Sacral nerve stimulation increases activation of the primary somatosensory cortex by anal canal stimulation in an experimental model.

BACKGROUND: Sacral and posterior tibial nerve stimulation may be used to treat faecal incontinence; however, the mechanism of action is unknown. The aim of this study was to establish whether sensory activation of the cerebral cortex by anal canal stimulation was increased by peripheral neuromodulation. METHODS: A multielectrode array was positioned over the right primary somatosensory cortex of anaesthetized rats. A brief burst of electrical stimulation was applied to either the left sacral root or the left posterior tibial nerve, and evoked potentials from anal canal stimulation were signal

Receptive field scatter, topography and map variability in different layers of the hindpaw representation of rat somatosensory cortex.

We recorded neurons extracellularly in layers II/III, IV, and V of the hindpaw representation of primary somatosensory cortex in anesthetized rats and studied laminar features of receptive fields (RFs) and representational maps. On average, RFs were smallest in layer IV and largest in layer V; however, for individual penetrations we found substantial deviations from this rule. Within the hindpaw representation, a distinct rostrocaudal gradient of RF size was present in all layers. While layer V RFs were generally largest independent of this gradient, layer IV RFs recorded caudally representing

Enrichment of mGluR7a in the presynaptic active zones of GABAergic and non-GABAergic terminals on interneurons in the rat somatosensory cortex.

The release of glutamate and GABA is modulated by presynaptic metabotropic glutamate receptors (mGluRs). We used immunocytochemical methods to define the location of the group III receptor mGluR7a in glutamatergic and GABAergic terminals innervating GABAergic interneurons and pyramidal cells. Immunoreactivity for mGluR7a was localized in the presynaptic active zone of both identified GABAergic and presumed glutamatergic terminals. Terminals innervating dendritic spines showed a variable level of receptor immunoreactivity, ranging from immunonegative to strongly immunopositive. The frequency of

Sensitivity of neurons in somatosensory cortex (S1) to cutaneous stimulation of the hindlimb immediately following a sciatic nerve crush.

While a large number of studies have examined receptive field alterations in the cerebral cortex after peripheral nerve injury, descriptions of neuronal sensitivity have been largely qualitative. In the present study, quantitative changes in minimal force thresholds evoking cortical responses were examined in somatosensory cortex (S1) of the rat after peripheral nerve injury. Cutaneous receptive fields were defined by conventional multi-unit recording techniques. Thresholds for evoking cortical responses were defined using Semmes-Weinstein monofilaments to stimulate the skin over the distal an

Tactile discrimination learning in the monkey: the effects of unilateral or bilateral removals of the second somatosensory cortex (area SII).

The tactile impairment in monkeys with unilateral removals of area SII is seen as possibly analogous to the rare condition of tactile agnosia. The lesions in a new series of animals with SII removals are described. The performance of 3 groups of monkeys is compared: no group differences were obtained on visual tasks (except retrieval of a moving target); minimal if any differences were obtained on inter-manual transfer of tactile learning; significant differences were found between animals with unilateral or bilateral removals of SII relative to unoperated animals at re-learning tactile discri

Long-term neuroplasticity of the face primary motor cortex and adjacent somatosensory cortex induced by tooth loss can be reversed following dental implant replacement in rats.

Tooth loss is common, and exploring the neuroplastic capacity of the face primary motor cortex (face-M1) and adjacent primary somatosensory cortex (face-S1) is crucial for understanding how subjects adapt to tooth loss and their prosthetic replacement. The aim was to test if functional reorganization of jaw and tongue motor representations in the rat face-M1 and face-S1 occurs following tooth extraction, and if subsequent dental implant placement can reverse this neuroplasticity. Rats (n = 22) had the right maxillary molar teeth extracted under local and general anesthesia. One month later, se

Two-photon in vivo imaging reveals cell type-specific mitophagy dynamic changes in mouse somatosensory cortex during aging.

Mitochondrial homeostasis is majorly maintained through mitochondrial autophagy (mitophagy). Recent research highlights the region- and cell type-specific nature of mitophagy during brain aging; however, these dynamics have largely remained unexplored in living brains. To address this gap, we conducted two-photon mt-Keima imaging in somatosensory cortical neurons and astrocytes in behaving male mice across two age groups, including 2-3-month-old (early-aged) and 18-20-month-old (old-aged) mice. We show reduced mitophagy in both cell types during aging, and we consistently found a higher level

Revisiting Face-to-Hand Area Remapping in the Human Primary Somatosensory Cortex after a Cervical Spinal Cord Injury.

