Reversal of haemorrhagic shock in rats by cholinomimetic drugs.

1. In an experimental model of haemorrhagic shock resulting in the death of all rats within 20-30 min, the intravenous (i.v.) injection of the tertiary amine cholinesterase inhibitor physostigmine (17-70 micrograms kg-1) induced a prompt, sustained and dose-dependent improvement of cardiovascular and respiratory function, with marked increase in the volume of circulating blood and survival of all treated animals, at least for the 2 h of observation. 2. Similar results were obtained with the i.v. injection of the cholinoceptor agonist oxotremorine (5-25 micrograms kg-1), while neostigmine (54 o

Neurons of C1 area mediate cardiovascular responses initiated from ventral medullary surface.

We sought to establish whether neurons of the C1 area of the rostral ventrolateral medulla (RVL) mediate changes in arterial pressure and heart rate evoked by topical application of drugs to the ventral medullary surface of the rat. Animals were anesthetized, paralyzed, and ventilated. The ventral surface was mapped with L-glutamate, and a restricted zone was identified from which L-glutamate, as well as kainic acid, bicuculline, strychnine, carbachol, or physostigmine, increased arterial pressure and heart rate. The hypertensive effects of carbachol and physostigmine were blocked by previous

The effects of cholinergic drugs support an avoidance learning hypothesis of brief footshock-induced analgesia.

Rats were tested for tail-flick responses and then immediately subjected to footshock for 30 sec. This procedure induced analgesia, i.e. prolonged the latency of the tail-flick response, which was maximal immediately after the shock and decayed to normal levels within 2 hr. No analgesia occurred if either the analgesia test before the shock or the shock itself was omitted. The dependence of the analgesia on the association between the test before the shock plus the shock, suggests that this was a form of avoidance learning. The effects of drugs injected immediately after the shock were determi

Acetylcholine potentiation of field stimulated rat vas deferens: a pre-synaptic muscarinic mechanism?

Acetylcholine (ACh) was found to markedly enhance the nerve stimulation induced twitch response of isolated, field-stimulated rat vas deferens (RVD). The ED200 (concentration which enhances the twitch response to 200% of control) for this potentiation was 6 X 10(-6)M with the maximum twitch response being increased by more than 3 fold (325 +/- 30%). Carbachol (ED200 = 8.5 X 10(-7)M) showed identical results. With each drug the potentiation was competitively antagonized by atropine (10(-7) - 10(-5)M). Physostigmine 10(-8) - 10(-6)M) both enhanced the basal twitch response (215 +/- 8% of control

Dissimilar effects on body temperature in the cat produced by guanosine 3',5'-monophosphate, acetylcholine and bacterial endotoxin.

1 Guanosine 3',5'-monophosphate (cyclic GMP) and N2-2'-O-dibutyryl guanosine 3',5'-monophosphate (db cyclic GMP) have been injected into the third cerebral ventricle (i.c.v.) of the unanaesthetized cat and the effects of rectal temperature and on behavioural and autonomic activities observed and compared with those of acetylcholine and physostigmine. 2 Acetylcholine (100 nmol) and physostigmine (100 nmol) injected together i.c.v. produced a rise in body temperature in cats at an environmental temperature of 20-24 degrees C, which was abolished by pretreatment i.c.v. with atropine (200 nmol). 3

Altered neural cholinergic receptor systems in cocaine-addicted subjects.

Changes in the brain's cholinergic receptor systems underlie several neuropsychiatric disorders, including Alzheimer's disease, schizophrenia, and depression. An emerging preclinical literature also reveals that acetylcoholine may have an important function in addictive processes, including reward, learning, and memory. This study was designed to assess alterations in cholinergic receptor systems in limbic regions of abstinent cocaine-addicted subjects compared with healthy controls. On three separate days, 23 1- to 6-week abstinent, cocaine- (and mostly nicotine-) addicted subjects and 22 sex

Treatment of opioid-induced delirium with acetylcholinesterase inhibitors: a case report.

A 55-year-old woman with advanced ovarian cancer and severe pain developed hypoactive delirium after an increase in her opioid dosage. Myoclonus and delirium improved dramatically with the intravenous injection of the acetylcholinesterase inhibitor physostigmine, and this improvement was maintained during the administration of donepezil, an oral medication with similar pharmacodynamic properties. Evidence for a disorder of cholinergic neurotransmission in opioid-induced delirium is discussed, as is the rationale for treatment with acetylcholinesterase inhibitors and other cholinomimetic agents

Pharmacologic management of Alzheimer-type dementia.

Diagnosis of Alzheimer-type dementia is made by excluding other causes of dementia. This type of dementia is not well understood, but affects one million persons in the United States. Ergoloid mesylates are sometimes useful. A promising approach may be increasing brain cholinergic activity through combinations of drugs, such as lecithin and physostigmine. Sleep disturbances and impulsivity may both respond to benzodiazepines. Other drugs are being studied, but nonpharmacologic strategies may accomplish more.

The effects of cholinergic agents on morphine-induced circling behavior in the intact mouse.

The effects of morphine on locomotor activity in mice was studied. It was shown that morphine in this species induces circling, a dose-dependent, stereotyped behavior. At high doses of morphine, mice engaged virtually exclusively in circling uninterrupted by other activities. The effect was modified by muscarinic agents. Blockade of muscarinic receptors with atropine potentiated circling while the muscarinic agonist, oxotremorine, attenuated it. The mixed muscarinic/nicotinic cholinergic agonist, physostigmine, had no effect on this behavior.

