Forskolin-loaded carboxymethyl chitosan and silk nonofiber hydrogels composite scaffolds improve skin regeneration.

Hydrogel is a kind of carrier of powerful therapeutic bioactive molecules. Here, carboxymethyl chitosan-silk nanofiber (CCSF) composite hydrogels loaded with a small molecule compound, Forskolin, were prepared to achieve sustained release of the small molecule drug for promoting wound healing. In vitro, investigations confirmed that the innovative composite dressing is non-toxic and cyto-compatible. Specifically, it can encourage the proliferation of human umbilical vein endothelial cells (HUVECs) and mouse dermis fibroblasts, and inhibit the accumulation of intracellular reactive oxygen speci

Fabrication of highly durable flame-retardant silk textiles by depositing phosphorus/nitrogen compounds.

The development of highly durable flame-retardant (FR) silk textiles has significant practical implications. This study presents an innovative in situ deposition technique for imparting durable functionality to silk textiles. Insoluble FR compounds were formed within silk fibers by reacting tetrakis(hydroxymethyl) phosphine sulfate and dicyandiamide with a phosphate ester compound. The chemical structure of the insoluble FR compounds, surface morphology, physical properties, combustion behavior, washing resistance and flame retardancy mechanism of modified silk fabrics were explored. The modif

Osteoimmunity-Regulating biospun 3D silk scaffold for bone regeneration in critical-size defects.

INTRODUCTION: Silk-based biomaterials have received a great deal of attention in tissue engineering research for bone repair. Current silk-based materials are typically derived from silk protein solutions, but the limited solubility and solution stability of silk protein solution, coupled with problems such as high preparation cost and low productivity, which severely restrict the application of silk-based materials. OBJECTIVE: To address the challenges associated with the complex extraction process and inferior mechanical properties of silk protein or silk fiber-based materials in bone scaff

Peptide-Laden Silk Fibroin Microneedles Modulate Inflammation and Promote Myocardial Repair In Vitro.

Myocardial infarction is a major cause of death and disability worldwide and poses a serious threat to global public health. Chronic ischemia and hypoxic environment induce excessive inflammatory response, leading to heart failure and even death. Therefore, inhibiting excessive inflammation and promoting tissue repair are important for myocardial repair. Based on this, natural polymeric Silk Fibroin Methacryloyl microneedles loaded with Tc peptide drugs (SilMA MNs) were fabricated for the MI therapy by photocuring and micro-molding techniques, which facilitate the protection of Tc peptides tha

Hemostatic and Ultrasound-Controlled Bactericidal Silk Fibroin Hydrogel via Integrating a Perfluorocarbon Nanoemulsion.

Excessive blood loss and infections are the prominent risks accounting for mortality and disability associated with acute wounds. Consequently, wound dressings should encompass adequate adhesive, hemostatic, and bactericidal attributes, yet their development remains challenging. This investigation presented the benefits of incorporating a perfluorocarbon nanoemulsion (PPP NE) into a silk-fibroin (SF)-based hydrogel. By stimulating the β-sheet conformation of the SF chains, PPP NEs drastically shortened the gelation time while augmenting the elasticity, mechanical stability, and viscosity of th

Black phosphorus/silk fibroin films hamper filamentous and invasive growth of Candida albicans.

The ability of fungi and bacteria to form biofilms on surfaces poses a serious threat to health and a problem in industrial settings. In this work, we investigated how the surface stiffness of silk fibroin (SF) films is modulated by the interaction with black phosphorus (BP) flakes, quantifying the morphogenesis of C. albicans cells. Raman and infrared (IR) spectroscopies, along with scanning transmission electron microscopy, allowed us to quantify the thickness and diameter of BP flakes dispersed in the SF matrix (e.g., 5.5 nm in thickness and 20 μm in diameter), as well as an increase in bet

Wild Silkworm Cocoon Waste Conversion into Tough Regenerated Silk Fibers by Solution Spinning.

Wild silkworm silk fibers have garnered attention owing to their softness, natural color, lightweight, and excellent mechanical properties. Because most wild silkworm cocoons obtained are pierced or dirty after the eclosion process, it is difficult to reel the long filament from the pierced cocoons to use as textile materials. Therefore, damaged wild silkworm cocoons are typically removed during the industrial process. Artificial silk spinning has been developed to transform domesticated silkworm silk solutions into regenerated silk fibers. However, regenerated fibers derived from wild silkwor

Insights into the material properties of dragline spider silk affecting Schwann cell migration.

Dragline silk of Trichonephila spiders has attracted attention in various applications. One of the most fascinating uses of dragline silk is in nerve regeneration as a luminal filling for nerve guidance conduits. In fact, conduits filled with spider silk can measure up to autologous nerve transplantation, but the reasons behind the success of silk fibers are not yet understood. In this study dragline fibers of Trichonephila edulis were sterilized with ethanol, UV radiation, and autoclaving and the resulting material properties were characterized with regard to the silk's suitability for nerve

Spatiotemporal Regulation of Injectable Heterogeneous Silk Gel Scaffolds for Accelerating Guided Vertebral Repair.

