhinally for the discrimination between odor enantiomers over a course of about 10 to 11 days. Results showed that training-induced perceptual gain was restricted to the trained nostril yet partially generalized to untrained odor enantiomers in a structure- rather than quality- based manner. In other words, learning enha

Potentiation and depression of synaptic transmission in the olfactory cortex of the guinea-pig.

1. The extracellular field potentials of the olfactory cortex evoked by stimulation of the lateral olfactory tract (l.o.t.) were studied in in vitro preparations from the olfactory cortex. The field potentials comprised an initial diphasic wave - the l.o.t. compound action potential - followed by a negative wave of about 10 msec duration which in turn was followed by a low amplitude positive wave of long duration (100 msec or more). In this paper, the size of the negative field potential (extracellularly recorded EPSP) has been studied during and after periods of repetitive stimulation of the

The Effect of Oral Theophylline on Traumatic Anosmia: A Prospective, Randomized Clinical Trial.

OBJECTIVE: Theophylline has been theorized to promote neural olfactory signaling and sensory axonal regeneration. In this study, our objective was to investigate the effect of oral theophylline on the treatment of traumatic anosmia. METHODS: We enrolled patients who had complained of traumatic complete anosmia. Eligible patients were randomly assigned to 2 groups. In the theophylline group, oral theophylline (400 mg bid) was prescribed for 6 months and patients also underwent olfactory training (OT). In the OT group, patients performed OT only for 6 months. The bilateral and unilateral olfact

Bioelectronic nose for ultratrace odor detection via brain-computer interface with olfactory bulb electrode arrays.

Rapid and accurate detection of hazardous volatile compounds is crucial for public health and environmental safety. While conventional methods, including electronic noses, typically exhibit detection thresholds in the parts-per-million (ppm) range, many harmful substances pose risks at parts-per-billion (ppb) concentrations or lower. To address this challenge, we leverage the exceptional sensitivity of the mammalian olfactory system, specifically that of Rattus norvegicus (lab rat), which has evolved to detect and discriminate a vast array of odors at extremely low concentrations. In this stud

Part Two: Perspectives on Olfactory and Gustatory Dysfunction Pathophysiology, Management, and Relevance to COVID-19: Current Approaches.

CONTEXT: The phenomena of olfactory and gustatory dysfunction disorders (OGD) are hardly unique to COVID-19. However, the widespread incidence of OGD as sequelae of COVID-19 has provoked rejuvenated interest in these long existing, but poorly studied maladies. OBJECTIVE: This second of a three-part review discusses past and current approaches for treatment of OGD, not restricted to those that COVID-19 has caused, with the intention to lay a foundation for consideration of new paradigms for evaluation and management of OGD. DESIGN: The researcher performed a narrative review by searching data

Anesthesia Resistant Memories in Drosophila, a Working Perspective.

Memories are lasting representations over time of associations between stimuli or events. In general, the relatively slow consolidation of memories requires protein synthesis with a known exception being the so-called Anesthesia Resistant Memory (ARM) in Drosophila. This protein synthesis-independent memory type survives amnestic shocks after a short, sensitive window post training, and can also emerge after repeated cycles of training in a negatively reinforced olfactory conditioning task, without rest between cycles (massed conditioning-MC). We discussed operational and molecular mechanisms

Early olfactory pre-conditioning during sensitive developmental periods is associated with enhanced detection performance in working dogs.

Early olfactory imprinting has been proposed as a strategy to enhance the performance of detection dogs, yet empirical evidence from operational contexts remains limited. This study evaluated the effects of early exposure to target odours on four core detection variables: sensitivity under tight-containment conditions, reaction distance, work endurance, and detection reliability. Twenty operational adult dogs served as controls, and four dogs underwent an early-learning protocol beginning during sensitive stages of neurosensory development. All dogs completed parallel test batteries under equi

An olfactory discrimination procedure for mice.

This paper describes an olfactory discrimination procedure for mice that is inexpensively implemented and leads to rapid discrimination learning. Mice were first trained to dig in small containers of sand to retrieve bits of buried chocolate. For discrimination training, two containers were presented simultaneously for eight trials per session. One container held sand mixed with cinnamon, and the other held sand mixed with nutmeg. Both containers were baited with chocolate buried in the sand. One odor was designated S+, and mice were allowed to dig and retrieve the chocolate from this containe

Interventions for the prevention of persistent post-COVID-19 olfactory dysfunction.

BACKGROUND: Loss of olfactory function is well recognised as a symptom of COVID-19 infection, and the pandemic has resulted in a large number of individuals with abnormalities in their sense of smell. For many, the condition is temporary and resolves within two to four weeks. However, in a significant minority the symptoms persist. At present, it is not known whether early intervention with any form of treatment (such as medication or olfactory training) can promote recovery and prevent persisting olfactory disturbance. This is an update of the 2021 review with four studies added. OBJECTIVES:
