When Long-Lasting Food Selectivity Leads to an Unusual Genetic Diagnosis: A Case Report.

Hereditary fructose intolerance is an autosomal recessive disorder of fructose metabolism caused by catalytic deficiency of aldolase B enzyme [1]. The disease is typically expressed when fructose- and sucrose-containing foods are first introduced in the diet; acute manifestations include nausea, vomiting, abdominal distress, and symptomatic hypoglycemia [1,2]. Chronic fructose ingestion eventually leads to poor feeding, growth retardation and gradual liver and/or renal failure [3,4]. Some patients may remain undiagnosed until adulthood because of a self-protective avoidance of sweet tasting fo

Genetic polymorphism directs IL-6 expression in fibroblasts but not selected other cell types.

Interleukin (IL)-6 blockade is an effective treatment for rheumatoid arthritis (RA), and synovial fibroblasts are a major IL-6 producer in the inflamed joint. We found that human RA and osteoarthritis (OA) synovial fibroblasts derived from independent donors reproducibly segregated into low, medium, and high IL-6 producers, independent of stimulus, cell passage, or disease state. IL-6 expression pattern correlated strongly with total mRNA expression, not mRNA stability, suggesting transcriptional rather than posttranscriptional regulation. High-fibroblast IL-6 expression was significantly asso

Strong genetic evidence for a selective influence of GABAA receptors on a component of the bipolar disorder phenotype.

Despite compelling evidence for a major genetic contribution to risk of bipolar mood disorder, conclusive evidence implicating specific genes or pathophysiological systems has proved elusive. In part this is likely to be related to the unknown validity of current phenotype definitions and consequent aetiological heterogeneity of samples. In the recent Wellcome Trust Case Control Consortium genome-wide association analysis of bipolar disorder (1868 cases, 2938 controls) one of the most strongly associated polymorphisms lay within the gene encoding the GABA(A) receptor beta1 subunit, GABRB1. Renin activity and angiotensin I concentration in genetically selective inbred line of hypertensive mice.

Blood pressure lowering kallikrein-kinin and blood pressure raising renin-angiotensin systems play a major role in the maintenance of normal blood pressure. In a previous study, we have shown that a kallikrein-like prorenin converting enzyme (PRCE C) is elevated in the submandibular gland tissue of a mouse line (BPH) that was genetically selected and inbred for high blood pressure in comparison to normotensive line (BPN) that was derived from the ancestors of BPH line. In the present investigation we wanted to find out if elevated levels of PRCE C were involved in the modulation of tissue (loc

Genetic diversity in RHD and RHCE genes among a selected Kenyan blood donor population.

BACKGROUND: Serologic typing for ABO and RhD is standard in transfusion services, with extended serology and genotyping performed to reduce red cell alloimmunization risk. In Kenya, RH typing is limited to RhD, and genotyping is unavailable. This study used RHD/RHCE genotyping to predict phenotypes and their distribution in a Kenyan blood donor population. STUDY DESIGN AND METHODS: A total of 191 donors (114 D-, 74 D+, and 3 weak D) from the Kenya National Blood Transfusion Service were selected. Next-generation sequencing was performed on DNA extracts using a targeted blood group sequencing

Whole-genome resequencing reveals population genetic structure and selective sweeps in Bombus waltoni.

BACKGROUND: Bombus waltoni is a typical alpine bumblebee species and an important indicator species of the Qinghai-Tibetan Plateau. However, whole-genome studies of bumblebees from this region remain limited. In this study, we investigated the genetic diversity, population structure, and adaptive genomic variation of B. waltoni, providing essential genomic resources for understanding its population genetic structure and signatures of local adaptation. RESULTS: Based on whole-genome SNP data, genetic diversity was relatively similar across B. waltoni populations, with expected heterozygosity (

Investigating the Genetic Association of Selected Candidate Loci with Alopecia Areata Susceptibility in Jordanian Patients.

Background and Objectives: Alopecia areata (AA) is a common cell-mediated autoimmune disease of the hair follicle that results in hair loss patches, affecting males and females of all ages and ethnicities. Although its etiology is not fully understood, AA is hypothesized to have a multifactorial basis with a strong genetic association. This study aims to replicate the genetic association of several risk loci in the Jordanian population for the first time. Materials and Methods: Genomic DNA samples of 152 patients with AA and 150 control individuals were extracted from EDTA blood tubes collecte

Precision patient selection for improved detection of circulating genetically abnormal cells in pulmonary nodules.

Circulating genetically abnormal cells (CACs) have emerged as a promising biomarker for the early diagnosis of lung cancer, particularly in patients with pulmonary nodules. However, their performance may be suboptimal in certain patient populations. This study aimed to refine patient selection to improve the detection of CACs in pulmonary nodules. A retrospective analysis was conducted on 241 patients with pulmonary nodules who had undergone pathological diagnosis through surgical tissue specimens. Utilizing consensus clustering analysis, the patients were categorized into three distinct clust

Brain monoamine levels and behaviour of young and adult chickens genetically selected on feather pecking.

Severe feather pecking (SFP) in chickens is a detrimental behaviour with possibly neurochemical deficits at its base. Recent neurological studies depicted conflicting results on the role of serotonin (5-hydroxytryptamine, 5-HT) and dopamine (DA) in the development and display of feather pecking. We studied brain monoamine levels and behaviour in domestic chickens divergently genetically selected on feather pecking behaviour, the Low Feather Pecking (LFP) and High Feather Pecking (HFP) lines, both at a young age and when adult, to elucidate the role of 5-HT and DA in feather pecking. Genetic disruption of the On visual pathway affects cortical orientation selectivity and contrast sensitivity in mice.

