Critical Role for Phosphatidylinositol-3 Kinase Vps34/PIK3C3 in ON-Bipolar Cells.

PURPOSE: Phosphatidylinositol-3-phosphate (PI(3)P), and Vps34, the type III phosphatidylinositol 3-kinase primarily responsible for its production, are important for function and survival of sensory neurons, where they have key roles in membrane processing events, such as autophagy, endosome processing, and fusion of membranes bearing ubiquitinated cargos with lysosomes. We examined their roles in the most abundant class of secondary neurons in the vertebrate retina, the ON-bipolar cells (ON-BCs). METHODS: A conditional Vps34 knockout mouse line was generated by crossing Vps34 floxed mice wit

Perivascular mural cells of the mouse choroid demonstrate morphological diversity that is correlated to vasoregulatory function.

OBJECTIVE: Perivascular mural cells of the choroid have been implicated in physiological functioning as well as in retinal disease pathogenesis. However details regarding their form and function are not well understood. We aim to characterize choroidal mural cells in the adult mouse choroid in terms of their distribution and morphology, and correlate these to their contractile behavior. METHODS: Sclerochoroidal flat-mounted explants were prepared from albino transgenic mice in which the α-smooth muscle actin (α-SMA) promoter drives the expression of green fluorescent protein (GFP). α-SMA-expr

Progressive loss of vision caused by asymptomatic pituitary macroadenoma: role of OCT.

INTRODUCTION: Most pituitary adenomas are clinically inactive. In patients with long-standing compression of the optic chiasm, ganglion cells may undergo axonal degeneration. Spectral domain optical coherence tomography (SD-OCT) is able to identify retinal nerve fiber layer (RNFL) and ganglion cell loss in the retina. We present a case in which SD-OCT was used to diagnose an asymptomatic pituitary macroadenoma. CLINICAL CASE: A 48-year-old female presented with progressive vision loss in both eyes. SD-OCT identified atrophy of the ganglion cell and nerve layers, with preservation of outer lay

The nob2 mouse, a null mutation in Cacna1f: anatomical and functional abnormalities in the outer retina and their consequences on ganglion cell visual responses.

Glutamate release from photoreceptor terminals is controlled by voltage-dependent calcium channels (VDCCs). In humans, mutations in the Cacna1f gene, encoding the alpha1F subunit of VDCCs, underlie the incomplete form of X-linked congenital stationary night blindness (CSNB2). These mutations impair synaptic transmission from rod and cone photoreceptors to bipolar cells. Here, we report anatomical and functional characterizations of the retina in the nob2 (no b-wave 2) mouse, a naturally occurring mutant caused by a null mutation in Cacna1f. Not surprisingly, the b-waves of both the light- and

Proton-mediated feedback inhibition of presynaptic calcium channels at the cone photoreceptor synapse.

Generation of center-surround antagonistic receptive fields in the outer retina occurs via inhibitory feedback modulation of presynaptic voltage-gated calcium channels in cone photoreceptor synaptic terminals. Both conventional and unconventional neurotransmitters, as well as an ephaptic effect, have been proposed, but the intercellular messaging that mediates the inhibitory feedback signal from postsynaptic horizontal cells (HCs) to cones remains unknown. We examined the possibility that proton concentration in the synaptic cleft is regulated by HCs and that it carries the feedback signal to

P2 component latency of fVEP as a bioindicator for clinical and diagnostic use in visual pathologies.

PURPOSE: The signaling of flash visual evoked potential (fVEP) derives from the retina, but how retinal activity influences fVEP remains unclear. This work aimed to decipher the specific retinal kinetic contributions to fVEP response. METHODS: Monocular and simultaneous recordings of flash VEP and electroretinogram were performed. Healthy and adult mice C57BL/6J were used. The right eye was injected intravitreally with 1 μL of PBS containing 25 mM APB, 10 mM Bicuculline, 30 mM DNQX, 100 mM Glutamate, 100 mM GABA, 5 mM TPMPA, or 25 mM HEPES. Autocrine protective mechanisms of human granulocyte colony-stimulating factor (G-CSF) on retinal ganglion cells after optic nerve crush.

