Evolution of the primate trypanolytic factor APOL1.

ApolipoproteinL1 (APOL1) protects humans and some primates against several African trypanosomes. APOL1 genetic variants strongly associated with kidney disease in African Americans have additional trypanolytic activity against Trypanosoma brucei rhodesiense, the cause of acute African sleeping sickness. We combined genetic, physiological, and biochemical studies to explore coevolution between the APOL1 gene and trypanosomes. We analyzed the APOL1 sequence in modern and archaic humans and baboons along with geographic distribution in present day Africa to understand how the kidney risk variants

Neuropathogenesis of Japanese encephalitis in a primate model.

BACKGROUND: Japanese encephalitis (JE) is a major cause of mortality and morbidity for which there is no treatment. In addition to direct viral cytopathology, the inflammatory response is postulated to contribute to the pathogenesis. Our goal was to determine the contribution of bystander effects and inflammatory mediators to neuronal cell death. METHODOLOGY/PRINCIPAL FINDINGS: Material from a macaque model was used to characterize the inflammatory response and cytopathic effects of JE virus (JEV). Intranasal JEV infection induced a non-suppurative encephalitis, dominated by perivascular, inf

Coagulation dysregulation as a barrier to xenotransplantation in the primate.

PURPOSE OF REVIEW: The ability to generate pigs expressing a human complement regulatory protein (hCRP) and/or pigs in which the alpha1,3-galactosyltransferase gene has been knocked out (GT-KO) has largely overcome the barrier of hyperacute rejection of a pig organ transplanted into a primate. However, acute humoral xenograft rejection (AHXR), presenting as microvascular thrombosis and/or consumptive coagulopathy, remains a major hurdle to successful xenotransplantation. This review summarizes recent studies of the coagulation problems associated with xenotransplantation, and discusses potenti

Parastrongylus cantonensis in a nonhuman primate, Florida.

Parastrongylus (= Angiostrongylus) cantonensis is a parasitic nematode of Norway rats throughout tropical regions. This parasite is neurotropic and causes disease and death in humans and other mammals. We report the first identification of P. cantonensis as the cause of a debilitating neurologic disease in a captive primate in Florida.

A primer of primate pathology: lesions and nonlesions.

Nonhuman primates are important laboratory animals for biomedical, pharmacology, and toxicology research. To effectively use primates as models, their gross and histologic anatomy, physiology and natural history, as well as common health problems and the source from which the primate is obtained, must be known and understood by pathologists involved in study design and/or interpretation. The first very important lesson in the "primer" is: there is no such thing as a generic monkey. Brand names (ie, species and subspecies) are important. Several taxonomic groups of primates are used in research

Teeth, brains, and primate life histories.

This paper explores the correlates of variation in dental development across the order Primates. We are particularly interested in how 1) dental precocity (percentage of total postcanine primary and secondary teeth that have erupted at selected absolute ages and life cycle stages) and 2) dental endowment at weaning (percentage of adult postcanine occlusal area that is present at weaning) are related to variation in body or brain size and diet in primates. We ask whether folivores have more accelerated dental schedules than do like-sized frugivores, and if so, to what extent this is part and pa

Clusterin protein diversity in the primate eye.

PURPOSE: The clusterin gene encodes a multi-functional protein that has been identified in different tissues, including a number of different eye tissues, primarily in the mouse and to a much lesser extent in humans. Clusterin has been implicated in a number of cellular processes such as lipid transport, membrane integrity, apoptosis, and neurodegeneration, all of which could be important to the biology of the eye. In the current communication, we provide data that confirms the expression of clusterin in a number of different human eye tissues and establishes the expression profile of this gen

The role of estrogen in the maintenance of primate pregnancy.

OBJECTIVE: The aim of this study was to determine the role of estrogen in pregnancy maintenance in baboons by suppressing estrogen synthesis through administration of a highly specific nonsteroidal aromatase inhibitor, CGS 20267. STUDY DESIGN: CGS 20267 was administered subcutaneously at maximal dosages of 2.0 mg/d to pregnant baboons (n = 24) daily beginning on either day 30 (n = 8), day 60 (n = 8), or day 100 (n = 8) of gestation (normal length of gestation is 184 days) until animals miscarried or were delivered abdominally on days 160 through 168 of gestation. A nonhuman primate model of panic disorder.

