Simultaneous Detection of HEV, HAstV and SaV in Bivalve Shellfish Using a Novel Real-Time RT-qPCR Method.

Hepatitis E virus (HEV), human astrovirus (HAstV), and sapovirus (SaV) are pathogens associated with foodborne disease outbreaks. We developed a rapid and sensitive quadruple real-time fluorescence quantitative PCR (RT-qPCR) method for the simultaneous detection of HEV, HAstV, and SaV, using MS2 phage as a process control virus. We optimized the experimental parameters, detection limits for HEV, HAstV, SaV, and MS2 RNA were 103 copies/μL, 103 copies/μL, 102 copies/μL, and 103 copies/μL, respectively, with intra-method and inter-method coefficients of variation below 3.0%, indicating good repro

Implications of gut microbiota-mediated epigenetic modifications in intestinal diseases.

Intestinal diseases are highly prevalent, affecting millions worldwide and significantly contributing to global morbidity. The treatment of complex disorders, such as inflammatory bowel disease (IBD) and colorectal cancer (CRC), remains challenging due to multifactorial etiologies, diverse patient responses, and the limitations of current therapeutic strategies. Although the gut microbiota clearly plays a role in regulating the onset of intestinal diseases, few studies have explored the epigenetic factors by which the microbiota contributes to disease development. Here, the latest insights int

First report of whole genome sequence of septicemic Pasteurella multocida serovar B:2 'Soron' strain isolated from swine.

Pig pasteurellosis, caused by Pasteurella multocida, is an acute infection that also has economic implications for pig farmers. We report the complete genome sequence of a P. multocida, serovar B:2 'Soron' strain isolated from the blood of a pig that had died of pasteurellosis in India. The isolate was not found to be haemorrhagic septicaemia (HS) specific B:2 by the PCR assay. The genome of 'Soron' strain is a single circular chromosome of 2,272,124 base pairs in length and contains 2014 predicted coding regions, 4 ribosomal RNA operons, and 52 tRNAs. It has 1812 protein-coding genes that wer

The Borrelia burgdorferi RelA/SpoT Homolog and Stringent Response Regulate Survival in the Tick Vector and Global Gene Expression during Starvation.

As the Lyme disease bacterium Borrelia burgdorferi traverses its enzootic cycle, alternating between a tick vector and a vertebrate host, the spirochete must adapt and persist in the tick midgut under prolonged nutrient stress between blood meals. In this study, we examined the role of the stringent response in tick persistence and in regulation of gene expression during nutrient limitation. Nutritionally starving B. burgdorferi in vitro increased the levels of guanosine tetraphosphate (ppGpp) and guanosine pentaphosphate (pppGpp), collectively referred to as (p)ppGpp, products of the bifuncti

Modular toolkit to facilitate molecular manipulations in mycobacteria.

Gene expression in mycobacteria relies on both integrative and episomal Escherichia coli-mycobacterium shuttle vectors. However, existing vectors have limited flexibility for adding epitope tags, selectable markers, and fluorescent reporters. To overcome this, we created a modular toolkit consisting of 32 integrative and eight episomal E. coli-mycobacterium shuttle vectors. Each vector features N- or C-terminal tags: SBP-HA-S, 3×-FLAG, GFP, and mCherry. The integrative vectors contain four different phage systems (L5, Giles, MS6, and Tweety), each with a distinct antibiotic resistance marker,

RmTIL, a cysteine-rich protein as a putative microbiota modulator from Rhipicephalus microplus tick.

Rhipicephalus microplus, also known as the "cattle tick", presents worldwide distribution in tropical and subtropical regions and causes significant economic losses in livestock activity. At the present work, an immunogenic cysteine-rich protein produced in salivary glands was identified through an association of phage display and third generation sequencing (TGS) high-throughput techniques. This protein presenting a trypsin inhibitor-like (TIL) domain from R. microplus, named here as RmTIL, was expressed as a recombinant protein and submitted to a functional characterization as a serine prote

An information-dynamics model of protein-templated DNA synthesis: From qualitative observations to quantitative predictions.

Dr.Pujuan Deng et al. (2026) showed that the bacterial anti-phage reverse transcriptase Drt3b synthesizes strictly alternating poly(AC) DNA without any nucleic acid template. We recast this mechanism within an information-dynamics framework in which the active pocket is a two-state automaton in constraint space and the free-nucleotide pool is real space, coupled by Boltzmann-weighted stochastic selection. The framework yields a closed-form master curve for the alternation-defect rate, from which the wild-type strict alternation and the partial fidelity loss of E26A follow directly. The E26A en

Antimicrobial resistance in plant-associated microbial communities: mechanisms, ecology, and one health implications.

Antimicrobial resistance (AMR) has emerged as a major global challenge threatening human, animal, plant and the environment health. Although AMR has been extensively studied in clinical and veterinary sectors, its emergence and dissemination in plant-based agricultural systems remains comparatively underexplored despite of the widespread use of antibiotics, fungicides and other antimicrobial compounds in crop production. These practices exerts selective pressure on plant associated microbial communities, promoting the evolution and dissemination of antimicrobial resistance genes (ARGs) across

Molecular mechanism of the type 2 defense-associated reverse transcriptase.

Defense-associated reverse transcriptase (DRT) systems play a crucial role in prokaryotic defense mechanisms against phage infections. Among the DRT family, DRT2, DRT3, and DRT9 systems employ protein-noncoding RNA (ncRNA) co-regulatory mechanisms to execute defense functions. Here, we focus on the DRT2 system from Klebsiella pneumoniae, which consists of a reverse transcriptase (RT) and an essential ncRNA component. Using biochemical and structural approaches, we determine the structure of the DRT2 system and reveal detailed interaction modes between the DRT2-RT protein and the ncRNA, especia

Predicting clinical outcome of Escherichia coli O157:H7 infections using explainable machine learning.

