Taurine in gerbil retina: changes during ischemia reperfusion/insult (I.R.I.) and aging.

The retina contains a high amount of taurine which suggests a role in retinal function. The mongolian gerbil is well studied in stroke because of its incomplete circle of Willis. Two groups of gerbils were used: one served as control and the other was subjected to unilateral left carotid occlusion during 30 minutes, followed by 60 minutes of reperfusion. Gerbils were selected by ocular fundus and only sensitive gerbils were retained for the experimentation. We studied the level of taurine in gerbil retina of different ages: 3, 9, 15 and 24 months old (sham operated and ischemic groups). Level

Intrathecal GABA, glycine, taurine or beta-alanine elicits dyskinetic movements in mice.

Dyskinetic, writhing-like movements, similar to those produced in mice after an intraperitoneal (IP) injection of acetic acid, were elicited by intrathecal (IT) injection of GABA, glycine, taurine or beta-alanine. Baclofen and muscimol failed to produce this behavior. While acetic acid-induced writhing is inhibited by narcotic and nonnarcotic compounds, GABA-induced writhing was found to be insensitive to pretreatment with either morphine or capsaicin. Moreover, acetic acid-induced writhing does not appear to involve GABAergic transmission as IT injections of nipecotic acid did not alter the i

In Vivo Measurement of Taurine With Optimized Double-Quantum Filtering Magnetic Resonance Spectroscopy.

PURPOSE: To enhance the accuracy and reproducibility for in vivo taurine quantification using the double-quantum filtering (DQF) technique. METHODS: Simulations and phantom experiments were conducted at both 7 T and 3 T to determine the optimal parameter τ by calculating the taurine purity index among other neighboring metabolites. The frequency-selective read-pulse bandwidth and the dephasing/rephasing gradient moments were further adjusted to reduce residual water side band interference. Rat models of C6 gliomas were compared to normal brain for taurine level at 7 T. Metabolomics was conduc

An orthogonal mammalian gene circuit for selective sensing of taurine-conjugated bile acids.

Beyond their traditional role as dietary lipid emulsifiers, bile acids (BAs) function as pleiotropic signaling molecules that regulate metabolism, inflammation, and diverse disease processes. Taurine-conjugated bile acids (Tau-BAs) and the glycine-to-taurine conjugation ratio (G/T ratio) have emerged as sensitive biomarkers of multiple pathological conditions, including chronic liver diseases, metabolic disorders, and neurodegenerative diseases. However, existing bile acid-responsive sensors and synthetic circuits largely rely on endogenous receptors or signaling pathways, thereby lacking true

Protection of multiple aspects of Alzheimer's disease pathology using dietary supplementation with taurine.

As Alzheimer’s disease (AD) continues to rise amongst the aging population, preventative measures such as dietary or lifestyle changes represent an attractive option to mitigate the burden. Taurine, known for its antioxidant and anti-inflammatory properties, may also play a neuroprotective role. This study investigates the protective effects of taurine supplementation in 5xFAD mice. Taurine was administered through drinking water at doses of 0, 500, 1000, 2000, 4000 mg/kg/day, with no change in water consumption or body mass was observed. Postmortem markers of neuroinflammation using cytokine

Hypothesis: Taurine therapy of nephropathic cystinosis may correct the deficiencies of cysteamine therapy.

Untreated nephropathic cystinosis is a lethal autosomal recessive disease. The current specific therapy, cysteamine, ameliorates the renal function loss, but does not alter the renal Fanconi syndrome, short stature, muscle weakness, male infertility, and other concerns. The primary biochemical/physiological defect in cystinosis is failure to supply cysteine to mTOR via cystinosin. This leads mTOR to react in starvation mode, which stops cell differentiation, leading to proximal tubule loss, and ultimately renal failure. It also increases apoptosis and autophagocytosis rates, which may contribu

Taurine attenuates activation of hepatic stellate cells by inhibiting autophagy and inducing ferroptosis.

BACKGROUND: Liver fibrosis is a compensatory response during the tissue repair process in chronic liver injury, and finally leads to liver cirrhosis or even hepatocellular carcinoma. The pathogenesis of hepatic fibrosis is associated with the progressive accumulation of activated hepatic stellate cells (HSCs), which can transdifferentiate into myofibroblasts to produce an excess of the extracellular matrix (ECM). Myofibroblasts are the main source of the excessive ECM responsible for hepatic fibrosis. Therefore, activated hepatic stellate cells (aHSCs), the principal ECM producing cells in the

Taurine rescues intervertebral disc degeneration by activating mitophagy through the PINK1/Parkin pathway.

Intervertebral disc degeneration (IDD) is a common cause of low back pain and disability. Recent studies have highlighted the critical role of mitochondrial dysfunction in the progression of IDD. In this study, we investigated the therapeutic potential of taurine in delaying IDD through the activation of mitophagy via the PINK1/Parkin pathway. Our in vitro and in vivo experiments demonstrate that taurine treatment significantly enhances mitophagy, reduces oxidative stress, delays cell senescence, and promotes the removal of damaged mitochondria in nucleus pulposus cells (NPC). Additionally, ta

Intervention Effect of Taurine on LPS-Induced Intestinal Mechanical Barrier Injury in Piglets.

Taurine has the advantages of being safe, highly efficient, chemically stabile, and biologically active, together with having versatile functions. Presently, it is employed as a veterinary feed additive in animal research. The tight junctions that constitute the intestinal epithelial cells are the most critical structures for ensuring regular and uninterrupted digestion and absorption of food by the intestinal mucosa, while at the same time resisting invasions by toxins. The purpose of this study was to investigate the protective effect and mechanism of taurine action on intestinal mechanical

Altered bile acid glycine : taurine ratio in the progression of chronic liver disease.

