Conditional hyperthermia in response to atropine associated with a hypothermic drug.

In a set of experiments designed to examine learned associations between drug states, atropine sulfate (10 mg/kg) elicited a conditional hyperthermia in rats following ten treatment sessions in which the drug was paired with either chlorpromazine hydrochloride (10 mg/kg) or ethanol (2.3 g/kg), both hypothermia-inducing agents. Atropine methyl nitrate, a quaternary analogue with similar peripheral effects but little central action, was ineffective as a cue in this situation. These experiments demonstrated that a sequential pairing of drug states can yield a change in an organism's response to t

Detecting the onset of accelerated long-term forgetting: evidence from temporal lobe epilepsy.

Accelerated long-term forgetting (ALF) refers to a slowly developing anterograde amnesia in which material is retained normally over short delays but then forgotten at an abnormally fast rate over days to weeks. Such long-term memory impairment is not detected by standard clinical tests. This study analysed ALF in a temporal lobe epileptic, RY. Key issues addressed were: (i) the timeframe of ALF onset; (ii) whether disruption of memory consolidation during sleep is a necessary requirement for precipitating ALF; (iii) the effectiveness of repeated recall in limiting the impact of ALF. RY's memo

Prosopagnosia and object agnosia without covert recognition.

Investigations of the visual recognition abilities of the patient M.S. are reported. M.S. is unable to achieve overt recognition of any familiar faces, and many everyday objects. In Task 1 he showed semantic priming from name primes but not from face primes in a name recognition task. In Task 2 he showed no advantage in learning true (face + correct name) rather than untrue (face + someone else's name) pairings of faces and names. In Task 3 semantic priming of lexical decision was only found for object picture primes that M.S. was able to recognize overtly. In Task 4 faster matching of photogr

Associative learning in flying phobia.

BACKGROUND AND OBJECTIVES: Modern learning theories suggest that particularly strong associative learning contributes to the etiology and maintenance of anxiety disorders, thus explaining why some individuals develop an anxiety disorder after a frightening (conditioning) event, whereas others do not. However, associative learning has rarely been investigated experimentally in specific phobias. The current study investigated associative learning in patients with flying phobia and healthy controls using a modified version of Olson and Fazio's associative learning paradigm (Olson & Fazio, 2001).

Time-course of attentional bias for positive social words in individuals with high and low social anxiety.

BACKGROUND: Although accumulating research demonstrates the association between attentional bias and social anxiety, the bias for positive stimuli has so far not been adequately studied. AIMS: The aim is to investigate the time-course of attentional bias for positive social words in participants with high and low social anxiety. METHOD: In a modified dot-probe task, word-pairs of neutral and positive social words were randomly presented for 100, 500, and 1250 milliseconds in a nonclinical sample of students to test their attentional bias. RESULTS: Non-significant interaction of Group × Expo

A longitudinal study of category-specific agnosia.

We report a 12-year longitudinal case study on a 60-year-old male patient (DW) with category-specific agnosia. The extent to which DW's impairment has changed over time was evaluated using identical tests at time 1 (1988) and time 2 (2000). In particular, we assessed his ability to identify pictures and real objects, to draw from memory, and to access stored semantic information about living and non-living things. The principal findings were: (i). DW was significantly better at identifying real objects in comparison with line drawings. (ii). DW presented with a category-specific impairment for

Recent advances in the phencyclidine model of schizophrenia.

OBJECTIVE: Phencyclidine (PCP, "angel dust") induces a psychotomimetic state that closely resembles schizophrenia. As opposed to amphetamine-induced psychosis, PCP-induced psychosis incorporates both positive (e.g., hallucinations, paranoia) and negative (e.g., emotional withdrawal, motor retardation) schizophrenic symptoms. PCP-induced psychosis also uniquely incorporates the formal thought disorder and neuropsychological deficits associated with schizophrenia. The purpose of the present paper is to review recent advances in the study of the molecular mechanisms of PCP action and to describe

Relational memory structure among prior neutral episodes guides Pavlovian threat responses.

Learning about danger through exposure to harmful events relies on Pavlovian conditioning, which enables individuals to prepare for potential threats when encountering warning signals in the future. Sensory preconditioning further shows that defensive significance can extend to neutral cues associated with a subsequently conditioned stimulus, even when those cues have never themselves been paired with an aversive outcome. Here, we asked whether this principle extends to a relational memory structure assembled across separate neutral experiences, and whether cues closer to the conditioned threa

Using facial expressions as CSs and fearsome and disgusting pictures as UCSs: affective responding and evaluative learning of fear and disgust in blood-injection-injury phobia.

Two experiments examine use of an evaluative conditioning (EC) paradigm in the acquisition of fear and disgust responding to neutral facial expressions. In Experiment 1, 60 participants were randomly assigned to one of three evaluative learning conditions in which neutral facial expressions were paired with fearsome, disgusting, or neutral pictures. No statistically significant differences were detected between the three conditions. However, significant differences emerged within subjects as post-exposure of fear and disgust ratings were higher among expressions that were paired with pictorial

A Novel Drug Repositioning Approach Based on Collaborative Metric Learning.

