Disruption of microtubular cytoskeleton induced by cryptogein, an elicitor of hypersensitive response in tobacco cells.

The dynamics of microtubular cytoskeleton were studied in tobacco (Nicotiana tabacum cv Xanthi) cells in response to two different plant defense elicitors: cryptogein, a protein secreted by Phytophthora cryptogea and oligogalacturonides (OGs), derived from the plant cell wall. In tobacco plants cryptogein triggers a hypersensitive-like response and induces systemic resistance against a broad spectrum of pathogens, whereas OGs induce defense responses, but fail to trigger cell death. The comparison of the microtubule (MT) dynamics in response to cryptogein and OGs in tobacco cells indicates tha

Cloning and functional analysis of three genes encoding polygalacturonase-inhibiting proteins from Capsicum annuum and transgenic CaPGIP1 in tobacco in relation to increased resistance to two fungal pathogens.

Polygalacturonase-inhibiting proteins (PGIPs) are plant cell wall glycoproteins that can inhibit fungal endopolygalacturonases (PGs). The PGIPs directly reduce the aggressive potential of PGs. Here, we isolated and functionally characterized three members of the pepper (Capsicum annuum) PGIP gene family. Each was up-regulated at a different time following stimulation of the pepper leaves by Phytophthora capcisi and abiotic stresses including salicylic acid, methyl jasmonate, abscisic acid, wounding and cold treatment. Purified recombinant proteins individually inhibited activity of PGs produce

Genome shuffling enhances biocontrol abilities of Streptomyces strains against two potato pathogens.

AIMS: To employ the genome shuffling technique for improving the phenotype of a biocontrol control agent of the genus Streptomyces. METHODS AND RESULTS: Two rounds of genome shuffling (GS) were carried out with Streptomyces melanosporofaciens EF-76, a geldanamycin producer. Six fusants that showed optimized in vitro antagonistic activity against Streptomyces scabies or Phytophthora infestans, two important pathogens of potato crops, were selected. All selected fusants retained the capacity to produce geldanamycin, but none overproduced this antibiotic. The higher antagonism ability appeared t

Spatiotemporal evolution of a pine wilt disease model integrating infectious disease transmission and predator-prey interactions.

Previous modeling studies on pine wilt disease have largely been confined to either standalone epidemic models or predator-prey models, overlooking the coupling between these two mechanisms within the ecosystem. A coupled model is developed that integrates predator-prey interactions with the transmission mechanism of pine wilt disease, thereby enabling a more accurate representation of the spatiotemporal dynamics of biological control and disease transmission. The theoretical analysis primarily establishes that, under given threshold conditions, several key subsystems are persistent, and furth

Late blight resistance of Julius Kühn Institute pre-breeding potato clones: a genome-wide association study.

BACKGROUND: Late blight caused by Phytophthora infestans remains one of the most devastating diseases in potato cultivation, and durable resistance requires the combination of multiple resistance loci. The Julius Kühn Institute (JKI) has maintained a late blight resistance breeding program for more than 60 years and has developed extensive pre-breeding material with largely unknown resistance genetics. To characterize the genetic basis of this resistance, field trials were conducted over three years with 200 genotypes, including pre-breeding clones and cultivars. Genotypes were inoculated with

Potato β-aminobutyric acid receptor IBI1 manipulates VOZ1 and VOZ2 transcription factor activity to promote disease resistance.

Upon infection with nonpathogenic microorganisms or treatment with natural or synthetic compounds, plants exhibit a more rapid and potent response to both biotic and abiotic stresses. However, the molecular mechanisms behind this phenomenon, known as defense priming, are poorly understood. β-minobutyric acid (BABA) is an endogenous stress metabolite that enhances plant tolerance to various abiotic stresses and primes plant defense responses, providing the ability to resist a variety of pathogens (broad-spectrum resistance). In this study, we identified an aspartyl-tRNA synthetase (AspRS), StIB

Elevated CO2 drives nonlinear trait responses and reshapes growth-virulence trade-offs in a plant pathogen.

Atmospheric CO2 is rising at unprecedented rates. While its physiological effects on plants are well-documented, the direct evolutionary impact on plant pathogens remains unknown. We investigated how CO2 interacts with other abiotic factors to shape the evolutionary potential of pathogens. We combined controlled CO2 gradient experiments with quantitative genetics to assess the interactive effects of CO2, temperature and geography on Phytophthora infestans. We measured intrinsic growth rate and aggressiveness, the two key pathogen traits, across seven geographically distinct populations and par

Characterization of bridged guaiane-type sesquiterpenoids from Stellera chamaejasme and their antifungal activities against phytopathogenic fungi.

Plant pathogenic fungi pose a serious threat to global agricultural productivity, driving the need for novel and eco-friendly antifungal agents. An investigation into the antifungal constituents from the roots of Stellera chamaejasme led to the isolation of nine new guaiane-type sesquiterpenoids, named stelleroids D-L (1-3 and 8-13), along with eight known analogues. Their structures, featuring extensively modified bridged scaffolds and diverse oxidative patterns, were elucidated by comprehensive spectroscopic analysis and X-ray crystallography. Enhanced eugenol biosynthesis through GmEGS overexpression confers broad-spectrum pathogen protection in soybean.

Soybean is a major source of plant protein and oil, yet its production is severely constrained by diverse pathogen infections. Here, we identify eugenol as a pathogen-induced defense metabolite in soybean with broad-spectrum antimicrobial activity against fungal, oomycete, and bacterial pathogens. Pathogen-associated molecular patterns (PAMPs), including Flg22, Chitin, and Elicitin, strongly induced soybean eugenol synthase (GmEGS) genes and promoted eugenol accumulation. Phylogenetic and expression analyses identified five GmEGS genes, among which GmEGS1a, GmEGS1c, and GmEGS2a were strongly i

Stacking of antimicrobial genes in potato transgenic plants confers increased resistance to bacterial and fungal pathogens.

