Deep metagenomic characterization of gut microbial community and function in preeclampsia.

Preeclampsia (PE) is a pregnancy complication characterized by severe hypertension and multiple organ damage. Gut microbiota has been linked to PE by previous amplicon sequencing studies. To resolve the PE gut microbiota in a higher taxonomy resolution, we performed shotgun metagenomic sequencing on the fecal samples from 40 early-onset PE and 37 healthy pregnant women. We recovered 1,750 metagenome-assembled genomes (representing 406 species) from the metagenomic dataset and profiled their abundances. We found that PE gut microbiota had enriched in some species belonging to Blautia, Pauljense

Investigating microbiota differences across chronic pancreatitis, influenced by lifestyle and genetic determinants.

BACKGROUND: Chronic pancreatitis (CP) is a multifaceted and irreversible inflammatory condition that impacts both endocrine and exocrine pancreatic functions. Whilst there is evidence of a general intestinal microbial dysbiosis, differences across CP resulting from diverse triggers remains poorly elucidated. METHODS: In this study, we investigated how different CP triggers, namely alcohol-induced CP (AIP) and CFTR-related CP (CFRP), might undermine differences across microbiota structure and predicted function. Faecal samples from 25 CP participants were collected, alongside 9 healthy controls

Members of the human gut microbiota involved in recovery from Vibrio cholerae infection.

Given the global burden of diarrhoeal diseases, it is important to understand how members of the gut microbiota affect the risk for, course of, and recovery from disease in children and adults. The acute, voluminous diarrhoea caused by Vibrio cholerae represents a dramatic example of enteropathogen invasion and gut microbial community disruption. Here we conduct a detailed time-series metagenomic study of faecal microbiota collected during the acute diarrhoeal and recovery phases of cholera in a cohort of Bangladeshi adults living in an area with a high burden of disease. We find that recovery

Associations of Gut Microbiota Composition and Fecal Metabolomic Profiles in Patients With Acquired Premature Ejaculation: A Cross-Sectional Pilot Study.

Premature ejaculation (PE) is a prevalent male sexual disorder with an incompletely understood pathogenesis and limited effective therapeutic strategies. Although the gut microbiota has been implicated in several diseases, its association with PE remains unclear. Metabolomic analyses and metagenomic sequencing were performed to compare the intestinal microbiota profiles between 20 patients with PE and 20 healthy controls, investigating the association between PE and gut microbiota. Comprehensive analysis revealed distinct microbial signatures between PE and control groups. The PE group exhibit

Enterococcus faecium HDRsEf1 induces changes in gut microbiota and metabolites to maintain host health.

Gut microbiota plays a pivotal role in host immunity and overall health. Enterococcus faecium HDRsEf1 (Ef1), a novel potential probiotic, has been reported to enhance immune defense against pathogenic invasion; however, its effects on inflammation and gut microbiota remain poorly understood. This study aimed to evaluate the impact of Ef1 on host health, gut microbiota, and fecal metabolite profiles using DSS-induced colitis in mice and nursery pig models. In mice, Ef1 effectively mitigated growth retardation and organ damage caused by DSS-induced inflammation. In nursery pigs, Ef1 administrati

Stability and iron affinity of Donkey-hide gelatin hydrolysates: Implications for improving iron bioavailability and gut microbiota modulation.

Iron deficiency anemia (IDA) is a prevalent nutritional disorder primarily treated with inorganic iron supplements. However, these interventions are often limited by poor bioavailability and gastrointestinal side effects. Food-derived protein hydrolysates have emerged as promising dietary strategies to enhance iron absorption and mitigate adverse effects. This study evaluated the efficacy of dietary supplementation with Donkey-hide gelatin hydrolysates (DH) in alleviating IDA in a rat model. DH-Fe exhibited superior binding affinity and stability, retaining more soluble iron fraction under div

Cholecystectomy-related gut microbiota dysbiosis exacerbates colorectal tumorigenesis.

Cholecystectomy represents the most prevalent biliary surgical procedure for gallbladder abnormalities. Growing evidence suggests that cholecystectomy is associated with an elevated risk of colorectal cancer. However, the underlying mechanism remains elusive. Here we show that cholecystectomy exacerbates colorectal tumorigenesis in both AOM/DSS and APCmin/+ mice models. Metagenomic sequencing and targeted metabolomics show that cholecystectomy leads to a decrease of Bifidobacterium breve (B. breve) and an increase of Ruminococcus gnavus (R. gnavus), along with increased levels of glycoursodeox

Melatonin Confers Protection Against Multidrug-Resistant Bacterial Infections in Aged Mice Via Microbiota-Derived Butyrate.

Aging is associated with increased susceptibility to bacterial infections, particularly multidrug-resistant (MDR) strains, which often result in antibiotic treatment failure and high mortality rates in the elderly. However, effective preventive and therapeutic strategies remain limited. Herein, we showed that aged mice exhibited higher susceptibility to colistin-resistant Salmonella enterica serotype Typhimurium and methicillin-resistant Staphylococcus aureus compared to young mice. Notably, pre-supplementation with melatonin, a hormone markedly reduced in the aging gut, effectively restricted

Metagenome-assembled microbial genomes from Parkinson's disease fecal samples.

