BACKGROUND: Virulent Newcastle disease virus (NDV, avian Avulavirus-1, APMV-1) induces a highly contagious and lethal systemic disease in gallinaceous poultry. APMV-1 antibody detection is used for surveillance and to control vaccination, but is hampered by cross-reactivity to other subtypes of avian Avulaviruses. Data are lacking concerning the applicability of NDV V proteins as differential diagnostic marker to distinguish vaccinated from virus-infected birds (DIVA strategy).
METHODS: Full length and C-terminally truncated nucleocapsid (NP) protein, and the unique C-terminal regions of the

The oncoprotein Bcr-Abl stimulates prosurvival pathways and suppresses apoptosis from its exclusively cytoplasmic locale, but when targeted to the mitochondrial compartment of leukemia cells, Bcr-Abl was potently cytotoxic. Therefore, we designed a protein construct to act as a mitochondrial chaperone to move Bcr-Abl to the mitochondria. The chaperone (i.e., the 43.6 kDa intracellular cryptic escort (iCE)) contains an EGFP tag and two previously characterized motifs: (1) an optimized Bcr-Abl binding motif that interacts with the coiled-coil domain of Bcr (ccmut3; 72 residues), and (2) a crypti

Real-time fluorescence imaging of viral proteins in living cells provides a valuable means to study virus-host interactions. The challenge of generating replication-competent fluorescent influenza A virus is that the segmented genome does not allow fusion of a fluorescent protein gene to any viral gene. Here, we introduced the tetracysteine (TC) biarsenical labeling system into influenza virus in order to fluorescently label viral protein in the virus life cycle. We generated infectious influenza A viruses bearing a small TC tag (CCPGCC) in the loop/linker regions of the NS1 proteins. In the b

Meningococcal factor H-binding protein (fHbp) is a promising antigen that is part of two vaccines in clinical development. The protein specifically binds human complement factor H (fH), which downregulates complement activation on the bacterial surface and enables the organism to evade host defenses. In humans, the vaccine antigen forms a complex with fH, which may affect anti-fHbp antibody repertoire and decrease serum bactericidal activity by covering important fHbp epitopes. In a recent study, fHbp residues in contact with fH were identified from a crystal structure. Two fHbp glutamate resi

Adoptive T cell therapies have markedly improved outcomes in hematologic malignancies but their efficacy in solid tumors can be diminished by a hostile tumor microenvironment that impedes sustained therapeutic responses. Beyond challenges such as limited trafficking and antigen heterogeneity, engineered T cells face suppressive myeloid and stromal populations, inhibitory checkpoint ligand interactions, and metabolically hostile niches that collectively diminish effector function and persistence. To overcome these barriers, a new generation of fusion protein-based costimulatory strategies has e

Escalating complexity in water pollution has catalyzed a shift toward chain-integrated management, encompassing source monitoring, targeted removal, and end-point purification. Protein nanofibrils (PNFs), self-assembled from abundant proteins, are ideal candidates for these challenges due to their rich functional groups, mild synthesis, and tunability. This review examines PNFs' potential in chain-integrated water treatment technologies. First, the evolution of PNFs reveals their advantages over conventional nanomaterials such as cellulose nanofibrils and carbon nanotubes, including simpler pr

Background/Objectives: African Swine Fever (ASF) represents one of the most serious threats to animal health and global food security. The causative agent of ASF is the African swine fever virus (ASFV), a DNA virus belonging to the Asfarviridae family. Here, we describe ex vivo results for an original anti-ASFV vaccine approach based on the cellular immune response induced by extracellular vesicles (EVs) engineered to express four ASFV proteins. EV engineering was achieved by expressing a DNA vector encoding a biologically inactive HIV-1 Nef protein (Nefmut), which exhibits unusually high effi

The accurate identification of multiple proteins and effective eradication of bacteria hold paramount importance for clinical diagnosis and oral disease prophylaxis. Although molybdenum disulfide (MoS2) nanomaterials possess exceptional physicochemical properties that offer distinct advantages in bioanalysis and antibacterial fields, their low photogenerated carrier mobility and limited active site density have restricted their widespread application. Herein, we report the synthesis of Fe-doped MoS2 (Fe-MoS2) nanomaterials via a facile one-step solvothermal method. The incorporation of Fe pres

Rhabdomyolysis-induced acute kidney injury (RM-AKI) is mediated primarily by myoglobin (Mb) toxicity, yet effective targeted therapies remain unavailable. Through computational structural modeling, we uncovered that the haptoglobin β-subunit (Hpβ) can bind Mb, forming a structurally stable complex. We then engineered a 55 kDa recombinant GST-Hpβ fusion protein and demonstrated that this engineered construct exhibits robust binding affinity for free Mb (17.8 kDa), thereby generating a stable 72.8 kDa GST-Hpβ-Mb complex. Significantly, this complex surpasses the molecular size threshold of the g