Solanum tuberosum plants were transformed with three genetic constructions expressing the Nicotiana tabacum AP24 osmotine, Phyllomedusa sauvagii dermaseptin and Gallus gallus lysozyme, and with a double-transgene construction expressing the AP24 and lysozyme sequences. Re-transformation of dermaseptin-transformed plants with the AP24/lysozyme construction allowed selection of plants simultaneously expressing the three transgenes. Potato lines expressing individual transgenes or double- and triple-transgene combinations were assayed for resistance to Erwinia carotovora using whole-plant and tub

The multifunctional enzyme CYP71B15 (PHYTOALEXIN DEFICIENT3) converts cysteine-indole-3-acetonitrile to camalexin in the indole-3-acetonitrile metabolic network of Arabidopsis thaliana.

Accumulation of camalexin, the characteristic phytoalexin of Arabidopsis thaliana, is induced by a great variety of plant pathogens. It is derived from Trp, which is converted to indole-3-acetonitrile (IAN) by successive action of the cytochrome P450 enzymes CYP79B2/B3 and CYP71A13. Extracts from wild-type plants and camalexin biosynthetic mutants, treated with silver nitrate or inoculated with Phytophthora infestans, were comprehensively analyzed by ultra-performance liquid chromatography electrospray ionization quadrupole time-of-flight mass spectrometry. This metabolomics approach was combi

Synthesis and fungicidal activity of novel 1,1-diaryl-2-azolylethanols with a pyridine moiety.

To investigate the effect of pyridine heterocycle on the activity of triazole alcohol fungicides, 31 pyridine-containing 2-azolyl-1,1-diarylmethanol derivatives were designed and synthesized using flutriafol as the lead compound and pyridine diaryl ketone molecular building blocks as the basis. The in vitro fungicidal activity against 10 plant pathogenic fungi was determined, and the results showed that the target compounds exhibited broad-spectrum inhibitory effects on the tested pathogens. Among them, the imidazole-substituted compounds I-24, I-26 and I-27 displayed remarkable fungicidal act

ROS and Oxidative Response Systems in Plants Under Biotic and Abiotic Stresses: Revisiting the Crucial Role of Phosphite Triggered Plants Defense Response.

Phosphite (Phi) is a chemical analog of orthophosphate [HPO4 3-]. It is a systemic pesticide generally known to control the prevalence of oomycetes and soil-borne diseases such as Phytophthora, Pythium, and Plasmopora species. Phi can also control disease symptoms and the spread of pathogenic bacteria, fungi, and nematodes. Phi plays critical roles as a fungicide, pesticide, fertilizer, or biostimulator. Overall, Phi can alleviate the severity of the disease caused by oomycete, fungi, pathogenic bacteria, and nematodes (leave, stem, fruit, tuber, and root) in various plants (vegetables, fruits

Liquid Fungal Cocultivation as a Strategy to Access Bioactive Metabolites.

Fungi are a rich source of bioactive compounds. Fungal cocultivation is a method of potentiating chemical interactions and, consequently, increasing bioactive molecule production. In this study, we evaluated the bactericidal, antiprotozoal, and cathepsin V inhibition activities of extracts from axenic cultures of 6 fungi (Fusarium guttiforme, Pestalotiopsis diospyri, Phoma caricae-papayae, Colletotrichum horii, Phytophthora palmivora, and C. gloeosporioides) that infest tropical fruits and 57 extracts obtained by their cocultivation. Our results reveal that fungal cocultivation enhances the bi

MicroRNA396a-5p and -3p induce tomato disease susceptibility by suppressing target genes and upregulating salicylic acid.

Plants have evolved a variety of mechanisms to perceive and resist the assault of pathogens. The biotrophs, necrotrophs and hemibiotrophs are types of plant pathogens that activate diverse salicylic acid (SA) and jasmonic acid (JA) signaling pathways. In this study we showed that the expressions of miR396a-5p and -3p in Solanum lycopersicum (S. lycopersicum) were both down-regulated after infection by hemibiotroph Phytophthora infestans (P. infestans) and necrotroph Botrytis cinerea (B. cinerea) infection. Overexpression of miR396a-5p and -3p in transgenic tomato enhanced the susceptibility of

Plant-derived antifungal agent poacic acid targets β-1,3-glucan.

A rise in resistance to current antifungals necessitates strategies to identify alternative sources of effective fungicides. We report the discovery of poacic acid, a potent antifungal compound found in lignocellulosic hydrolysates of grasses. Chemical genomics using Saccharomyces cerevisiae showed that loss of cell wall synthesis and maintenance genes conferred increased sensitivity to poacic acid. Morphological analysis revealed that cells treated with poacic acid behaved similarly to cells treated with other cell wall-targeting drugs and mutants with deletions in genes involved in processes

Identification of the pepper SAR8.2 gene as a molecular marker for pathogen infection, abiotic elicitors and environmental stresses in Capsicum annuum.

Pepper ( Capsicum annuum L.) SAR8.2 genes, designated CASAR82A, B and C, which are induced by all the biotic and abiotic stresses, were isolated from a pepper cDNA library constructed with the mRNAs from pepper plants infected with Xanthomonas campestris pv. vesicatoria. The pepper CASAR82A, B and C gene products, which are very similar to each other in amino acid sequences, have 43-50% homology with those of tobacco SAR8.2 genes. The CASAR8.2 genes were not constitutively expressed in any of the organs of healthy pepper plants. In contrast, the CASAR82A gene was locally or systemically induce
