Strain-dependent differences in vertebral bone mass, serum osteocalcin, and calcitonin in calcium-replete and -deficient mice.

The effects of dietary calcium intake on vertebral bone mass, composition, and turnover (calcium deposition and resorption) were determined in 10- and 14-week-old C57BL/6 (small) and SENCAR (large) mice. Total vertebral mass, percent ash, calcium, magnesium, and phosphorus were higher in SENCAR mice than in C57BL/6 mice at 10 weeks of age and after being fed 0.02% or 0.6% dietary calcium for 4 additional weeks. Relative calcium deposition was higher in C57BL/6 than in SENCAR mice, while relative calcium resorption was similar in both strains. The rate of resorption was higher in mice fed 0.02%

Stage-dependent analysis of IL-6, TGF-β, osteocalcin, and sRANKL in the synovial fluid of dogs with osteoarthritis.

Osteoarthritis (OA) is traditionally considered a degenerative joint disease. However, morphological similarities between OA progression and fracture healing suggest that OA may be a dysregulated bone repair process. This study investigated the morphological and biochemical parallels between OA and bone healing in dogs following cranial cruciate ligament rupture. To differentiate between biochemical profiles at different stages of the disease, a structured OA scoring system was developed based on radiographic and additional intraoperative findings. Synovial fluid samples from the OA cohort wer

Serum N-terminal osteocalcin is a good indicator for estimating responders to hormone replacement therapy in postmenopausal women.

To estimate the response to hormone replacement therapy (HRT) by bone metabolic markers, 36 patients with postmenopausal osteoporosis or osteopenia were studied to assess the correlation between percent baseline changes in lumbar bone mineral density (BMD) after 12 months and those in various bone metabolic markers after 3, 6, and 12 months of HRT. All the patients were treated with 0.625 mg of conjugated estrogen and 2.5 mg of medroxyprogesterone per day and continued for 12 months. BMD was significantly increased up to 4.19 +/- 0.87% after 6 months and 4.93 +/- 1.27% after 12 months of HRT (

Different pattern of alkaline phosphatase, osteopontin, and osteocalcin expression in developing rat bone visualized by in situ hybridization.

Alkaline phosphatase (AP), osteopontin (OP), and osteocalcin (OC) are expressed during osteoblastic differentiation. However, previous studies suggested differences in the timing and possibly the site of their expression. In this study we used in situ hybridization to follow the distribution of these osteoblastic markers during bone development. Frozen sections of neonatal rat long bones and calvariae were hybridized with 35S-labeled RNA probes complementary to the AP, OP, and OC mRNAs. Controls included sections hybridized with the sense (mRNA) probes or pretreated with RNase. Positive cells

Salivary osteocalcin: Reactivity to acute social stress and associations with concomitant stress biomarkers in healthy African Americans.

Animal studies suggest that osteocalcin (OCN) may play an important role in the early steps of parasympathetic nervous system withdrawal to initiate the autonomic nervous system (ANS) component of the fight-or-flight response to stress. However, the OCN response to stress has not yet been well-characterized in humans. This study examined reactivity of OCN, measured in saliva, in response to acute social stress in healthy African Americans, and considered associations of OCN reactivity with well-established biomarkers associated with stress-reactivity of the autonomic, endocrine, and immune sys

Hypoxia signaling pathway activation in osteoblasts suppresses osteocalcin production but does not alter male fertility in mice.

INTRODUCTION/OBJECTIVE: Bone can contribute to systemic and extra-skeletal processes through bone-derived hormones. The osteoblast-specific protein osteocalcin (OCN) has been reported to positively regulate male fertility, by directly favoring testicular testosterone biosynthesis. We previously demonstrated that aberrant hypoxia-inducible factor (HIF) signaling in osteoprogenitors drastically changes bone properties associated with suppressed OCN expression. This led us to question whether hypoxia signaling in bone could be an upstream determinant of male fertility, by indirectly impacting tes

Osteocalcin modulates hippocampal activities and influences glycophagy within astrocytes in a chronic unpredictable mild stress mouse model.

Osteocalcin (OCN), an active substance produced by bone tissue, has demonstrated potential in alleviating depressive symptoms and exerting extensive regulatory effects on glial cells in animals by participating in the communication between bone and brain, while its precise mechanisms within the central nervous system (CNS) still not fully understood. In this study, we validated the antidepressant effects of OCN in depressive mice induced by chronic unpredictable mild stress (CUMS), and conducted a comprehensive assessment of the functional impacts of OCN in hippocampus using proteomic analysis

Type 2 Diabetes Is Causally Associated With Reduced Serum Osteocalcin: A Genomewide Association and Mendelian Randomization Study.

Recent advances indicate that bone and energy metabolism are closely related. However, little direct evidence on causality has been provided in humans. We aimed to assess the association of three bone-related biomarkers-25 hydroxyvitamin D (25OHD), parathyroid hormone (PTH), and osteocalcin (OCN)-with several metabolic phenotypes and investigate any causal relevance to the associations using a Mendelian randomization (MR) study. Serum 25OHD, PTH, and total OCN were measured at baseline in 5169 eligible Chinese participants in Changfeng study. Partial correlation and bivariate GREML analysis we

Effects of Oral Dosage of Lead Acetate II on Osteocalcin Gene Expression in Rat Mesenchymal Stem Cells.

BACKGROUND: Lead (Pb) is a heavy metal that has devastating effects on many animal tissues. In this study we investigated the effects of orally-dosed lead acetate II on osteocalcin gene (osteocalcin) expression in mesenchymal stem cells grown in an osteogenic medium. Osteocalcin is an abundant bone matrix differentiation protein. METHODS: Twelve male Wistar rats were divided into three groups of four rats each. Two groups were fed orally with 50 or 100 ppm of lead acetate II with libitum feed and water for two months. The control group was fed with libitum feed and water only. Rats were eutha

Synergistic effect of extracellularly supplemented osteopontin and osteocalcin on stem cell proliferation, osteogenic differentiation, and angiogenic properties.

A high demand for functional bone grafts is being observed worldwide, especially due to the increased life expectancy. Osteoinductive components should be incorporated into functional bone grafts, accelerating cell recruitment, cell proliferation, angiogenesis, and new bone formation at a defect site. Noncollagenous bone matrix proteins, especially osteopontin (OPN) and osteocalcin (OC), have been reported to regulate some physiological process, such as cell migration and bone mineralization. However, the effects of OPN and OC on cell proliferation, osteogenic differentiation, mineralization,

In vivo determination of vitamin d function using transgenic mice carrying a human osteocalcin luciferase reporter gene.

Vitamin D is an essential factor for ossification, and its deficiency causes rickets. Osteocalcin, which is a noncollagenous protein found in bone matrix and involved in mineralization and calcium ion homeostasis, is one of the major bone morphogenetic markers and is used in the evaluation of osteoblast maturation and osteogenic activation. We established transgenic mouse line expressing luciferase under the control of a 10-kb osteocalcin enhancer/promoter sequence. Using these transgenic mice, we evaluated the active forms of vitamins D2 and D3 for their bone morphogenetic function by in vivo
