Assimilation dynamics of xylem-transported CO2 by woody tissue photosynthesis revealed with 11C- and 13C-labelling.

A considerable quantity of locally respired CO2 in stems can be assimilated via woody tissue photosynthesis (Pwt). Hence, Pwt locally provides carbon fuelling radial stem growth and therefore plays a major role in the stem carbon balance. It is, however, not straightforward to quantify or visualize its dynamic behaviour, especially on small spatial scales. To assess Pwt we traced xylem-transported 11CO2 in detached branches of Populus tremula L. and used positron autoradiography to visualize and quantify its dynamics on a submillimetre scale, while manipulating light exposure to the branch. Photochemical reflectance index and its relation to photosynthetic characteristics under dynamic light environment.

The photochemical reflectance index (PRI) is a normalized reflectance index that is expected to be useful for estimating photosynthetic activity based on remote-sensing images. Experimental and theoretical studies have examined how the PRI is related to photosynthesis, but they have been based on observations under steady-state light conditions. Photosynthetic systems display differential temporal responsiveness when exposed to variation in light intensity. Here, we examined the responses of the CO2 assimilation rate (A), quantum yield of PSII photochemistry (ФP), nonphotochemical quenching (N

A single gene underlies the dynamic evolution of poplar sex determination.

Although hundreds of plant lineages have independently evolved dioecy (that is, separation of the sexes), the underlying genetic basis remains largely elusive1. Here we show that diverse poplar species carry partial duplicates of the ARABIDOPSIS RESPONSE REGULATOR 17 (ARR17) orthologue in the male-specific region of the Y chromosome. These duplicates give rise to small RNAs apparently causing male-specific DNA methylation and silencing of the ARR17 gene. CRISPR-Cas9-induced mutations demonstrate that ARR17 functions as a sex switch, triggering female development when on and male development wh

Fate of carbon tetrachloride during phytoremediation with poplar under controlled field conditions.

The fate of carbon tetrachloride (CT) during phytoremediation with poplar was assessed by examining the transpiration of CT from leaves, diffusion from soil, tree trunks, and surface roots, and accumulation of chloride ion in soil and plant tissues. Feedwater containing 12-15 mg/L CT was added to the field test beds planted with poplar, and over 99% of the CT was removed. No significant amount of CT was transpired or diffused into the air, and no significant amount of CT-chlorine accumulated in the tree tissues. Chloride ion accumulated in the soil accounted for all of the CT-chlorine removed.

Lack of antagonism between salicylic acid and jasmonate signalling pathways in poplar.

Salicylic acid (SA) and jasmonic acid (JA) often play distinct roles in plant defence against pathogens. Research from Arabidopsis thaliana has established that SA- and JA-mediated defences are more effective against biotrophs and necrotrophs, respectively. These two hormones often interact antagonistically in response to particular attackers, with the induction of one leading to suppression of the other. Here, we report a contrasting pattern in the woody perennial Populus: positive SA-JA interplay. Using genetically engineered high SA lines of black poplar and wild-type lines after exogenous

The Potential of the Ni-Resistant TCE-Degrading Pseudomonas putida W619-TCE to Reduce Phytotoxicity and Improve Phytoremediation Efficiency of Poplar Cuttings on A Ni-TCE Co-Contamination.

To examine the potential of Pseudomonas putida W619-TCE to improve phytoremediation of Ni-TCE co-contamination, the effects of inoculation of a Ni-resistant, TCE-degrading root endophyte on Ni-TCE phytotoxicity, Ni uptake and trichloroethylene (TCE) degradation of Ni-TCE-exposed poplar cuttings are evaluated. After inoculation with P. putida W619-TCE, root weight of non-exposed poplar cuttings significantly increased. Further, inoculation induced a mitigation of the Ni-TCE phytotoxicity, which was illustrated by a diminished exposure-induced increase in activity of antioxidative enzymes. Consi

[Progress on whole genome sequencing in woody plants].

In recent years, the number of sequencing data of plant whole genome have been increasing rapidly and the whole genome sequencing has been also performed widely in woody plants. However, there are a set of obstacles in investigating the whole genome sequencing in woody plants, which include larger genome, complex genome structure, limitations of assembly, annotation, functional analysis, and restriction of the funds for scientific research. Therefore, to promote the efficiency of the whole genome sequencing in woody plants, the development and defect of this field should be analyzed. The three

Allergenic pollen in the atmosphere of Kayseri, Turkey.

Airborne pollen are important allergens that cause sensitization in allergic rhinoconjunctivitis and asthma. Our aim was to detect the pollen in the atmosphere of Kayseri, to present a pollen calendar, and to detect the allergenic level of these pollen by performing skin tests on patients. Atmospheric pollen were collected by Durham gravimetric samplers in Kayseri between March and November in the years 1996 and 1997. In our study, we observed pollen belonging to 43 different taxa. The total number of pollen per cm2 was found to be 1,330.8 in 1996 and 1,182.5 in 1997. Most of the pollen were f

Betula pendula: A Promising Candidate for Phytoremediation of TCE in Northern Climates.

Betula pendula (Silver birch) trees growing on two contaminated sites were evaluated to assess their capacity to phytoscreen and phytoremediate chlorinated aliphatic compounds and heavy metals. Both locations are industrially-contaminated properties in central Sweden. The first was the site of a trichloroethylene (TCE) spill in the 1980s while the second was polluted with heavy metals by burning industrial wastes. In both cases, sap and sapwood from Silver birch trees were collected and analyzed for either chlorinated aliphatic compounds or heavy metals. These results were compared to analyses

Fulminating fungal sinusitis caused by Valsa sordida, a plant pathogen, in a patient immunocompromised by acute myeloid leukemia.

