Expression of beta 1-integrins, H-CAM (CD44) and LECAM-1 in primary gastro-intestinal B-cell lymphomas as compared to the adhesion receptor profile of the gut-associated lymphoid system, tonsil and peripheral lymph node.

beta 1-Integrins (VLA-1 to -6) are cell-surface molecules binding to matrix molecules such as collagen, fibronectin and laminin. VLA-4 is the human homologue to the murine Peyer's patch homing receptor mediating cell/cell adhesion required for lymphocyte extravasation or "homing". Other structures which have a homing-receptor function through recognition of venular endothelium are H-CAM (CD44) and LECAM-1 (LAM-1, human MEL-14 equivalent). In order to elucidate whether these adhesion receptors are expressed in primary gastro-intestinal malignant B-cell lymphomas (GI BmL) which, in this case, mi

Does Basal Cell Carcinoma Arise from a Precursor Lesion?

Basal cell carcinoma (BCC) is driven in nearly all cases by mutations that constitutively activate upstream Hedgehog (HH) signaling, either through loss-of-function mutations in PTCH1 or gain-of-function mutations in SMO. Unlike other skin cancers such as squamous cell carcinoma, a clinically apparent precursor lesion for BCC has not been identified. Recent genomic analyses of both human and mouse tumors suggest that BCC formation requires not only PTCH1 or SMO mutations but also additional genetic changes. These findings imply that some BCCs may follow a stepwise tumor progression model, wher

The sigma-1 receptor behaves as an atypical auxiliary subunit to modulate the functional characteristics of Kv1.2 channels expressed in HEK293 cells.

Expression of Kv1.2 within Kv1.x potassium channel complexes is critical in maintaining appropriate neuronal excitability and determining the threshold for action potential firing. This is attributed to the interaction of Kv1.2 with a hitherto unidentified protein that confers bimodal channel activation gating, allowing neurons to adapt to repetitive trains of stimulation and protecting against hyperexcitability. One potential protein candidate is the sigma-1 receptor (Sig-1R), which regulates other members of the Kv1.x channel family; however, the biophysical nature of the interaction between

Heterogeneity of GABA(A) receptor-mediated responses in the human IMR-32 neuroblastoma cell line.

The gamma-aminobutyric acid (GABA) response profiles of IMR-32 human neuroblastoma cells were examined using whole-cell patch clamp and RT-PCR techniques. GABA activated a concentration-dependent and bicuculline-sensitive current, and RT-PCR revealed the expression of multiple GABA(A) receptor subunit mRNAs (alpha(1), alpha(3), alpha(4), beta(1), beta(3), gamma(2), and delta). A pharmacological profile of the GABA-induced current was derived using several subunit-selective agents. Diazepam, which requires the presence of a gamma subunit in order to modulate GABA(A) receptor-mediated responses,

A novel type of nicotinic receptor in the rat central nervous system characterized by patch-clamp techniques.

We present a functional characterization of a neuronal nicotinic receptor in the CNS using patch-clamp techniques and a preparation of acutely isolated neurons from the medial habenular nucleus of 10- to 20-d-old rats. The salient pharmacological and electrophysiological properties of this nicotinic response are (1) its association with a channel that is relatively nonselective for cations and has a unitary conductance of 26.2 (+)+/- 5pS at room temperature; (2) its insensitivity to alpha-bungarotoxin and to neuronal bungarotoxin; (3) its activation by the ganglionic nicotinic agonists nicotin

Nanomicelles-in-coaxial nanofibers with exit channels as a transdermal delivery platform for smoking cessation.

Smoking is a life-threatening habit; that is why many nicotine-replacement therapies (NRTs), which include chewing gums, nicotine patches, lozenges, mouth sprays, inhalers and nasal sprays that are usually administered for 8-12 weeks, have been reported for smoking cessation. We report the fabrication of patches comprising nanomicelles-in-coaxial nanofibers (NFs) for the transdermal delivery of varenicline (VAR) tartrate, a partial agonist of the α4β2 receptor subtype, for smoking cessation. The cores of the fabricated coaxial NF structures are composed of polyethylene oxide, VAR-loaded Pluron

Real-World Experience With TRBC1 Immunohistochemistry Across Cutaneous T-Cell Lymphoma Subtypes: A Large Cohort Study.

