AMP-activated protein kinase and metabolic regulation in cold-hardy insects.

Winter survival for many insects depends on cold hardiness adaptations as well as entry into a hypometabolic diapause state that minimizes energy expenditure. We investigated whether AMP-activated protein kinase (AMPK) could be involved in this adaptation in larvae of two cold-hardy insects, Eurosta solidaginis that is freeze tolerant and Epiblema scudderiana that uses a freeze avoidance strategy. AMPK activity was almost 2-fold higher in winter larvae (February) compared with animals collected in September. Immunoblotting revealed that phosphorylation of AMPK in the activation loop and phosph

Metabolic circuitry of NADH buffering reveals a vulnerability in redox-adaptive cancer cells.

Mitochondrial dysfunction elevates cellular NADH/NAD+ ratios, inducing reductive stress that impairs biosynthesis and cell viability. However, the mechanisms by which cells buffer excess NADH to maintain redox homeostasis remain unclear. In this study, we identify a mitochondrial-cytosolic metabolic circuitry involving pyruvate carboxylase (PC), malate dehydrogenases (MDH1/2), and malic enzyme 1 (ME1) that mitigates NADH overload by coupling anaplerotic flux with NADH oxidation. Under reductive stress induced by electron transport chain (ETC) dysfunction, oxaloacetate derived from PC is conver

Metagenomic and Metabolomic Insights into Volatile Flavor Changes and Microbial Community Shifts in Physalis pubescens L. Fermentation by Lactiplantibacillus plantarum.

Physalis pubescens L. is a seasonal fruit with high nutritional value but a short shelf life that limits its processing and utilization. This study integrated metagenomics and metabolomics to investigate the comparative effects of Lactiplantibacillus plantarum fermentation on volatile flavor metabolites and microbial community composition of P. pubescens by comparing initial (0 h) and post-fermentation (24 h) states. After 24 h of fermentation, 1316 volatile compounds were putatively identified by GC-MS, with 592 metabolites significantly changed and 501 upregulated and 91 downregulated. Key f

Dietary berberine can ameliorate glucose metabolism disorder of Megalobrama amblycephala exposed to a high-carbohydrate diet.

Blunt snout bream (Megalobrama amblycephala) were randomly assigned into three diets: normal-carbohydrate diet (NCD, 30% carbohydrate, w/w), high-carbohydrate diet (HCD, 43% carbohydrate), and HCB (HCD supplemented with 50 mg/kg berberine (BBR)). After 10 weeks' feeding trial, the results showed that higher levels of plasma glucose, triglyceride, and total cholesterol were observed in HCD-fed fish than in NCD-fed fish, while HCB feeding significantly ameliorated this effect. Moreover, HCB feeding remarkably reversed HCD-induced hepatic glycogen and lipid contents. In insulin signaling, BBR inc

Kaempferol ameliorates hyperglycemia through suppressing hepatic gluconeogenesis and enhancing hepatic insulin sensitivity in diet-induced obese mice.

Obesity-associated insulin resistance (IR) is a major risk factor for developing type 2 diabetes and an array of other metabolic disorders. In particular, hepatic IR contributes to the increase in hepatic glucose production and consequently the development of fasting hyperglycemia. In this study, we explored whether kaempferol, a flavonoid isolated from Gink go biloba, is able to regulate hepatic gluconeogenesis and blood glucose homeostasis in high-fat diet-fed obese mice and further explored the underlying mechanism by which it elicits such effects. Oral administration of kaempferol (50 mg/k

Indoleamine 2,3-dioxygenase depletes tryptophan, activates general control non-derepressible 2 kinase and down-regulates key enzymes involved in fatty acid synthesis in primary human CD4+ T cells.

Indoleamine 2,3-dioxygenase (IDO) is expressed in antigen-presenting cells and exerts immunosuppressive effects on CD4(+) T cells. One mechanism is through the inhibition of aerobic glycolysis. Another prerequisite for T-cell proliferation and differentiation into effector cells is increased fatty acid (FA) synthesis. The effect of IDO on enzymes involved in FA synthesis was evaluated in primary human cells both in mixed lymphocyte reactions in the presence or not of the IDO inhibitor 1-dl-methyl-tryptophan, and in stimulated CD4(+) T cells in the presence or not of the general control non-der

Metabolic acclimation of rice cultivars via cold stress-induced carbohydrate accumulation.