In this article we report on the reliable production of several features of panic attacks following subcutaneous administration of sodium lactate to nonhuman primates. Unrestrained monkeys were evaluated by an observer without knowledge of the subjects' treatment with either sodium lactate or a dextrose control solution. The lactate produced temporally circumscribed episodes of agitation, wariness, and motor responses, normally elicited under stressful or threatening conditions. In an initial pharmacological intervention, we found that pretreatment with imipramine blocked the response to lacta

A nonhuman primate model of Gilbert's syndrome.

A Bolivian population of squirrel monkeys, Saimiri sciureus, exhibits several features of Gilbert's syndrome in man, and is proposed as a nonhuman primate model of the condition. The Bolivian population was found to have higher fasting (40.6 +/- 2.7 microM; mean +/- S.E.) and postcibal (9.9 +/- 0.9 microM) plasma unconjugated bilirubin concentrations (p less than 0.001) than a closely related Brazilian population (fasting 5.5 +/- 0.7 microM); postcibal (2.4 +/- 0.7 microM). After intravenous administration of [3H]bilirubin as a tracer dose or at 3.4 mumoles per kg body weight, there was delaye

Ciliary process microvasculature of the primate eye.

Using methylmethacrylate microvascular luminal castings, we studied the three-dimensional angioarchitecture of the primate ciliary process with the scanning electron microscope. We found that the ciliary processes are served by vessels that radiate anteriorly and posteriorly from the "major arterial circle" of the iris. The anterior arterioles possess focal constrictions and supply the anterior and marginal aspects of the major ciliary processes as well as interprocess networks that connect contiguous processes. The posterior arterioles lack focal constrictions and supply the minor ciliary pro

A primate model of human corneal transplantation.

Human donor corneas were used for penetrating keratoplasty in one eye of each of 12 rhesus monkeys. In six animals, a 9.5-mm cornea was sutured into a 9.0-mm recipient bed by means of interrupted 10-0 nylon sutures. Six other animals received a 6.5-mm cornea in a 6.0-mm bed. Biomicroscopy, pachymetry, and specular microscopy revealed two distinct healing patterns. Of the six eyes receiving the smaller grafts, five showed prompt, stable clearing and thinning of the grafts with endothelial cell densities ranging from 850 to 1600 cells/mm2 Two of the six animals receiving larger grafts developed

Rupture of an experimentally induced aneurysm in a primate.

Cerebral aneurysms are extremely rare in all animals except man. We report the occurrence of a carotid-ophthalmic aneurysm in a cynomolgous monkey two months following accidental occlusion of the ipsilateral cervical internal carotid artery. This was documented angiographically. The aneurysm was deliberately ruptured by the insertion and withdrawal of fine needles. This produced a large subarachnoid hemorrhage visualized on CT scan. There was a progressive severe fall in cerebral blood flow and moderate vasospasm. The neurological status deteriorated on the tenth day and the animal was sacrifi

Anterior collateral circulation in the primate eye.

Sequential microdissection and scanning electron microscopy of whole primate ocular lumenal methyl methacrylate corrosion castings were performed to determine the anatomic basis of collateral arterial blood flow to the anterior uvea. Collateral anastomoses were observed at three sites: (1) the episclera where the anterior ciliary arteries interconnect at the limbus to form the episcleral circle; (2) the ciliary muscle where anastomoses between the perforating anterior ciliary and long posterior ciliary arteries comprise the intramuscular circle, the most extensive of the three collateral arter

Argentine hemorrhagic fever: a primate model.

Experimental Junin virus infection of a New World primate, Callithrix jacchus, was evaluated. The virus produced anorexia, loss of weight, thrombocytopenia, leukopenia, and hemorrhagic and neurological symptoms and terminated in death. Virus was recovered from urine, blood samples and all tissues taken at autopsy. These preliminary observations show that several aspects of the experimental disease in C. jacchus are quite similar to severe natural Argentine hemorrhagic fever of man.

Type D primate retroviruses: a review.

The prototype virus of the type D retroviruses is the Mason-Pfizer monkey virus (MPMV). MPMV was originally isolated from a breast carcinoma of a female rhesus monkey (an Old World monkey). MPMV is of obvious importance in that it is the only retrovirus thus far isolated from a mammary tumor of a primate and has been shown to have transforming potential for primate cells in vitro. Subsequent to the isolation of MPMV viruses morphologically and immunologically indistinguishable from MPMV have been isolated from normal placenta and lactating mammary glands of other rhesus monkeys in captivity.