T-cell receptor β-chain constant region 1 (TRBC1) immunohistochemistry identifies clonal αβ T-cell populations on tissue sections, but its real-world performance across cutaneous T-cell lymphoma (CTCL) and related infiltrates is uncharacterized. The analytic cohort comprised 665 biopsies (566 patients) with paired T-cell receptor (TCR) clonality testing, classified clinicopathologically as mycosis fungoides (MF; MF-Patch, MF-Plaque, MF-Tumor, and MF-Folliculotropic); MF or Sézary syndrome; primary cutaneous small or medium T-cell lymphoproliferative disorders (LPDs); other CTCL-cutaneous LPDs;

Localized secretion of ATP and opioids revealed through single Ca2+ channel modulation in bovine chromaffin cells.

In bovine chromaffin cells, the Ca2+ channels involved in exocytosis are effectively inhibited by ATP and opioids that are coreleased with catecholamines during cell activity. This autocrine loop causes a delay in Ca2+ channel activation that is quickly removed by preceding depolarizations. Changes in Ca2+ channel gating by secreted products thus make it possible to correlate Ca2+ channel activity to secretory events. Here, using cell-attached patch recordings, we found a remarkable correlation between delayed Ca2+ channel openings and neurotransmitter secretion induced by either local or whol

Nociceptor-derived CGRP enhances dermal type I conventional dendritic cell function to drive autoreactive CD8+ T cell responses in vitiligo.

Vitiligo is an autoimmune disease characterized by depigmented patches of skin. Autoreactive CD8+ T cells kill melanocytes in vitiligo, but the immunopathogenesis remains elusive and ideal drug targets are lacking. Through single-cell and spatial transcriptomic analysis of vitiligo lesional skin, we found that conventional type 1 dendritic cells (cDC1s) primed CD8+ T cells and highly expressed the neuropeptide calcitonin gene-related peptide (CGRP) receptor. Deletion of Nav1.8+ nociceptors, cDC1-specific ablation of the CGRP receptor, or treatment with a CGRP receptor antagonist (rimegepant) a

Shox2 is required for the proper development of the facial motor nucleus and the establishment of the facial nerves.

BACKGROUND: Axons from the visceral motor neurons (vMNs) project from nuclei in the hindbrain to innervate autonomic ganglia and branchial arch-derived muscles. Although much is known about the events that govern specification of somatic motor neurons, the genetic pathways responsible for the development of vMNs are less well characterized. We know that vMNs, like all motor neurons, depend on sonic hedgehog signaling for their generation. Similarly, the paired-like homeobox 2b (Phox2b) gene, which is expressed in both proliferating progenitors and post-mitotic motor neurons, is essential for t

Multiple (familial) trichoepitheliomas: a clinicopathological and molecular biological study, including CYLD and PTCH gene analysis, of a series of 16 patients.

Multiple familial trichoepitheliomas (MFT) constitute an autosomally inherited syndrome possibly related to Brooke-Spiegler syndrome (BSS). Although some early studies suggested a role for the PTCH gene on chromosome 9q22.3 in the etiopathogenesis of MFT, recent studies of occasional patients with the MFT clinical phenotype identified mutations in the CYLD gene on chromosome 16q12-q13, a gene responsible for BSS. A systematic investigation of PTCH and CYLD mutations in patients with MFT has never been performed. Our main objective was to collect a reasonably large series of patients with MFT t

Noggin null allele mice exhibit a microform of holoprosencephaly.

Holoprosencephaly (HPE) is a heterogeneous craniofacial and neural developmental anomaly characterized in its most severe form by the failure of the forebrain to divide. In humans, HPE is associated with disruption of Sonic hedgehog and Nodal signaling pathways, but the role of other signaling pathways has not yet been determined. In this study, we analyzed mice which, due to the lack of the Bmp antagonist Noggin, exhibit elevated Bmp signaling. Noggin(-/-) mice exhibited a solitary median maxillary incisor that developed from a single dental placode, early midfacial narrowing as well as abnor

Two new patients with Curry-Jones syndrome with trichoblastoma and medulloblastoma suggest an etiologic role of the sonic hedgehog-patched-GLI pathway.

