Congenital stationary night blindness with myopia: a clinico-pathologic study.

The left eye of a 77 year old male patient was enucleated because of absolute glaucoma. This eye showed subnormal electroretinogram (ERG) and oscillatory potentials preoperatively, but the other eye showed Schubert-Bornschein type ERG and monophasic dark adaptation curve. Light and electron microscopic studies of the left eye showed a normal arrangement of discs of rod outer segments, normal synaptic ends of the photoreceptors, and complete loss of ganglion cells. From the subnormal ERG in the left eye we assumed it was originally Schubert-Bornschein type ERG but inverted to subnormal type ERG

Early photoreceptor outer segment loss and retinoschisis in Cohen syndrome.

PURPOSE: To describe early structural and functional retinal changes in a patient with Cohen syndrome. METHODS: A 13-month-old Caucasian girl of Irish and Spanish ancestry was noted to have micrognathia and laryngomalacia at birth, which prompted a genetic evaluation that revealed biallelic deletions in COH1 (VPS13B) (a maternally inherited 60-kb deletion involving exons 26-32 and a paternally inherited 3.5-kb deletion within exon 17) consistent with Cohen syndrome. She underwent a complete ophthalmic examination, full-field flash electroretinography and retinal imaging with spectral domain o

Stimulatory effect of taurine on calcium ion uptake in rod outer segments of the rat retina is independent of taurine uptake.

Taurine stimulates ATP-dependent Ca(2+) uptake in the rat rod outer segments (ROS). This stimulation has been linked to the function of the cyclic nucleotide-gated cation channel, implying an important physiologic role for taurine in visual signal transduction. Calmodulin (CaM) has been reported to affect taurine transport in the choroid plexus and also to inhibit the cyclic nucleotide-gated channel; thus, the effects of the competitive CaM inhibitors trifluoperazine (TFP) and N-(8-aminooctyl)-5-iodonaphthalene-1-sulfonamide (J-8) were studied on Ca(2+) and taurine uptake in the rat ROS. Knockout of Nr2e3 prevents rod photoreceptor differentiation and leads to selective L-/M-cone photoreceptor degeneration in zebrafish.

Mutations in the photoreceptor cell-specific nuclear receptor gene Nr2e3 increased the number of S-cone photoreceptors in human and murine retinas and led to retinal degeneration that involved photoreceptor and non-photoreceptor cells. The mechanisms underlying these complex phenotypes remain unclear. In the hope of understanding the precise role of Nr2e3 in photoreceptor cell fate determination and differentiation, we generated a line of Nr2e3 knockout zebrafish using CRISPR technology. In these Nr2e3-null animals, rod precursors undergo terminal mitoses but fail to differentiate as rods. Rod

Immunocytochemical demonstration of interphotoreceptor retinoid-binding protein in cerebellar medulloblastoma.

Previously, immunoreactive rod-opsin and S-antigen (arrestin), two highly characteristic markers of retinal photoreceptors and pinealocytes, were shown to be present in certain medulloblastoma cells. It, thus, has been suggested that such cells differentiate along the photoreceptor lineage. This is corroborated in the present immunocytochemical investigation using antibodies against another photoreceptor-cell marker, the interphotoreceptor retinoid-binding protein (IRBP). As shown in preparations of human retina and pineal organ, IRBP can be successfully demonstrated in formalin-fixed and para

Permissive glycan support of photoreceptor outer segment assembly occurs via a non-metabolic mechanism.

PURPOSE: We have previously demonstrated that in RPE-deprived retinas, lactose, galactose, and structurally related glycans support the proper assembly of nascent photoreceptor outer segment membranes. Other glycans such as mannose and glucose have no effect on this process. While the ability to support outer segment assembly is highly specific, all of the permissive glycans we have tested are able to enter metabolic pathways within the retinal cells, thus the mechanism by which the glycans are functioning is still ambiguous. The present study was undertaken to determine if permissive glycan-m

A truncated form of the cGMP-gated cation channel β-subunit lacking the GARP2 region transiently restores rod structure and function.

The cGMP-gated channel β-subunit is required for phototransduction, disc morphogenesis, and structural integrity of rod cells. Its structural role may be mediated by its GARP2 (glutamic acid rich protein) domain. To assess domain functions, we generated transgenic mice expressing a truncated protein lacking the N-terminal region on a Cngb1 null background. In these mice, light microscopy showed normal disk morphology up to 6 months, and cryo-electron tomography revealed disc-to-plasma membrane connections similar to WT. However, the truncated protein was insufficient to prevent progressive deg

A comparison of some photoreceptor characteristics in the pineal and retina. II. The Djungarian hamster (Phodopus sungorus).

A rod-specific antiserum was used to immunolabel elements within the retina and pineal of the adult Djungarian hamster and Welsh Mountain sheep. In the retina immunostaining was localized to the outer segments and perikarya of photoreceptor cells, while in the pineal limited numbers of labelled pinealocytes were scattered throughout the gland. An enzyme-linked immunosorbent assay (ELISA) was then used to obtain a quantitative measure of rod opsin in total eye and pineal extracts from the Djungarian hamster. Total rod opsin (+/- SEM) in the eye was measured by absorbance spectroscopy (1.88 +/-

Abnormal photoresponses and light-induced apoptosis in rods lacking rhodopsin kinase.