Computational drug repositioning, which is an efficient approach to find potential indications for drugs, has been used to increase the efficiency of drug development. The drug repositioning problem essentially is a top-K recommendation task that recommends most likely diseases to drugs based on drug and disease related information. Therefore, many recommendation methods can be adopted to drug repositioning. Collaborative metric learning (CML) algorithm can produce distance metrics that capture the important relationships among objects, and has been widely used in recommendation domains. By ap

Kaliotoxin, a Kv1.1 and Kv1.3 channel blocker, improves associative learning in rats.

Olfactory associative learning was used to investigate the involvement of Kv channels containing Kv1.1 and Kv1.3 alpha-subunits in learning and memory. Kaliotoxin (KTX), a specific inhibitor of these Kv channels, was injected intracerebroventricularly in the rat brain, at a dose of 10 ng that did not disturb the rats' locomotor activity or drinking behaviour. In the first paradigm (odour-reward training), KTX improved learning but not information consolidation. Moreover, KTX increased the long-term retrieval of an odour-reward association tested by a reversal test 1 month after the odour-rewar

Equation learning to identify nano-engineered particle-cell interactions: an interpretable machine learning approach.

Designing nano-engineered particles capable of the delivery of therapeutic and diagnostic agents to a specific target remains a significant challenge. Understanding how interactions between particles and cells are impacted by the physicochemical properties of the particle will help inform rational design choices. Mathematical and computational techniques allow for details regarding particle-cell interactions to be isolated from the interwoven set of biological, chemical, and physical phenomena involved in the particle delivery process. Here we present a machine learning framework capable of el

In utero alcohol exposure: functional and structural brain damage.

Pregnant rats were intubated with alcohol throughout gestation. Control animals received vehicle and were pair-fed. At birth, all offspring were surrogate-fostered to nontreated dams. At 11 weeks of age, alcohol-exposed animals did not perform as well in a two-way shock avoidance task. The brains of these animals also exhibited significant deficits in dendritic structure in the area of the hippocampus. These behavioral observations may be a counterpart to the learning deficits associated with fetal alcohol effects, e.g., fetal alcohol syndrome in humans and animals, and the neuroanatomical obs

Associations of clinical features in neurofibromatosis 1 (NF1).

Neurofibromatosis 1 (NF1), an autosomal dominant disease, exhibits extreme clinical variability. This variability greatly increases the burden for affected families and impairs our ability to understand the pathogenesis of NF1. Recognition of heterogeneity within a disease may provide important pathogenic insights, therefore we tested clinical data from three large sets of NF1 patients for evidence that certain common features are more likely to occur in some NF1 patients than in others. Clinical information on 4,402 patients with NF1 was obtained from three independent databases. We examined

Retrograde facilitation under midazolam: the role of general and specific interference.

In a double-blind, placebo-controlled experiment that used midazolam, a benzodiazepine that creates temporary amnesia, we compared acquisition and retention of paired associates of different types. Some word pairs were studied before the injection of saline or midazolam, and two lists of word pairs were studied after the injection. Critical comparisons involved retention of pairs that were practiced on all three lists, pairs studied on only one list, and pairs that involved recombining cue and response terms from one list to the next, as a function of drug condition. Previous research with ben

COVID-19-related distress is associated with analogue PTSD symptoms after exposure to an analogue stressor.

Background: The COVID-19 outbreak in early 2020 was associated with an immediate increase in mental health problems in a significant percentage of the general population. Therefore, it is crucial to investigate how the COVID-19 pandemic - as a psychosocial stressor - affected the aetiological processes of mental disorders. Previous research has shown that stress potentiates associative (fear) learning and analogue symptoms of posttraumatic stress disorder (PTSD) and that analogue PTSD symptoms can emerge in response to associative learning. Objective: We investigated whether distress in respon

High-definition tDCS of the temporo-parietal cortex enhances access to newly learned words.

Learning associations between words and their referents is crucial for language learning in the developing and adult brain and for language re-learning after neurological injury. Non-invasive transcranial direct current stimulation (tDCS) to the posterior temporo-parietal cortex has been suggested to enhance this process. However, previous studies employed standard tDCS set-ups that induce diffuse current flow in the brain, preventing the attribution of stimulation effects to the target region. This study employed high-definition tDCS (HD-tDCS) that allowed the current flow to be constrained t

Pre- and postnatal propylthiouracil-induced hypothyroidism impairs synaptic transmission and plasticity in area CA1 of the neonatal rat hippocampus.

Thyroid hormones are essential for neonatal brain development. It is well established that insufficiency of thyroid hormone during critical periods of development can impair cognitive functions. The mechanisms that underlie learning deficits in hypothyroid animals, however, are not well understood. As impairments in synaptic function are likely to contribute to cognitive deficits, the current study tested whether thyroid hormone insufficiency during development would alter quantitative characteristics of synaptic function in the hippocampus. Developing rats were exposed in utero and postnatall