The human gut microbiome composition has been linked to Parkinson's disease (PD). However, knowledge of the gut microbiota on the genome level is still limited. Here we performed deep metagenomic sequencing and binning to build metagenome-assembled genomes (MAGs) from 136 human fecal microbiomes (68 PD samples and 68 control samples). We constructed 952 non-redundant high-quality MAGs and compared them between PD and control groups. Among these MAGs, there were 22 different genomes of Collinsella and Prevotella, indicating high variability of those genera in the human gut environment. Microdiv

Integrated Gut Microbiota and Urine Metabolite Analyses of T2DM with NAFLD Rat Model.

Globally 80% type 2 diabetes mellitus (T2DM) patients suffer nonalcoholic fatty liver disease (NAFLD). The interplay of gut microbiota and endogenous metabolic networks has not yet been reported in the setting of T2DM with NAFLD. As such, this study utilized 16S rRNA gene sequencing to assess the changes in intestinal flora and nuclear magnetic resonance spectroscopy (1H NMR) to identify potential metabolites in a T2DM with NAFLD rat model. Spearman correlation analysis was performed to explore the relationship between gut microbiota and metabolites. Results revealed that among T2DM with NAFLD

Paneth Cell-Derived Lysozyme Defines the Composition of Mucolytic Microbiota and the Inflammatory Tone of the Intestine.

Paneth cells are the primary source of C-type lysozyme, a β-1,4-N-acetylmuramoylhydrolase that enzymatically processes bacterial cell walls. Paneth cells are normally present in human cecum and ascending colon, but are rarely found in descending colon and rectum; Paneth cell metaplasia in this region and aberrant lysozyme production are hallmarks of inflammatory bowel disease (IBD) pathology. Here, we examined the impact of aberrant lysozyme production in colonic inflammation. Targeted disruption of Paneth cell lysozyme (Lyz1) protected mice from experimental colitis. Lyz1-deficiency diminishe

Effect of Bifidobacterium infantis NLS super strain in symptomatic coeliac disease patients on long-term gluten-free diet - an exploratory study.

Bifidobacterium infantis NLS super strain (B. infantis NLS-SS) was previously shown to alleviate gastrointestinal symptoms in newly diagnosed coeliac disease (CD) patients consuming gluten. A high proportion of patients following a gluten-free diet experiences symptoms despite dietary compliance. The role of B. infantis in persistently symptomatic CD patients has not been explored. The aim of the study was to evaluate the effect of B. infantis NLS-SS on persistent gastrointestinal symptoms in patients with CD following a long-term GFD. We conducted a randomised, cross-over, double-blind, place

The human microbiome in cancer: Not just a sidekick anymore.

The human microbiome is increasingly recognized as a dynamic element in cancer biology. Studies across breast, prostate, lung, colorectal, and cervical tumors reveal that microbial communities influence carcinogenesis, immune regulation, and treatment outcomes. When the balance of these microorganisms is altered, inflammation becomes chronic, metabolism is disrupted, and signaling pathways such as NF-κB, IL6-STAT3, and β-catenin are activated. Bacterial metabolites and genotoxins, including colibactin and bile acids, may damage DNA and reshape the epigenetic landscape. Distinct microbial profi

A synbiotic combination of arabinogalactan and Bacteroides thetaiotaomicron ameliorates ulcerative colitis in mice.

Inflammatory bowel disease (IBD) is a chronic disease associated with excessive inflammation and gut microbiota dysbiosis. Synbiotic intervention represents a novel management strategy for IBD. However, there remains a critical need to identify new synbiotic combinations and to elucidate their effects on microbial structure and metabolic profiles. This study investigated the therapeutic potential of a combination of arabinogalactan (AG) and Bacteroides thetaiotaomicron (BT) in a dextran sulfate sodium (DSS)-induced colitis mouse model. AG-BT treatment ameliorated colitis-associated phenotypes,

Structural elucidation of polysaccharide from fresh Areca catechu L. and its mitigation effect on DSS-induced colitis in mice.

Ulcerative colitis (UC) is an inflammatory bowel disease and has emerged as a public health challenge worldwide. The present study aimed to isolate polysaccharide from fresh Areca catechu L. (FAP1a) and to investigate its potential mechanism for ameliorating UC symptoms in a mouse model of UC. The results indicated that the molecular weight of FAP1a was approximately 65 kDa and consisted of arabinose, mannose and galactose, with the main chain probably consisting of →3,6)-β-Gal-(1 → and →2,4,6)-β-Man-(1→. FAP1a effectively attenuated weight loss and colonic tissue damage in mice. FAP1a modulat

The Gut-Joint Connection: Microbiome's Role in Rheumatic Disease.

The human gut microbiome is a pivotal regulator of systemic immunity and a central factor in the pathogenesis of rheumatic diseases. An imbalance in this microbial community, known as "dysbiosis," can trigger and perpetuate autoimmune responses through the "gut-joint axis." A key mechanism underpinning this connection is increased intestinal permeability ("leaky gut"), which facilitates the translocation of microbial products like lipopolysaccharide into the systemic circulation, thereby provoking chronic inflammation. Concurrently, dysbiosis disrupts the critical homeostatic balance between p
