Penetration of chloroform, trichloroethylene, and tetrachloroethylene through human skin.

In vitro dermal absorption was measured for three volatile organic compounds in dilute aqueous solution through freshly prepared and previously frozen human skin. The permeability coefficients at 26 degrees C for chloroform (0.14 cm/h) and trichloroethylene (0.12 cm/h) were similar but much larger than that for tetrachloroethylene (0.018 cm/h). Storage of the skin at -20 degrees C did not significantly affect the penetration of these chemicals. The dermal absorption of chloroform through freshly prepared human skin was not changed significantly by pretreatment of the skin with commonly used co

Domain Knowledge Constrained Symbolic Regression for Optimising Dermal Drug Formulations.

PURPOSE: Interpretable dynamic models that remain physically admissible under extrapolation are essential for dermal formulation optimisation. Mechanistic models are often labour-intensive to derive, whereas unconstrained data-driven models may violate physical constraints and degrade beyond observed conditions. METHODS: We developed a domain knowledge constrained symbolic regression (DKC-SR) framework to discover compact, interpretable dynamic models for cumulative release directly from time-series experiments. Domain knowledge was embedded through a restricted operator set and feasibility c

Incorporating a dermal absorption route into high throughput toxicokinetic modeling.

BACKGROUND: Dermal absorption of chemicals represents an important route of exposure in pharmaceutical, occupational, and environmental settings. Thousands of chemicals with little toxicity or toxicokinetic (TK) data are in use. However, in many cases, in vitro bioactivity data are available. While it is not feasible to collect in vivo TK data, TK may be estimated using high-throughput methods. OBJECTIVE: This study developed a generalized physiologically-based TK (PBTK) dermal exposure model for in vitro-in vivo extrapolation (IVIVE). This model estimates dermal exposures that result in syst

Size-dependent transdermal absorption of cadmium sulphide quantum dots: an ex-vivo study using Franz diffusion cells.

Cadmium sulphide (CdS) quantum dots (QDs) are semiconductor nanomaterials extensively used in optoelectronic and biomedical applications, raising concerns regarding potential health hazards associated with dermal exposure. This study investigated the size-dependent transdermal absorption of N-acetylcysteine (NAC)-capped CdS QDs using an ex-vivo human skin model and Franz diffusion cells. Three QD sizes (3.4, 5.8 and 6.7 nm) were synthesised in aqueous solution, characterised by UV-Vis and fluorescence spectroscopy, and applied to intact and damaged human skin. Cadmium permeation into the recep

Are lateral lipid-phase diffusion coefficients pertinent to dermal absorption?

The skin's outer barrier layer, the stratum corneum (SC), is comprised of keratin-rich corneocytes, connected by proteinaceous corneodesmosomes and separated by nonpolar, lamellar lipids. The lipids are inherently anisotropic due to their lamellar organization. There is growing acceptance that this structural anisotropy carries over into transport properties including diffusive mass transport, which is consequently characterized by two diffusion coefficients, D‖lipfor lateral diffusion along the lipid lamellae and D⊥lipfor transverse diffusion across them. Many microscopic models of SC transpo

Study on the synthesis, physicochemical properties and transdermal absorption of scutellarein prodrugs.

In this research, we aimed to improve the lipid solubility and transdermal absorbability of scutellarein through a prodrug strategy, and then improve its efficacy for treating psoriasis. Firstly, we designed and synthesized 19 prodrugs of scutellarein, including AC (alkylcarbonyl), AAC (alkylaminocarbonyl) and AOC (alkyloxycarbonyl) series. Then, we selected three prodrugs (AC-4, AAC-2 and AOC-SEC-4) with proper log P and favorable solubility for subsequent transdermal absorption study, and the results demonstrated the three prodrugs significantly improved the transdermal flux and the drug ret

Finite Dose In Vitro Permeation Testing: Significance of Occluding the Donor compartment, a Case study.

The evaporation of the solvent induces significant changes in formulation, directly impacting its performance. The performance of topical products is determined by the interplay between their inherent quality attributes and the transformations that occur due to solvent evaporation when applied to the skin in clinically relevant doses. To accurately assess, in vivo performance, it is advisable to apply smaller doses to the skin and keep the donor compartment open to enable evaporation of solvents while carrying out in vitro permeation tests. This manuscript highlights the critical role of solve

Establishing a General Atomistic Model for the Stratum Corneum Lipid Matrix Based on Experimental Data for Skin Permeation Studies.

