Effects of amisulpride and aripiprazole on progressive-ratio schedule performance: comparison with clozapine and haloperidol.

Clozapine and some other atypical antipsychotics (e.g. quetiapine, olanzapine) have been found to exert a characteristic profile of action on operant behaviour maintained by progressive-ratio schedules, as revealed by Killeen's Mathematical Principles of Reinforcement model of schedule-controlled behaviour. These drugs increase the value of a parameter that expresses the 'incentive value' of the reinforcer (a) and a parameter that is inversely related to the organism's 'motor capacity' (δ). This experiment examined the effects of two further atypical antipsychotics, aripiprazole and amisulprid

The effect of prefeeding on fixed-ratio pausing is jointly determined by past and upcoming reinforcer magnitudes.

Pausing within multiple fixed-ratio schedules differing in reinforcer magnitude is jointly controlled by both past and upcoming conditions of reinforcement. Abrupt shifts from a just-received large reinforcer to a signaled upcoming small reinforcer (i.e., a negative incentive shift) produce marked disruptions in responding, as indexed by extended pausing. The purpose of this experiment was to determine if reducing the level of food deprivation via prefeeding enhanced these disruptive effects. Five Long Evans rats lever-pressed according to a fixed-ratio schedule. Half of the components ended i

Conditions sufficient for the production of oral cocaine or lidocaine self-administration in preference to water.

Groups of rats were given a chronic history of drinking cocaine solutions of different concentrations in daily, 3-h schedule induced polydipsia sessions. Animals failed to develop a preference for cocaine solution to concurrently presented water. Schedule-induction conditions were maintained, and the animals were divided into separate groups, drinking either cocaine or lidocaine placed in a highly acceptable vehicle (glucose-saccharin solution). Animals preferred their respective drug solutions to concurrently presented water, and these preferences remained stable after the glucose-saccharin v

AM6527, a neutral CB1 receptor antagonist, suppresses opioid taking and seeking, as well as cocaine seeking in rodents without aversive effects.

Preclinical research has demonstrated the efficacy of CB1 receptor (CB1R) antagonists in reducing drug-taking behavior. However, clinical trials with rimonabant, a CB1R antagonist with inverse agonist profile, failed due to severe adverse effects, such as depression and suicidality. As a result, efforts have shifted towards developing novel neutral CB1R antagonists without an inverse agonist profile for treating substance use disorders. Here, we assessed AM6527, a CB1R neutral antagonist, in addiction animal models. Our findings revealed that AM6527 did not affect cocaine self-administration u

Effects of nicotine on schedule-controlled behaviour. Role of fixed-interval length and modification by mecamylamine and chlorpromazine.

The effects of nicotine, alone and in combination with mecamylamine and chlorpromazine, were studied in one group of rats exposed to a fixed-interval 30 sec schedule of food reinforcement, and a second group exposed to a fixed-interval 120 sec schedule. For both groups nicotine increased overall response rates in a dose-related fashion up to a maximum at 0.3 mg/kg. Examination of the within-interval response patterns showed that nicotine tended to increase the low level response rates in the early and middle parts of the interval and decrease, or increase to a proportionally smaller extent, th

Quantitative analysis of the effects of some "atypical" and "conventional" antipsychotics on progressive ratio schedule performance.

RATIONALE: Performance on progressive ratio schedules has been proposed as a means of assessing the effects of drugs on the value or "efficacy" of reinforcers. A mathematical model affords a basis for quantifying the effects of drugs on progressive ratio schedule performance. According to this model, the relation between response rate and ratio size is described by a bitonic (inverted-U) function. One parameter of the function, alpha, expresses the motivational or "activating" effect of the reinforcer (duration of activation of responding produced by the reinforcer), whereas another parameter,

Variable-ratio schedules of timeout from avoidance: effects of d-amphetamine and morphine.

Rats were trained on concurrent schedules in which pressing one lever postponed shock and pressing the other lever produced periods of signaled timeout from avoidance on variable-ratio schedules. These procedures generated high rates of timeout-reinforced responding and provided a baseline for studying the effects of drugs on behavior maintained by different types of negative reinforcement (shock postponement vs. timeout). Morphine (2.5 to 10.0 mg/kg) reduced behavior maintained by timeout at doses that increased or had no effect on avoidance responding. In contrast, d-amphetamine (0.125 to 2.

Functional assessment and treatment of perseverative speech about restricted topics in an adolescent with Asperger syndrome.

A functional analysis showed that a 14-year-old boy with Asperger syndrome displayed perseverative speech (or "restricted interests") reinforced by attention. To promote appropriate speech in a turn-taking format, we implemented differential reinforcement (DR) of nonperseverative speech and DR of on-topic speech within a multiple schedule with stimuli that signaled the contingencies in effect and who was to select the topic. Both treatments reduced perseverative speech, but only DR of on-topic speech increased appropriate turn taking during conversation. Treatment effects were maintained when

Effects of drugs on responding under concurrent fixed-interval schedules and concurrent fixed-ratio schedules.