Spinal cord injury (SCI) leads to profound disruptions in sensorimotor processing. Seminal research in nonhuman primates suggests this sensory deprivation causes functional remapping in the primary somatosensory cortex (S1), where somatotopic representations of deprived body parts, such as the hand in cervical SCI, become responsive to touch on intact body parts, such as the face. However, evidence for such remapping in humans remains inconclusive. We investigated face-to-hand remapping in 16 chronic cervical SCI patients (15 male, 1 female) and 21 able-bodied controls (19 male, 2 female) usin

Transcranial Intermittent Theta-Burst Stimulation Reverses Neurodegeneration in the Somatosensory Motor Cortex after Spinal Cord Transection in Rats.

BACKGROUND: Complete spinal cord injury (SCI) leads to a disconnection between the brain and the body below the injury level, resulting in the functional silencing, degeneration, and apoptosis of sensorimotor cortex (SMC) neurons, which is of crucial importance to the pathological process. METHODS: In this study, a rat model of spinal cord transection was employed to explore the activation of neurons in the SMC and the reversal of neurodegeneration after the rats were treated with transcranial intermittent theta-burst stimulation (T-iTBS). RESULTS: The results demonstrated that the expressio

Multi-Omics Analysis Unveils Nsun5-Mediated Molecular Alterations in the Somatosensory Cortex and Its Impact on Pain Sensation.

Nsun5 assumes a pivotal role in the regulation of RNA methylation, and its deficiency has been linked to the advancement of hepatocellular carcinoma, gliomas, tetralogy of Fallot, cognitive deficits in Williams-Beuren syndrome, and brain development. This underscores Nsun5's significant involvement in the nervous system. In this study, we present evidence of Nsun5's influence on the structure of the primary somatosensory cortex. Through comprehensive multi-omics analyses, we unveil a spectrum of systematically altered genes and proteins, collectively engaged in the orchestration of translation

Somatosensory cortex and central amygdala regulate neuropathic pain-mediated peripheral immune response via vagal projections to the spleen.

Pain involves neuroimmune crosstalk, but the mechanisms of this remain unclear. Here we showed that the splenic T helper 2 (TH2) immune cell response is differentially regulated in male mice with acute versus chronic neuropathic pain and that acetylcholinergic neurons in the dorsal motor nucleus of the vagus (AChDMV) directly innervate the spleen. Combined in vivo recording and immune cell profiling revealed the following two distinct circuits involved in pain-mediated peripheral TH2 immune response: glutamatergic neurons in the primary somatosensory cortex (GluS1HL)→AChDMV→spleen circuit and

Can individually targeted and optimized multi-channel tDCS outperform standard bipolar tDCS in stimulating the primary somatosensory cortex?

BACKGROUND: Transcranial direct current stimulation (tDCS) has emerged as a non-invasive neuro-modulation technique. Most studies show that anodal tDCS increases cortical excitability, however, with variable outcomes. Previously, we have shown in computer simulations that our multi-channel tDCS (mc-tDCS) approach, the distributed constrained maximum intensity (D-CMI) method can potentially lead to better controlled tDCS results due to the improved directionality of the injected current at the target side for individually optimized D-CMI montages. OBJECTIVE: In this study, we test the applicat

Activation of Bilateral Secondary Somatosensory Cortex With Right Hand Touch Stimulation: A Meta-Analysis of Functional Neuroimaging Studies.

Background: Brain regions involved in processing somatosensory information have been well documented through lesion, post-mortem, animal, and more recently, structural and functional neuroimaging studies. Functional neuroimaging studies characterize brain activation related to somatosensory processing; yet a meta-analysis synthesis of these findings is currently lacking and in-depth knowledge of the regions involved in somatosensory-related tasks may also be confounded by motor influences. Objectives: Our Activation Likelihood Estimate (ALE) meta-analysis sought to quantify brain regions that

Rapid synaptic remodeling in the adult somatosensory cortex following peripheral nerve injury and its association with neuropathic pain.

