Interoceptive autonomic regulation in typical development and autism spectrum disorder: A computational model integrating multiple physiological systems.

BACKGROUND: Interoceptive cardiovascular signals, including heart rate (HR) and blood pressure (BP), arise from coordinated sympathetic (SNS) and parasympathetic (PSNS) regulation and contribute to affective and cognitive processes. Although atypical autonomic nervous system (ANS) modulation has been reported in autism spectrum disorder (ASD), the dynamical structure underlying branch-specific coordination remains insufficiently characterized. OBJECTIVE: To estimate latent ANS regulatory structure in typically developing (TD) and ASD individuals using a computational modeling framework. METH

Anti-ganglioside complex antibody profiles in a recurrent complicated case of GQ1b-seronegative miller fisher syndrome and Bickerstaff brainstem encephalitis: a case report.

BACKGROUND: Guillain-Barré syndrome (GBS), Miller Fisher syndrome (MFS) and Bickerstaff brainstem encephalitis (BBE) are a group of autoimmune neurological disorders (GBS spectrum disorder) that rarely recur. Recently, anti-ganglioside complex antibodies (GSC-Abs) were identified in patients with GBS spectrum disorder. However, there has been no case report describing GSC-Abs profiles in a recurrent case showing different phenotypes. CASE PRESENTATION: We report the case of a 33-year-old male patient with GQ1b-seronegative BBE-GBS after two prior episodes of MFS-GBS. Our patient showed ophtha

Obstructive and Central Sleep Apnea and the Risk of Incident Atrial Fibrillation in a Community Cohort of Men and Women.

BACKGROUND: Previous studies have documented a high prevalence of atrial fibrillation (AF) in individuals with obstructive sleep apnea (OSA). Central sleep apnea (CSA) has been associated with AF in patients with heart failure. However, data from prospective cohorts are sparse and few studies have distinguished the associations of obstructive sleep apnea from CSA with AF in population studies. METHODS AND RESULTS: We assessed the association of obstructive sleep apnea and CSA with incident AF among 2912 individuals without a history of AF in the SHHS (Sleep Heart Health Study), a prospective,

High resolution respirometry of permeabilized skeletal muscle fibers in the diagnosis of neuromuscular disorders.

High resolution respirometry in combination with the skinned fiber technique offers the possibility to study mitochondrial function routinely in small amounts of human muscle. During a period of 2 years, we investigated mitochondrial function in skeletal muscle tissue of 13 patients (average age = 5.8 years). In all of them, an open muscle biopsy was performed for diagnosis of their neuromuscular disorder. Mitochondrial oxidation rates were measured with a highly sensitive respirometer. Multiple substrate-inhibitor titration was applied for investigation of mitochondrial function. About 50 mg

Characterization of a UQCRC1 variant in a patient with progressive weakness, pain and sleep issues reveals a functional mitochondrial defect restored by mitochondrial transplantation.

Primary mitochondrial defects underlie the heterogeneity of many rare inherited disorders. Pathogenic variants that disrupt the function of the multi-subunit protein complexes of the mitochondrial respiratory chain contribute to a range of neurological phenotypes and other clinical manifestations. These variants are also thought to contribute to the onset and progression of numerous more common neurodegenerative conditions such as Parkinson's and Alzheimer's disease. Here we describe an individual affected with progressive muscle weakness and pain harboring a paternally inherited missense vari

Breathing stimulation mediated by 5-HT1A and 5-HT3 receptors within the preBötzinger complex of the adult rabbit.

Serotonin (5-HT) has been reported to play excitatory effects on respiration by acting on preBötzinger complex (preBötC) neurons in neonatal or juvenile rodents. However, whether its action is circumscribed to the preBötC and present in other animal species, particularly in adult preparations, is unknown. We investigated the respiratory role of 5-HT within the preBötC and neighbouring respiration-related regions. Experiments were performed on α-chloralose-urethane anesthetized, vagotomized, paralyzed and artificially ventilated rabbits making use of bilateral microinjections (30-50 nl). 5-HT c

[Apraxia of the larynx].