We describe a case in which a patient immunocompromised by acute myeloid leukemia experienced acute fulminating invasive fungal sinusitis followed by pneumonia and then death. Though the microbiology of the pneumonia could not be directly investigated, nasal lesions revealed fungal mycelium. Valsa sordida was consistently cultured from a biopsied sample. The fungus was identified to the genus level based on morphology in culture and DNA sequence homology, and then was placed at species level by means of phylogenetic analysis of the nuclear ribosomal internal transcribed spacer region. The fung

Coupling recycled K2CO3 from first-round activation with Ca(OH)2 for facilitating development of mesopores for enhancing tetracycline adsorption.

Activation with K2C2O4 generates K2CO3 that could be recycled for second-round activation. However, activation with either K2C2O4 or K2CO3 produces activated carbon (AC) with mainly micropores. Herein, recycled K2CO3 reacted with Ca(OH)2 to generate KOH, CaO and CO2 for activation of poplar, cellulose and lignin. The results indicated that K2CO3 + Ca(OH)2 showed superior cracking capability, generating AC from poplar with higher SBET (1822.1 versus 1179.4 m2/g with K2CO3 or 1255.6 m2/g with K2C2O4) and higher meso-porosity (43.3 % versus 10.0 % with recycled K2CO3 and 3.1 % with K2C2O4). Leaf beetles employ tryptophan to detoxify the chemical defenses of poplar trees.

Herbivorous insects have evolved many fascinating adaptations to overcome the chemical defenses of their host plants. This study employed targeted and untargeted metabolomics, coupled with isotope labeling, to shed light on the metabolism of salicinoids-potent antiherbivore phenolic defenses present in the Salicaceae family-in the poplar-specialized leaf beetle, Chrysomela tremulae. C. tremulae was found to produce a range of metabolites from salicortin and utilize the essential amino acid tryptophan and its breakdown products, namely kynurenine, kynurenic acid, and 4-hydroxyquinoline, to form

Transformation of [14C] trichloroethylene by poplar suspension cells.

Trichloroethylene (TCE) is one of the most prevalent environmental contaminants, and it poses an expensive remediation problem. Phytoremediation has been investigated as a potential tool for the removal of TCE from ground water and soil, and has shown promise in preliminary trials. However, the fate of TCE in plants is largely unknown. Radiolabel studies showed that once taken up and transformed, most of the TCE is incorporated into plant tissue as a non-volatile, un-extractable residue. We describe here an assay for TCE transformation by poplar suspension cells. Using this assay, it was shown

Belowground Fungal Volatile Exposure Induces Systemic Metabolic Reprogramming in Poplar.

Although trees encounter diverse soil-associated fungi, it is unclear how they adjust their physiology in response to volatile organic compounds (VOCs) from fungi with contrasting ecological strategies before physical contact. Here, we tested whether VOCs emitted by fungi are sufficient to induce systemic, fungus-dependent metabolic responses in poplar. Populus × canescens roots were exposed to VOCs from a pathogen (Heterobasidion annosum), a saprotroph (Postia placenta), or an ectomycorrhizal mutualist (Laccaria bicolor) for 6 weeks in a contact-free pot-in-pot system. Annotation survey and life cycle transcriptomics of transcription factors in rust fungi (Pucciniales) identify a possible role for cold shock proteins in dormancy exit.

Fungi of the order Pucciniales are obligate plant biotrophs causing rust diseases. They exhibit a complex life cycle with the production of up to five spore types, infection of two unrelated hosts and an overwintering stage. Transcription factors (TFs) are key regulators of gene expression in eukaryote cells. In order to better understand genetic programs expressed during major transitions of the rust life cycle, we surveyed the complement of TFs in fungal genomes with an emphasis on Pucciniales. We found that despite their large gene numbers, rust genomes have a reduced repertoire of TFs comp

Effects of heat and drought stress on post-illumination bursts of volatile organic compounds in isoprene-emitting and non-emitting poplar.

Over the last decades, post-illumination bursts (PIBs) of isoprene, acetaldehyde and green leaf volatiles (GLVs) following rapid light-to-dark transitions have been reported for a variety of different plant species. However, the mechanisms triggering their release still remain unclear. Here we measured PIBs of isoprene-emitting (IE) and isoprene non-emitting (NE) grey poplar plants grown under different climate scenarios (ambient control and three scenarios with elevated CO2 concentrations: elevated control, periodic heat and temperature stress, chronic heat and temperature stress, followed by

A mass balance study of the phytoremediation of perchloroethylene-contaminated groundwater.

A mass balance study was performed under controlled field conditions to investigate the phytoremediation of perchloroethylene (PCE) by hybrid poplar trees. Water containing 7-14 mg L(-1) PCE was added to the test bed. Perchloroethylene, trichloroethylene, and cis-dichloroethylene were detected in the effluent at an average of 0.12 mg L(-1), 3.9 mg L(-1), and 1.9 mg L(-1), respectively. The total mass of chlorinated ethenes in the water was reduced by 99%. Over 95% of the recovered chlorine was as free chloride in the soil, indicating near-complete dehalogenation of the PCE. Coupling two mercury resistance genes in Eastern cottonwood enhances the processing of organomercury.

Eastern cottonwood (Populus deltoides Bartr. ex Marsh.) trees were engineered to express merA (mercuric ion reductase) and merB (organomercury lyase) transgenes in order to be used for the phytoremediation of mercury-contaminated soils. Earlier studies with Arabidopsis thaliana and Nicotiana tabacum showed that this gene combination resulted in more efficient detoxification of organomercurial compounds than did merB alone, but neither species is optimal for long-term field applications.