Undiagnosed central anticholinergic syndrome may lead to dangerous complications.

This report describes two cases of central anticholinergic syndrome, the first after general anaesthesia and the other during a prolonged stay in the intensive care unit. The symptoms in both patients resolved soon after physostigmine administration. There was a delay in the diagnosis of central anticholinergic syndrome, which resulted in acute lung injury and unanticipated intensive care unit admission. It is suggested that in cases of abnormal mental recovery after anaesthesia or sedation, the diagnosis of central anticholinergic syndrome should be considered.

Role of cholinergic mechanism in the hypoglycemic action of B. pertussis vaccine.

The present work aims to study the role of the cholinergic mechanism in the hypoglycemic action of B. pertussis vaccine in rats. The vaccine resembles insulin in that the hypoglycemia produced is mainly due to the increase conversion of glucose to glycogen in the liver and muscle. Both the hypoglycemia as well as glycogen deposition was partially blocked by atropine. On the other hand the vaccine potentiated the fall in the blood sugar and deposition of glycogen in the tissues produced by physostigmine. This suggests the possible involvement of the cholinergic system in the hypoglycemic action

Microinjection of kynurenic acid and lidocaine into the dorsal motor nucleus of vagus evokes airway smooth muscle relaxation in ferrets.

The dorsal motor nucleus of vagus (DMV) provides a critical source of parasympathetic motor output to the airway smooth muscle; however, the central neurotransmitter mechanisms that regulate this activity remain incompletely understood. This study investigated how blockade of excitatory glutamatergic transmission or transient functional suppression of the DMV modulates vagally mediated airway constriction in anaesthetized ferrets (n = 10), divided into two experimental groups (n = 5 per group). Animals were maintained under alpha-chlorose anesthesia and mechanically ventilated. The pressure in

Anticholinesterase agents affect contrast gain of the cat cortical visual evoked potential.

The effects of the anticholinesterase agents physostigmine, pyridostigmine, diisopropylfluorophosphate and soman on the cortical visual evoked potential (VEP) were examined using phase reversed sine wave gratings of varying contrasts. All 4 agents decreased the slope (gain) of the response vs log contrast function while the intercept with the noise level (threshold, an indicator of contrast sensitivity) was unaffected. These results suggest that anticholinesterases modify the VEP by changing a contrast gain control mechanism.

In vitro inhibition of goat brain acetylcholinesterase by pure and commercial anticholinesterase pesticides.

In vitro inhibition of goat cerebellar acetylcholinesterase by pure and commercial anticholinesterase pesticides clearly indicates a remarkably high inhibitory effect of commercial carbamate and organophosphate pesticides containing a lower percentage of the respective active ingredients comparable to that of the known anticholinesterase agents such as DFP and physostigmine. It may be presumed that injudicious use of commercial formulations conduce severe toxicity in the nontarget mammalian species, namely, goat, and this response of the brain acetylcholinesterase may be utilized as a reliable

Multiple cholinesterase inhibitors have antidepressant-like properties in the mouse forced swim test.

There is high clinical interest in improving the pharmacological treatment of individuals with Major Depressive Disorder (MDD). This neuropsychiatric disorder continues to cause significant morbidity and mortality worldwide, where existing pharmaceutical treatments such as selective serotonin reuptake inhibitors often have limited efficacy. In a recent publication, we demonstrated an antidepressant-like role for the acetylcholinesterase inhibitor (AChEI) donepezil in the C57BL/6J mouse forced swim test (FST). Those data added to a limited literature in rodents and human subjects which suggests

Impaired and spared cholinergic functions in the hippocampus after lesions of the medial septum/vertical limb of the diagonal band with 192 IgG-saporin.

To lesion the cholinergic input to the hippocampus, rats received injections of 192 IgG-saporin into the medial septum/vertical limb of the diagonal band (MS/VDB). The lesions produced near-total loss of choline acetyltransferase (ChAT)-positive neurons in the MS/VDB. The loss was accompanied, however, by only partial decreases (to 40% of control levels) in acetylcholine (ACh) release in the hippocampus. Moreover, ACh release in the hippocampus increased when lesioned and control rats were tested on a spontaneous alternation task, indicating that there was significant residual cholinergic func

Enhancement of purinergic neurotransmission by galantamine and other acetylcholinesterase inhibitors in the rat vas deferens.

Galantamine, a mild acetylcholinesterase inhibitor and an allosteric ligand of nicotinic receptors, enhanced in a concentration-dependent manner the amplitude of purinergic twitch contractions of the electrically stimulated rat vas deferens (0.2 Hz, 1 ms, 60 V). Other acetylcholinesterase inhibitors also increased the twitches, showing a hierarchy of potencies of galantamine>physostigmine>tacrine>rivastigmine=donepezil. The potentiations seem to be unrelated to the ability to inhibit acetylcholinesterase, since the hierarchy of potencies to block the enzyme in vas deferens was tacrine>physosti

Is the input to a GABAergic or cholinergic synapse the sole asymmetry in rabbit's retinal directional selectivity?

We examined contrast, direction of motion, and concentration dependencies of the effects of GABAergic and cholinergic antagonists, and anticholinesterases on responses to movement of On-Off directionally selective (DS) ganglion cells of the rabbit's retina. The drugs tested were curare and hexamethonium bromide (cholinergic antagonists), physostigmine (anticholinesterase), and picrotoxin (GABAergic antagonist). They all reduced the cells' directional selectivity, while maintaining their preferred-null axis. However, cholinergic antagonists did not block directional selectivity completely even
