Esophageal tissue engineering: a new approach for esophageal replacement.

A number of congenital and acquired disorders require esophageal tissue replacement. Various surgical techniques, such as gastric and colonic interposition, are standards of treatment, but frequently complicated by stenosis and other problems. Regenerative medicine approaches facilitate the use of biological constructs to replace or regenerate normal tissue function. We review the literature of esophageal tissue engineering, discuss its implications, compare the methodologies that have been employed and suggest possible directions for the future. Stem cell and tissue engineering therapies for ocular regeneration.

The eye is a relatively small but very complex organ. It is responsible for vision. Most of its cells are terminally differentiated, and several pathologies affecting those cells lead to vision loss and eventual blindness. Several years ago, a group of cells, located in the limbus, was identified as having the capacity of self-renewal and later on found to feed the renewal of the corneal epithelial layer. Since then, this niche of stem cells has been studied in order to provide clues that can be valuable for the regeneration of ocular structures. The worldwide shortage of donors, increased ris

A poloxamine-polylysine acrylate scaffold for modular tissue engineering.

A new polymer, poloxamine-polylysine acrylate (PPA), was synthesized for tissue-engineering applications. Polylysine was used to confer endothelial cell attachment properties to the poloxamine-based polymer, while the acrylate groups made it photo-cross-linkable. The PPA polymer was synthesized in three reaction steps. (1) Polylysine was acrylated using N-hydroxysuccinimide acrylic acid ester and the reaction product was characterised by (1)H-NMR. (2) The hydroxyl groups on poloxamine were activated by tresylation with tresyl chloride and the reaction product (tresylated poloxamine) was charac

Intervertebral disk tissue engineering using biphasic silk composite scaffolds.

Scaffolds composed of synthetic, natural, and hybrid materials have been investigated as options to restore intervertebral disk (IVD) tissue function. These systems fall short of the lamellar features of the native annulus fibrosus (AF) tissue or focus only on the nucleus pulposus (NP) tissue. However, successful regeneration of the entire IVD requires a combination approach to restore functions of both the AF and NP. To address this need, a biphasic biomaterial structure was generated by using silk protein for the AF and fibrin/hyaluronic acid (HA) gels for the NP. Two cell types, porcine AF

Stem cells for skin tissue engineering and wound healing.

The tremendous ability of the skin's epidermis to regenerate is due to the presence of epidermal stem cells that continuously produce keratinocytes, which undergo terminal differentiation to a keratinized layer that provides the skin's barrier properties. The ability to control this process in vitro has made it possible to develop various types of tissue-engineered skin grafts, some of which are among the first tissue-engineered products to ever reach the market. In the past 30 years, these products have been applied with some success to the treatment of chronic skin wounds such as diabetic an

Tracheal regeneration after partial resection: a tissue engineering approach.

OBJECTIVES: The aims of this study are to investigate the efficiency of a tissue engineering approach to partial tracheal reconstruction and to improve epithelialization of the reconstructed trachea. The trachea must be resected in some cases of cancer or trauma. Various restructuring techniques are used, with no consensus on the best approach. Two problems that arise when treating tracheal defects by conventional techniques are an inability to regenerate ciliated epithelium at the reconstructed site and having to perform multiple procedures to achieve the desired repair. This study is designe

Mesenchymal stem cells for tissue engineering and regenerative medicine.

Mesenchymal stem cells (MSCs) exist in bone marrow and other musculoskeletal tissues. These cells contribute to the homeostasis of musculoskeletal tissue as well as support for the growth and differentiation of primitive hemopoietic cells. Recent advancements in tissue engineering and regenerative medicine have highlighted MSCs as a potential source of cells which would differentiate to a variety of tissue tailored to individual needs. This paper briefly outlines the current status of MSCs, focusing on their biological characteristics and potential for clinical applications.

Neural tissue engineering: a self-organizing collagen guidance conduit.