Phosphorylation is thought to be an essential first step in the prompt deactivation of photoexcited rhodopsin. In vitro, the phosphorylation can be catalyzed either by rhodopsin kinase (RK) or by protein kinase C (PKC). To investigate the specific role of RK, we inactivated both alleles of the RK gene in mice. This eliminated the light-dependent phosphorylation of rhodopsin and caused the single-photon response to become larger and longer lasting than normal. These results demonstrate that RK is required for normal rhodopsin deactivation. When the photon responses of RK-/- rods did finally tur

Retinal morphology in spinocerebellar ataxia type 1 (SCA1) mice: A stereological analysis across different age groups.

Spinocerebellar ataxia type 1 (SCA1) affects not only the cerebellum but also the retina; however, retinal pathology remains poorly characterised in murine models of SCA1. To fill this gap, we performed a comprehensive stereological analysis of the retinal structure of SCA1154Q/2Q knock-in mice and their healthy SCA12Q/2Q littermates at 6 and 10 months of age. We compared animals across genotypes at each age and across ages within each genotype. Using unbiased stereology, we quantified the total retinal volume, volumes of individual retinal layers, total photoreceptor numbers, numbers of rods

[Electron microscopic and morphometric study of the effects of ENKAD preparation on photoreceptor cells of the retina in Campbell rats with hereditary retinal dystrophy].

The drug , a complex of ribonucleotides, which was found to improve the vision in some patients with retinitis pigmentosa, stimulates the morphogenesis of rod outer segments (ROS) in mutant Campbell rats with inherent retinal dystrophy. Four litters of neonatal rats, which being subdivided into groups, were injected subcutaneously with either 0.1 ml of 3.5% drug (experimental group) or 0.1 ml of 0.9% NaCl (control group) daily for 6-7 days. The results were checked in posterior area of the retina at Day 7 by the method of electron microscopy and morphometry. The drug treatment resulted in the

Cell-specific localization of β-synuclein in the mouse retina.

β-synuclein, a member of the synuclein family, is frequently co-expressed with α-synuclein in the neural system, where it serves to inhibit abnormal aggregation of α-synuclein in neurodegenerative diseases. Beyond its role in pathological conditions, β-synuclein plays various functions independently of α-synuclein. In our investigation, we discovered a broader expression of β-synuclein in the mouse retina compared to α-synuclein. This widespread pattern implies its potential significance in the retina. Through detailed examination via light- and electron-microscopic immunocytochemistry, we ide

The retina of Spalax ehrenbergi: novel histologic features supportive of a modified photosensory role.

PURPOSE: The retina of the blind mole rat Spalax ehrenbergi was compared with other vertebrate photosensitive organs in an attempt to correlate its histologic organization with a presumptive nonvisual photoreceptor role. METHODS: The eyes of eight adult animals were analyzed by light and electron microscopy, using conventional staining and immunolabeling with antibodies against phototransduction proteins and calretinin. RESULTS: Rods accounted for most of the photoreceptor cells in the Spalax retina, although their morphology is dissimilar to that of sighted mammals, in that they contained o

The melatonin antagonist luzindole protects retinal photoreceptors from light damage in the rat.

PURPOSE: Systemic administration of melatonin can increase retinal light damage in the rat. The role of retinal melatonin receptors in modulating light-damage susceptibility was investigated by intravitreally injecting the melatonin receptor antagonist luzindole into rats. METHODS: Nine Sprague-Dawley albino rats 8 to 9 weeks of age were kept in 50 lux cyclic light for at least 7 days before receiving an intravitreal injection of 1 microl 1 mM luzindole in one eye and 1 microl vehicle in the other eye. The injection was given just before the beginning of the normal 12-hour dark phase. At the

[Characteristics and functions of melanin in retinal pigment epithelium].

Melanin granules in the retinal pigment epithelium (RPE) have many important functions which are not yet completely understood. Melanin in the RPE protects the cell from damage caused by oxidative stress. This pigment acts as a free radical sink and diminishes cytotoxic lipid peroxidation. Thus, melanin protects against light toxicity and against cytotoxic effects caused by ocular inflammation. Many enzymes, e.g. superoxide dismutase or carboanhydrase, are only activated in the presence of zinc. Melanin can store zinc and release it when required. The absence of melanin in patients with oculoc

Syrian hamster prion protein (PrP(C)) is expressed in photoreceptor cells of the adult retina.

PrP(C), the cellular isoform of the prion protein (PrP) serves as a precursor to abnormal PrP isoforms which accumulate in diseases such as scrapie in sheep, and Creutzfeldt-Jakob disease in humans. Since prions can replicate in photoreceptors we surmised that PrP(C) must be expressed in these cells. Accordingly, monoclonal antisera directed against two epitopes of hamster PrP(C) produced retinal immunostaining in hamsters, and in mice bearing a hamster PrP transgene. Immunostaining was most prominent in the inner and outer segments of rod photoreceptors, coinciding with the earliest site of p

Ultrastructural and Electrophysiological Determination of the Patency of Monkey Cone Outer Segment Membranes.

Cone photoreceptors support the high-acuity color vision characteristic of many primates, including humans. In several non-mammalian species, such as amphibians, the light-sensitive disk membranes within cone outer segments (COSs) are fully open to the extracellular space and continuous (patent) with the plasma membrane. In contrast, the extent of such patency in mammalian COSs has remained unresolved. The open disk architecture represents a fundamental distinction from the closed, discrete disks that dominate a rod OS (ROS), with important implications for outer segment renewal and phototrans