Shiga toxin-producing Escherichia coli (STEC) O157:H7 is a globally dispersed zoonotic pathogen capable of causing severe disease outcomes, including bloody diarrhoea and haemolytic uraemic syndrome. While variations in Shiga toxin subtype are well-recognized drivers of disease severity, many unexplained differences remain among strains carrying the same toxin profile. We applied explainable machine learning (ML) approaches - Random Forest and Extreme Gradient Boosting - to whole-genome sequencing data from 1,030 STEC O157:H7 isolates to predict patient clinical outcomes, using data collected o

[Construction of a cDNA library for Sparganum mansoni and screening of diagnostic antigen cadidates].

OBJECTIVE: To construct a cDNA library of Sparganum mansoni and immunoscreen antigen candidates for immunodiagnosis of sparganosis mansoni. METHODS: Total RNA was extracted from S. mansoni, and reversely transcribed into cDNA, which was ligated into the phage vector. These recombinant vectors were packaged in vitro to construct the SMART cDNA library of S. mansoni. Then, the cDNA library was immunoscreened with sera from patients with sparganosis mansoni to yield positive clones. The inserted fragments of positive clones were sequenced and subjected to homology analyses, and the structure and

Performance of UV disinfection and the microbial quality of greywater effluent along a reuse system for toilet flushing.

This paper examines the microbial quality of treated RBC (Rotating Biological Contactor) and MBR (Membrane Bioreactor) light greywater along a continuous pilot-scale reuse system for toilet flushing, quantifies the efficiency of UV disinfection unit, and evaluates the regrowth potential of selected microorganisms along the system. The UV disinfection unit was found to be very efficient in reducing faecal coliforms and Staphylococcus aureus. On the other hand, its efficiency of inactivation of HPC (Heterotrophic Plate Count) and Pseudomonas aeruginosa was lower. Some regrowth occurred in the re

Efficient induction of mucosal and systemic immune responses by virus-like particles administered intranasally: implications for vaccine design.

Intranasal (i.n.) immunization aims to induce local as well as systemic immune responses. In the present study, we assessed a vaccine platform based on virus-like particles (VLP) derived from the RNA phage Qbeta for i.n. immunization. We found that both i.n. and subcutaneous (s.c.) administration of Qbeta-VLP elicited strong and comparable specific IgG responses in serum and lung. Surprisingly, both routes also induced high levels of specific IgA in serum. In contrast, only i.n. administration of Qbeta-VLP resulted in local IgA production in the lung. Efficient induction of B cell responses by

Molecular cloning of an echinococcal microtrichal antigen immunoreactive in Echinococcus multilocularis disease.

A cDNA expression library of the larval stage of the cestode worm Echinococcus multilocularis has been established in the phage lambda ZAPII system. By immunoscreening with pooled sera from patients with alveolar echinococcosis an immunoreactive clone, termed pEM13, was isolated. EM13 was expressed using the expression vector pGEX-3X, resulting in the synthesis of a glutathione S-transferase fusion protein. 82% of the sera from 28 patients suffering from E. multilocularis disease had antibodies against EM13, whereas none of the 55 sera obtained from Echinococcus granulosus-infected patients an

Phi29 polymerase based random amplification of viral RNA as an alternative to random RT-PCR.

BACKGROUND: Phi29 polymerase based amplification methods provides amplified DNA with minimal changes in sequence and relative abundance for many biomedical applications. RNA virus detection using microarrays, however, can present a challenge because phi29 DNA polymerase cannot amplify RNA nor small cDNA fragments (<2000 bases) obtained by reverse transcription of certain viral RNA genomes. Therefore, ligation of cDNA fragments is necessary prior phi29 polymerase based amplification. We adapted the QuantiTect Whole Transcriptome Kit (Qiagen) to our purposes and designated the method as Whole Tr

Peptide targeting of lysophosphatidylinositol-sensing GPR55 for osteoclastogenesis tuning.

BACKGROUND: The G-protein-coupled receptor GPR55 has been implicated in multiple biological activities, which has fuelled interest in its functional targeting. Its controversial pharmacology and often species-dependent regulation have impacted upon the potential translation of preclinical data involving GPR55. RESULTS: With the aim to identify novel GPR55 regulators, we have investigated lysophosphatidylinositol (LPI)-induced GPR55-mediated signal transduction. The expression system for wild-type and mutated GPR55 was HeLa cells silenced for their endogenous receptor by stable expression of a

Novel Strategies to Profile SARS-CoV-2 and Human Lung Proteome: Inflammatory Pathways in the Spotlight.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of COVID-19, has caused widespread morbidity and mortality worldwide. SARS-CoV-2 infection triggers innate and adaptive immune responses, but excessive cytokine release can drive hyperinflammation, acute respiratory distress syndrome and poor clinical outcomes. Although serological and molecular assays, such as ELISA and RT-qPCR, remain central to COVID-19 diagnostics, they have limited capacity to reveal host-pathogen interactions at the tissue level. Therefore, profiling the human lung proteome offers a powerfu

A systematic review and critical appraisal of metagenomic and culture studies in hidradenitis suppurativa.

Hidradenitis suppurativa (HS), also known as acne inversa, is a chronic inflammatory skin disease with still largely unknown pathogenesis. While infectious organisms have been identified in lesions of the disease since the 1980s, questions remain over the role that bacteria and microbiome play. Recent studies using 16S ribosomal RNA gene sequencing and larger culture-based studies have begun to paint a clearer picture of the microbial world of HS. With this systematic review, we summarize all the work that has been done to date in HS bacteriology, analyse potential pitfalls and limitations of
