Antiviral activity of silymarin against chikungunya virus.

The mosquito-borne chikungunya virus (CHIKV) causes chikungunya fever, with clinical presentations such as severe back and small joint pain, and debilitating arthritis associated with crippling pains that persist for weeks and even years. Although there are several studies to evaluate the efficacy of drugs against CHIKV, the treatment for chikungunya fever is mainly symptom-based and no effective licensed vaccine or antiviral are available. Here, we investigated the antiviral activity of three types of flavonoids against CHIKV in vitro replication. Three compounds: silymarin, quercetin and kae

Alginate-based ferrofluid and magnetic microsphere thereof.

The Fe(3)O(4) ferrofluids have been prepared using sodium alginate (Na-AL) as a stabilizing agent. The alginate can prevent the aggregation of magnetic nanoparticles and hence contributed to higher stability for the ferrofluids. Furthermore, the alginate component in the ferrofluids was crosslinked by Ca(2+) to produce magnetic microspheres. The swelling behavior of magnetic microspheres showed a pH-dependence, and hence determined the drug release process under various pH conditions. The presence of the Fe(3)O(4) nanoparticles made the magnetic microspheres swell more easily. Meanwhile, the s

Acute liver acetaminophen toxicity in rabbits and the use of antidotes: a metabonomic approach in serum.

The metabonomic approach has been widely used in toxicology to investigate mechanisms of toxicity. In the present study alterations in the metabolic profiles, monitored by (1)H-NMR spectroscopy, on serum samples in acetaminophen (APAP)-induced liver injury in rabbits were examined. Furthermore, the effect of the established antidote N-acetylcysteine (NAC) and the proposed antidotes silybinin (SIL), cimetidine (CIM) and SIL/CIM was also investigated. A single dose of APAP (2 g kg(-1) b.w., i.g.) was administered to rabbits and APAP combined with the antidotes SIL, CIM and NAC. Animals were sacr

The flavonoid silibinin decreases glucose-6-phosphate hydrolysis in perfused rat hepatocytes by an inhibitory effect on glucose-6-phosphatase.

BACKGROUND/AIMS: The flavonoid silibinin has been reported to be beneficial in several hepatic disorders. Recent evidence also suggests that silibinin could be beneficial in the treatment of type 2 diabetes, owing to its anti-hyperglycemic properties. However, the mechanism(s) underlying these metabolic effects remains unknown. METHODS: The effects of silibinin on liver gluconeogenesis were studied by titrating hepatocytes from starved rats with sub-saturating concentrations of various exogenous substrates in a perifusion system. Hepatocytes from fed rats were also used to investigate glycoge

Silibinin alleviates lung fibrosis by targeting Annexin A6 to suppress endoplasmic reticulum stress and oxidative stress.

INTRODUCTION: Endoplasmic reticulum (ER) stress and oxidative stress are closely linked in driving the TGF-β1-mediated proliferation and myofibroblast differentiation of lung fibroblasts during pulmonary fibrosis. Silibinin (SIL), a bioactive compound derived from Silybum marianum l., has been proven to suppress inflammatory responses in a bleomycin (BLM) mouse model, but the protective mechanisms and specific molecular target through which SIL acts in pulmonary fibrosis remain unclear. OBJECTIVES: This study aimed to identify the direct molecular target of SIL, and investigate the mechanisms

Evaluation of anti-cancer potency of silibinin on murine renal carcinoma RenCa cells in an animal model with an intact immune system.

Silibinin is a flavonoid extract isolated from milk thistle and has been proved to be a promising chemotherapeutic drug for cancer. However, most of those studies were performed on the human cancer cells, where the effects of silibinin could only be observed on an animal model with a deficient immune system. RenCa cells were isolated from a murine spontaneous renal cell carcinoma, which resembles many features of human renal cell carcinoma, and have been used to establish animal models with a sound immune response. Herein, we investigated the anti-cancer effects of silibinin on RenCa cells, re

Protective role of silibinin against myocardial ischemia/reperfusion injury-induced cardiac dysfunction.

