Oral propranolol in early stages of retinopathy of prematurity.

OBJECTIVE: To assess the effect of oral propranolol on the progression of early stages of retinopathy of prematurity (ROP) in very low birth weight (VLBW) infants. METHODS: We analyzed VLBW infants with ROP (stages 2-3, zones II-III). Newborns received oral propranolol (0.5 mg/kg/dose q8h), and were monitored throughout the treatment period for possible side effects. Propranolol was administered until regression of ROP. A historic control group of patients with equivalent ROP was used. We compared characteristics of both groups and the progression of retinopathy. RESULTS: Forty-seven newborn

Propranolol therapy for Tardive dyskinesia: A retrospective examination.

OBJECTIVE: To examine the tolerability and effectiveness of propranolol in treating tardive dyskinesia (TD). BACKGROUND: TD is a disabling, often irreversible, movement disorder that results from chronic therapy with dopamine receptor blocking drugs. There are no treatments currently approved for this disorder. Propranolol, a β-adrenergic blocker, has been reported to alleviate TD in case series and reports. METHODS: Retrospective database search of the Emory movement disorder clinic for TD patients treated with propranolol. All patients identified with at least one follow-up evaluation had

Propranolol impairs the closure of pressure ulcers in mice.

AIMS: β-Adrenoceptors modulate acute wound healing; however, few studies have shown the effects of β-adrenoceptor blockade on chronic wounds. Therefore, this study investigated the effect of β1-/β2-adrenoceptor blockade in wound healing of pressure ulcers. MAIN METHODS: Male mice were daily treated with propranolol (β1-/β2-adrenoceptor antagonist) until euthanasia. One day after the beginning of treatment, two cycles of ischemia-reperfusion by external application of two magnetic plates were performed in skin to induce pressure ulcer formation. KEY FINDINGS: Propranolol administration reduce

Corticosterone and propranolol's role on taste recognition memory.

Taste recognition is a robust procedure to study learning and memory processes, as well as the different stages involved in them, i.e. encoding, storage and recall. Considerable evidence indicates that adrenal hormones and the noradrenergic system play an important role in aversive and appetitive memory formation in rats and humans. The present experiments were designed to characterize the effects of immediate post training corticosterone (Experiment 1) and propranolol administration (Experiment 2 and 3) on taste recognition memory. Administration of a high dose of corticosterone (5mg/kg, sc)

Propranolol-induced relaxation in the rat basilar artery.

Propranolol is a non-selective beta-adrenergic receptor blocker used in the treatment of cardiovascular diseases and migraine prophylaxis. Although it has been shown that propranolol dilates the peripheral arteries of rat, its action in the central nervous system vasculature has not been investigated. In this study, the effects of propranolol in rat basilar artery were investigated. Basilar arteries from male Wistar rats were examined in a myograph system. The relaxant effects of propranolol, pindolol, atenolol, pizotifen and methysergide were examined in basilar arteries precontracted by sero

Use of propranolol to treat multicentric airway haemangioma.

OBJECTIVE: To report our experience of using propranolol to treat an infantile airway haemangioma. METHODS: A five-week-old girl presented with upper airway obstruction. Having started systemic steroids, concurrent propranolol therapy was commenced. Propranolol was given with close monitoring of the blood pressure, pulse and capillary glucose level. The dose of propranolol was gradually increased to 2 mg/kg total daily dose, with simultaneous reduction and withdrawal of steroids. RESULTS: Prior to propranolol treatment, laryngotracheobronchoscopy revealed an extensive haemangioma extending f

Hyperkalemia complicating propranolol treatment of an infantile hemangioma.

Propranolol treatment was recently reported to be successful for the management of severe infantile hemangioma. Known adverse effects of propranolol treatment include transient bradycardia, hypotension, hypoglycemia, and bronchospasm (in patients with underlying spastic respiratory illnesses), which led to a general recommendation to gradually increase propranolol dosage and closely monitor patients' hemodynamics at the onset of therapy. To date, no serious or unexpected adverse effects that required specific intervention have been reported. In this report, we describe the case of a 17-week-ol

Orthodeoxia and platypnea in liver cirrhosis: effects of propranolol.

Chronic liver disease is well known to be associated with pulmonary abnormalities. Hypoxemia, clubbing, cyanosis and hyperventilation are common. The hypoxemia in cirrhotic patients has several causes: diffuse shunts due to intrapulmonary arteriolar vasodilatation, impaired hypoxic vasoconstriction, impaired matching of ventilation to perfusion, pleural effusions and diaphragmatic dysfunction. Because of gravity, shifting of blood to the dilated precapillary beds of the lung bases results in an increased hypoxemic dyspnea when the patient is in the upright position, also known as orthodeoxia a

[Comparative study of theophylline and propranolol in essential tremor].

