The effect of trovafloxacin, ciprofloxacin and ceftriaxone on cytokine production of human peripheral blood mononuclear cells (PBMCs) was examined. PBMC responses were measured after stimulation with lipopolysaccharide (LPS), lipoteichoic acid (LTA) or killed or viable Streptococcus pneumoniae and Haemophilus influenzae. Trovafloxacin inhibited the production of tumour necrosis factor alpha (TNF-alpha), interleukin-1beta (IL-1beta), IL-6 and IL-8 by PBMCs after stimulation with either LPS or LTA by 83%. Similar inhibition occurred in PBMCs incubated with killed or live bacteria and trovafloxac

Group B streptococcal antigens stimulated tumor necrosis factor alpha (TNF-alpha), interleukin-1 (IL-1), and IL-6 production in human blood cultures in a concentration- and time-dependent fashion. The minimal concentrations of type-specific polysaccharides, lipoteichoic acid, and group-specific polysaccharide required to produce these effects were, respectively, 0.01, 1, and 10 microg/ml. Cell separation experiments indicated that monocytes were the cell type mainly responsible for cytokine production. Time course studies indicated that TNF-alpha was released before the other cytokines. TNF-al

Mesenchymal stem cells (MSC) are able to stimulate the regeneration of injured tissue. Since bacterial infections are common complications in wound healing, bacterial pathogens and their components come into direct contact with MSC. The interaction with bacterial structures influences the proliferation, differentiation and migratory activity of the MSC, which might be of relevance during regeneration. Studies on MSC migration in response to bacterial components have shown different results depending on the cell type. Here, we analyzed the migration rate and chemotaxis of human adipose-derived

Bacterial infections of bones remain a serious complication of endoprosthetic surgery. These infections are difficult to treat, because many bacterial species form biofilms on implants, which are relatively resistant towards antibiotics. Bacterial biofilms elicit a progressive local inflammatory response, resulting in tissue damage and bone degradation. In the majority of patients, replacement of the prosthesis is required. To address the question of how the local inflammatory response is linked to bone degradation, tissue samples were taken during surgery and gene expression of the macrophage

The cell wall of Gram-positive bacteria contains lipoteichoic acid (LTA) and peptidoglycan (PepG), which synergise to cause shock and organ failure in animals, and to activate human blood to release proinflammatory cytokines. The structural elements within LTA and PepG that are essential for the observed synergism are discussed.

Group B streptococci (GBS) are the major cause of neonatal pneumonia, sepsis, and meningitis. Steps considered to be important in the pathogenesis of this infection include colonization of the rectum and vagina of the mother, aspiration of GBS into the fetal lung during or just prior to delivery, and invasion of GBS into pulmonary epithelial cells. We have previously demonstrated that GBS can invade pulmonary epithelial cells both in vivo and in vitro. Adherence of GBS to epithelial cells may play an important role in colonization of the rectum and vagina and constitute a first step in invasio

Enterococci are opportunistic pathogens notorious for causing a variety of infections. While both Enterococcus faecalis and Lactobacillus crispatus are commensal residents of the vaginal tract, the molecular mechanisms that enable E. faecalis to take advantage of a vaginal biome with lower counts of lactobacilli to colonize the vaginal tract and induce aerobic vaginitis remain unknown. Here, we show that L. crispatus eradicates E. faecalis in a contact-independent manner. Using transposon sequencing to identify E. faecalis OG1RF transposon (Tn) mutants that are either under-represented or over

Group B Streptococcus (GBS; Streptococcus agalactiae) is a leading cause of sepsis in neonates and pregnant mothers worldwide. Whereas the hyper-virulent serogroup III clonal cluster 17 has been associated with neonatal disease and meningitis, serogroup III ST283 was recently implicated in invasive disease among non-pregnant adults in Asia. Here, through comparative genome analyses of invasive and non-invasive ST283 strains, we identified a truncated DNA-binding regulator of a two-component system in a non-invasive strain that was homologous to Bacillus subtilis bceR, encoding the bceRSAB resp

Background: Antimicrobial resistance is at alarming levels. Methicillin-resistant Staphylococcus aureus is a clinical concern and has been classified as one of the so-called ESKAPE pathogens. The increase in resistance coupled with the lack of new agents in development requires innovative ways of developing new antimicrobials. Methods: Here, we investigate an approved drug (Zafirlukast), which has been previously reported to have activity against Mycobacterium, for activities against other bacterial strains. Antibacterial activity was assessed using standard MIC testing along with checkerboard

Phagocytosis of microbes through their surface glycans by mononuclear phagocytes (MNPs) is an important process in triggering inflammatory responses and subsequent acquired immunity. However, it is not fully understood how glycan-dependent phagocytosis is related to production of inflammatory cytokines, and especially the production of interleukin (IL)-12, which plays a key role in activation of natural killer cells and differentiation of naive T cells into T helper 1 (Th1). If microbes are considered to be glycan-presenting particles with an appropriate size, assessment of the responses of ph

