Intestinal permeability is increased in patients with advanced cirrhosis.

BACKGROUND/AIMS: The dysfunction of the intestinal barrier is a factor that has been related to bacterial translocation from lumen to extra-intestinal sites and consequently to the development of spontaneous bacterial peritonitis. The aim of this study was to investigate if the alterations of the intestinal barrier in cirrhosis are related with the degree of liver failure and associated with other clinical complications. METHODOLOGY: Intestinal permeability was assessed by means of the lactulose/mannitol test in 79 cirrhotic and 25 controls subjects. They received 10 g of lactulose and 5 g of

[Fluid resuscitation in conditions with disorders of capillary permeability].

Fluid resuscitation remains a cornerstone in the treatment of various types of circulatory failure. Alterations in microvascular permeability are a hallmark of a number of inflammatory conditions including sepsis, septic shock, burns and the acute respiratory distress syndrome. As a result, the loss of plasma fluid into the interstitial space leads to hypovolaemia and tissue hypoperfusion. Administration of large volumes of fluids in often necessary to restore the nutritional blood flow to tissues. This strategy, however, involves the risk of interstitial edema formation, which in turn may fur

Design and construction techniques for permeable reactive barriers.

Adequate site characterization, bench-scale column testing, and hydrogeologic modeling formed the basis for the design and construction of permeable reactive barriers for groundwater remediation at various sites, such as Dover Air Force Base, DE and Naval Air Station, Moffett Field, CA. Dissolved chlorinated solvents, such as perchloroethylene (PCE) and trichloroethylene (TCE), have been the focus at many sites because the passive nature of the reactive barrier operation makes such barriers particularly useful for treating groundwater contaminants that can persist in the aquifer for several ye

Role of progesterone in capillary permeability in hyperstimulated rats.

The role of progesterone in capillary permeability, which may be causally related to the pathophysiology of ovarian hyperstimulation syndrome (OHSS), was investigated in immature rats. A total of 96 female Wistar rats aged 22 days were given 10 IU of equine chorionic gonadotrophin daily for 4 consecutive days, and given 30 IU of human chorionic gonadotrophin on the fifth day to produce hyperstimulated manifestations. On the sixth day, groups of 12 rats each received RU486 at a dose of 0, 1, 2.5, 5, 10, 15 or 20 mg/kg (groups 1-7), or RU486 at 5 mg/kg combined with 6alpha-methyl-17alpha-hydroxy

Effect of vitamin E on peroxidation and permeability of the peritoneum.

Because of the evidence that peritoneal macrophages are activated during peritoneal dialysis, we hypothesised that the injury of the peritoneum is, at least in part, dependent on the intraperitoneal generation of free radicals. The aim of the study was to evaluate the effect of vitamin E on the peroxidation and permeability of the peritoneum during chronic peritoneal dialysis in rats. Supplementation of the intraperitoneally infused saline with vitamin E decreased the peroxidation of peritoneum estimated as the malondialdehyde (MDA) level in rats' omentum. However the permeability of the perit

[Permeability of the gastrointestinal tract to macromolecules in experimental anaphylaxis].

Rats were sensitized with single or multiple intraperitoneal administration of chick's egg ovalbumin (OA) or DNP-conjugated bovine serum albumin (DNP-BSA) on aluminium. The animals were challenged 14-28 days thereafter by intragastric or intravenous administration of respective antigens. Gastrointestinal barrier permeability was tested with macromolecular probes: OA and polyethylene glycol (PEG)-4000. The increase of permeability was revealed in animals sensitized with DNP-BSA and challenged intragastrically. The morphological view of intestinal damage included epithelial edema, exfoliation on

Mechanisms and regulation of water permeability in renal epithelia.

Water transport occurs in all biological membranes. A few selected membranes in the kidney, amphibian urinary bladder, and erythrocyte have very high water permeability and are thought to contain specialized water transporting units termed "water channels." The known biophysical properties of membranes containing water channels are a high osmotic water permeability coefficient (Pf), an osmotic-to-diffusional water permeability coefficient ratio (Pf/Pd) greater than unity, a low activation energy (Ea), and inhibition by mercurial compounds. The biochemical and molecular characteristics of water

Vapor-permeable membrane therapy for ulcers of osteomyelitis.