Understanding the permeation of drugs through the intercellular lipid matrix of the stratum corneum layer of skin is crucial for effective transdermal delivery. Molecular dynamics simulations can provide molecular insights into the permeation process. In this study, we developed a new atomistic model representing the multilamellar arrangement of lipids in the stratum corneum intercellular space for permeation studies. The model was built using ceramides in extended conformation as the backbone along with free fatty acids and cholesterol. The properties of the equilibrated model were in agreeme

Cutaneous pharmacokinetics of a volatile drug post-application to the skin.

This research explored whether in vitro release and skin permeation tests, combined with in vitro and in vivo stratum corneum (SC) sampling, can quantify the "input rate" of a volatile drug into and through the skin. Two topical methyl salicylate (MeSA) products were studied. Drug release from the formulations across a silicone membrane was similar, with ~ 65% of the drug load being released in 6 h. In vitro porcine skin permeation tests showed that ~ 50% of the dose crossed the skin in 24 h from both products. In SC sampling experiments, the mass of MeSA in the SC at one 'uptake' and three 'c

Development of nano-liposomal human growth hormone as a topical formulation for preventing uvb-induced skin damage.

Due to its involvement in skin maintenance and repair, topical administration of recombinant human growth hormone (rhGH) is an interesting strategy for therapeutic purposes. We have formulated and characterized a topical rhGH-loaded liposomal formulation (rhGH-Lip) and evaluated its safety, biological activity, and preventive role against UVB-induced skin damage. The rhGH-Lip had an average size and zeta potential of 63 nm and -33 mV, respectively, with 70 % encapsulation efficiency. The formulation was stable at 4 °C for at least one year. The SDS-PAGE and circular dichroism results showed no

Immediate dry decontamination using efficient absorbent materials is beneficial following skin exposure to low-volatile toxic chemicals.

In a chemical mass casualty incident requiring skin decontamination, dry removal using absorbent materials may be beneficial to enable immediate decontamination. The efficacy of absorbent materials has therefore been evaluated, alone or procedures including both dry and wet decontamination, following skin exposure to two low volatile toxic chemicals using an in vitro human skin penetration model. Additionally, removal using active carbon wipes was evaluated with or without the Dahlgren Decon solution. All dry decontamination procedures resulted in a significantly decreased skin penetration rat

Breaking boundaries: the advancements in transdermal delivery of antibiotics.

Transdermal drug delivery systems (TDDS) for antibiotics have seen significant advances in recent years that aimed to improve the efficacy and safety of these drugs. TDDS offer many advantages over other conventional delivery systems such as non-invasiveness, controlled-release pattern, avoidance of first-pass metabolism. The objective of this review is to provide an overview on the recent advances in the TDDS of different groups of antibiotics including β-lactams, tetracyclines, macrolides, and lincosamides, utilized for their effective delivery through the skin and to explore the challenges

Desonide Nanoemulsion Gel for Transdermal Absorption Drug Delivery: Pharmacodynamic and Safety Evaluation.

BACKGROUND: When administered transdermally, desonide is ineffective due to its poor solubility. As a new transdermal delivery system, nanoemulsion gel has demonstrated significant advantages for drug delivery over conventional formulations. We have established desonide nanoemulsion gel (DES NE gel) for better transdermal absorption, but its efficacy and safety still need to be evaluated. This study aims to provide additional evidence demonstrating the improved pharmacodynamics and safety of transdermal delivery of Desonide via nanoemulsion gel. METHODS: Pharmacodynamics and safety of Desonid

3D human foreskin model for testing topical formulations of sildenafil citrate.

Sildenafil citrate is an approved drug used for the treatment of erectile dysfunction and premature ejaculation. Despite a widespread application, sildenafil citrate shows numerous adverse cardiovascular effects in high-risk patients. Local transdermal drug delivery of this drug is therefore being explored as an interesting and noninvasive alternative administration method that avoids adverse effects arised from peak plasma drug concentrations. Although human and animal skin represents the most reliable models to perform penetration studies, they involve a series of ethical issues and restrict

The dose-duration effect on cutaneous pharmacokinetics of metronidazole from topical dermatological formulations in Yucatan mini-pigs.

Dermal microdialysis (dMD) permits the investigation of cutaneous pharmacokinetics (cPK) for topical dermatological drug products (TDDP). dMD involves probe implantation into the dermis and a sample collection system that restricts subjects' movements for the experimental duration. A truncated dose-duration, by TDDP removal at predetermined time-points, may help to adequately characterize the cPK in a relatively short time. The goals of this study were to: assess and compare the dose-duration effect on the dermal exposure of metronidazole (MTZ) containing TDDPs; and characterize MTZ dermal eli