Curry-Jones syndrome (OMIM #601707) is a rare multiple malformation disorder of unknown etiology, associated with brain and skull abnormalities, polysyndactyly, and defects of the eyes, skin and gastrointestinal tract. We report on two new cases of Curry-Jones syndrome with previously unreported features, including benign and malignant neoplasms. The first patient had typical features of Curry-Jones syndrome as well as multiple intra-abdominal smooth muscle hamartomas and trichoblastoma of the skin. The second patient was born with occipital meningoceles and developed a desmoplastic medullobla

Early effects of PRL on ion conductances in CHO cells expressing PRL receptor.

Chinese hamster ovary (CHO-K1) cells were stably transfected with prolactin (PRL) receptor cDNA. These cells (CHO-E32) expressed the long form of functional PRL receptor. Using microfluorimetric and patch-clamp techniques, we have investigated the effects of PRL on intracellular Ca2+ concentration ([Ca2+]i) and membrane ion conductances. Exposure of CHO-E32 cells to 5 nM PRL resulted in an increase in [Ca2+]i. Two types of response were observed: 1) a stimulation of Ca2+ entry and 2) an intracellular Ca2+ mobilization. As PRL inhibited voltage-activated Ca2+ current, the PRL-induced Ca2+ incre

Spider venom of Araneus opens and desensitizes glutamate channels in chick spinal cord neurones.

Whole cell currents induced by the excretions of venom glands from the spider Araneus diadematus (ASV) were tested in neurones from the embryonic spinal cord of the chick in culture by the patch clamp technique. ASV in a dose of one bite in 1 ml induced a large membrane current of the same polarity as the excitatory amino acids, which decreased during long-lasting application. The effects were, to a large extent, rapidly reversible. It is suggested that ASV activates and desensitizes the receptor channels for excitatory amino acids.

EPS-8 regulates human malignant melanoma development by activating the Hedgehog pathway via degradation of Ptch1.

BACKGROUND: The epidermal growth factor receptor pathway substrate 8 (EPS-8) is a tumor-associated antigen that is frequently overexpressed in various types of human solid tumors and is associated with aggressiveness and poor prognosis. The role of EPS-8 in cutaneous malignant melanoma and its potential mechanism remains unknown. METHODS: EPS-8 expression, mutation immune infiltration, and the tumor microenvironment in melanoma were analyzed using various databases. Clinical samples were collected and melanoma cell viability, apoptosis and protein levels were detected using cell counting kit-

Sonic hedgehog pathway genes are expressed and transcribed in the adult mouse epididymis.

One role of the hedgehog (hh) signaling pathway during development is to assist in establishing pattern orientation in the embryo. The structure and function in the adult epididymis is highly patterned, and since the sonic hedgehog (Shh) pathway is known to be functional in the developing male tract and the expression of other pattern-influencing genes has recently been found in the adult epididymis, we have examined the adult mouse epididymis for Shh pathway molecules. Examination was at both the gene and protein level. Shh, the secreted signal molecule, patched (Ptc), its membrane receptor,

Defects in mouse mammary gland development caused by conditional haploinsufficiency of Patched-1.

In vertebrates, the hedgehog family of cell signaling proteins and associated downstream network components play an essential role in mediating tissue interactions during development and organogenesis. Loss-of-function or misexpression mutation of hedgehog network components can cause birth defects, skin cancer and other tumors. The mammary gland is a specialized skin derivative requiring epithelial-epithelial and epithelial-stromal tissue interactions similar to those required for development of other organs, where these interactions are often controlled by hedgehog signaling. We have investi

Dissecting the Impact of Genetic Background on Oncogenic Response to Radiation Exposure in the Ptch1+/- Mouse Model.

Medulloblastoma (MB) is a common primary brain cancer in children. The sonic hedgehog (SHH) pathway is indispensable for the normal development of the cerebellum, and MB is often caused by persistent SHH activation owing to mutations in pathway components. Patched1 (PTCH1) is the primary receptor for the SHH ligand and a negative regulator of the SHH signal transduction pathway. Mice heterozygous for the Ptch1 gene (Ptch1+/-) are predisposed to MB development. Irradiation of newborn Ptch1+/- mice dramatically increases MB occurrence. A genetic background carrying the Ptch1 mutation significant