OBJECTIVES: This study aimed to investigate the role of mitochondrial dysfunction in the pathogenesis of Enterococcus faecalis and its lipoteichoic acid (LTA)-induced refractory apical periodontitis, and then to evaluate whether the modulation of mitochondrial dynamics with Mdivi-1 could alleviate the ensuing inflammatory response..
METHODS: An LTA-induced macrophage model was established to simulate the inflammatory environment of apical periodontitis. Changes in inflammatory factors, mitochondrial morphology, dynamics-related proteins, autophagy markers, and reactive oxygen species (ROS) we

Balb/c-mice were intravenously infected with 5 X 10(4) viable cells of Listeria monocytogenes SLCC 4013. Liver, lung and spleen of the animals showed heavy organ colonization 48 h after infection. Pretreatment of experimental animals with staphylococcal lipoteichoic acid (LTA; 2 mg i.p. 7 and 3 days before challenge) significantly reduced the bacterial count in these organs. In vitro, peritoneal macrophages from LTA-treated mice significantly reduced listerial viability as compared to control cells (p less than 0.0025). In vitro, exposure of macrophages from non-treated animals to LTA did not

This study aimed to quantify LPS and LTA levels in root canals requiring endodontic retreatment due to secondary or persistent infection and evaluate the relationship between Gram-positive and Gram-negative virulence factors and clinical symptoms and signs. Samples were collected from the root canals of 40 patients who needed endodontic retreatment and had radiographic evidence of apical periodontitis. Lipopolysaccharides (LPS) and lipoteichoic acid (LTA) were quantified using Limulus amebocyte lysate and enzyme-linked immunosorbent assay tests, respectively. LPS and LTA were detected in all s

BACKGROUND: High-fat diet, microbial alterations and lipopolysaccharide (LPS) are thought to cause enteric diabetic neuropathy and intestinal dysmotility. However, the role of the gut microbiota, lipoteichoic acid (LTA) from Gram-positive bacteria and short-chain fatty acids (SCFAs) in the development of diabetic enteric neuropathy and intestinal dysmotility is not well understood. Our aim was to examine the role of the gut microbiota, LTA and SCFAs in the development of diabetic enteric neuropathy and intestinal dysmotility.
METHODS: We fed germ-free (GF) and conventionally raised (CR) mice

Lipoteichoic acids (LTAs) from various bacterial species, including Staphylococcus aureus, Streptococcus pyogenes, Streptococcus pneumoniae, Enterococcus faecalis, and Listeria monocytogenes, were examined for the ability to induce secretory and cellular responses in a pure population of bone marrow-derived mononuclear phagocytes. Some of the highly purified LTAs, in particular LTAs from Bacillus subtilis, S. pyogenes, E. faecalis, and Enterococcus hirae, were able to affect each of the macrophage parameters measured, i.e., reductive capacity, secretion of tumor necrosis factor and nitrite, an

The colonization of mucosal cells in the oral cavity of newborn infants was studied at various intervals after birth in an attempt to define the nature of the epithelial binding sites for group A streptococci and their lipoteichoic acid (LTA). Stained smears of buccal mucosal cells showed that the average number of naturally acquired bacteria/cell was zero to one in infants less than one day old, one to four in infants one day old, and 11-19 in infants two days old. Samples of the same mucosal cells were incubated with group A streptococci, and the average number of streptococci bound per cell

Methicillin resistant Staphylococcus aureus (MRSA) bacteremia has a high rate of morbidity and mortality. Multiple clinical studies have demonstrated improved outcomes when MRSA bacteremia is treated with dual antibiotic therapy that includes a β-lactam antibiotic such as cefazolin. This is a paradox as MRSA should be inherently resistant to this class of antibiotics. We report on a serendipitous observation of a phenotype where MRSA became sensitive to cefazolin when cultured in a physiologic relevant media of fetal bovine serum as well as in synovial fluid. This could be observed across mult

BACKGROUND: Phage therapy is a promising approach for infections caused by multidrug-resistant Staphylococcus aureus, but its clinical development is limited by knowledge gaps in pharmacokinetic/pharmacodynamic (PK/PD) and potential emergence of resistance.
METHODS: We performed in vitro time-kill assays to study the effect of three anti-staphylococcal phages (V1SA019, V1SA020, V1SA022) against two S. aureus strains (SH1000 and USA300). Resistance emergence was monitored through phenotyping and whole-genome sequencing of resistant clones.Bacteria and phage counts were analyzed by pharmacokine

BACKGROUND AND OBJECTIVES: The imminent threat of antibiotic resistance has spurred studies of nonconventional antimicrobial approaches. Gallium utilization is a promising and emerging approach to treating a variety of resistant bacteria using "Trojan horse" strategies to disrupt iron-dependent processes and biofilms. This study utilized experimental evolution to test the evolvability of gallium resistance in Staphylococcus aureus and resistance traits potentially correlated with metals, antibiotics and polyfluorinated compounds, as well as its genomics foundations.
METHODS: Whole-genome sequ

Aspiration of bile into the cystic fibrosis (CF) lung has emerged as a prognostic factor for reduced microbial lung biodiversity and the establishment of often fatal, chronic pathogen infections. Staphylococcus aureus is one of the earliest pathogens detected in the lungs of children with CF, and once established as a chronic infection, strategies for its eradication become limited. Several lung pathogens are stimulated to produce biofilms in vitro in the presence of bile. In this study, we further investigated the effects of bile on S. aureus biofilm formation. Most clinical S. aureus strains