Disturbances of vocal function in patients with apraxia of speech are well known in the literature and have often been described. Although apraxia of phonation is presumed, only 1 case is mentioned in the literature. We present a 51-year-old man (with missing signs of dysarthria and aphasia after spontaneous right temproparietal hemorrhagia) who had aphonia and missing respiration during speaking, whereas articulation was nearly undisturbed. After differential diagnostic exclusion of other central disorders of phonation we classify this patient as having apraxia of phonation or laryngeal aprax

[Intravenous general anesthesia with low doses of ketamine in performing complete oral hygiene in the outpatient practice].

A variant of total anesthesia with modified doses of ketamine was developed and tried in 1305 patients in an outpatient setting. Low doses of ketamine hydrochloride (initial dose 25-70 mg, repeated one 20-40 mg) do not lead to hemodynamic shifts, disorders of spontaneous respiration, and long postanesthetic depression. At the same time the level of anesthesia and analgesia attained is sufficient for complete sanitization of the oral cavity. The suggested method extends the indications to total anesthesia in stomatological interventions on an outpatient basis, is simple for anesthesiologist, an

Management of severe pediatric head trauma.

Except for prevention, nothing can alter the primary damage to the central nervous system tissues and blood vessels caused by the impact of traumatic forces over a few milli-seconds. However, damage to nervous system tissues secondary to transient reversible brain dysfunction may occur and lead to failure of respiration and circulation. Brain swelling and intracranial hypertension can develop and interfere with oxygen delivery and cellular metabolism of vital central nervous structures. A team approach with simultaneous treatment of the various disorders and recurrent evaluation is the hallmar

Hepatic bioenergetics and metabolism in mitochondrial disease: insights from the Ndufs4 KO mouse model.

INTRODUCTION: Mitochondrial complex (CI) deficiency frequently manifests as a severe neurometabolic disorder called Leigh syndrome (LS). Research on the Ndufs4 knockout (KO) mouse model has identified neuronal vulnerability to CI deficiency as a major driver of the disease, yet its effects on hepatic function remain unclear. Considering the importance of the liver, and its interconnection with the brain, in regulating whole-body metabolic balance, further investigation into the effects of whole-body Ndufs4 KO on the liver is warranted. OBJECTIVES: This study investigated liver bioenergetics a

Respiratory disorders and atopy in cotton, wool, and other textile mill workers in Denmark.

A cross-sectional study of respiratory disorders and atopy in Danish textile industry workers was conducted to survey respiratory symptoms throughout the textile industry, to estimate the association of these disorders with atopy, and to study dose-response relationships within the cotton industry. Workers at cotton mills, a wool mill, and a man-made fiber (MMF) mill were examined. Four hundred nine (90%) of the 445 workers participated in this survey, i.e., 253, 62, and 94 workers at the cotton mills, the wool mill, and the MMF mill, respectively. An interview designed to assess the prevalenc

[Self-ventilation at home in chronic respiratory insufficiency].

Non-invasive intermittent "self-ventilation" employing a nose mask is considered to be an efficient means of treating chronic respiratory failure. After several hours of treatment with a respirator, 10 patients (four with kyphoscoliosis, three with COLD, two with neuromuscular disorders, and one with sequela of Tbc) experienced reversal of the chronic respiratory failure and an improvement in respiratory muscle force.

Upper airway function and dysfunction in respiration.

1. The upper airway not only provides a passage for air to be breathed in and out of the lungs, but it also heats, humidifies and filters the air and is involved in cough, swallowing and speech. 2. The complex muscle structure of the upper airway that produces speech and swallowing in humans also modulates respiratory airflow throughout the respiratory cycle, but is vulnerable to functional problems that may compromise respiration. 3. Even in normals, there is some collapse of the upper airway and increased upper airway resistance during sleep. 4. A substantial proportion of people suffer from

A small‑molecule HDAC/PDE modulator activates human adipocyte UCP1 and resolves inflammatory signaling.

BACKGROUND: Obesity-associated inflammation in white adipose tissue (WAT) drives insulin resistance and type 2 diabetes. While UCP1-mediated thermogenesis is a therapeutic target, most browning agents lack anti-inflammatory activity. We aimed to identify small molecules that induce browning and suppress inflammation in human adipocytes. METHODS: Human cohorts and cell line models were analyzed for inflammatory markers. We generated an in-house multicomponent reaction (MCR)-based chemical library and developed a proprietary high-throughput screening platform to identify UCP1 activators in human
