Specific targeting of neurotoxic side effects and pharmacological profile of the novel cancer stem cell drug salinomycin in mice.

UNLABELLED: Salinomycin is a polyether antibiotic which effectively eliminates a variety of cancer stem cells and chemotherapy-resistant tumor cells in vitro and in vivo. One important caveat for its clinical application is the paucity of preclinical pharmacological and safety data. In the present study, we thus aimed to elucidate pharmacokinetic properties of salinomycin and to assess the side effect profile of chronic treatment with this compound in C57Bl/6 mice. In addition, we tested whether neurotoxic side effects can be prevented by interference with the intracellular calcium homeostasis

3D printed biomimetic PEEK/HA scaffolds induce cranial bone regeneration.

The repair of critical-sized cranial defects resulting from trauma, congenital malformations, or surgery remains a significant clinical challenge. Polyetheretherketone (PEEK) is a commonly used material for cranial defect repair in clinical practice. However, its application is severely limited by drawbacks such as poor structural adaptability, bioinertness, and insufficient osseointegration, which significantly compromise clinical outcomes. In this study, we biomimicked the structure and composition of natural bone to fabricate a biomimetic PEEK/hydroxyapatite (HA) scaffold via a 3D printing

Quality-of-life assessment in the randomized JBCRG-M06/EMERALD study of eribulin plus dual HER2 blockade in HER2-positive locally advanced or metastatic breast cancer.

BACKGROUND: Although taxanes are a mainstay treatment for locally advanced or metastatic breast cancer (LABC/MBC), they often impair quality of life (QoL). Treatments that avoid taxane-related QoL deteriorations would be valuable. METHODS: The JBCRG-M06/EMERALD trial (NCT03264547, UMIN000027938) compared eribulin with a taxane, each combined with trastuzumab and pertuzumab, in patients with human epidermal growth factor receptor type 2 (HER2)-positive LABC/MBC. QoL was assessed using the European Organization for Research and Treatment of Cancer Quality of Life Questionnaire Module C30 (EORTC

Combining conventional and digital technologies in immediate mandibular implant overdenture fabrication: A dental technique.

Immediate complete dentures provide patients with improved esthetics and reasonable function following dental extractions. Denture base reinforcement in implant overdentures helps distribute occlusal stress and reduces the risk of denture fracture. This technique describes the fabrication of a digitally designed and subtractively manufactured denture base, a conventional compression-molded denture base for an immediate implant overdenture, and polyetheretherketone frameworks step-by-step. The integration of techniques facilitated a predictable transition to a mandibular implant overdenture for

Design of Eudragit E100 Films Incorporating Lipoic Acid as a Permeability-Enhancing Platform for Prednisolone Delivery in the Treatment of Ophthalmic Diseases.

Prednisolone (PRED), a widely used corticosteroid for the treatment of ocular inflammation, has limited bioavailability due to its poor water solubility. In this study, Eudragit E100-based films incorporating polyethylene glycol 400 (PEG) and α-lipoic acid (ALA) as plasticizers were developed and characterized to improve the ocular delivery and corneal permeability of PRED. ALA, which is also known for its antioxidant and therapeutic properties in ocular surface disorders, plays a dual role by enhancing both the mechanical properties and drug delivery performance of the films. The E100:PEG:ALA

Combined effects of okadaic acid and pectenotoxin-2, 13-desmethylspirolide C or yessotoxin in human intestinal Caco-2 cells.

Lipophilic phycotoxins are secondary metabolites produced by phytoplanktonic species. They accumulate in filtering shellfish and can cause human intoxications. Humans can be exposed to combinations of several phycotoxins. The toxicological effects of phycotoxin mixtures on human health are largely unknown. Published data on phycotoxin co-exposure show that okadaic acid (OA) is simultaneously found with pectenetoxin-2 (PTX-2), 13-desmethylspirolide C (also known as SPX-1), or yessotoxin (YTX). Therefore, the aim of this study was to examine the effects of three binary mixtures, OA/PTX-2, OA/SPX