We report a novel implantable device that will deliver a tethered aligned collagen guidance conduit containing Schwann cells into a peripheral nerve injury site. Cells (Schwann cells and fibroblasts) incorporated into tethered rectangular collagen gels contracted and resulted in uniaxial alignment. This tissue-engineered construct was tested in three-dimensional culture and demonstrated the ability to guide neurite extension from dissociated dorsal root ganglia. A silicone tube was adapted to provide tethering sites for an implantable construct such that uniaxial cell-generated tension resulte

Bioreactors for tissue engineering--a new role for perfusionists?

Tissue engineering is an exciting new area of medicine with rapid growth and expansion over the last decade. It has the potential to have a profound impact on the practice of medicine and influence the economic development in the industry of biotechnology. In almost every specialty of medicine, the ability to generate replacement cells and develop tissues will change the focus from artificial organs and transplantation to growing replacement organs from the patient's own stem cells. Once these organs are at a size that requires perfusion to maintain oxygen and nutrient delivery, then automated

Tissue engineering by cocultivating human elastic chondrocytes and keratinocytes.

To date, there is no optimal way to reconstruct an external ear in cases of microtia or after trauma or burns damaging the external ear. However, success in the area of tissue engineering has indicated that autologous elastic cartilage produced in vitro might be of great importance in the future treatment of these patients. In the present study we have engineered human, elastic cartilage in vitro by culturing chondrocytes in fibrin glue. Furthermore, the engineered elastic cartilage was seeded with human keratinocytes to investigate the possibility of combining these two tissues into one integ

Advancing vascularization with VEGF-bound hydrogels in tissue engineering.

Tissue vascularization continues to be a significant and unresolved challenge in in vitro tissue engineering. A promising approach involves binding vascular endothelial growth factor (VEGF) to engineered hydrogels, which stimulates endothelial cells (ECs) to form functional blood vessels. VEGF plays a central role in vascular development from embryogenesis to adulthood by promoting new blood vessel formation and regulating EC proliferation, migration, survival, differentiation, and vascular remodeling. However, vascularization outcomes depend not only on the presence of VEGF but also on its re

Advanced polymers for kidney and urinary tract tissue engineering.

Kidney tissue engineering has emerged as a promising strategy to address the growing clinical burden of end-stage renal disease (ESRD), for which current treatments such as hemodialysis and kidney transplantation remain limited by donor shortage, cost, and long-term complications. Central to these approaches is the development of advanced biomaterials capable of recapitulating the complex structural, mechanical, and biochemical microenvironment of native renal tissue. Among available material platforms, polymers and hydrogels have received significant attention due to their tunable properties,

Hydrogel Implementing Drug Delivery in Cranial Bone Tissue Engineering.

Cranial bone defects remain a significant clinical challenge due to their limited intrinsic regenerative capacity and the complexity of coordinating osteogenesis, angiogenesis, and immune responses within a confined and poorly vascularised environment. Conventional approaches, including autologous grafts and synthetic implants, provide structural support but fail to actively modulate the biological processes required for effective bone regeneration. In this context, hydrogel-based systems have emerged as versatile platforms for localized and controlled drug delivery in cranial bone tissue engi

Mechanobiological engineering strategies for oral and maxillofacial tissue regeneration.

Mechanical forces are central determinants of development, homeostasis, and pathology in oral and maxillofacial tissues, yet most regenerative strategies continue to prioritize biochemical signaling over mechanical control. In anatomically and functionally heterogenous tissues such as dentin-pulp, periodontium, temporomandibular joint, oral mucosa, and salivary glands, mechanics is not a secondary cue but a primary regulator of cell fate, immune responses, and tissue integration. This review reframes oral and maxillofacial regeneration through a mechanobiological engineering lens, summarizing

Regenerative and tissue engineering strategies for segmental mandibular reconstruction.

Segmental mandibular reconstruction following major ablative surgery, tumor resection, trauma, or osteonecrosis remains a formidable clinical challenge due to the region's complex anatomical, functional, and aesthetic demands. Although autologous vascularized bone grafts, such as the fibula free flap, remain the gold standard, they are limited by donor-site morbidity, restricted vertical height, and geometric incongruence. Bone tissue engineering offers a promising alternative by combining biomaterial scaffolds, osteogenic cells, and bioactive molecules to regenerate functional bone. Advances