This study investigated the role of autocrine mechanisms in the anti-apoptotic effects of human granulocyte colony-stimulating factor (G-CSF) on retinal ganglion cells (RGCs) after optic nerve (ON) crush. We observed that both G-CSF and G-CSF receptor (G-CSFR) are expressed in normal rat retina. Further dual immunofluorescence staining showed G-CSFR immunoreactive cells were colocalized with RGCs, Müller cells, horizontal and amacrine cells. These results confirm that G-CSF is an endogenous ligand in the retina. The semi-quantitative RT-PCR finding demonstrated the transcription levels of G-CS

Effects of neonatally induced strabismus on binocular responses in cat area 18.

Responses to binocular visual stimulation were compared in cortical area 18 of normal cats and in cats in which one eye was exodeviated by surgery early in postnatal life. In contrast to normal cats where most units (58%) were binocularly activated, relatively few units (10%) in strabismic cats were activated well by stimulation of either eye. Rather individual units were driven mainly via one eye or the other but not both. In addition, there was a tendency for more units to be driven well via the unoperated eye than via the exodeviated eye. Fewer cells preferring vertically oriented than hori

Visual fields and optical coherence tomography (OCT) in neuro-ophthalmology: Structure-function correlation.

Visual field (VF) testing is an essential component of the neurological examination. The differential diagnosis of VF defects depends on relating this measure of afferent visual function to the structure of the visual pathway and optical coherence tomography (OCT) is an invaluable tool for detailed structural evaluation of the optic nerve and retina. This review describes the ways in which interpretation of VF and OCT can be used together to increase the accuracy of the localization of lesions along the visual pathway. Lesions of the anterior visual pathway (originating in ganglion cells or ne

Wolfram syndrome 1 (Wfs1) gene expression in the normal mouse visual system.

Wolfram syndrome (OMIM 222300) is a neurodegenerative disorder defined by insulin-dependent diabetes mellitus and progressive optic atrophy. This syndrome has been attributed to mutations in the WFS1 gene, which codes for a putative multi-spanning membrane glycoprotein of the endoplasmic reticulum. The function of WFS1 (wolframin), the distribution of this protein in the mammalian visual system, and the pathogenesis of optic atrophy in Wolfram syndrome are unclear. In this study we made a detailed analysis of the distribution of Wfs1 mRNA and protein in the normal mouse visual system by using

Morphological study on the regeneration of the retina in the rainbow trout after ouabain-induced damage: evidence for dedifferentiation of photoreceptors.

The retina of the rainbow trout is capable of marginal regeneration after ouabain-induced degeneration (intraocular injection of 5 microliters 10(-4) M ouabain). In the central area where the pigment epithelium proliferates to a multicellular layer, the neural retina does not regenerate up to 182 days after injection of ouabain. The regeneration process begins in the marginal growth zone with an increase in the mitotic rate; the growth zone itself is not damaged after ouabain administration. The proliferate differentiates with time into a newly layered retina; this portion of the retina is cal

IL-1-mediated vitreous inflammation as an early indicator of retinal ganglion cell loss following acute optic nerve injury.

BACKGROUND: Retinal ganglion cell (RGC) degeneration in optic neuropathies is often preceded by neuroinflammatory changes, yet the earliest in vivo indicators of this process remain poorly defined. Vitreous hyperreflective foci (VHRFs) emerging within 24 h following optic nerve crush (ONC) might represent a promising early in vivo indicator of RGC loss. METHODS: VHRFs were longitudinally tracked by visible-light optical coherence tomography (vis-OCT) imaging post-ONC. Whole-eye sectioning, immunohistochemistry, and confocal imaging revealed the identity and migration of the VHRFs. RNAscope in