This study was conducted to comparatively investigate the accumulation of carbohydrates and transcript levels of genes encoding relevant enzymes in two rice (Oryza sativa L.) cultivars 'Hashemi' (cold-sensitive) and 'Kouhsar' (cold-tolerant) at 1, 3 and 6 days after cold stress (DAS) (4℃). The sinusoidal pattern of H2O2 and MDA in Hashemi under cold, compared with the steadily decreasing levels in Kouhsar indicates differing oxidative damage, reflected in 2.28- and 2.39-fold higher contents, respectively, at 6 DAS. Kouhsar had higher sucrose than Hashemi on all stress days, with a peak increas

Nitrogen Sources Reprogram Carbon and Nitrogen Metabolism to Promote Andrographolide Biosynthesis in Andrographis paniculata (Burm.f.) Nees Seedlings.

Carbon (C) and nitrogen (N) metabolisms participate in N source-regulated secondary metabolism in medicinal plants, but the specific mechanisms involved remain to be investigated. By using nitrate (NN), ammonium (AN), urea (UN), and glycine (GN), respectively, as sole N sources, we found that N sources remarkably affected the contents of diterpenoid lactone components along with C and N metabolisms reprograming in Andrographis paniculata, as compared to NN, the other three N sources raised the levels of 14-deoxyandrographolide, andrographolide, dehydroandrographolide (except UN), and neoandrog

Polystyrene nanoplastics induce lipid metabolism disorder and alter fatty acid composition in the hepatopancreas of Pacific whiteleg shrimp (Litopenaeus vannamei).

The impact of nanoplastics (NPs) on environmental pollution and aquatic organisms has gradually attracted attention, but there are relatively few reports of the effects of NPs on the lipid metabolism of crustaceans. In this study, we exposed Pacific whiteleg shrimp (Litopenaeus vannamei) to different concentrations of polystyrene NPs (0, 0.1, 1, 5, and 10 mg/L) for 28 days. We then evaluated the effects of NP exposure on metabolite content, histology, lipid metabolism-related enzyme activity, and gene expression. Our results showed that with increasing NPs concentrations and exposure time, (1)

Structural Insight of KSIII (β-Ketoacyl-ACP Synthase)-like Acyltransferase ChlB3 in the Biosynthesis of Chlorothricin.

Chlorothricin (CHL) belongs to a spirotetronate antibiotic family produced by Streptomyces antibioticus that inhibits pyruvate carboxylase and malate dehydrogenase. For the biosynthesis of CHL, ChlB3 plays a crucial role by introducing the 6-methylsalicylic acid (6MSA) moiety to ChlB2, an acyl carrier protein (ACP). However, the structural insight and catalytic mechanism of ChlB3 was unclear. In the current study, the crystal structure of ChlB3 was solved at 3.1 Å-resolution and a catalytic mechanism was proposed on the basis of conserved residues of structurally related enzymes. ChlB3 is a di

Genipin dismantles WSSV's metabolic support system by targeting PC and PEPCK in shrimp.

White spot syndrome virus (WSSV) is the most devastating pathogen in shrimp aquaculture, causing massive economic losses worldwide. Current control strategies are limited due to the lack of adaptive immunity in crustaceans. In this study, the antiviral mechanism of genipin, a natural compound with known anti-WSSV activity, was investigated using a dual proteomic approach in Pacific whiteleg shrimp (Litopenaeus vannamei). In vitro fluorescence analysis confirmed that genipin specifically suppressed the expression of WSSV structural proteins. Through a four-group design (control, WSSV-infected,

Short-term actions of arsenic trioxide on metabolic fluxes in the isolated perfused rat liver.

Arsenic trioxide has recently been approved for the treatment of acute promyelocytic leukemia, but it is also a highly toxic compound. The purpose of the present work was to obtain a general view about the acute effects of arsenic trioxide on liver metabolism using the isolated perfused rat liver, a system that preserves microcirculation and cell polarity. Hepatic lactate and alanine gluconeogenesis were inhibited by arsenic trioxide with IC50 values of 21.7 and 21.4 µM, respectively. Oxygen uptake was inhibited only at concentrations above the IC50 value for gluconeogenesis inhibition. In add

Zonated iron deposition in the periportal zone of the liver is associated with selectively enhanced lipid synthesis.