The retina signals stimulus contrast via parallel On and Off pathways and sends the information to higher visual centers. Here we study the role of the On pathway using mice that have null mutations in the On-specific GRM6 receptor in the retina (Pinto LH, Vitaterna MH, Shimomura K, Siepka SM, Balannik V, McDearmon EL, Omura C, Lumayag S, Invergo BM, Brandon M, Glawe B, Cantrell DR, Donald R, Inayat S, Olvera MA, Vessey KA, Kirstan A, McCall MA, Maddox D, Morgans CW, Young B, Pletcher MT, Mullins RF, Troy JB, Takahashi JS. Genetic analysis of metabolic traits in an intercross between body weight-selected chicken lines.

A network of four interacting loci has been reported previously to influence growth in two lines of chickens divergently selected for body weight at 56 days of age. Located on chromosomes 3 (Growth4), 4 (Growth6), 7 (Growth9), and 20 (Growth12), they explained nearly half of the difference in body weight at selection age between the two lines. The original study reported effects on body weight and fat deposition, but no attempts were made to explore the effects of the network on other phenotypes measured in the F(2) population. In this study we conducted further analyses to evaluate the specif

Effects of selected genetic polymorphisms in xeroderma pigmentosum complementary group D on gastric cancer.

DNA repair capacity (DRC) can be altered based on sequence variations in DNA repair genes, which may result in cancer susceptibility. The current study was to evaluate the association between genetic polymorphisms, including associated haplotypes of xeroderma pigmentosum complementary group D (XPD), and individual susceptibility to gastric cancer. Two-hundred-eight patients with gastric cancer and 339 healthy controls were enrolled in this study. Their genomic DNA was extracted from peripheral blood leukocytes. The genotypes at exon 6, 10 and 23 were identified by polymerase chain reaction (PC

Direct comparison of selected methods for genetic categorisation of Cryptosporidium parvum and Cryptosporidium hominis species.

A study was undertaken to compare the performance of five different molecular methods (available in four different laboratories) for the identification of Cryptosporidium parvum and Cryptosporidium hominis and the detection of genetic variation within each of these species. The same panel of oocyst DNA samples derived from faeces (n=54; coded blindly) was sent for analysis by: (i) DNA sequence analysis of a fragment of the HSP70 gene; (ii) DNA sequence analysis and the ssrRNA gene in laboratory 1; (iii) single-strand conformation polymorphism analysis of part of the ssrRNA; (iv) SSCP analysis

Genetic screens and selections for small molecules based on a synthetic riboswitch that activates protein translation.

Genetic selection provides the most powerful method to assay large libraries of biomolecules for function. However, harnessing the power of genetic selection for the detection of specific, nonendogenous small-molecule targets in vivo remains a significant challenge. The ability to genetically select for small molecules would provide a reaction-independent mechanism to clone biosynthesis genes from large DNA libraries and greatly facilitate the exploration of large libraries of mutant enzymes for improved synthetic capabilities including altered substrate specificities and enhanced regio- or st

Dimensions and disorders of adolescent adjustment: a quantitative genetic analysis of unselected samples and selected extremes.

One of the fundamental questions for developmental psychopathology concerns the etiological links between the normal and abnormal. To what extent do disorders differ quantitatively or qualitatively from variation in the normal range? Genetic research on the normal and the abnormal differs in terms of concepts, methods, statistics, and target audiences. An approach, referred to as "DF" analysis, provides a framework for integrating these two worlds of genetic analysis. We applied traditional correlational analyses as well as DF (DeFries & Fulker, 1985) analyses to mother and father ratings of a

Resistance to listeriosis in two lines of mice genetically selected for high and low antibody production.

Infection by the intracellular parasite Listeria monocytogenes was studied in two inbred lines of mice genetically selected for high and low antibody production against xenogeneic red blood cells. It was revealed that, during the early non-specific phase of infection, bacterial growth in tissues was significantly enhanced in high responder (HR) mice, as opposed to low responder (LR) mice. This is interpreted as the in vivo expression of a genetic impairment of the bactericidal activity of resident macrophages in this line of mice. After Day 2 of infection, the kinetics of bacterial growth in t

Comprehensive Clinical Genetics, Molecular and Pathological Evaluation Efficiently Assist Diagnostics and Therapy Selection in Breast Cancer Patients with Hereditary Genetic Background.

Using multigene panel testing for the diagnostic evaluation of patients with hereditary breast and ovarian cancer (HBOC) syndrome often identifies clinically actionable variants in genes with varying levels of penetrance. High-penetrance genes (BRCA1, BRCA2, CDH1, PALB2, PTEN, STK11, TP53) inform specific clinical surveillance and therapeutic decisions, while recommendations for moderate-penetrance genes (ATM, BARD1, BRIP1, CHEK2, MLH1, MSH2, MSH6, PMS2, EPCAM, NF1, RAD51C, RAD51D) are more limited. A detailed disease history, including pedigree data, helps formulate the most appropriate and p

Genetic polymorphism and natural selection of the erythrocyte binding antigen 175 region II in Plasmodium falciparum populations from Myanmar and Vietnam.

Plasmodium falciparum erythrocyte binding antigen 175 (PfEBA-175) plays essential role in erythrocyte invasion by the parasite and is a leading vaccine candidate. However, its genetic diversity in global isolates is a concern in developing an universal vaccine incorporating this protein. This study aimed to investigate genetic polymorphisms and natural selection of pfeba-175 region II (RII) in Myanmar and Vietnam P. falciparum isolates. Vietnam pfeba-175 RII displayed a low genetic polymorphism, while Myanmar pfeba-175 RII showed high levels of genetic diversity across the region. Point mutati
