Expression of UV-sensitive parapinopsin in the iguana parietal eyes and its implication in UV-sensitivity in vertebrate pineal-related organs.

The pineal-related organs of lower vertebrates have the ability to discriminate different wavelengths of light. This wavelength discrimination is achieved through antagonistic light responses to UV or blue and visible light. Previously, we demonstrated that parapinopsin underlies the UV reception in the lamprey pineal organ and identified parapinopsin genes in teleosts and frogs of which the pineal-related organs were reported to discriminate light. In this study, we report the first identification of parapinopsin in the reptile lineage and show its expression in the parietal eye of the green

Unexpected diversity and photoperiod dependence of the zebrafish melanopsin system.

Animals have evolved specialized photoreceptors in the retina and in extraocular tissues that allow them to measure light changes in their environment. In mammals, the retina is the only structure that detects light and relays this information to the brain. The classical photoreceptors, rods and cones, are responsible for vision through activation of rhodopsin and cone opsins. Melanopsin, another photopigment first discovered in Xenopus melanophores (Opn4x), is expressed in a small subset of retinal ganglion cells (RGCs) in the mammalian retina, where it mediates non-image forming functions su

Melanopsin expression in dopamine-melatonin neurons of the premammillary nucleus of the hypothalamus and seasonal reproduction in birds.

Melanopsin (OPN4) is a photoreceptive molecule regulating circadian systems in mammals. Previous studies from our laboratory have shown that co-localized dopamine-melatonin (DA-MEL) neurons in the hypothalamic premammillary nucleus (PMM) are putatively photosensitive and exhibit circadian rhythms in DAergic and MELergic activities. This study investigates turkey OPN4x (tOPN4x) mRNA distribution in the hypothalamus and brainstem, and characterizes its expression in PMM DA-MEL neurons, using in situ hybridization (ISH), immunocytochemistry (ICC), double-label ISH/ICC, and real time-PCR. The mRNA

Daily and seasonal variation in the spectral composition of light exposure in humans.

Light is considered the most potent synchronizer of the human circadian system and exerts many other non-image-forming effects, including those that affect brain function. These effects are mediated in part by intrinsically photosensitive retinal ganglion cells that express the photopigment melanopsin. The spectral sensitivity of melanopsin is greatest for blue light at approximately 480 nm. At present, there is little information on how the spectral composition of light to which people are exposed varies over the 24 h period and across seasons. Twenty-two subjects, aged 22+/-4 yrs (mean+/-SD)

The eye of the blind mole rat, Spalax ehrenbergi. Rudiment with hidden function?

The rudimentary eyes of the mole rat (Spalax ehrenbergi) are located under the skin and do not respond to light stimuli. However, removal of the eyes disturbs photoperiod perception in these animals. To help clarify the possibly remaining function of the eyes in this species, the authors studied their development and ultrastructure. In the early embryos the presumptive eye regions--the epithelium, lens vesicle, and optic cup--appear initially normal. As development progresses, the iris-ciliary body complex originates prematurely from the margin of the optic cup and shows a very rapid and massi

Mechanistic Insights into MYO1C-Mediated Rhodopsin Trafficking and Rod Photoreceptor Homeostasis.

Rhodopsin trafficking from the photoreceptor inner segment to the outer segment is essential for photoreceptor function, yet the molecular mechanism(s) regulating this process remain incompletely understood. MYO1C is an actin-based motor protein implicated in intracellular cargo trafficking. Here, we investigated its role in rhodopsin trafficking and photoreceptor cell homeostasis. In-silico docking identified a putative interaction between the MYO1C C-terminal region and the C-terminal region of rhodopsin containing the conserved VxPx ciliary trafficking motif. Biochemical studies confirmed t

Photoreceptor proteins and melatonin rhythm generating AANAT in the carp pineal: Temporal organization and correlation with natural photo-thermal cues.

We studied temporal organization of both the photoreceptor (rod-like opsin, alpha subunit of the G protein transducin or alpha-TD) and melatonin generating (AANAT) proteins in the same pineal of a tropical surface dwelling free-living carp Catla catla, and analyzed possible correlation between them as well as with natural photo-thermal variables in an annual cycle. The pineal from individual fish was collected at four different time points (06.00 h, 12.00 h, 18.00 h, and 24.00 h) in a 24.00 h cycle and the same was repeated in four distinct seasons in an annual reproductive cycle to study each

The Diverse Role of ipRGCs in Visual Perception Beyond Non-Image-Forming Functions.

The intrinsically photosensitive retinal ganglion cells (ipRGCs) are the third class of photoreceptors apart from rods and cones, containing the photopigment melanopsin and are characterized into different subtypes (M1-M6) that support conscious visual perception. They transmit the light information to the brain through complex circuits serving both non-image-forming and image-forming functions. Of the total population, there are four main ipRGCs, such as M2, M4, M5 and M6 that project substantially to the image-forming visual pathway and target the dorsal lateral geniculate nucleus (dLGN). Melanopsin activates divergent phototransduction pathways in intrinsically photosensitive retinal ganglion cell subtypes.

Melanopsin signaling within intrinsically photosensitive retinal ganglion cell (ipRGC) subtypes impacts a broad range of behaviors from circadian photoentrainment to conscious visual perception. Yet, how melanopsin phototransduction within M1-M6 ipRGC subtypes impacts cellular signaling to drive diverse behaviors is still largely unresolved. The identity of the phototransduction channels in each subtype is key to understanding this central question but has remained controversial. In this study, we resolve two opposing models of M4 phototransduction, demonstrating that hyperpolarization-activat

Machine learning for comprehensive prediction of high risk for Alzheimer's disease based on chromatic pupilloperimetry.