Structural and functional plastic changes in the primary somatosensory cortex (S1) have been observed following peripheral nerve injury that often leads to neuropathic pain, which is characterized by tactile allodynia. However, remodeling of cortical connections following injury has been believed to take months or years; this is not temporally correlated with the rapid development of allodynia and S1 hyperexcitability. Here we first report, by using long-term two-photon imaging of postsynaptic dendritic spines in living adult mice, that synaptic connections in the S1 are rewired within days fo

[Focal cortical dysplasia at the primary somatosensory cortex could manifest both intractable partial epilepsy and cortical reflex myoclonus].

We report a 31-year-old patient who suffered from the left foot motor seizure since 6 years of age. In addition to spontaneous seizures, the seizures tended to be triggered by somatosensory stimulus to the left foot. Recently, he developed irregular myoclonic jerks of the left foot. Neurologically, he had mild impairment of fine movements of the left foot, but otherwise no weakness or sensory disturbance was noted. Interictal spikes were frequently recorded at the vertex region, where ictal discharges also started during the video-EEG monitoring. Electrical stimulation of the left tibial nerve

Altered stimulus-intensity encoding occurs in the primary somatosensory cortex of a central post-stroke pain rat model.

Central post-stroke pain (CPSP) is a chronic form of pain that develops after ventral posterolateral thalamus (VPL) lesions and may involve maladaptive plasticity within central somatosensory pathways. Although abnormalities in the primary somatosensory cortex (S1) are observed in patients and animal models, the mechanism by which thalamic lesions alter cortical processing of sensory stimuli remains unclear. Here, we investigated stimulus intensity encoding and dendritic remodeling in S1 using a VPL-lesion CPSP rat model. Behavioral assessments were conducted, and mechanical allodynia was obse

Layer-specific cellular composition of mouse primary somatosensory and human temporal cortex: a direct 3D confocal counting approach.

Accurate quantification of cellular composition is fundamental to understanding the structural and functional organization of the cerebral cortex. In the present study, we quantified the proportions of neurons, glia, and vascular cells (primarily endothelial cells) across the full cortical thickness (layers I-VI) of the mouse primary somatosensory cortex (S1HL) and the human temporal cortex (BA21) using immunocytochemical techniques and a direct 3D counting method. Over 25,000 cells in the mouse and 13,000 cells in the human cortex were individually identified and classified. For this purpose,

Rod microglia: elongation, alignment, and coupling to form trains across the somatosensory cortex after experimental diffuse brain injury.

BACKGROUND: Since their discovery, the morphology of microglia has been interpreted to mirror their function, with ramified microglia constantly surveying the micro-environment and rapidly activating when changes occur. In 1899, Franz Nissl discovered what we now recognize as a distinct microglial activation state, microglial rod cells (Stäbchenzellen), which he observed adjacent to neurons. These rod-shaped microglia are typically found in human autopsy cases of paralysis of the insane, a disease of the pre-penicillin era, and best known today from HIV-1-infected brains. Microglial rod cells

Multiple tooth-losses during development suppress age-dependent emergence of oscillatory neural activities in the oral somatosensory cortex.

Tooth and tooth-related organs play important roles in not only mastication, but also sensory perception in the oral region. In general, sensory neural inputs during the developmental period are required for the maturation of functions in the sensory cortex. However, whether maturations of oral somatosensory cortex (OSC) require certain levels of sensory input from oral regions has been unclear. The present study investigated the influence of multiple tooth-losses during the developmental period on age-dependent emergence of rhythmic activities of population neurons in the OSC. Gastric fundic distension activates fronto-limbic structures but not primary somatosensory cortex: a functional magnetic resonance imaging study.

INTRODUCTION: The brain representation of visceral stimulation bears important similarities to that of somatic stimulation. However, the role of the primary (S1) and secondary (S2) somatosensory cortices in mediating gastric sensation is uncertain. MATERIALS AND METHODS: Eighteen healthy, right-handed volunteers (age 32 years+/-6.5 years; 14 men) underwent dynamic assessment of the relationship between sensation and fundic barostat distending pressure and volume, and then brain functional magnetic resonance imaging (fMRI) during noxious fundic distension.