Osteoporotic vertebral fracture is jeopardizing the health of the aged population around the world, while the hypoxia microenvironment and oxidative damage of bone defect make it difficult to perform effective tissue regeneration. The balance of oxidative stress and the coupling of vessel and bone ingrowth are critical for bone regeneration. In this study, an injectable heterogeneous silk gel scaffold which can spatiotemporally and sustainedly release bone mesenchymal stem cell-derived small extracellular vesicles, HIF-1α pathway activator, and inhibitor is developed for bone repair and verteb

PREDICT: Precise deployment of Silk Vista Baby in confined territory: A technical note.

Flow diverters (FD) have become increasingly useful in treating complex intracranial aneurysms, particularly wide-necked and recurrent aneurysms. Their use has progressively expanded to smaller vessels beyond the circle of Willis (CoW), and Silk Vista Baby (SVB) is one such low-profile FD which stands out because of deliverability through a 0.017″ microcatheter and smoother navigability. Precise deployment of SVB, specifically, the proximal end, can be challenging in certain anatomical locations when the proximal landing zone is very short, limited by vessel bifurcation or important branches a

Eco-friendly silk fibroin/tannic acid coacervates for humid and underwater wood adhesives.

Bioadhesives derived from biomass are steadily gaining spotlight as substitutes for formaldehyde-based resins in the adhesive industry. However, there is a need to develop novel water-resistant bioadhesives with high adhesive and cohesive strengths because the currently available biomaterial-based adhesives have low mechanical strength. In this study, a complex coacervate was prepared easily by mixing silk fibroin and tannic acid to produce a bioadhesive with high adhesive and cohesive strengths as well as water resistance. The silk fibroin-tannic acid coacervate adhered well to various substr

Recombinant Spider Silk Gels Derived from Aqueous-Organic Solvents as Depots for Drugs.

Hydrogels are widely used in various biomedical applications, as they cannot only serve as materials for biofabrication but also as depots for the administration of drugs. However, the possibilities of formulation of water-insoluble drugs in hydrogels are rather limited. Herein, we assembled recombinant spider silk gels using a new processing route with aqueous-organic co-solvents, and the properties of these gels could be controlled by the choice of the co-solvent. The presence of the organic co-solvent further enabled the incorporation of hydrophobic drugs as exemplarily shown for 6-mercapto

Self-Healing, Self-Adhesive Silk Fibroin Conductive Hydrogel as a Flexible Strain Sensor.

Flexible and wearable hydrogel strain sensors have attracted tremendous attention for applications in human motion and physiological signal monitoring. However, it is still a great challenge to develop a hydrogel strain sensor with certain mechanical properties and tensile deformation capabilities, which can be in conformal contact with the target organ and also have self-healing properties, self-adhesive capability, biocompatibility, antibacterial properties, high strain sensitivity, and stable electrical performance. In this paper, an ionic conductive hydrogel (named PBST) is rationally desi

Membranes Prepared from Recombinant RGD-Silk Fibroin as Substrates for Human Corneal Cells.

A recombinant formulation of silk fibroin containing the arginine-glycine-aspartic acid (RGD) cell-binding motif (RGD-fibroin) offers potential advantages for the cultivation of corneal cells. Thus, we investigated the growth of corneal stromal cells and epithelial cells on surfaces created from RGD-fibroin, in comparison to the naturally occurring Bombyx mori silk fibroin. The attachment of cells was compared in the presence or absence of serum over a 90 min period and analyzed by quantification of dsDNA content. Stratification of epithelial cells on freestanding membranes was examined by con

Silk fibroin scaffolds loaded with angiogenic genes in adenovirus vectors for tissue regeneration.

Vascularization remains a critical challenge in dermal tissue regeneration. In this study, a vascular endothelial growth factor (VEGF165) and angiopoietin-1 (Ang-1) dual gene coexpression vector that encoded green fluorescent protein (GFP) was constructed from an arginine-glycine-aspartic acid-modified adenovirus. Silk fibroin (SF) scaffolds loaded with adenovirus vectors were fabricated by freeze-drying method. In vitro, the human endothelial-derived cell line EA.hy926 was infected with adenovirus vectors and then expressed GFP, secreted VEGF165 and Ang-1, and promoted cell proliferation effe

Precisely printable and biocompatible silk fibroin bioink for digital light processing 3D printing.

Although three-dimensional (3D) bioprinting technology has gained much attention in the field of tissue engineering, there are still several significant engineering challenges to overcome, including lack of bioink with biocompatibility and printability. Here, we show a bioink created from silk fibroin (SF) for digital light processing (DLP) 3D bioprinting in tissue engineering applications. The SF-based bioink (Sil-MA) was produced by a methacrylation process using glycidyl methacrylate (GMA) during the fabrication of SF solution.
