Structural characterization of a galectin isolated from the marine sponge Chondrilla caribensis with leishmanicidal potential.

BACKGROUND: Solving primary structure of lectins leads to an understanding of the physiological roles within an organism and its biotechnological potential. Only eight sponge lectins have had their primary structure fully determined. METHODS: The primary structure of CCL, Chondrilla caribensis lectin, was determined by tandem mass spectrometry. The three-dimensional structure was predicted and the protein-carbohydrate interaction analysed by molecular docking. Furthermore, the anti-leishmanial activity was observed by assays with Leishmania infantum. RESULTS: The amino acid sequence consists

Fuscasins A-D, Cycloheptapeptides from the Marine Sponge Phakellia fusca.

Four new cycloheptapeptides, fuscasins A-D (1-4), were isolated from the marine sponge Phakellia fusca collected from the South China Sea. Their planar structures were fully characterized by spectroscopic methods, and the absolute configurations of amino acid residues were determined using the advanced Marfey's method. Structurally, 1 is a unique cycloheptapeptide with a backbone bearing a pyrrolidine-2,5-dione unit. Among the isolated compounds, 1 exhibited potent growth-inhibitory activity against HepG2 cells with an IC50 value of 4.6 μM, whereas it did not show apparent inhibitory effects a

Formal Synthesis of Sarain A: Intramolecular Cycloaddition of an Eight-Membered Cyclic Nitrone to Construct the 2-Azabicyclo[3.3.1]nonane Framework.

An enantioselective route to the tetracyclic skeleton of sarain A has been developed. Asymmetric reduction of an ynone introduced a chiral center which was transferred to the contiguous tertiary stereogenic centers through an Ireland-Claisen rearrangement. The 2-azabicyclo[3.3.1]nonane framework was constructed by an unprecedented intramolecular cycloaddition of an eight-membered cyclic nitrone. Using the steric bias of the bicyclic system, the quaternary carbon atom was constructed by a stereoselective aldol reaction. Further ring formations were performed by ring-closing metathesis for the 1

Antimicrobial activity of marine sponge Clathria indica (Dendy, 1889).

Sponges are sessile filter feeders that have developed efficient defense mechanisms against foreign invaders such as viruses, bacteria or eukaryotic organisms. Antimicrobial peptides are known as major components of the innate immune defense system in marine invertebrates. The aim of the present work was to study the antimicrobial properties of the Indian sponge Clathria indica with special reference to the identification of antimicrobial peptides. Crude methanolic extract and its chloroform, n-butanol and aqueous fractions were tested against 16 human pathogens which include eleven bacteria w

Morphological models of radiate accretive growth and the influence of hydrodynamics.

In many marine sessile organisms (for example sponges and stony corals) the skeleton is formed by an accretive growth process, where layers of material are secreted on top of each other in a surface normal deposition process. In many of these organisms the growth process exhibits a strong morphological plasticity due to differences in exposure to water movement. In general, many of these organisms tend to form thin-branching growth forms under sheltered conditions, while the growth form gradually transforms into a more compact shape when the exposure of water movement increases. In this paper,

Cytotoxic Cytochalasans from Sponge-Derived Aspergillus sp. SCSIO 41044.

Two new cytochalasans (1-2), along with eight known ones (3-10) were isolated from sponge-derived Aspergillus sp. SCSIO 41044. The planar structures of 1-2 were elucidated through extensive spectroscopic analyses and their absolute configurations determined by modified Mosher's methods. Biological evaluation revealed that 5, 7, and 8 showed potent cytotoxicity against small cell lung cancers H446 and H1048, with IC50 values ranging from 0.0441 to 1.61 μM.

Descriptions of new and rare crabs associated with coral rubble and sponges from Kanyakumari, southeastern Arabian Sea, southern India (Decapoda: Brachyura: Epialtidae and Pilumnidae).

Three new brachyuran crab species are described from coral rubble and sponges collected in the southern Indian state of Tamil Nadu. The epialtid, Acanthophrys keeae n. sp., was obtained from coral rubble and is the first species from the genus reported from the Indian Ocean. It is most similar to A. costatus Griffin & Tranter 1986, from eastern Australia, but can be distinguished by the proportionately longer postorbital spine, more strongly granulated gastric region, relatively less inflated and narrower branchial region, sinuous outer margin of the basal antennal article which has the median

Evaluation of the In Vivo Biological Effects of Marine Collagen and Hydroxyapatite Composite in a Tibial Bone Defect Model in Rats.

One of the most promising strategies to improve the biological performance of bone grafts is the combination of different biomaterials. In this context, the aim of this study was to evaluate the effects of the incorporation of marine spongin (SPG) into Hydroxyapatite (HA) for bone tissue engineering proposals. The hypothesis of the current study is that SPG into HA would improve the biocompatibility of material and would have a positive stimulus into bone formation. Thus, HA and HA/SPG materials were produced and scanning electron microscopy (SEM) analysis was performed to characterize the sam

[Delayed posttraumatic high flow priapism: a case report].