Propranolol having been used for decades was compared with low dose theophylline and placebo in the treatment of essential tremor. Ten de novo patients with essential tremor were treated for three months in a blind cross-over study. Tremor assessed by volumetric method decreased significantly after one week propranolol (80 mg/day) administration, while it was reduced by theophylline (150 mg/day) after two weeks. There was no difference between the two treatments after four weeks. During the propranolol period tension and dizziness were observed in some cases. According the results both drugs a

[The Stevens-Johnson syndrome from maprotiline and propranolol?].

A wide variety of agents have been associated with erythema multiforme, particularly drugs and infectious agents. We describe a typical Stevens-Johnson's syndrome developing 15 days after initiation of propranolol and maprotiline therapy. Other etiologies have been excluded by appropriate investigations and the responsibility of each drug is discussed.

A comparative trial of flunarizine and propranolol in the prevention of migraine.

The calcium-entry blocker flunarizine (Sibelium; Janssen) was compared with the beta-adrenoreceptor-blocking agent propranolol in the prophylaxis of migraine. Fifty-eight patients were entered into a double-blind 4-month treatment trial. Patients in whom beta-blockers were contraindicated were excluded from the trial. At the end of the trial 28 patients had received 10 mg flunarizine at night during the study, 29 patients had received 60 mg propranolol 3 times a day and 1 patient was withdrawn. Both groups responded well; and there was a 4-fold drop in frequency of attacks. There was no signif

Propranolol and depression: evidence from the antihypertensive trials.

The relationship between propranolol and depression is a subject of controversy. Numerous case reports suggest that propranolol can cause depression, but two small prospective trials have failed to confirm this. The contemporary psychiatric literature is divided as to whether propranolol can cause depression. This study addresses this issue by re-analyzing side effect data from clinical trials of propranolol as an antihypertensive agent. A literature review was carried out and the data were analyzed using meta-analytic statistical techniques. Propranolol was found to cause depression as a side

[Hypercalcemia and hyperphosphatemia in thyrotoxoicosis and the therapeutic effect of propranolol].

Measured levels of serum calcium, phosphate, alkaline phosphatase, and urinary hydroxyproline were measured and calcium-phosphate product was calculated in 20 hyperthyroid patients and 20 normal controls. Eleven of the patients took propranolol 160 mg per day for 28 days. We found that the serum level of calcium was higher than that of normal controls. The incidence of hypercalcaemia in hyperthyroid patients was 10%. The serum level of phosphate and the calcium-phosphate product increased (P less than 0.01). Elevation of alkaline phosphatase and bone alkaline phosphatase were also observed (P

Supraventricular tachycardia treated with continuous infusions of propranolol.

Because oral therapy is often contraindicated in hospitalized patients we assessed the safety and efficacy of continuous intravenous propranolol infusions in nine patients with refractory supraventricular tachycardia. Standard pharmacokinetic formulas predicted a loading dose (52.2 +/- 38.3 micrograms/kg), steady-state plasma concentration, and the initial maintenance dose (16.1 +/- 16.2 micrograms/kg/hr; range 6.1 to 56.0 micrograms/kg/hr) to control heart rate. Subsequent maintenance doses (3.9 to 74.9 micrograms/kg/hr) were determined by clinical response. Heart rate decreased from 146 +/-

Psychiatric and neuropsychological response to propranolol in Graves' disease.

We describe the endocrine, psychiatric, and neuropsychological assessments of 10 untreated, newly diagnosed Graves' disease subjects who were studied longitudinally at three stages: hyperthyroid (stage 1), after 2 weeks of propranolol treatment (stage 2), and after 6 months of antithyroid treatment (stage 3). Major depression, generalized anxiety disorder, and hypomania were diagnosed at stage 1. Elevations on psychiatric symptom rating scales and in motor activity monitoring at stage 1 were significantly decreased at stage 2 and again at stage 3. Psychiatric improvements paralleled improvemen

Antihypertensive therapy with nitrendipine: comparison with hydrochlorothiazide and propranolol.

The calcium channel blockers are being used increasingly to treat essential hypertension. Nitrendipine is a second-generation dihydropyridine derivative that, though not yet approved in the United States, has been extensively evaluated both there and abroad. This report concerns two multicenter trials in which nitrendipine was compared to hydrochlorothiazide and propranolol. In the first study, which included 305 patients over 50 years of age with off-treatment sitting diastolic blood pressures of 95-114 mm Hg, nitrendipine 10 mg b.i.d. was compared to hydrochlorothiazide 50 mg q.d. The mean b

Therapeutic use of propranolol for intermittent explosive disorder.

Intermittent explosive disorder is a syndrome characterized by episodic sudden outbursts of verbal abuse and physical violence in response to minor provocations. Propranolol has been proposed as a promising treatment for this cause of violent behavior. Of eight Mayo Clinic patients with intermittent explosive disorder who had been treated with propranolol between 1983 and 1985, five had substantial diminution or complete remission of symptoms. This response confirms the previously published reports of the effectiveness of propranolol in the treatment of intermittent explosive disorder.
