Scrub typhus: vector competence of Leptotrombidium chiangraiensis chiggers and transmission efficacy and isolation of Orientia tsutsugamushi.

A total of 4,281 rodents, belonging to 20 species, was collected from three villages in Chiangrai Province, northern Thailand, from January 1994 to December 2001. The predominant species were Rattus rattus, R. losea and Bandicota indica, accounting for 96% of the total. More than 135,000 chigger mites were removed from wild rodents of which 1% were identified as Leptotrombidium chiangraiensis, a new species vector of scrub typhus. Five Orientia tsutsugamushi-infected L. chiangraiensis colonies established from chiggers removed from R. rattus and R. losea rodents. The prevalence of O. tsutsugam

Occurrence of Orientia tsutsugamushi in chiggers (Acari: Trombiculidae) and small animals in an orchard near Bangkok, Thailand.

The ecology of Orientia tsutsugamushi (Hayashi) was studied in chiggers and small animals in an orchard near Bangkok, Thailand. Small animals were trapped monthly between July and November 1992 and examined for the presence of O. tsutsugamushi and ectoparasitic chiggers. A total of 40 Rattus rattus (L.) and 16 Tupaia glis (Diard) was trapped. O. tsutsugamushi was isolated from liver and spleen samples of 30.8% of R. rattus and 18.6% of T. glis. Antibodies to O. tsutsugamushi were detected in 95% of R rattus, and IgG antibodies persisted for up to 10 mo after removing rats from potential reinfe

Nasopharyngeal Swabs for Orientia tsutsugamushi Detection in Doxycycline Treatment: A Prospective Cohort Study in Hainan, China.

Scrub typhus, caused by Orientia tsutsugamushi, remains a neglected cause of acute febrile illness. Molecular testing of blood supports early diagnosis, yet once doxycycline is started, blood qPCR positivity can drop rapidly, complicating short-term follow-up and relapse surveillance. We compared detection across multiple clinical specimens and evaluated nasopharyngeal swabs (NPSs) as noninvasive supplementary specimens during treatment initiation. In a prospective single-center cohort from Hainan, China, we enrolled 20 patients with scrub typhus. Blood, NPS, urine, and stool were collected be

Isolation of Orientia tsutsugamushi from Human Blood and Factors Effect In Vitro Isolation in South Korea.

INTRODUCTION: Orientia tsutsugamushi, the causative agent of scrub typhus, is endemic to South Korea. While diagnostics such as PCR and immunochromatographic tests (ICT) are widely used, pathogen isolation remains essential for antimicrobial resistance and genetic studies. METHODS: In this study, 444 blood samples were collected from febrile patients across 13 hospitals. Among 263 samples identified as scrub typhus-positive by at least one diagnostic method, 99 isolates were successfully cultured. Fisher's exact test and the Wilcoxon rank-sum test were used to estimate odds ratios. RESULTS:

Assessment of a Sensitive qPCR Assay Targeting a Multiple-Copy Gene to Detect Orientia tsutsugamushi DNA.

Scrub typhus is caused by an obligated intracellular organism, Orientia tsutsugamushi (Orientia). The disease was traditionally thought to be limited in the tsutsugamushi triangle. Recently, scrub typhus has been confirmed in areas outside the triangle. Serological diagnosis of scrub typhus relies on indirect immunofluorescence assay (IFA). Molecular assays such as PCR, qPCR, loop-mediated isothermal amplification, and recombinase polymerase amplification are often targeting a single copy gene. These assays are sensitive and specific, yet they are not broadly used in clinical settings possibly

Dissemination of Orientia tsutsugamushi, a Causative Agent of Scrub Typhus, and Immunological Responses in the Humanized DRAGA Mouse.

Scrub typhus is caused by Orientia tsutsugamushi, an obligated intracellular bacterium that affects over one million people per year. Several mouse models have been used to study its pathogenesis, disease immunology, and for testing vaccine candidates. However, due to the intrinsic differences between the immune systems in mouse and human, these mouse models could not faithfully mimic the pathology and immunological responses developed by human patients, limiting their value in both basic and translational studies. In this study, we have tested for the first time, a new humanized mouse model t

Inappropriateness of quinolone in scrub typhus treatment due to gyrA mutation in Orientia tsutsugamushi Boryong strain.

The use of quinolone for treatment of rickettsial diseases remains controversial. Recent clinical studies suggest that quinolone is not as effective as others in patients with rickettsial diseases including scrub typhus, although the mechanism is not well understood. In this study, we evaluated the mutation in gyrA associated with quinolone resistance. We prospectively enrolled scrub typhus patients, collected blood samples and clinical data from October, 2010 to November, 2011. Among the 21 patients enrolled, one initially received ciprofloxacin for 3 days but was switched to doxycycline due

Influence of Orientia tsutsugamushi infection on the developmental biology of Leptotrombidium imphalum and Leptotrombidium chiangraiensis (Acari: Trombiculidae).

Leptotrombidium chiangraiensis Tanskul & Linthicum, and Leptotrombidium imphalum Vercammen-Grandjean are important vectors of scrub typhus in rice field habitats in northern Thailand. The developmental biology of all stages of the life cycle of two generations of these species of mites infected with Orientia tsutsugamushi (Hayashi) and uninfected mites is reported. The development of the infected lines of both F1 and F2 L. chiangraiensis were significantly longer than their respective uninfected lines (P < 0.05). The developmental times of uninfected and infected F1 lines of L. imphalum were n

Genome-based construction of the metabolic pathways of Orientia tsutsugamushi and comparative analysis within the Rickettsiales order.

Orientia tsutsugamushi, the causative agent of scrub typhus, is an obligate intracellular bacterium that belongs to the order of Rickettsiales. Recently, we have reported that O. tsutsugamushi has a unique genomic structure, consisting of highly repetitive sequences, and suggested that it may provide valuable insight into the evolution of intracellular bacteria. Here, we have used genomic information to construct the major metabolic pathways of O. tsutsugamushi and performed a comparative analysis of the metabolic genes and pathways of O. tsutsugamushi with other members of the Rickettsiales o

Evaluation of an immunoglobulin M capture enzyme-linked immunosorbent assay for diagnosis of Orientia tsutsugamushi infection.

To differentiate scrub typhus from other acute febrile diseases, a rapid and reliable serological diagnosis is important. We developed an immunoglobulin M (IgM) capture enzyme-linked immunosorbent assay (ELISA) for diagnosis of recent Orientia tsutsugamushi infections in humans. The 56-kDa major outer membrane protein of O. tsutsugamushi is well known as the most immunodominant antigen in scrub typhus. The test is based on the use of the biotinylated recombinant 56-kDa protein of O. tsutsugamushi Boryong, Bor56, which was expressed as a fusion protein with a maltose-binding protein in Escheric