Currently there are no reliable biomarkers for early detection of Alzheimer's disease (AD) at the preclinical stage. This study assessed the pupil light reflex (PLR) for focal red and blue light stimuli in central and peripheral retina in 125 cognitively normal middle age subjects (45-71 years old) at high risk for AD due to a family history of the disease (FH+), and 61 age-similar subjects with no family history of AD (FH-) using Chromatic Pupilloperimetry coupled with Machine Learning (ML). All subjects had normal ophthalmic assessment, and normal retinal and optic nerve thickness by optical

Perinatal development of melanopsin expression in the mouse retina.

The melanopsin-expressing retinal ganglion cells are specialized in measuring irradiance for several functions, including daily photoentrainment and regulation of pupil size. In the present study, these cells were analyzed in mice during their perinatal period, from embryonic day (E) 15 to postnatal day (P) 1. Melanopsin expression was detected at E15 in cells that did not co-express the transcription factor Brn3a. Under light/dark (LD) cycles, the number of melanopsin-expressing cells did not change between E16 and E19, while a very significant increase was observed during the short interval

Presence of retina-specific proteins in the lamprey pineal complex.

The pineal complex of river lamprey reacted with the antisera raised against retina specific proteins including bovine opsin, chick visinin and frog light-sensitive cyclic GMP phosphodiesterase (PDE). Immunoreactive materials stained with anti-opsin were evenly located at the outer segment of photoreceptor cells in the pineal organ and also found in the parapineal organ. Although anti-visinin stained the pineal and parapineal photoreceptor cells, the immunopositive photoreceptor cells were observed only at the lateral portion and not at the medial portion of the pineal organ. No immunoreactive

Detecting irreversible cone photoreceptors injury caused by whole-body hyperthermia.

Whole-body hyperthermia (WBH) has been used as an adjunctive strategy for treating malignant and viral diseases. However, WBH can cause toxic reactions in the brain and affect cerebral function. The retina is an extension of the brain. This study concerned about the effects of WBH on murine retinas. Under general anesthesia, the C57BL/6J mice in the WBH group were maintained abdomen-down position on a thermostatic platform, the temperature of which ranged from 40 to 43 °C. Ultimately, murine rectal temperatures reached a recommended high point, approximately 42 °C. However, rectal temperatures

Cones and cone pathways remain functional in advanced retinal degeneration.

Most defects causing retinal degeneration in retinitis pigmentosa (RP) are rod-specific mutations, but the subsequent degeneration of cones, which produces loss of daylight vision and high-acuity perception, is the most debilitating feature of the disease. To understand better why cones degenerate and how cone vision might be restored, we have made the first single-cell recordings of light responses from degenerating cones and retinal interneurons after most rods have died and cones have lost their outer-segment disk membranes and synaptic pedicles. We show that degenerating cones have functio

The Allosteric Inhibitor Pentachloropseudilin Inhibits Myosin 1C ATPase Activity and Recapitulates Retinitis Pigmentosa Phenotypes in Mice.

Unconventional myosins are molecular motors that move along actin filaments in an ATPase-dependent manner, thereby influencing intracellular cargo transport. Dysfunction of myosins due to loss-of-function mutations causes human disease phenotypes such as deafness, retinitis pigmentosa, renal failure, and hypertrophic cardiomyopathy. However, some genetic models lacking unconventional myosins do not recapitulate the retinal phenotypes observed in humans, necessitating alternative approaches. We previously showed that the myosin motor protein MYO1C is required for the trafficking of rhodopsin in

Current understanding of photophobia, visual networks and headaches.

OBJECTIVE: To review clinical and pre-clinical evidence supporting the role of visual pathways, from the eye to the cortex, in the development of photophobia in headache disorders. BACKGROUND: Photophobia is a poorly understood light-induced phenomenon that emerges in a variety of neurological and ophthalmological conditions. Over the years, multiple mechanisms have been proposed to explain its causes; however, scarce research and lack of systematic assessment of photophobia in patients has made the search for answers quite challenging. In the field of headaches, significant progress has been

Light tunes long-term threat avoidance behavior in male mice.

Animals must constantly scan their environment for imminent threats to their safety. However, they must also integrate their past experiences across long timescales to assess the potential recurrence of new threats. Though visual inputs are critical for the detection of environmental danger, whether and how visual information shapes an animal's assessment of whether a new threat is likely to reappear in a given context is unknown. In this work, we developed a behavioral assessment of long-term threat avoidance behavior where animals will avoid a familiar location where they previously experien

Melanopsin's Phototransduction and Spiking Response Are Evident in the Photoreceptor-Directed Multifocal Electroretinogram.

Existing methods of measuring melanopsin's functional response in humans can only return a measure of a postcortical interpretation of the intrinsic photoresponse. There is physiological ex vivo evidence to suggest that the retinal activity of melanopsin can differ from its cortical interpretation. We developed an in vivo method for isolating the multifocal neuroretinal responses originating from melanopsin signaling in humans (seven males, one female). These electrical responses can be decomposed into three separable components; (1) a very fast positive potential with timing on the scale of m

Light-at-night exposure drives myopia via melanopsin signalling.

Myopia poses a growing global public health challenge, with its underlying mechanisms not yet fully understood. While insufficient daytime light exposure is a recognized protective factor, the role of nighttime light exposure remains contentious. Through a murine model, we uncover a novel melanopsin-dependent pathway by which light-at-night (LAN) exposure disrupts the expression of core circadian clock genes (including Arntl, Cry1, Per1, and Per2) and downregulates melanopsin expression in intrinsically photosensitive retinal ganglion cells (ipRGCs). These alterations are associated with axial