Pigeons were trained to respond under concurrent fixed-interval fixed-interval (concurrent FI FI) schedules with Fl values (in seconds) of 30/300, 60/240, 60/60, 240/60 and 300/30. A second group was trained to respond under concurrent fixed-ratio fixed-ratio (concurrent FR FR) with FR values of 10/40, 10/20, 20/20, 20/10 and 40/10. Under the concurrent Fl Fl schedules, pigeons responded much less often on the key with the higher reinforcement density than would be predicted basing on a perfect matching of the ratio of responses made to the ratio of reinforcers delivered, and the pigeons also

Amphetamine increases schedule-induced drinking reduced by negative punishment procedures.

RATIONALE: d-Amphetamine has been reported to increase schedule-induced drinking punished by lick-dependent signalled delays in food delivery. This might reflect a drug-behaviour interaction dependent on the type of punisher, because no such effect has been found when drinking was reduced by lick-contingent electric shocks. However, the anti-punishment effect of amphetamine could be mediated by other behavioural processes, such as a loss of discriminative control or an increase in the value of delayed reinforcers. OBJECTIVES: To test the effects of d-amphetamine on the acquisition and mainten

Nicotine enhances the ability of cues to control behavior and evoke dopamine release in the dorsolateral striatum.

Nicotine is one of the most widely used addictive substances, yet its primary reinforcing effects are relatively weak. Nicotine's ability to potentiate responding for conditioned reinforcers is thought to drive persistent drug use. Here, we show that nicotine failed to alter responding maintained by a primary sucrose reinforcer (under a variable-ratio [VR] 11 schedule) across a broad dose range (0.01-1.0 mg/kg). Yet, nicotine enhanced operant behavior under a second-order reinforcement schedule in which responses produced sucrose-associated cues and were only intermittently reinforced by sucro

Negative allosteric modulation of CB1 cannabinoid receptor signalling decreases intravenous morphine self-administration and relapse in mice.

The endocannabinoid system interacts with the reward system to modulate responsiveness to natural reinforcers, as well as drugs of abuse. Previous preclinical studies suggested that direct blockade of CB1 cannabinoid receptors (CB1R) could be leveraged as a potential pharmacological approach to treat substance use disorder, but this strategy failed during clinical trials due to severe psychiatric side effects. Alternative strategies have emerged to circumvent the side effects of direct CB1 binding through the development of allosteric modulators. We hypothesized that negative allosteric modula

Inactivation of the thalamic paraventricular nucleus promotes place preference and sucrose seeking in male rats.

RATIONALE: The experience of reward entails both positive affect and motivation. While the brain regions responsible for these distinct aspects of reward are dissociable from each other, the paraventricular nucleus of the thalamus (PVT) may play a role in both. OBJECTIVES: To investigate the role of the PVT in both affect and motivation, and to identify neuropeptides that might mediate these effects. METHODS: Male rats were tested for conditioned place preference following temporary inactivation of the anterior or posterior PVT with local injections of the GABAB and GABAA agonists, baclofen

Reinforcement History Dependent Effects of Low Dose Ethanol on Reward Motivation in Male and Female Mice.

Alcohol use disorders (AUDs) are more prevalent in men than in women, though AUD diagnoses in women are growing rapidly, making an understanding of sex differences in alcohol-related behaviors increasingly important. The development of AUDs involves the transition from casual, low levels of alcohol drinking to higher, maladaptive levels. The ability of low dose alcohol to drive reward and drug seeking may differ in males and females, and this could underlie differences in susceptibility to AUD. In this study we sought to determine whether a history of chronic, low dose ethanol exposure (0.5 g/

Kappa opioid agonists reduce oxycodone self-administration in male rhesus monkeys.

RATIONALE: Combinations of mu and kappa opioid receptor (KOR) agonists have been proposed as potential analgesic formulations with reduced abuse liability. The current studies extend previous work by investigating the typical KOR agonist, salvinorin A, and the atypical KOR agonist, nalfurafine, as deterrents of oxycodone self-administration using a progressive ratio (PR) schedule of reinforcement. METHODS: In separate experiments, adult male rhesus monkeys (N = 4/experiment) were trained under a PR schedule of reinforcement to self-administer cocaine (0.1 mg/kg/injection) and saline on altern

Sigma-1 receptor antagonist PD144418 suppresses food reinforced operant responding in rats.

Sigma-1 (σ1) receptors have been investigated for their involvement in learning, rewarding and motivational processes, particularly as it relates to substances of abuse. Few studies have examined the effects of σ1 receptor agonists and antagonists on the rewarding and motivational properties of natural reinforcers, such as food. Studies that have investigated σ1 receptor agonists and antagonists has produced conflicting results. σ1 receptor antagonist PD144418 has been found to produce a dose-dependent attenuation of locomotor activity induced by cocaine, and by itself, does not suppress basal

Heroin self-administration and reinstatement of heroin-seeking in adolescent vs. adult male rats.

RATIONALE: Heroin abuse is prevalent among teenagers, and early onset drug use might predict long-term drug dependence. However, adolescent sensitivity to drug reinforcement has not been explored thoroughly in animal models. OBJECTIVES: This study aimed to compare intravenous (i.v.) self-administration of heroin, as well as extinction and reinstatement of heroin-seeking, in adolescent vs. adult male rats. METHODS: Adolescent (35 days old at start) and adult (86 days old at start) male Sprague-Dawley rats spontaneously acquired lever pressing maintained by i.v. heroin infusions. In experiment
