GLP-1R agonist semaglutide protects human forebrain organoids from cholesterol-induced neurotoxicity.

INTRODUCTION: Dysregulated cholesterol metabolism represents a critical metabolic stressor in the central nervous system, contributing to neuronal injury across multiple neurological conditions, including Alzheimer's disease (AD). While glucagon-like peptide-1 receptor (GLP-1R) agonists show neuroprotective potential, their capacity to directly mitigate cholesterol-induced metabolic stress in human neural cells remains to be established. METHODS: Here, we modeled metabolic stress using human induced pluripotent stem cell-derived forebrain organoids exposed to cholesterol overload. The protect

Semaglutide and Its Potential Hepatoprotective Effects Against Acute Drug-Induced Liver Injury.

Drug-induced liver injury (DILI) is hepatic damage caused by medications, illegal drugs, herbal products, or dietary supplements and is related to inflammation, oxidative stress, and mitochondrial dysfunction. Manifestations can vary from isolated liver enzyme elevation to fulminant hepatic failure. As many well-established therapeutic options do not exist, it is crucial to explore new hepatoprotective agents. Semaglutide is a glucagon-like peptide-1 receptor agonist (GLP-1 RA), primarily used as an antidiabetic and anti-obesity agent, which manifests pleiotropic effects through activation of

Semaglutide ameliorates osteoarthritis progression through a weight loss-independent metabolic restoration mechanism.

Metabolic disorders have been recognized as a major contributor to the occurrence and progression of osteoarthritis (OA). Identifying novel therapeutic agents to ameliorate the progression of OA with metabolic disorder is crucial. In this study, we demonstrate that semaglutide (SG), a glucagon-like peptide-1 receptor (GLP-1R) agonist, exhibits strong chondroprotective effects in an OA mouse model with obesity, as evidenced by reduced pathological changes, including cartilage degeneration, osteophyte formation, synovial lesion, and pain sensitivity. A randomized pilot clinical study (ChiCTR2200

Late-life semaglutide treatment slows ageing and extends lifespan in female mice.

Pharmacological glucagon-like peptide-1 receptor (GLP-1R) activation reduces food intake and is an effective therapy for type 2 diabetes and obesity1. The use of GLP-1 medicines has revealed pleiotropic beneficial effects beyond glucose and weight control2-5, but little is known about the underlying basis of the pleiotropic effects. Here, treatment of 20-month-old female C57BL/6 mice with the GLP-1R agonist semaglutide for 3 months improved physiological function, attenuated hallmarks of ageing and modulated nutrient sensors and conserved genetic regulators of ageing. Continued treatment exten

Semaglutide-Induced Hepatic Injury: A Rare Case of Drug Induced Liver Injury.

Semaglutide, a glucagon-like peptide-1 (GLP-1) receptor agonist, is widely used for type 2 diabetes mellitus and has demonstrated hepatoprotective effects. However, this case highlights a rare instance of possible drug-induced liver injury (DILI) temporally linked to its use. A middle-aged male with well-controlled diabetes, social alcohol use, and no history of liver disease presented with asymptomatic elevations in alanine transaminase (ALT) and aspartate aminotransferase (AST). Extensive workup, including undetectable blood ethanol level, normal viral hepatitis panel, and unremarkable liver

Semaglutide treatment reverses HFD induced hippocampal microglia activation and improves cognitive dysfunction.

Long-term high-fat diets (HFD) induce obesity, neuroinflammation, and cognitive decline, increasing Alzheimer's disease (AD) risk. This study explores whether Semaglutide, a GLP-1 receptor agonist, mitigates these effects by modulating microglia via IGFBPL-1 and the PI3K/AKT pathway. In HFD-fed C57/BL6 mice, Semaglutide improved cognitive function, reduced hippocampal microglia activation, and decreased AD-like pathology (phospho-Tau, Aβ). IGFBPL-1, a neuroprotective factor downregulated by HFD and restored by Semaglutide. Direct IGFBPL-1 supplementation replicated Semaglutide's benefits, whil

Gastric ultrasound in patients receiving semaglutide: a prospective, multicentre, matched control study.

INTRODUCTION: Delayed gastric emptying is a known effect of glucagon-like peptide-1 receptor agonists, such as semaglutide. While tachyphylaxis has been reported, recent clinical data indicate an increased risk of residual gastric content on pre-operative gastric ultrasound in patients who are fasted. However, the limited available data do not control for several important potential confounders. METHODS: We conducted a prospective, multicentre, matched case-control study, enrolling patients receiving semaglutide treatment for diabetes mellitus or weight loss and having elective surgery. Sodium glycocholate liposome encapsulated semaglutide increases oral bioavailability by promoting intestinal absorption.

The aim of this study was to prepare sodium glycocholate liposomes (SGC-Lip) encapsulating semaglutide (Sml) to improve oral bioavailability and better exert hypoglycemic effect. In this paper, SGC-Lip was prepared by reverse-phase evaporation method with particle size around 140 nm, potential around -27 mV, rounded morphology and better stability. The hypoglycemic and intestinal uptake effects of SGC-Lip and cholesterol-containing liposomes (CH-Lip) were comparatively investigated in rats, and the oral safety of SGC-Lip was examined by cytotoxicity assay. The results indicate that SGC-Lip can

Nanoparticles targeting the intestinal Fc receptor enhance intestinal cellular trafficking of semaglutide.

Semaglutide is the first oral glucagon-like peptide-1 (GLP-1) analog commercially available for the treatment of type 2 diabetes. In this work, semaglutide was incorporated into poly(lactic-co-glycolic acid)-poly(ethylene glycol) (PLGA-PEG) nanoparticles (NPs) to improve its delivery across the intestinal barrier. The nanocarriers were surface-decorated with either a peptide or an affibody that target the human neonatal Fc receptor (hFcRn), located on the luminal cell surface of the enterocytes. Both ligands were successfully conjugated with the PLGA-PEG via maleimide-thiol chemistry and there

Subcutaneous Semaglutide during Breastfeeding: Infant Safety Regarding Drug Transfer into Human Milk.

Postpartum mothers and their healthcare providers often face the challenge of limited data regarding the safety of drug therapies during lactation. Pregnancy can lead to sustained weight gain, and obesity can negatively impact both physical and psychological well-being. The introduction of GLP-1 agonists to augment weight loss has become a topic of interest for many postpartum mothers. Our study aims to investigate the transmission of semaglutide into human milk in the first steps to ensure the safety and health of both lactating mothers and their breastfed infants. Semaglutide quantification

Semaglutide protects against diabetes-associated cardiac inflammation via Sirt3-dependent RKIP pathway.

BACKGROUND AND PURPOSE: Glucagon-like peptide-1 receptor agonists (GLP-1RAs) exert cardiovascular benefits in diabetic patients, but the underlying mechanisms remain incompletely understood. Semaglutide, a novel long-acting GLP-1RA, has shown a reduced risk of cardiovascular events. Based on these results, we investigated the therapeutic potential of semaglutide in diabetic cardiomyopathy and sought to elucidate the underlying mechanisms. EXPERIMENTAL APPROACH: Mice with diabetes induced by high-fat diet/streptozotocin were treated with semaglutide. The mechanisms underlying the cardioprotect
