Silk sericin/PLGA electrospun scaffolds with anti-inflammatory drug-eluting properties for periodontal tissue engineering.

Periodontal disease is associated with chronic inflammation and destruction of the soft and hard tissues in the periodontium. Scaffolds that would enable cell attachment and proliferation while at the same time providing a local sustained anti-inflammatory action would be beneficial in restoring or reversing disease progression. In the current study, silk sericin, a natural protein derived from the silkworm cocoons, was electrospun with poly lactide-co-glycolic acid (PLGA) and ketoprofen, and the composite scaffolds were assessed for their physicochemical and mechanical properties, as well as

Biomimetic approach to articular cartilage tissue engineering using carbon nanotube-coated and textured polydimethylsiloxane scaffolds.

There is a significant need to understand the complexity and heterogeneity of articular cartilage to develop more effective therapeutic strategies for diseases such as osteoarthritis. Here, we show that carbon nanotubes (CNTs) are excellent candidates as a material for synthetic scaffolds to support the growth of chondrocytes-the cells that produce and maintain cartilage. Chondrocyte morphology, proliferation, and alignment were investigated as nanoscale CNT networks were applied to macroscopically textured polydimethylsiloxane (PDMS) scaffolds. The application of CNTs to the surface of PDMS-b

From Soft to Hard Biomimetic Materials: Tuning Micro/Nano-Architecture of Scaffolds for Tissue Regeneration.

Failure of tissues and organs resulting from degenerative diseases or trauma has caused huge economic and health concerns around the world. Tissue engineering represents the only possibility to revert this scenario owing to its potential to regenerate or replace damaged tissues and organs. In a regeneration strategy, biomaterials play a key role promoting new tissue formation by providing adequate space for cell accommodation and appropriate biochemical and biophysical cues to support cell proliferation and differentiation. Among other physical cues, the architectural features of the biomateri

Carbon Nanotube Polymer Scaffolds as a Conductive Alternative for the Construction of Retinal Sheet Tissue.

With the great success of graphene in the biomedical field, carbon nanotubes have attracted increasing attention for different applications in ophthalmology. Here, we report a novel retinal sheet composed of carbon nanotubes (CNTs) and poly(lactic-co-glycolic acid) (PLGA) that can enhance retinal cell therapy. By tuning our CNTs to regulate the mechanical characteristics of retina sheets, we were able to improve the in vitro viability of retinal ganglion cells derived from human-induced pluripotent stem cells incorporated into CNTs. Engrafted retinal ganglion cells displayed signs of regenerat

Fabrication of 3D porous silk scaffolds by particulate (salt/sucrose) leaching for bone tissue reconstruction.

Silk fibroin is a biomaterial being actively studied in the field of bone tissue engineering. In this study, we aimed to select the best strategy for bone reconstruction on scaffolds by changing various conditions. We compared the characteristics of each scaffold via structural analysis using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), the swelling ratio, water uptake, porosity, compressive strength, cell infiltration and cell viability (CCK-8). The scaffolds had high porosity with good inter pore connectivity and showed high compressive strength and mod

Bone Tissue Engineering by Using Calcium Phosphate Glass Scaffolds and the Avidin-Biotin Binding System.

Highly porous and interconnected scaffolds were fabricated using calcium phosphate glass (CPG) for bone tissue engineering. An avidin-biotin binding system was used to improve osteoblast-like cell adhesion to the scaffold. The scaffolds had open macro- and micro-scale pores, and continuous struts without cracks or defects. Scaffolds prepared using a mixture (amorphous and crystalline CPG) were stronger than amorphous group and crystalline group. Cell adhesion assays showed that more cells adhered, with increasing cell seeding efficiency to the avidin-adsorbed scaffolds, and that cell attachmen

Fabrication and characterization of biomimetic collagen-apatite scaffolds with tunable structures for bone tissue engineering.

The objective of the current study is to prepare a biomimetic collagen-apatite scaffold for improved bone repair and regeneration. A novel bottom-up approach has been developed, which combines a biomimetic self-assembly method with a controllable freeze-casting technology. In this study, the mineralized collagen fibers were generated using a simple one-step co-precipitation method which involved collagen self-assembly and in situ apatite precipitation in a collagen-containing modified simulated body fluid (m-SBF). The precipitates were then subjected to controllable freeze casting, forming sca

Natural Polymer-Based Scaffolds for Bone Tissue Engineering: A Review of Materials, Function, and Translation.

Scaffold-based bone tissue engineering (BTE) represents a pivotal strategy in regenerative medicine, offering substantial promise for repairing damaged bone. These scaffolds provide essential structural support and promote new bone formation by mimicking the native extracellular matrix (ECM). The rational design of an ideal scaffold requires careful balancing of biomaterial properties with biomechanical performance. Natural polymers have gained considerable attention in this context due to their inherent bioactivity, biocompatibility, and tunable mechanical properties. Their key advantage lies

Melt electrowriting-enabled hybrid platforms: Toward functional biomimetic scaffolds for cellular interaction and tissue regeneration.