Silibinin is a traditional medicine and utilized for liver protection with antioxidant, anti-inflammation and anti-apoptosis properties. However, its role in myocardial I/R injury and the mechanism involved is currently unknown. In the present study, Silibinin treatment improves cardiac function and limits infarct size, and subsequently inhibits fibrotic remodeling in mice with myocardial I/R injury. Mechanistically, silibinin reduces cardiomyocytes apoptosis, attenuates mitochondrial impairment and endoplasmic reticulum (ER) stress, alleviates ROS generation, neutrophil infiltration and cytok

Isolated Silymarin Flavonoids Increase Systemic and Hepatic Bilirubin Concentrations and Lower Lipoperoxidation in Mice.

Bilirubin is considered to be one of the most potent endogenous antioxidants in humans. Its serum concentrations are predominantly affected by the activity of hepatic bilirubin UDP-glucuronosyl transferase (UGT1A1). Our objective was to analyze the potential bilirubin-modulating effects of natural polyphenols from milk thistle (Silybum marianum), a hepatoprotective herb. Human hepatoblastoma HepG2 cells were exposed to major polyphenolic compounds isolated from milk thistle. Based on in vitro studies, 2,3-dehydrosilybins A and B were selected as the most efficient compounds and applied either

The cytotoxicity of OPA-modified CdSe/ZnS core/shell quantum dots and its modulation by silibinin in human skin cells.

Quantum dots (QDs) have emerged as alternative or complementary tools to organic fluorescent dyes currently used in bioimaging. QDs hold several advantages over conventional fluorescent dyes including greater photostability and a wider range of excitation/emission wavelengths. However, recent work suggests that QDs exert deleterious effects on cellular processes which could obscure bioassay results. This study examined the toxicity of octylamine-poly(acrylic acid) (OPA) modified CdSe/ZnS quantum dots (QDs) and a pharmacological means of preventing QD-induced cell death. Cell viability and the

Effect of silibinin on the pharmacokinetics of pyrazinamide and pyrazinoic acid in rats.

Pyrazinamide (PZA) is widely used in combination with other drugs in chemotherapy for tuberculosis. However, the dose-related liver injury is the main adverse effect of PZA and its metabolite [pyrazinoic acid (PA)]. Silibinin is the main flavonoid extracted from milk thistle (Silybum marianum), and it displays hepatoprotective properties. This study investigates the pharmacokinetics of PZA and PA and their interaction with silibinin in rats. The parallel study design was divided into six groups: PZA alone, PZA + long-term silibinin exposure, PZA + concomitant short-term silibinin exposure, PA

Development of pH-responsive gelatin/PVP nanogel by gamma radiation for controlled delivery of silibinin.

Silibinin (SB) is a natural polyphenolic flavonoid with recognized health and therapeutic potential. SB has poor aqueous solubility, which limits its bioavailability and therapeutic effectiveness. To overcome these limitations, a pH-responsive gelatin/polyvinylpyrrolidone (Gel/PVP) nanogel was synthesized via γ-irradiation (5 kGy) for controlled SB delivery. SB/PVP solid dispersions were first prepared by solvent evaporation to enhance dissolution, then incorporated into the preformed Gel/PVP nanogel in the presence of N-hydroxysuccinimide (NHS), yielding the SB/Gel/PVP formulation. Silibinin Attenuates Experimental Periodontitis by Downregulation of Inflammation and Oxidative Stress.

Periodontitis is an oral microbiota-induced inflammatory disease, in which inflammation and oxidative stress play a critical role. Silibinin (SB), a Silybum marianum-derived compound, exhibits strong anti-inflammatory and antioxidative properties. We adopted a rat ligature-induced periodontitis model and a lipopolysaccharide- (LPS-) stimulated human periodontal ligament cells (hPDLCs) model to evaluate the protective effects of SB. In the in vivo model, SB reduced alveolar bone loss and apoptosis of PDLCs in the periodontal tissue. SB also maintained the expression of nuclear factor-E2-related

Hepatoprotective effect of 2,3-dehydrosilybin on carbon tetrachloride-induced liver injury in rats.

The aim of this study was to investigate the protective effect of 2,3-dehydrosilybin (DHS) against carbon tetrachloride (CCl(4))-induced liver injury in rats. Administration of DHS significantly attenuated the levels of serum aspartate aminotransferase, alanine aminotransferase, and liver lipid peroxidation in CCl(4)-treated rats. Moreover, we showed that DHS prevented DNA damage and decreased the protein levels of γ-H2AX, which is a specific DNA damage marker, in CCl(4)-treated rat livers. DHS also markedly increased the activity of antioxidant enzymes, such as superoxide dismutase, catalase,