Two patients with chronic ulcerations secondary to arterial insufficiency of the distal extremities were found to have concomitant osteomyelitis immediately beneath the ulcers. An adjunctive would care regimen is described in which local excision or debridement of the involved bone with subsequent systemic antibiotic therapy was the initial treatment. The resultant cutaneous defect was then covered with a vapor-permeable membrane and allowed to close by secondary intention. Rapid and complete healing occurred in both cases in spite of the compromised arterial supply to both areas. This techniq

Intestinal permeability in patients with eczema and food allergy.

Polyethylene glycol (PEG) was used as a probe molecule to investigate intestinal absorption in eight patients with eczema and evidence of food allergy and ten with eczema alone. In both groups absorption of molecules of larger molecular weight was greater than in normal subjects but absorption of molecules of low molecular weight was normal. There was no difference in absorption between eczema patients with or without food allergy. These results suggest that there is an intestinal mucosal defect in eczema which exists whether or not there is coexistent food allergy. Half the patients with ecze

Endonasal venular permeability in rats exposed to the cold.

Sixteen rats were killed 22 hrs after being injected with a dose of 4 mCi(198)Au mixed with 16 mg of inert colloidal gold/100g body wt. The anterior nasal mucosa of the septum and turbinates in 10 rats which were injected and kept in a temperature of 5-7 degrees C showed mean radioactive counts 2-3 times higher than those recorded in the same areas in 6 control rats kept in ordinary room temperature (22 degrees C). In all sixteen rats the highest counts were obtained in the RES organs, namely the liver, spleen, adrenals and the bone marrow, in descending order. Other tissues revealed no signif

Quantitative molecular approach to the permeability changes of excitation.

Functional relationships, available from only a few monolayer studies, can be applied to a relatively simple model of the excitable membrane to give permeability-potential curves quite similar to the conductance-potential curves obtained experimentally in voltage-clamped giant axons. Contrary to the usual view in terms of "carrier systems," the present model considers the permeability to sodium and potassium to be reduced by the increase in the surface pressure induced by large lipophilic cations and anions in the outer layer of the lipoidal bimolecular leaflet constituting the living membrane

A physical interpretation of the phenomenological coefficients of membrane permeability.

A "translation" of the phenomenological permeability coefficients into friction and distribution coefficients amenable to physical interpretation is presented. Expressions are obtained for the solute permeability coefficient omega and the reflection coefficient sigma for both non-electrolytic and electrolytic permeants. An analysis of the coefficients is given for loose membranes as well as for dense natural membranes where transport may go through capillaries or by solution in the lipoid parts of the membrane. Water diffusion and filtration and the relation between these and capillary pore ra

Permeability of the Ehrlich ascites tumor cell to water.

The osmotic permeability coefficient for water has been measured for the Ehrlich mouse ascites tumor cell. Measurements were made of the rate of cell shrinkage in hyperosmotic solutions of NaCI, a functionally impermeable solute. During the first 9 months of weekly serial transplantation the mean was 6.4 micro(3)/micro(3)/atm. +/- 0.8 (S.E.). By the end of the 2nd year the permeability coefficient was much lower and averaged 1.6 +/- 0.09. There were no significant differences in the volume of the tumor cells which could explain the discrepancy on the basis of a change in the volume to surface

PERMEABILITY OF THE HUMAN PLACENTA TO ANTIBODIES : A QUANTITATIVE STUDY.

The ratio of the titers of various antibodies, namely, hemagglutinins and syphilitic reagin, in the maternal blood to that of the corresponding antibody in the cord blood was found to be relatively constant, falling somewhere between 8 and 16. This figure may be considered the "index of permeability" of human placenta to antibodies, or the coefficient of distribution of antibodies between maternal and cord blood. The possible application of these findings to the study of the placental permeability to sensitizing antibodies (or reagins) is discussed.

Development of Cell-Permeable Adenylosuccinate Lyase Inhibitor.

Abnormal adenylosuccinate lyase (ADSL) activity is associated with cancer and neurodevelopmental processes. However, a cell-permeable ADSL inhibitor is not yet available. Our high-throughput screen identified NF-449 as a potential lead compound. To improve cell permeability of the lead compound, fragments of NF-449 were synthesized. This fragment, 2,2'-(1,3-phenylenebis(carbonylimino))-bisbenzenesulfonate, competitively inhibits purified human ADSL with an inhibitory constant of 0.4 micromolar. Its triethylammonium salt inhibited ADSL in HeLa cells with an IC50 of 0.4 micromolar. While this co