Melt electrowriting (MEW) is a high-resolution additive manufacturing technique that enables the precise deposition of micro- to submicron-scale fibers, allowing the fabrication of highly ordered 3D fibrous scaffolds. Owing to its unique capability to program scaffold architecture and mechanical properties, MEW has attracted increasing attention for engineering personalized, tissue-specific constructs. However, MEW alone remains insufficient to replicate the hierarchical structural organization and multifunctional complexity of native tissues. To overcome these limitations, MEW is increasingly

PTGFRN- and IL-4-overexpressing exosome-functionalized Janus fibrous scaffolds for enhanced heterogeneous tissue regeneration.

Bone defects caused by trauma, infection, or tumor removal are often combined with soft tissue injury. This creates a major clinical problem because there are no effective methods to repair both hard and soft tissues at the same time. This study presents a strategy that combines physical and biological signals based on materiobiology and immune-guided regeneration. A Janus electrospun nanofibrous scaffold loaded with immunomodulatory exosomes (ExoPTGFRN/IL-4) was designed and fabricated. The scaffold has different surface structures and mechanical properties on each side, and it can regulate i

High-Performance Regenerated Silk Fibers as Building Blocks of Tissue Scaffolds: The European THOR Project.

The European Pathfinder THOR project envisages the creation of a vascularized fragment of tissue that can be implanted in a patient using regenerated silk fibers as its building blocks. The selection of regenerated silk as the main building block of the scaffold relies heavily on its outstanding biocompatibility in comparison with either other artificial polymeric fibers or even natural silk fibers. Additionally, regenerated fibers produced through the Dynamic Dope Destabilization Spinning (D3STM) process are shown to exhibit high mechanical performance as reflected in values of strain at brea

Engineering Smart Scaffolds for Osteomyelitis: Integrating Nanotechnology, Drug Delivery, and Tissue Regeneration-A Narrative Review.

Osteomyelitis is a severe, progressive bone infection affecting approximately 21.8 per 100,000 individuals in the United States and remains a major challenge in orthopedic and reconstructive medicine. Staphylococcus aureus (S. aureus), responsible for nearly 75% of cases, is the most common causative pathogen. Current gold-standard management involves culture-directed antibiotic therapy administered for 4-6 weeks, often combined with surgical debridement and the use of antibiotic-loaded cement spacers. Despite these interventions, treatment frequently fails to achieve complete infection eradic

Cryoelectrospun Elastin-Alginate Scaffolds Support In Vitro 3D Epithelial-Stromal Cocultures for Salivary Tissue Engineering.

Bioengineered functional salivary tissues can advance regenerative therapies, preclinical drug testing, and the fundamental understanding of salivary gland dysfunction. Current salivary tissue models are typically Matrigel-based, hydrogel-based or scaffold-free organoid systems, with limited physiological relevance or mimicry of cell-cell and cell-extracellular matrix (ECM) interactions. We previously developed elastin-alginate cryoelectrospun scaffolds (CES) that resemble the topography and viscoelastic properties of healthy salivary ECM, and validated their potential for stromal cell culture

Advances and Challenges of Tissue Vascular Scaffolds and Supercritical Carbon Dioxide Technology in Cardiovascular Diseases.

BACKGROUND: Atherosclerosis often leads to ischemic heart disease and peripheral artery disease. Traditional revascularization technique such as bypass grafting using autologous vessels are commonly employed. However, limitations arise when patients lack suitable grafts due to underlying diseases or previous surgeries, prompting the need to substitute vessel grafts. Due to the high biocompatibility of decellularized products (grafts or scaffolds) prepared using supercritical carbon dioxide (ScCO2), it has been widely applied in decellularization-related technologies in recent years. Therefore,

Endotoxin, not DNA, determines the host response and tissue regeneration behavior of acellular biologic scaffolds.

Established quantitative standards for assessing decellularization of biologic scaffolds based on residual DNA levels have been well-documented and widely acknowledged. However, post-implantation complications, such as fever and seroma, are commonly observed which negatively impact clinical outcomes. The presence of cellular debris following decellularization or using source tissues that are naturally high in endotoxin may contribute to the host response to a biologic scaffold. In the study, several multi-step decellularization methods were used to decellularize small intestinal submucosa (SIS

Optimization of Gelatin-Based Scaffolds for Soft Tissue Regeneration: In Vitro and In Vivo Performance.

In this study, promising compositions of cross-linked gelatin-based scaffolds were tested in vitro and in vivo. Our aim was to utilize a solid matrix that is suitable for medical applications, and to be regulated as a medical device as a soft tissue implant. Three different cross-linkers were used in vitro, and the optimal composition was chosen for in vivo testing. The surfaces of the scaffolds were observed with SEM, and, in the case of divinyl sulfone (DVS), small cracks appeared, and the structure was rigid. With the use of poly(ethylene glycol) diglycidyl ether (PEGDE), the surface was fo

Cyto- and bio-compatibility assessment of plasma-treated polyvinylidene fluoride scaffolds for cardiac tissue engineering.

As part of applications dealing with cardiovascular tissue engineering, drop-cast polyvinylidene fluoride (PVDF) scaffolds have been treated by cold plasma to enhance their adherence to cardiac cells. The scaffolds were treated in a dielectric barrier device where cold plasma was generated in a gaseous environment combining a carrier gas (helium or argon) with/without a reactive gas (molecular nitrogen). We show that an Ar-N2 plasma treatment of 10 min results in significant hydrophilization of the scaffolds, with contact angles as low as 52.4° instead of 132.2° for native PVDF scaffolds. Corr