Targeting amino acid metabolism has emerged as a promising strategy for cancer therapy. To investigate the feasibility of tryptophan restriction as a dietary intervention strategy to suppress tumor growth in vivo, this study developed a novel tryptophan-depleted protein (TDP) through protein engineering combined with the Pichia pastoris expression system, for dietary intervention to suppress tumor growth. Based on the human serum albumin (HSA) scaffold, we designed the TDP by mutating tryptophan at position 238 to phenylalanine (Phe), and successfully expressed it in P. pastoris. Following pur

In this study, a novel Immunoglobulin G (IgG) binding, Programmed Cell Death-Ligand 1 (PD-L1) targeting affibody, IgBD-ZPD-L1, was constructed by genetically fusing the PD-L1-specific affibody ZM1 with the IgG-binding domain (IgBD). IgBD-ZPD-L1 could simultaneously bind to IgG, PD-L1 receptor protein, and PD-L1-positive tumor cells. Furthermore, the fusion of IgBD significantly prolonged the in vivo circulation half-life of IgBD-ZPD-L1 by binding to endogenous IgG, enhancing tumor accumulation and immunotherapeutic effects. Subsequently, anti-angiogenic/immunotherapeutic and dual-targeted comb

Conventional osteogenic platforms utilize active growth factors to repair bone defects that are extensive in size, but they can adversely affect patient health. Here, an unconventional osteogenic platform is reported that functions by promoting capture of inactive osteogenic growth factor molecules to the site of cell growth for subsequent integrin-mediated activation, using a recombinant fragment of latent transforming growth factor beta-binding protein-1 (rLTBP1). It is shown that rLTBP1 binds to the growth-factor- and integrin-binding domains of fibronectin on poly(ethyl acrylate) surfaces,

Periodontitis is an oral disease caused by bacterial infection that has stages according to the severity of tissue destruction. The advanced stage of periodontitis presents irreversible destruction of soft and hard tissues, which finally results in loss of teeth. When conventional treatment modalities show limited results, tissue regeneration therapy is required in patients with advanced periodontitis. In the present study, we aimed to evaluate the effect of bone marrow-derived mesenchymal stem cells (BM-MSCs) delivering bone morphogenetic protein 7 (BMP7) on tissue regeneration in a periodont

The rapid emergence and spread of escaping mutations of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has significantly challenged our efforts in fighting against the COVID-19 pandemic. A broadly neutralizing reagent against these concerning variants is thus highly desirable for the prophylactic and therapeutic treatments of SARS-CoV-2 infection. We herein report a covalent engineering strategy on protein minibinders for potent neutralization of the escaping variants such as B.1.617.2 (Delta), B.1.617.1 (Kappa), and B.1.1.529 (Omicron) through in situ cross-linking with the spik

αB-Crystallin is a protein chaperone with anti-apoptotic and anti-inflammatory activity that is apically secreted in exosomes by polarized human retinal pigment epithelium. A 20 amino acid mini-peptide derived from residues 73-92 of αB-crystallin protects human retinal pigment epithelial (RPE) cells from oxidative stress, a process involved in the progression of age-related macular degeneration (AMD). Unfortunately, due to its small size, its development as a therapeutic requires a robust controlled release system. To achieve this goal, the αB-crystallin peptide was re-engineered into a protei

BACKGROUND & AIMS: The optimal protein requirement in critically ill patients with acute kidney injury (AKI) remains undefined. Evidence supporting individualized protein prescription guided by protein catabolic rate (PCR) is currently limited. This study aims to estimate protein requirements using PCR, identify its determinants, and evaluate the association between protein intake relative to PCR and clinical outcomes.
METHODS: This study is a post hoc analysis of the open-label, randomized controlled ENGINE trial, which enrolled 80 critically ill patients with AKI. PCR was assessed in all pa

Heterologous protein secretion in filamentous fungi is often constrained by limitations in signal peptide recognition and intracellular trafficking. Aspergillus oryzae, a food-grade industrial fungus, has a robust native secretory system. However, its capacity for recombinant protein secretion remains suboptimal. Here, we developed a two-step, carrier-free engineering strategy to enhance protein secretion in A. oryzae. We identified endogenous signal peptides among highly secreted proteins using a green fluorescent protein (GFP) reporter. The oryzin signal peptide SPAoalp1 increased GFP secret

Mesenchymal stem cell (MSC) therapies hold immense promise for regenerative medicine, yet their clinical translation is hindered by inefficient delivery and poor homing at injury sites. Here, we exploited a modular protein-nucleic acid nanodevice for non-genetic engineering of MSCs, which enables the cells to dock onto pathological collagen-rich tissues. The protein-nucleic acid nanodevice integrates two key modules: a multi-functional fusion protein for collagen binding and clickable covalent conjugation between protein and DNA, and a MSC-specific binding DNA nanostructure. A common bottleneck challenge for many therapeutic proteins lies in their short plasma half-lives, which often makes the treatment far less compliant or even disables achieving sufficient therapeutic efficacy. To address this problem, we introduce a novel drug delivery strategy based on the genetic fusion of an albumin binding domain (ABD) and an anti-neonatal Fc receptor (FcRn) affibody (AFF) to therapeutic proteins. This ABD-AFF fusion strategy can provide a synergistic effect on extending the plasma residence time by, on one hand, preventing the rapid glomerular filtration via ABD-mediated