A 21-year-old male complained of persisting erection. A urethral balloon catheter had been for 3 weeks after indwelt urethral injury by a skateboard, and he was hospitalized because of penile erection persisting after removing the catheter. High flow priapism was suspected by intracavernous blood gas study and color Doppler ultrasound study. Selective internal pudendal arteriography revealed a leakage of contrast medium at the base of penis. He was treated with selective embolization of bilateral internal genital arteries using gelatin sponges and achieved detumescence. Normal potency was evid

Topology-driven mechanical performance in architected cellular materials: insights from bioinspired glass sponge lattices.

Architected lattice materials inspired by biological structures are frequently described as bioinspired, yet the underlying functional principles governing their mechanical response are not always explicitly isolated. The hexactinellid spongeEuplectella aspergillumexhibits a distinctive skeletal organization based on a periodic square unit subdivided into four sub-squares, where two opposite regions are reinforced by paired diagonal struts while the remaining corners remain non-reinforced. This alternating reinforcement pattern introduces spatial heterogeneity in stiffness and connectivity at

Rapid Clearance of Tetracapsuloides bryosalmonae Spores by Freshwater Sponge Ephydatia muelleri: Potential Implications for Controlling Proliferative Kidney Disease in Salmonids.

Host-parasite interactions are influenced by both abiotic and biotic factors. While abiotic drivers, particularly temperature, have received considerable attention in recent years due to global climate change, the role of biotic factors remains comparatively underexplored. The malacosporean parasite Tetracapsuloides bryosalmonae (Tb), which causes proliferative kidney disease in salmonids, is a well-documented example, where elevated temperature and eutrophication have been shown to intensify disease prevalence and severity. In this study, we investigated whether freshwater sponges, known for

The UHPLC-Orbitrap MS/MS and Network Pharmacology Strategies Reveal the Active Antioxidants of Bleeding Sap from Sponge Gourd in Treating Tuberculosis.

The bleeding sap of Luffa cylindrica (L.) Roem has been used for the treatment of tuberculosis since the record of Supplements to Compendium of Materia Medica. The active components and possible mechanism of it are yet ambiguous. Hence, this study is focused on investigating the possible mechanism underpinning this effect on the perspective of the antioxidant ingredients from the bleeding sap. Through organic solvents extraction, HPLC fractionation, DPPH trials evaluation, and UHPLC-Orbitrap tandem MS identification, a total of 37 compounds were identified from the bleeding sap with the strong

Selection of sponge-associated bacteria with high potential for the production of antibacterial compounds.

The potential of sponge-associated bacteria for the biosynthesis of natural products with antibacterial activity was evaluated. In a preliminary screening 108 of 835 axenic isolates showed antibacterial activity. Active isolates were identified by 16S rRNA gene sequencing and selection of the most promising strains was done in a championship like approach, which can be done in every lab and field station without expensive equipment. In a competition assay, strains that inhibited most of the other strains were selected. In a second round, the strongest competitors from each host sponge competed

Sponge grade body fossil with cellular resolution dating 60 Myr before the Cambrian.

An extraordinarily well preserved, 600-million-year (Myr)-old, three-dimensionally phosphatized fossil displaying multiple independent characters of modern adult sponges has been analyzed by SEM and synchrotron X-ray tomography. The fossilized animal (Eocyathispongia qiania gen. et sp. nov.) is slightly more than 1.2 mm wide and 1.1 mm tall, is composed of hundreds of thousands of cells, and has a gross structure consisting of three adjacent hollow tubes sharing a common base. The main tube is crowned with a large open funnel, and the others end in osculum-like openings to the exterior. The ex

Anti-parasitic compounds from Streptomyces sp. strains isolated from Mediterranean sponges.

Actinomycetes are prolific producers of pharmacologically important compounds accounting for about 70% of the naturally derived antibiotics that are currently in clinical use. In this study, we report on the isolation of Streptomyces sp. strains from Mediterranean sponges, on their secondary metabolite production and on their screening for anti-infective activities. Bioassay-guided isolation and purification yielded three previously known compounds namely, cyclic depsipeptide valinomycin, indolocarbazole alkaloid staurosporine and butenolide. This is the first report of the isolation of valino

Induced Dysbiosis Modifies Physiological Responses to Warming in a Temperate Sponge.

Marine sponges host complex and specific microbiomes, yet the role of these microbial symbionts in sponge responses to acute environmental stress is not fully understood. We investigated how altering the microbiome influences physiological responses to temperature in the temperate calcareous sponge Rowella lancifera under simulated marine heatwave (MHW) conditions. Sponges were treated with broad-spectrum antibiotics to disrupt their microbiome and then exposed to sub-lethal MHW temperatures. Microbial community composition was analysed using 16S rRNA gene sequencing, including assessing diver