BACKGROUND AND AIM: The onset and progression of chronic liver disease (CLD) is a multistage process spanning years or several decades. Some bile acid (BA) features are identified as indicators for CLD progression. However, BAs are highly influenced by various factors and are stage and/or population specific. Emerging evidences demonstrated the association of structure of conjugated BAs and CLD progression. Here, we aimed to investigate the alteration of conjugated BAs and identify new features for CLD progression. METHODS: Based on liquid chromatography-mass spectrometry platform, 15 BAs wer

Taurine Enhances Iron-Related Proteins and Reduces Lipid Peroxidation in Differentiated C2C12 Myotubes.

Taurine is a nonproteinogenic amino sulfonic acid in mammals. Interestingly, skeletal muscle is unable to synthesize taurine endogenously, and the processing of muscular taurine changes throughout ageing and under specific pathophysiological conditions, such as muscular dystrophy. Ageing and disease are also associated with altered iron metabolism, especially when there is an excess of labile iron. The present study addresses the question of whether taurine connects cytoprotective effects and redox homeostasis in a previously unknown iron-dependent manner. Using cultured differentiated C2C12 m

Urinary Taurine Excretion and Risk of Late Graft Failure in Renal Transplant Recipients.

Taurine is a sulfur containing nutrient that has been shown to protect against oxidative stress, which has been implicated in the pathophysiology leading to late graft failure after renal transplantation. We prospectively investigated whether high urinary taurine excretion, reflecting high taurine intake, is associated with low risk for development of late graft failure in renal transplant recipients (RTR). Urinary taurine excretion was measured in a longitudinal cohort of 678 stable RTR. Prospective associations were assessed using Cox regression analyses. Graft failure was defined as the sta

Effect of Taurine on iNOS-Mediated DNA Damage in Drug-Induced Renal Injury.

Owing to an outstanding wide antitumor spectrum and excellent anti-tumor effect cisplatin has been used in chemotherapy for malignant tumor. However, cisplatin has strong side effects such as renal injury. Taurine has been found to protect against inflammatory tissue damage in a variety of experimental models. The aim of the present study was to investigate the effect of taurine against iNOS dependent DNA damage in cisplatin-induced renal injury in rats. With the help of a rat model of drug-induced kidney damage, we have assessed the nephrotoxic effects of different doses of cisplatin in the p

Serum Taurine and Stroke Risk in Women: A Prospective, Nested Case-Control Study.

BACKGROUND: Taurine (2-aminoethanesulfonic acid), a conditionally essential sulfur-containing amino acid, is mainly obtained from diet in humans. Experimental studies have shown that taurine's main biological actions include bile salt conjugation, blood pressure regulation, anti-oxidation, and anti-inflammation. METHODS: We conducted a prospective case-control study nested in the New York University Women's Health Study, a cohort study involving 14,274 women enrolled since 1985. Taurine was measured in pre-diagnostic serum samples of 241 stroke cases and 479 matched controls. RESULTS: There

Taurine in adipocytes prevents insulin-mediated H2O2 generation and activates Pka and lipolysis.

Among many actions assigned to taurine (Tau), the most abundant amino acid in numerous mammalian tissues, it prevents high-fat diet-induced obesity with increasing resting energy expenditure. To sustain this Tau action, the goal of the present study was to explore the acute effects of Tau on baseline and on adrenaline, insulin and their second messengers to modulate lipolysis in white adipose tissue (WAT) cells from rat epididymis. The Tau effects in this topic were compared with those recorded with Gly, Cys and Met. Tau on its own did not modify baseline lipolysis. Tau raised isoproterenol- a

Prophylactic and therapeutic effects of taurine against aluminum-induced acute hepatotoxicity in mice.

Aluminum is a well known neurotoxin and a possible candidate of hepatotoxins to humans. Using natural antioxidants against metal-induced hepatotoxicity is a modern approach. In the present study, Aluminum (AlCl(3)) intoxication (a single injection of 25mg Al(3+)/kg, i.p.) for 24h in mice resulted in elevations in serum alanine aminotransferase activity and serum tumor necrosis factor and hepatic malondialdehyde levels. Aluminum reduced the activities of glutathione peroxidase, glutathione S-transferase, quinone oxidoreductase, and catalase in liver. In addition, Al caused hepatic hemorrhage, c

Taurine bromamine (TauBr)--its role in immunity and new perspectives for clinical use.

This review is an attempt to summarize our knowledge about taurine bromamine (TauBr) properties, its role in innate immunity and its therapeutic potential.TauBr and taurine chloramine (TauCl) are major haloamines generated by eosinophils and neutrophils at a site of inflammation. Both haloamines share anti-inflammatory and anti-oxidant properties. TauBr, similarly to TauCl, decreases the production of proinflammatory mediators. Their anti-inflammatory and anti-oxidant activities are enhanced by their ability to induce the expression of heme oxygenase-1 (HO-1). TauCl is more stable than TauBr.

Electro-acupuncture improves epileptic seizures induced by kainic acid in taurine-depletion rats.

Electro-acupuncture (EA) partially inhibited epilepsy with great success. The biological basis underlying EA anti-convulsion remained uncertain, which resulted in limited application and slow improvement of acupuncture. Our previous study indicated that taurine may play an inhibitory role against epilepsy as an inhibitory amino acid in the central nervous system and EA may inhibit epilepsy via up-regulating the expression of taurine transporter to increase the release of taurine. Involvement of taurine in kainic acid (KA)-induced epilepsy and anti-convulsion of EA was further addressed on taur