BACKGROUND AND AIMS: Disorders in liver lipid metabolism have been implicated in a range of metabolic conditions, including fatty liver and liver cancer. Altered lipid distribution within the liver, shifting from the pericentral to the periportal zone under pathological circumstances, has been observed; however, the underlying mechanism remains elusive. Iron, an essential metal, exhibits a zonal distribution in the liver similar to that of lipids. Nevertheless, the precise relationship between iron and lipid distribution, especially in the pericentral and periportal zones, remains poorly under

Fat Induces Glucose Metabolism in Nontransformed Liver Cells and Promotes Liver Tumorigenesis.

Hepatic fat accumulation is associated with diabetes and hepatocellular carcinoma (HCC). Here, we characterize the metabolic response that high-fat availability elicits in livers before disease development. After a short term on a high-fat diet (HFD), otherwise healthy mice showed elevated hepatic glucose uptake and increased glucose contribution to serine and pyruvate carboxylase activity compared with control diet (CD) mice. This glucose phenotype occurred independently from transcriptional or proteomic programming, which identifies increased peroxisomal and lipid metabolism pathways. HFD-fe

Mold Exposure and Mitochondrial Antibodies.

CONTEXT: Some studies have reported that airborne levels of mold above 150-1000 spores per cubic meter are sufficient to cause a wide range of human health problems. In trying to understand the mechanism of injury to the mitochondria that trigger an autoimmune response, researchers have learned that pyruvate carboxylase is a major site of antigenicity for antimitochondrial antibodies (AMA). Antibodies to mitochondria are found in over 90% of patients with primary biliary cirrhosis and are occasionally found in patients with other autoimmune diseases. OBJECTIVES: This study intended to examine

The Mechanism by Which Amentoflavone Improves Insulin Resistance in HepG2 Cells.

BACKGROUND: The aim of this study was to explore the mechanism by which amentoflavone (AME) improves insulin resistance in a human hepatocellular liver carcinoma cell line (HepG2). METHODS: A model of insulin resistant cells was established in HepG2 by treatment with high glucose and insulin. The glucose oxidase method was used to detect the glucose consumption in each group. To determine the mechanism by which AME improves insulin resistance in HepG2 cells, enzyme-linked immunosorbent assay (ELISA) and western blotting were used to detect the expression of phosphatidyl inositol 3-kinase (PI3

Sublethal indoxacarb exposure alters pheromone production and ovarian development in the yellow peach moth, Conogethes punctiferalis.

The Conogethes punctiferalis, a major pest of peach and other fruit trees, causes significant damage by boring into fruits during its larval stage. Indoxacarb, a sodium channel blocker insecticide, has been widely applied in agriculture and horticulture for pest control, particularly against larval pests. However, its effects on adult insects remain largely unexplored. Present study employed C. punctiferalis as model to investigate the toxic effects of indoxacarb on adult moths. The results showed that sublethal exposure to indoxacarb significantly reduced the release of sex pheromones, reduce

Lack of liver glycogen causes hepatic insulin resistance and steatosis in mice.

Disruption of the Gys2 gene encoding the liver isoform of glycogen synthase generates a mouse strain (LGSKO) that almost completely lacks hepatic glycogen, has impaired glucose disposal, and is pre-disposed to entering the fasted state. This study investigated how the lack of liver glycogen increases fat accumulation and the development of liver insulin resistance. Insulin signaling in LGSKO mice was reduced in liver, but not muscle, suggesting an organ-specific defect. Phosphorylation of components of the hepatic insulin-signaling pathway, namely IRS1, Akt, and GSK3, was decreased in LGSKO mi

Mechanism of oestrogen and progesterone effects on lipid and carbohydrate metabolism: alteration in the insulin: glucagon molar ratio and hepatic enzyme activity.

As in women receiving oestrogens the administration of 17beta-oestradiol to ovariectomized female rats caused a rise in fasting plasma triglycerides and a fall in plasma glucose. Progesterone, on the other hand, had no significant effects. In the oestradiol treated rats, the portal vein basal insulin levels were slightly reduced. Oestradiol, however, had a marked suppressive effect on the alpha cells of the pancreas resulting in a greater reduction in basal glucagon and impaired glucagon response to alanine infusions. The relative insulin to glucagon (I/G) molar concentration ratio in portal v
